目的:探讨分泌型卷曲相关蛋白5(SFRP5)基因rs10748709位点单核苷酸多态性与妊娠期糖尿病(GDM)及相关临床、肥胖指标的关系。方法:用Taqman探针法对150例GDM孕妇(GDM组)和168例同期健康孕妇(对照组)rs10748709位点进行基因分型。依据BMI... 目的:探讨分泌型卷曲相关蛋白5(SFRP5)基因rs10748709位点单核苷酸多态性与妊娠期糖尿病(GDM)及相关临床、肥胖指标的关系。方法:用Taqman探针法对150例GDM孕妇(GDM组)和168例同期健康孕妇(对照组)rs10748709位点进行基因分型。依据BMI分层,采用logistic回归分析不同遗传模型下超重/肥胖组(BMI≥24kg/m^(2))与正常/低体重组(BMI<24kg/m^(2))SFRP5基因多态性与GDM风险的关联,并比较不同基因型的临床、肥胖指标。结果:GDM组与对照组rs10748709位点在不同遗传模型中基因型及等位基因频率差异均无统计学意义(P>0.05)。超重/肥胖组rs10748709位点在隐性模型中与GDM风险升高有关(GG vs AG+AA,OR=6.00,95%CI为1.68~21.38);正常/低体重组未发现rs10748709位点与GDM存在关联(P>0.05)。携带GG基因型的超重/肥胖GDM孕妇2h OGTT、TG、体脂肪及内脏脂肪水平明显高于携带AG+AA基因型的孕妇(P均<0.05),其余指标未见明显差异(P均>0.05)。结论:SFRP5基因rs10748709位点多态性可能与GDM发生风险无直接相关性,但SFRP5基因可能通过超重/肥胖间接导致GDM的发生。 展开更多

Rural sustainable development:A case study of the Zaozhuang Innovation Demonstration Zone in China 被引量:1

Sustainable development is the central theme of modern global development.With the arrival of the urban era,the vulnerability and instability of rural areas have significantly increased,and rural sustainable development faces serious challenges.To address these issues,the study took the Zaozhuang Innovation Demonstration Zone in China under the National Sustainable Development Agenda as a case,combined with economic,social and land use data during 2016-2020,and applied Granger causality test method to explore the theoretical and practical pathways of“innovation-driven rural sustainable development”.The results showed that rural sustainable development and economic sustainability displayed a trend of synergistic change,with“explosive”growth from 2018 to 2020.The social sustainability steadily increased from 2016 to 2020.Ecological and spatial sustainability continuously declined during the study period.Moreover,the rural innovation capacity of the Zaozhuang Innovation Demonstration Zone displayed rapid growth during 2016-2020.Although the rural innovation capacity of the Zaozhuang Innovation Demonstration Zone has rapidly improved,it has a weak driving effect on rural sustainable development and economic sustainability.There are two primary challenges that must be overcome to ensure the rural sustainable development of the Zaozhuang Innovation Demonstration Zone.The first challenge is the imbalance among the multi-dimensional relationships in the process of rural sustainable development,and the second challenge is the weakening of rural innovation capacity to drive rural sustainable development.To overcome these challenges,this study proposed a systematic pathway for rural sustainable development in the Zaozhuang Innovation Demonstration Zone from multidimensions,such as policy actions,technologies,projects,and institutional guarantees,and formed a universal and representative“Zaozhuang model”.This study expands the theoretical foundation of rural sustainable development and provides theoretical and practical support for innovation-driven rural sustainable development. 展开更多

