In this paper, we introduce the control system design,including human–machine interfaces, alarm system, historical data archiving system, and the software for access control. The software development followed the standard of Capability Maturity Model Integration(CMMIò) Level3, and the software had passed a third-party test, which indicated that the functionality and the reliability could fulfill the requirements of the radiation protection system.展开更多
AIM To explore the role and mechanism of total flavone of Abelmoschus manihot(TFA) on epithelial-mesenchymal transition(EMT) progress of Crohn's disease(CD) intestinal fibrosis.METHODS First,CCK-8 assay was perfor...AIM To explore the role and mechanism of total flavone of Abelmoschus manihot(TFA) on epithelial-mesenchymal transition(EMT) progress of Crohn's disease(CD) intestinal fibrosis.METHODS First,CCK-8 assay was performed to assess TFA on the viability of intestinal epithelial(IEC-6) cells and select the optimal concentrations of TFA for our further studies.Then cell morphology,wound healing and transwell assays were performed to examine the effect of TFA on morphology,migration and invasion of IEC-6 cells treated with TGF-β1.In addition,immunofluorescence,real-time PCR analysis(q RT-PCR) and western blotting assays were carried out to detect the impact of TFA on EMT progress.Moreover,western blotting assay was performed to evaluate the function of TFA on the Smad and MAPK signaling pathways.Further,the role of co-treatment of TFA and si-Smad or MAPK inhibitors has been examined by q RTPCR,western blotting,morphology,wound healing andtranswell assays.RESULTS In this study,TFA promoted transforming growth factor-β1(TGF-β1)-induced(IEC-6) morphological change,migration and invasion,and increased the expression of epithelial markers and reduced the levels of mesenchymal markers,along with the inactivation of Smad and MAPK signaling pathways.Moreover,we revealed that si-Smad and MAPK inhibitors effectively attenuated TGF-β1-induced EMT in IEC-6 cells.Importantly,co-treatment of TFA and si-Smad or MAPK inhibitors had better inhibitory effects on TGF-β1-induced EMT in IEC-6 cells than either one of them.CONCLUSION These findings could provide new insight into the molecular mechanisms of TFA on TGF-β1-induced EMT in IEC-6 cells and TFA is expected to advance as a new therapy to treat CD intestinal fibrosis.展开更多
BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease with undefined pathogenesis.Non-SMC condensin I complex subunit D2(NCAPD2)and non-SMC condensin II complex subunit D3(NCAPD3)pl...BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease with undefined pathogenesis.Non-SMC condensin I complex subunit D2(NCAPD2)and non-SMC condensin II complex subunit D3(NCAPD3)play pivotal roles in chromosome assembly and segregation during both mitosis and meiosis.To date,there has been no relevant report about the functional role of NCAPD2 and NCAPD3 in UC.AIM To determine the level of NCAPD2/3 in intestinal mucosa and explore the mechanisms of NCAPD2/3 in UC.METHODS Levels of NCAPD2/3 in intestinal tissue were detected in 30 UC patients and 30 healthy individuals with in situ hybridization(ISH).In vitro,NCM60 cells were divided into the NC group,model group,si-NCAPD2 group,si-NCAPD3 group and si-NCAPD2+si-NCAPD3 group.Inflammatory cytokines were measured by ELISA,IKK and NF-κB were evaluated by western blot,and IKK nucleation and NF-κB volume were analyzed by immunofluorescence assay.RESULTS Compared with expression in healthy individuals,NCAPD2 and NCAPD3 expression in intestinal tissue was significantly upregulated(P<0.001)in UC patients.Compared with levels in the model group,IL-1β,IL-6 and TNF-αin the si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 groups were significantly downregulated(P<0.01).IKK and NF-κB protein expression in the si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 groups was significantly decreased(P<0.01).Moreover,IKK nucleation and NF-κB volume were suppressed upon si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 transfection.CONCLUSION NCAPD2/3 is highly expressed in the intestinal mucosa of patients with active UC.Overexpression of NCAPD2/3 promotes the release of pro-inflammatory cytokines by modulating the IKK/NF-κB signaling pathway.展开更多
