Methods: A modified bipedal rat model was established using a brachial plexus rhizotomy approach to imitate human upright posture. The experimental animals were randomly divided into three groups: control, vertical vibration, and whole-body vibration. Gene expression in degeneration of the intervertebral discs was assessed by reverse transcription-quantitative polymerase chain reaction. Results: The expression of aggrecan, Col1α1, Col2α1, and decorin were shown to be up-regulated in 14-week-old rats in the vertical vibration and whole-body vibration groups, whereas biglycan and versican expression was down-regulated in 14-week-old rats of the two experimental groups. Furthermore, biglycan and versican expression levels were shown to be lower in the whole-body vibration group than in the vertical vibration group(P<0.05). Conclusions: This in-vivo study demonstrated that vibrations can influence the expression of anabolic genes. Furthermore, whole-body vibrations seem to have a greater effect in this regard than vertical vibrations. A new method is expected to relieve the low back pain of the patients through our research.展开更多

However, it is becoming increasingly appreciated that memory cells are also capable of mediating the pathology associated with autoimmune disorders and transplant rejection, and may pose a significant barrier to future clinical advancement in immunoregulation. Therefore, understanding the unique properties of memory lymphocytes (as compared to their naive precursors) is a major area of investigation. Here, we focus on one of those singular properties of memory T cells (TM)-rapid recall. As will be discussed in more detail, rapid recall refers to the ability of quiescent TM cells to efficiently and robustly express‘effector functions' following stimulation. Studies that have advanced our understanding of TM cells' rapid recall using CD4^+ T cells have been expertly reviewed elsewhere, so we will focus primarily on studies of CD8^+ T cells. We will first review the different ways that CD8^+ TM cells can be generated, followed by discussing how this influences their functional properties in the settings of immune protection and pathology. Then, rapid recall ability will be discussed, with emphasis placed on what is currently known about the mechanisms that underlie this unique property of TM cells.展开更多

The transition probabilities of the Markov chain are assumed to be piecewise time-varying, and the persistent dwell-time switching rule,as a more general switching rule, is adopted to describe this variation characteristic. Afterwards, based on the classical Lyapunov stability theory, a Lyapunov function is established, in which the information about the Markov jump feature of the system mode and the persistent dwell-time switching of the transition probabilities is considered simultaneously.Furthermore, via using the stochastic analysis method and some advanced matrix transformation techniques, some sufficient conditions are obtained such that the estimation error system is mean-square exponentially stable with an H_(∞) performance level, from which the specific form of the estimator can be obtained. Finally, the rationality and effectiveness of the obtained results are verified by a numerical example.展开更多

文摘BACKGROUND Immunotherapy for advanced gastric cancer has attracted widespread attention in recent years. However, the adverse reactions of immunotherapy and its relationship with patient prognosis still need further study. In order to determine the association between adverse reaction factors and prognosis, the aim of this study was to conduct a systematic prognostic analysis.By comprehensively evaluating the clinical data of patients with advanced gastric cancer treated by immunotherapy,a nomogram model will be established to predict the survival status of patients more accurately.AIM To explore the characteristics and predictors of immune-related adverse reactions(irAEs)in advanced gastric cancer patients receiving immunotherapy with programmed death protein-1(PD-1)inhibitors and to analyze the correlation between irAEs and patient prognosis.METHODS A total of 140 patients with advanced gastric cancer who were treated with PD-1 inhibitors in our hospital from June 2021 to October 2023 were selected.Patients were divided into the irAEs group and the non-irAEs group according to whether or not irAEs occurred.Clinical features, manifestations, and prognosis of irAEs in the two groups were collected and analyzed.A multivariate logistic regression model was used to analyze the related factors affecting the occurrence of irAEs,and the prediction model of irAEs was established. The receiver operating characteristic(ROC)curve was used to evaluate the ability of different indicators to predict irAEs.A Kaplan-Meier survival curve was used to analyze the correlation between irAEs and prognosis. The Cox proportional risk model was used to analyze the related factors affecting the prognosis of patients.RESULTS A total of 132 patients were followed up,of whom 63(47.7%)developed irAEs.We looked at the two groups’clinical features and found that the two groups were statistically different in age≥65 years,Ki-67 index, white blood cell count, neutrophil count, and regulatory T cell(Treg)count(all P<0.05).Multivariate logistic regression analysis showed that Treg count was a protective factor affecting irAEs occurrence(P=0.030).The ROC curve indicated that Treg+Ki-67+age(≥65 years)combined could predict irAEs well(area under the curve=0.753,95%confidence interval:0.623-0.848,P=0.001).Results of the Kaplan-Meier survival curve showed that progressionfree survival(PFS)was longer in the irAEs group than in the non-irAEs group(P=0.001).Cox proportional hazard regression analysis suggested that the occurrence of irAEs was an independent factor for PFS(P=0.006).CONCLUSION The number of Treg cells is a separate factor that affects irAEs in advanced gastric cancer patients receiving PD-1 inhibitor immunotherapy.irAEs can affect the patients’PFS and result in longer PFS.Treg+Ki-67+age(≥65 years old)combined can better predict the occurrence of adverse reactions.