猪硒蛋白W的原核表达及IgY多克隆抗体制备 被引量:2

试验旨在优化硒蛋白W(selenoprotein W,SelW)的原核表达系统,并评估SelW的免疫原性和基于IgY抗体检测猪体内SelW的可行性。将获得的猪SelW基因序列密码子进行优化、合成并连接至pET-32a(+)表达载体中,构建原核表达重组质粒pET32a(+)-Se... 试验旨在优化硒蛋白W(selenoprotein W,SelW)的原核表达系统,并评估SelW的免疫原性和基于IgY抗体检测猪体内SelW的可行性。将获得的猪SelW基因序列密码子进行优化、合成并连接至pET-32a(+)表达载体中,构建原核表达重组质粒pET32a(+)-SelW,转化E.coli BL21(DE3)宿主菌中,进行IPTG诱导表达,SDS-PAGE鉴定重组猪SelW的表达情况,并免疫产蛋鸡制备抗SelW的IgY多克隆抗体,通过间接ELISA检测IgY抗体滴度,采用Western blotting检测IgY识别抗原的特异性。SDS-PAGE结果显示,SelW基因在原核细胞中成功表达,得到了大小约为33 ku的重组蛋白,IPTG最佳诱导浓度及时间分别为0.5 mmol/L和6 h;可溶性分析结果显示,重组蛋白SelW主要以包涵体形式存在。间接ELISA检测结果显示,免疫后45 d的IgY多克隆抗体的效价可达1∶51 200。Western blotting检测结果显示,重组蛋白具有良好的免疫原性,所制备的IgY抗体与SelW的亲和力较高。本试验成功构建并优化了SelW蛋白的原核表达系统,提高了SelW蛋白的表达量,获得了具有良好免疫原性的SelW蛋白,制备的IgY抗体能特异性识别猪肌肉组织中的SelW,可用于检测猪组织中SelW的表达、预防硒中毒和缺硒性疾病的监测等,为进一步探究SelW的生物学功能奠定基础。 展开更多

针对服装定制信息物理生产系统(cyber-physical production system,CPPS)中离散事件、连续过程、随机因素、时延特性以及冲突现象等混杂特性的建模问题,将智能工厂CPPS系统视为一类混杂系统,分析CPPS系统中的混杂特性,基于HSTPN(hybrid ... 针对服装定制信息物理生产系统(cyber-physical production system,CPPS)中离散事件、连续过程、随机因素、时延特性以及冲突现象等混杂特性的建模问题,将智能工厂CPPS系统视为一类混杂系统,分析CPPS系统中的混杂特性,基于HSTPN(hybrid stochastic timed petri net)建立服装定制智能工厂CPPS的事件层和物理层模型,并基于HSTPNSim软件对该模型仿真.分析了不同材质服装熨烫的连续过程,系统时延以及工艺合格率对客户满意度的影响,并与红领集团的个性化定制生产节奏进行对比.仿真结果表明,服装定制CPPS系统中熨烫过程可实现对不同材质服装熨烫过程的描述,且通过对面辅料采购与设计过程并行、增加防刮保护以及设备升级等措施可提高顾客的满意度.因此,HSTPN模型可有效描述服装定制CPPS. 展开更多

Simulation and Experiment Based on FSMLC Method with EUPI Hysteresis Compensation for a Piezo-Driven Micro Position Stage

Micro/nano positioning technologies have been attractive for decades in industrial and scientific applications fields. The actuators have inherent hysteresis that can cause system unexpected behave in some extend. In this research, the authors used extented unparallel Prandtl-Ishlinshii(EUPI)models to represent the input-output relationship of a piezo-driven micro position stage. Integral inverse(I-I) compensator is used for compensating the hysteresis characteristics of the micro positioning stage and compared with direct inverse(D-I) compensator and inverse model(I-M) compensator. However,the accuracy and the robustness of the I-I compensator are worse when there is noisy in the system,a novel sliding-mode-like-control with EUPI(SMLC-EUPI) method was proposed and analyzed by different trajectory tracking experiments in Matlab environment. Though the above strategies can alleviate most deviation, the adjustment of the SMLC’s parameters is very complex. So the fuzzy method is used to adjust these parameters and be verified by trajectory tracking experiments. Finally,for validating the proposed control method, the paper did the corresponding experiment in microscope with CMOS and obtained convincing results. 展开更多
