The study shows that turbid water under the action of the waves...In this paper, the characteristics of density current under the action of waves are described with the help of flume experiment and theoretical analysis. The study shows that turbid water under the action of the waves can present three types of motion, i. e. significant stratification, fragile stratification and strong mixing. The motion gf turbid;,ater presents significant stratification when (H/D)/root Delta rho/rho less than or equal to 4.5, generally this state is known as density current. The formulas of motion velocity, thickness, and discharge of density current moving on horizontal bottom are derived by use of basic equations such as momemtum equation, equation of energy conservation and continuity equation of fluid. The time-average velocity and the thickness of density current under the action of waves have a relationship with such parameters as relative density (Delta rho/rho), wave height (H), and water depth (D). When these parameters are determined, the time-average thickness and motion velocity of density current are also determined. The relative thickness of density current (D-t/D) decreases with the increase of Delta rho/rho and increases with the increase of H/D. On the other hand, the motion velocity of density current increases with the increase of Delta rho/rho and decreases with the increase of the relative thickness (D-t/D) of density current. It is shown that the calculated results are in agreement with those of the flume experiment.展开更多

The optimal staining solution consists of 54-1...We report here a fast staining method for single-stranded DNA fixed on nitrocellulose membrane using M-meso-letra(3-N-methylpyridyl)-porphrin (M-TMPyP, M: Mn3, Fe3, Pt3). The optimal staining solution consists of 54-108 μg/ml M-TMPyP in 25~200 mmol/l. phosphate buffer solution (pH 5.0~8.0).When DNA dot on NC membrane is treated with the staining solution at room temperature for 1~2 minutes with continuous stirring, it turns to brown-yellow.10pg per dot of single-stranded DNA is visible. It is a useful tool to sludy the DNA blotting, and for recognition of the special bands of DNA in a complete electrophoretogram of DNA blot. The staining mechanism is also discussed.The staining techniques of DNA play a important role in DNA blotting,display of DNA elcctophoretogram and location of the special DNA band from it.Since R.J. Fiel and co-workers[1] reported that meso-tetra(4-N-methylpyridyl) porphyrin can intercalale in calf thymus DNA. the intercalation between TMPyP-4 and DNA in solution has been intensively investigated by several groups[2~8],but none of them concerned with the interaction of metalloporphyrin and filterbound DNA. The electrophoresis separation was performed at 24℃ in a fused-silica capillary(51.2cm×50 μmi.d., 38.5cm to UV detector)using 20mmol/L borax-20mmol/L H3PO4(pH 8.5) as the background electrolyte at 24KV applied voltage. Inosine samples were led into the capillary electrokinetically at 10KV for 15 seconds. In the range of 20μg/mL to 300μg/mL, inosine can be quantitatively determined with a deduction limit of 2.1μg/mL and a RSD of 2.92%. Hypoxanthine was used as the internal standard.展开更多

In order to reflect wave energy loss accurately, the Bretchneider-Reid formula is quoted and the friction coefficient in the formula is discussed in this paper. The calculation results indicate that the wave becomes a little damped at the mouth of Kemena River influenced by the topography and bottom friction, and the wave at the east beach is higher than that at the west beach, because the east beach extends out.展开更多

The former caused Fe and Mg concentrated in the finer grain-size fractions, displaying synchronous variations, while the latter made Fe and Mg separated, leading to Fe retained in the weathered section and Mg leached out. Therefore, Fe/Mg ratios in the acid insoluble fraction of loess and paleosols can eliminate the effect of wind sorting and serve as an excellent proxy record on intensity of weathering-pedogenesis. Based on calculation, leaching percentage of Mg in the paleosol S1 from the Luochuan, Xifeng and Huanxian sections is 15%, 11% and 2%, respectively, and on average 9% for the paleosols S2—S14 from the Luochuan section, with the highest value amounting to 22% in S5-1, suggesting the strongest weathering-pedogenesis.展开更多
