Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological damage.Hypoxic preconditioning has been shown to impr...Cardiac arrest can lead to severe neurological impairment as a result of inflammation,mitochondrial dysfunction,and post-cardiopulmonary resuscitation neurological damage.Hypoxic preconditioning has been shown to improve migration and survival of bone marrow–derived mesenchymal stem cells and reduce pyroptosis after cardiac arrest,but the specific mechanisms by which hypoxia-preconditioned bone marrow–derived mesenchymal stem cells protect against brain injury after cardiac arrest are unknown.To this end,we established an in vitro co-culture model of bone marrow–derived mesenchymal stem cells and oxygen–glucose deprived primary neurons and found that hypoxic preconditioning enhanced the protective effect of bone marrow stromal stem cells against neuronal pyroptosis,possibly through inhibition of the MAPK and nuclear factor κB pathways.Subsequently,we transplanted hypoxia-preconditioned bone marrow–derived mesenchymal stem cells into the lateral ventricle after the return of spontaneous circulation in an 8-minute cardiac arrest rat model induced by asphyxia.The results showed that hypoxia-preconditioned bone marrow–derived mesenchymal stem cells significantly reduced cardiac arrest–induced neuronal pyroptosis,oxidative stress,and mitochondrial damage,whereas knockdown of the liver isoform of phosphofructokinase in bone marrow–derived mesenchymal stem cells inhibited these effects.To conclude,hypoxia-preconditioned bone marrow–derived mesenchymal stem cells offer a promising therapeutic approach for neuronal injury following cardiac arrest,and their beneficial effects are potentially associated with increased expression of the liver isoform of phosphofructokinase following hypoxic preconditioning.展开更多
2,5-Furandicarboxylic acid(FDCA)is a promising biomass-derived polymeric monomer that serves as an attractive alternative to terephthalic acid derived from fossil resources.However,the green and efficient production o...2,5-Furandicarboxylic acid(FDCA)is a promising biomass-derived polymeric monomer that serves as an attractive alternative to terephthalic acid derived from fossil resources.However,the green and efficient production of FDCA through the oxidation of 5-hydroxymethylfurfural(HMF)and its derivatives is still rudimentary under base-free conditions.In this work,oxygen-vacancy-rich Mn Oxwas prepared and displayed a strong adsorption and anchoring ability to Ru species that mainly exposed the(210)plane of RuO_(2),bringing about highly dispersed and active interfacial Ru-O-Mn structures.Experimental results and density functional theory calculations confirm that these above features greatly facilitate the adsorption/activation of oxygen and the dehydrogenation-oxidation of HMF/5-methoxymethylfurfural(MMF),which enables an efficient FDCA production under base-free and mild conditions.Notably,a desirable FDCA yield of 86.56%was still obtained from concentrated HMF(10 wt%)under base-free conditions over oxygen-vacancy-rich Mn Oxsupported Ru Ox(1.0 MPaO_(2),120℃,6 h).This work delineates a facile catalyst preparation strategy for HMF/MMF oxidation,and might open a new avenue for the green synthesis of FDCA under base-free conditions.展开更多
The Solar Upper Transition Region Imager(SUTRI)onboard the Space Advanced Technology demonstration satellite(SATech-01),which was launched to a Sun-synchronous orbit at a height of~500 km in 2022 July,aims to test the...The Solar Upper Transition Region Imager(SUTRI)onboard the Space Advanced Technology demonstration satellite(SATech-01),which was launched to a Sun-synchronous orbit at a height of~500 km in 2022 July,aims to test the on-orbit performance of our newly developed Sc/Si multi-layer reflecting mirror and the 2k×2k EUV CMOS imaging camera and to take full-disk solar images at the Ne VII 46.5 nm spectral line with a filter width of~3 nm.SUTRI employs a Ritchey-Chrétien optical system with an aperture of 18 cm.The on-orbit observations show that SUTRI images have a field of view of~416×416 and a moderate spatial resolution of~8″without an image stabilization system.The normal cadence of SUTRI images is 30 s and the solar observation time is about16 hr each day because the earth eclipse time accounts for about 1/3 of SATech-01's orbit period.Approximately15 GB data is acquired each day and made available online after processing.SUTRI images are valuable as the Ne VII 46.5 nm line is formed at a temperature regime of~0.5 MK in the solar atmosphere,which has rarely been sampled by existing solar imagers.SUTRI observations will establish connections between structures in the lower solar atmosphere and corona,and advance our understanding of various types of solar activity such as flares,filament eruptions,coronal jets and coronal mass ejections.展开更多