文摘Non-oxidative conversion of methane to olefins, aromatics and hydrogen(MTOAH) has been reported recently over metal single sites such as iron and platinum. The reaction was proposed to involve catalytic activation of methane followed by gas phase C-C coupling of methyl radicals. This study using H atom Rydberg Tagging time-of-flight technique provides direct experimental evidence for the formation of hydrogen radicals during MTOAH reaction over a catalytic quartz wall reactor containing embedded iron species(denoted as Fe-reactor).Fe-reactor gives 7.3% methane conversion at 1273 K with 41.2% selectivity toward C2(ethane,ethylene and acetylene) and 31.8% toward BTX(benzene, toluene and xylene),respectively. The enhancing effects of hydrogen radicals on overall MTOAH performance are validated by cofeeding hydrogen donor benzene, which provides an additional route of methane activation apart from catalytic activation.

文摘Objective: To assess the effects of axial vibrations on gene expression and lumbar intervertebral disc degeneration in vivo. A new method is expected to relieve the low back pain of the patients through our research.
文摘The functional roles of memory B and T lymphocytes underlie the phenomenal success of prophylactic vaccinations, which have decreased morbidities and mortalities from infectious diseases globally over the last 50 years. Then, rapid recall ability will be discussed, with emphasis placed on what is currently known about the mechanisms that underlie this unique property of TM cells.

文摘Objective Increased expression of CDCA7 is associated with a poorer prognosis in patients with non-small-cell lung cancer(NSCLC).This study was performed to investigate the effects of down-regulating cell division cycle-associated 7(CDCA7)gene expression on the proliferation, migration, invasion, cell cycle and apoptosis of human NSCLC cell lines. Methods Cultured A549 and NCI-H292 cells were transfected with si NC(control group)or si CDCA7(experimental group).Cell activity was detected using the CCK-8 and a Real Time Cell Analyzer(RTCA).Cell migration and invasion were also measured by RTCA.In addition, flow cytometry was used to assess the cell cycle progression and apoptosis in the cells, and Western blotting was used to detect the expression level of proteins in key signaling pathways. Results Compared with the cells transfected with si NC,the cell proliferation was significantly reduced(P<0.05),the migration and invasion were decreased, and the cell cycle was blocked in the G0/G1 phase(P<0.05)in the cells transfected with si CDCA7.The number of cells undergoing apoptosis was also increased(P<0.05).Western blotting revealed that P-ERK,Cyclin-D1,Vimentin and Bcl-2 were all downregulated. However, the expression of E-cadherin was not affected by the down-regulation of CDCA7,suggesting that it is upstream of this gene. Conclusion Silencing the CDCA7 gene inhibited the proliferation, migration and invasion of A549 and NCI-H292 cells, hindered the cell cycle transition to the S phase, and promoted cell apoptosis. These findings indicate that CDCA7 may represent a new therapeutic target for NSCLC.

