Elaidic acid(EA)stimulation can lead to endoplasmic reticulum stress(ERS),accompanied by a large release of Ca^(2+),and ultimately the activation of NLRP3 inflammasome in Kupffer cells(KCs).Mitochondrial instability or dysfunction may be the key stimulating factors to activate NLRP3 inflammasome,and sustained Ca^(2+)transfer can result in mitochondrial dysfunction.We focused on KCs to explore the damage to mitochondria by EA.After EA stimulation,cells produced an oxidative stress(OS)response with a significant increase in ROS release.Immunoprecipitation experiments and the addition of inhibitors revealed that the increase in the level of intracellular Ca^(2+)led to Ca^(2+)accumulation in the mitochondrial matrix via mitochondria-associated membranes(MAMs).This was accompanied by a significant release of m ROS,loss of MMP and ATP,and a significant increase in mitochondrial permeability transition pore opening,ultimately leading to mitochondrial instability.These findings confirmed the mechanism that EA induced mitochondrial Ca^(2+)imbalance in KCs via MAM,ultimately leading to mitochondrial dysfunction.Meanwhile,EA induced OS and the decrease of MMP and ATP in rat liver,and significant lesions were found in liver mitochondria.Swelling of the inner mitochondrial cristae and mitochondrial vacuolization occurred,with a marked increase in lipid droplets. 展开更多

Previous studies have shown that trans fatty acids(TFA) are associated with several chronic diseases,the gut microbiota is directly influenced by dietary components and linked to chronic diseases.Our research investigated the effects of elaidic acid(EA),a typical TFA,on the gut microbiota to understand the underlying mechanisms of TFA-related chronic diseases.16S rDNA gene sequencing on faecal samples from Sprague-Dawley rats were performed to explore the composition change of the gut microbiota by EA gavage for 4 weeks.The results showed that the intake of EA increased the abundance of well-documented harmful bacteria,such as Proteobacteria,Anaerotruncus,Oscillibacter and Desulfovibrionaceae.Plus,EA induced translocation of lipopolysaccharides(LPS) and the above pathogenic bacteria,disrupted the intestinal barrier,led to gut-liver axis derangement and TLR4 pathway activation in the liver.Overall,EA induced intestinal barrier damage and regulated TLR4-MyD88-NF-κB/MAPK pathways in the liver of SD rats,leading to the activation of NLRP3 inflammasome and inflammatory liver damage. 展开更多

Geochronology and geochemistry of the W-Mo-ore-related granitic rocks from eastern Ningzhen,lower Yangtze river belt,eastern China

Here we present zircon U–Pb–Hf and wholerock major and trace element studies of eastern Ningzhen W-Mo-ore-related magmatic rocks,Yushan and Longwangshan granitic rocks,to constrain their form timing,magma sources,and tectonic settings.The results showed that the two plutons were formed in the Early Cretaceous with;Pb/;U ages of 107.8±1.2 and 105.2±1.5 Ma(;Pb/;U),respectively.The trapped/residual zircons are mainly distributed in 2.0–2.5 Ga.The two intrusions are characterized by high silicon(68.60–73.99%),high aluminum(13.56–15.02%),high Mg#(47–55),high Sr,Sr/Y,LaN/YN,and low Yb,falling into high Mg#adakitic rock region.The zirconεHf(t)values of the two intrusions range from-24.8 to-13.2,indicating an ancient continental crust in their magma sources.The average Ti-inzircon temperature is 689°C,slightly higher than those of other high-Mg adakitic rocks in the lower Yangtze River belt,but lower than those of high-Mg adakitic rocks in the Southern Tanlu Fault(STLF).Zircon Ce;/Ce;show low oxygen fugacity(LWS-1:3–400,average 92;sample ZYS-4:9–382,average 93).These geochemical features indicate a thickened lower continental crust in the Eastern Ningzhen region in a subduction setting.Comparing the geochemical characteristics of the eastern Ningzhen to the western Ningzhen and other areas in the Lower Yangtze River Metallogenic Belt(LYRMB)and the high-Mg ore-barren adakitic rocks of the STLF,we propose that the magmatic rocks from eastern Ningzhen may be mainly from a thickened lower continental crust that hybridized with a very small part of mantle sources,while the west Ningzhen magmatic rocks may have experienced a higher degree of mantle contaminations in their source.The metallogenic differences between the eastern(W–Mo)and western(Cu–Fe–Pb–Zn)parts of Ningzhen also indicate different proportions of crustal materials in their magma source. 展开更多

Hyperpolarized^(3)He nuclei have emerged as a significantly important approach in quantum precision measurement techniques,with extensive applications in fundamental physics,magnetometry,metrology,and beyond.In this study,we report on the design and implementation of a^(3)He polarization system at the China Mianyang Research Reactor(CMRR),utilizing the metastabilityexchange optical pumping(MEOP)method.We employed a Merritt coil system consisting of four square coils to furnish a uniform holding field.We deployed a 2 W fiber laser to pump the metastable^(3)He atoms and conducted free induction decay(FID)detection of the polarized^(3)He nuclei using both pickup coil and optical methods.For the optical method,we used a50 m W linearly polarized distributed Bragg reflector(DBR)laser as the probe.We applied transverse light absorption polarimetry to measure the absolute nuclear polarization of the ground-state^(3)He.We have developed cell fabrication capabilities at the CMRR,and cells at various pressures ranging from 100 to 1000 Pa have been fabricated and evaluated.For a typical borosilicate cell with 100 Pa pressure,the absolute polarization is measured as Pn≈70%,and the transverse relaxation time is estimated as T2≈0.5 s.Moreover,we constructed a few aluminosilicate cells,each carefully filled with pure^(3)He at a pressure of 100 Pa.Subsequently,we conducted a comprehensive evaluation of their performance in the context of MEOP. 展开更多

Bipolar membrane electrodialysis(BMED)is considered a state-of-the-art technology for the conversion of salts into acids and bases.However,the low concentration of base generated from a traditional BMED process may limit the viability of this technology for a large-scale application.Herein,we report an especially designed multistage-batch(two/three-stage-batch)BMED process to increase the base concentration by adjusting different volume ratios in the acid(Vacid),base(Vbase),and salt compartments(Vsalt).The findings indicated that performance of the two-stage-batch with a volume ratio of Vacid:Vbase:Vsalt=1:1:5 was superior in comparison to the threestage-batch with a volume ratio of Vacid:Vbase:Vsalt=1:1:2.Besides,the base concentration could be further increased by exchanging the acid produced in the acid compartment with fresh water in the second stage-batch process.With the two-stage-batch BMED,the maximum concentration of the base can be obtained up to 3.40 mol∙L^(-1),which was higher than the most reported base production by BMED.The low energy consumption and high current efficiency further authenticate that the designed process is reliable,cost-effective,and more productive to convert saline water into valuable industrial commodities. 展开更多
