To analyze the effect of blade number on the performance of hydraulic turbines during the transient stage in which theflow rate is not constant,six hydraulic turbines with different blade numbers are considered.The in...To analyze the effect of blade number on the performance of hydraulic turbines during the transient stage in which theflow rate is not constant,six hydraulic turbines with different blade numbers are considered.The instantaneous hydraulic performance of the turbine and the pressure pulsation acting on the impeller are investigated numerically by using the ANSYS CFX software.The ensuing results are compared with the outcomes of experimental tests.It is shown that thefluctuation range of the pressure coefficient increases with time,but the corresponding range for the transient hydraulic efficiency decreases gradually when theflow velocity transits to larger values.During the transition to smallflow velocity,thefluctuation range of the pressure coefficient gradually decreases as time passes,but the correspondingfluctuation range of its transient hydraulic efficiency gradually becomes larger.Thefluctuation range in the Z9 case is small during the transition.The main frequency of transient hydraulic efficiency pulsation is equal to the blade frequency.At the main frequency,Z7 has the largest amplitude of the hydraulic efficiency pulsation,Z10 has the smallest amplitude,and the difference between Z7 and Z9 is limited.As the number of blades grows,the pressure pulsation during the transition process gradually decreases,but the pressure pulsation of Z10 at the volute tongue is larger.In the steady state,Z9 has the highest efficiency and in the transient stage,the pressure coefficientfluctuation range is small.Accordingly,for the hydraulic turbine Z9,the performance is optimal.展开更多

Adoptive cellular immunotherapy with chimeric antigen receptor(CAR)T cells has emerged as a novel modality for treating relapsed and/or refractory B-cell non-Hodgkin lymphoma(B-NHL).With increasing approval of CAR T-c...Adoptive cellular immunotherapy with chimeric antigen receptor(CAR)T cells has emerged as a novel modality for treating relapsed and/or refractory B-cell non-Hodgkin lymphoma(B-NHL).With increasing approval of CAR T-cell products and advances in CAR T cell therapy,CAR T cells are expected to be used in a growing number of cases.However,CAR T-cell-associated toxicities can be severe or even fatal,thus compromising the survival benefit from this therapy.Standardizing and studying the clinical management of these toxicities are imperative.In contrast to other hematological malignancies,such as acute lymphoblastic leukemia and multiple myeloma,anti-CD19 CAR T-cell-associated toxicities in B-NHL have several distinctive features,most notably local cytokine-release syndrome(CRS).However,previously published guidelines have provided few specific recommendations for the grading and management of toxicities associated with CAR T-cell treatment for B-NHL.Consequently,we developed this consensus for the prevention,recognition,and management of these toxicities,on the basis of published literature regarding the management of anti-CD19 CAR T-cell-associated toxicities and the clinical experience of multiple Chinese institutions.This consensus refines a grading system and classification of CRS in B-NHL and corresponding measures for CRS management,and delineates comprehensive principles and exploratory recommendations for managing anti-CD19 CAR T-cell-associated toxicities in addition to CRS.展开更多
In order to analyze the response of a hydraulic turbine to a variation in the operating conditions,different laws of variation in time of the massflow rate have been considered.After validating the overall numerical fr...In order to analyze the response of a hydraulic turbine to a variation in the operating conditions,different laws of variation in time of the massflow rate have been considered.After validating the overall numerical framework through comparison with relevant experiments,the performances of the considered turbine have been analyzed from afluid-dynamic point of view.The results show that different time profiles of the massflow rate(in this work,for simplicity,referred to as“transition functions”)have a varying influence on the transient behavior of the turbine.When a quadratic function is considered for the case of largeflow,the transient head and torque increase gradually with time,thefluctuation amplitude of the transient hydraulic efficiency at the main frequency is the largest,and thefluctuation amplitude of the radial force is the smallest.For the smallflow case,the time profile with exponential nature leads to the best results.The transient head and torque decrease gradually with time,the pulsation amplitude of the transient hydraulic efficiency is the largest at the main frequency,and the pulsation amplitude of the radial force is the smallest.展开更多
In order to study the effect of splitter blades on the internal and external sound field of the hydraulic turbine,the paper chose a centrifugal pump with a specific speed ns=33 reversed as a hydraulic turbine as the r...In order to study the effect of splitter blades on the internal and external sound field of the hydraulic turbine,the paper chose a centrifugal pump with a specific speed ns=33 reversed as a hydraulic turbine as the research object,and added the short blades on the original impeller to form a new splitter impeller.Based on the Re-Normalization Group(RNG)k-εturbulence model to conduct numerical simulation for the hydraulic turbine,this thesis calculated the internal and external acoustic field by means of the acoustic boundary element(BEM)and finite element(FEM)and analyzed the noise radiation characteristics of the two models under different working conditions.The results show that the blade frequency is the main factor affecting the inlet and outlet sound pressure,and the optimized model decreases the inlet and outlet sound pressure levels by 6.84 and 7.24 dB in optimal working conditions.Rotor-stator interaction is the main reason for the flow-induced noise of hydraulic turbine volute appearing,the optimized model can effectively reduce the impeller and volute rotor-stator interaction and the flow-induced noise of volute.Outfield maximum sound pressure appears at the inlet and the volute tongue,which decreased by 1.84,6.07,and 5.24 dB at each operating condition.To sum up,splitter blades can improve hydraulic characteristics and flow field noise in the hydraulic turbine.展开更多
Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. No...Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. Combined analyses of laboratory biomarkers with their clinical manifestations before and after salvage treatment showed that the persistent immunosurveillance mediated by CAR-T-19 cells would inevitably potentiate the leukemia-killing effectiveness of subsequent chemotherapy in patients who showed relapse after CAR-T-19-induced remission.展开更多