文摘Janus amphiphilic polymer nanosheets(JAPNs)with anisotropic morphology and distinctive perfor-mance have aroused widespread interest. However, due to the difficulty in synthesis and poor dispersion stability,JAPNs have been scarcely reported in the field of enhancing oil recovery(EOR).Herein,a kind of organic-based flexible JAPNs was prepared by paraffin emulsion methods. The lateral sizes of JAPNs were ranging from hundreds of nanometers to several micrometers and the thickness was about 3 nm.The organic-based nanosheets were equipped with remarkably flexible structures, which could improve their injection performance. The dispersion and interfacial properties of JAPNs were studied systematically.By modification of crosslinking agent containing multiple amino groups, the JAPNs had excellent hydro-philicity and salt resistance compared with conventional inorganic or composite nanosheets. The settling time of nanosuspension with NaCl and CaCl_(2) at a low salinity of 1000 mg/L was over 240 h.The value could also remain 124 h under the salinity of 10,000 mg/L NaCl.With the dual functionalities of Janus amphiphilic nature and nanoparticles'Pickering effect,JAPNs could change rock wettability and form emulsions as"colloidal surfactants",In particular,a new technology called optical microrheology was pioneered to explore the destabilization state of nanosuspensions for the first time.Since precipitation lagged behind aggregation, especially for stable suspension systems, the onset of the unstable behavior was difficult to be detected by conventional methods, which should be the indicator of reduced effec-tiveness for nanofluid products. In addition, the oil displacement experiments demonstrated that the JAPNs could enhance oil recovery by 17.14%under an ultra-low concentration of 0.005%and were more suitable for low permeability cores. The findings can help for a better understanding of the material preparation of polymer nanosheets. We also hope that this study could shed more light on the nano-flooding technology for EOR.

文摘With the rising needs of better prediction of the load-displacement performance of grouted anchors in an era of developing large-scale underground infrastructures, the existing methods in literature lack an accurate analytical model for the real-life projects or rigorous understanding of the parameters such as grouting pressures. This paper proposes Fast ICA-MARS as a novel data-driven approach for the prediction of the load-displacement performance of uplift-resisting grouted anchors. The hybrid and data-driven Fast ICA-MARS approach integrates the multivariate adaptive regression splines(MARS)technique with the Fast ICA algorithm which is for Independent Component Analysis(ICA).A database of 4315 observations for 479 different anchors from 7 different projects is established. The database is then used to train, validate and compare the Fast ICA-MARS approach with the classical MARS approach. The developed Fast ICA-MARS model can provide more accurate predictions than MARS.Moreover, the developed Fast ICA-MARS model is easy to interpret since the evaluation of the parameter importance of the independent components can be conducted along with the considerations of the correlations with the original variables. It is noteworthy to point out that the grouting pressures play a central role in the proposed model, which is considered of paramount importance in engineering practices but has not been properly taken into account in any prior analytical or empirical predictive models for the load-displacement relationships.

文摘We investigate the problem of H_(∞) state estimation for discrete-time Markov jump neural networks. Finally, the rationality and effectiveness of the obtained results are verified by a numerical example.

文摘This paper is concerned with the global stabilization of state-dependent switching neural networks(SDSNNs)viadiscontinuous event-triggered control with network-induced communication delay.Aiming at decreasing triggering times,a discontinuous event-trigger scheme is utilized to determine whether the sampling information is required to be sent outor not.Meanwhile, under the effect of communication delay, the trigger condition and SDSNNs are transformed into twotractable models by designing a fictitious delay function. Then, using the Lyapunov–Krasovskii stability theory, someinequality estimation techniques, and extended reciprocally convex combination method, two sufficient criteria are established for ensuring the global stabilization of the resulting closed-loop SDSNNs,respectively.A unified framework isderived that has the ability to handle the simultaneous existence of the communication delay, the properties of discontinuousevent-trigger scheme,as well as feedback controller design.Additionally, the developed results demonstrate a quantitativerelationship among the event trigger parameter, communication delay, and triggering times. Finally, two numerical examples are presented to illustrate the usefulness of the developed stabilization scheme.
文摘BACKGROUND Central pontine myelinolysis(CPM)usually occurs during rapid correction of serum osmolality, typically with brainstem lesions presenting uniform signals following enhancement on magnetic resonance imaging(MRI).We report a case of CPM caused by diabetes, which was characterized by glioma-like imaging features and the patient responded well to corticosteroids.CASE SUMMARY A 49-year-old man with type 2 diabetes was admitted due to numbness and weakness for 6 mo with progressive aggravation for 2 wk.His complete blood count, serum electrolytes, renal and liver function parameters were within the normal range.MRI showed mass lesions in the brainstem, with unusually inhomogeneous signal intensity after contrast-enhanced scans. His symptoms worsened after hypoglycemic therapy.Due to his clinical history and examination results,CPM was considered the most likely diagnosis.Treatment with corticosteroids was administered with a methylprednisolone pulse in the acute phase followed by dose tapering.During the 8-mo follow-up period, his clinical symptoms and imaging features significantly improved.CONCLUSION Diabetes could rarely be accompanied by CPM,and patients who experience this neurological complication could benefit from corticosteroid treatment.Clinicians should recognize the special relationship between diabetes and CPM,and improve awareness of early identification and appropriate treatment.