In this work,the factors affecting asphaltenes deposition in high-temperature and high-pressure wells were studied using backscattered light and PVT equipment customized to suit the well conditions.In an examination o...In this work,the factors affecting asphaltenes deposition in high-temperature and high-pressure wells were studied using backscattered light and PVT equipment customized to suit the well conditions.In an examination of the intensity of backscattered light,it was revealed that there exists a linear relationship between temperature and asphaltene precipitation within a specific temperature range.Within this range,a decrease in temperature tends to accelerate asphaltene precipitation.However,the impacts of pressure and gas-oil ratio are more pronounced.The pressure depletion induces the asphaltenes to precipitate out of the solution,followed by the formation of flocs below the bubble point.In addition,an increase in the gas-oil ratio causes a more severe asphaltene deposition,shifting the location of asphaltenes to deep well sections.展开更多

Objective:To investigate the correlation between the change in expression of the tumor associated macrophage (TAM) markers CD68 and CD206 and the clinicopathological parameters.Methods: We retrospectively analyzed 28 ...Objective:To investigate the correlation between the change in expression of the tumor associated macrophage (TAM) markers CD68 and CD206 and the clinicopathological parameters.Methods: We retrospectively analyzed 28 PGI-DLBCL patients by immunohistochemical stainning. The surface expression of biomarkers CD68 and CD206 on TAMs in samples obtained at diagnosis was compared with that in specimens obtained at first relapse. Based on the change in expression levels of CD68 and CD206 from diagnosis to relapse, patients were categorized into three groups:'increased','stable' and'decreased'. Results: A significant difference was observed between the three groups with respect to CD68 and CD206 expressions (P=0.012) and there existed a consistency between the two (P=0.008). The three groups showed a negative correlation with age, sex, stage, B symptoms, diagnosis, ECOG performance status, and IPI scores, while a positive correlation was observed with the site of lesion of CD206 expression (P=0.032). The time to relapse (TTR) in the three groups of CD68 and CD206 was not significantly different (P=0.23,P=0.818).Conclusion: Our study suggests that increase in CD206 and decrease in CD68 expression at relapse indicates M1-TAM polarization into M2-TAM, which plays an important role in PGI-DLBCL relapse. Furthermore, M2-TAM polarization may be associated with the site of Primary gastric diffuse large B lymphoma.展开更多
Taxifolin is a flavonoid commonly found in fruits and vegetables with a variety of bioactivities.However,it is unknown whether taxifolin has positive effects on food preservation.In the present study,the antibacterial...Taxifolin is a flavonoid commonly found in fruits and vegetables with a variety of bioactivities.However,it is unknown whether taxifolin has positive effects on food preservation.In the present study,the antibacterial activity and mechanism of taxifolin on two foodborne bacteria,Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus),were investigated for the first time.Besides,its applications in milk preservation were evaluated.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of taxifolin on E.coli were 1.11 mg/mL and 2.22 mg/mL,while for S.aureus were 0.556 mg/mL and 1.11 mg/mL,respectively.Furthermore,taxifolin exhibited strong synergistic antibacterial activity with tetracycline.Taxifolin functions against the two strains of bacteria through damaging bacterial cell wall and membrane integrity,inhibiting bacterial biofilm formation,causing adversity and resulting in the increase of superoxide dismutase and alkaline phosphatase activities in bacteria.Moreover,morphological changes in bacteria were observed after exposure to taxifolin,including the formation of aggregates,irregular forms,wrinkled cell surfaces,and shriveling of cells.During milk storage,taxifolin significantly delayed the increase of acidity and the loss of lactose in milk through inhibiting the proliferation of bacteria.Hence,in addition to health-promoting bioactivities,taxifolin also has positive effects on food preservation because of antibacterial activity.展开更多