Gibberella ear rot(GER)caused by Fusarium graminearum(teleomorph Gibberella zeae)is a common maize disease that not only severely reduces grain yield but also contaminates maize grain with mycotoxins.We investigated t...Gibberella ear rot(GER)caused by Fusarium graminearum(teleomorph Gibberella zeae)is a common maize disease that not only severely reduces grain yield but also contaminates maize grain with mycotoxins.We investigated the molecular mechanism underlying the host defense responses against pathogen infection using comparative transcriptomic analysis.We injected F.graminearum spore suspensions into plants of resistant(IBM-81)and a susceptible(IBM-85)maize inbred line after pollination and performed RNA-seq 48,72,and 96 h after inoculation.Respectively 487 and 410 differentially expressed genes(DEGs)were induced in the resistant and susceptible lines across three time points,indicating that a stronger defense response was activated in the resistant than in the susceptible line.Among them,198 genes commonly induced in the two lines were subjected to pathway analysis,revealing that most of the DEGs were closely associated with defense and a wide range of metabolic activities.DEGs associated with pathogenesis-related protein 1(PR1)and regulation of salicylic acid were significantly enriched during F.graminearum infection,suggesting that these DEGs play dominant roles in maize resistance to GER.Our results provide a resource for future gene discovery and facilitate elucidation of the complex defense mechanisms involved in resistance to GER.展开更多

Therefore, the Solar Upper Transition Region Imager(SUTRI) was proposed by Chinese scientists and launched in 2022 July. Right now, the first imaging observation of the upper transition region around 46.5 nm has been carried out by SUTRI. To ensure the spectral and temporal resolution of the SUTRI telescope, we have developed a narrowband Sc/Si multilayer. Based on the extreme ultraviolet(EUV)reflectivity measurements, the multilayer structure has been modified for ensuring the peak position of reflectivity was at 46.5 nm. Finally, the narrowband Sc/Si multilayer was successfully deposited on the hyperboloid primary mirror and secondary mirrors. The deviation of multilayer thickness uniformity was below than 1%, and the average EUV reflectivity at 46.1 nm was 27.8% with a near-normal incident angle of 5°. The calculated bandwidth of the reflectivity curve after primary and secondary mirrors was 2.82 nm, which could ensure the requirements of spectral resolution and reflectivity of SUTRI telescope to achieve its scientific goals.展开更多

Microwave ablation(MWA)status monitoring in real time plays a key role in assessment of therapeutic fectiveness.As a novel realtime assessment method,near infrared spectroscopy(NIRs)was used to evaluate the ablation e...Microwave ablation(MWA)status monitoring in real time plays a key role in assessment of therapeutic fectiveness.As a novel realtime assessment method,near infrared spectroscopy(NIRs)was used to evaluate the ablation eficacy.MWA experiments were carried out on in vitroporcine livers.An optical measurement system for biological tissue is developed by our lab tomonitor reduced scattering coefficient(μ')at 690 nm of the coagulation zones.It is noted that p's of liver tissue,which increases as the liver tissue being ablated,is clearly related with the co-agulation status.μ's of normal tissue and coagulated tissue is 3-5 and 17-19 cm-1,respectively.Continuous changes ofμ's demonstrate that optical parameter can be used as an efficacy evaluation factor because it essentially indicates the degree of thermal damage.Compared withtemperat ure,optical parameter is more sensitive and accurate,which is promising for real-timetherapeutic eficacy assessment in MWA.展开更多
Understanding the underlying mechanisms and links between genome evolution and adaptive innovations stands as a key goal in evolutionary studies.Poplars,among the world’s most widely distributed and cultivated trees,...Understanding the underlying mechanisms and links between genome evolution and adaptive innovations stands as a key goal in evolutionary studies.Poplars,among the world’s most widely distributed and cultivated trees,exhibit extensive phenotypic diversity and environmental adaptability.In this study,we present a genus-level super-pangenome comprising 19 Populus genomes,revealing the likely pivotal role of private genes in facilitating local environmental and climate adaptation.Through the integration of pangenomes with transcriptomes,methylomes,and chromatin accessibility mapping,we unveil that the evolutionary trajectories of pangenes and duplicated genes are closely linked to local genomic landscapes of regulatory and epigenetic architectures,notably CG methylation in gene-body regions.Further comparative genomic analyses have enabled the identification of 142202 structural variants across species that intersect with a significant number of genes and contribute substantially to both phenotypic and adaptive divergence.We have experimentally validated a∼180-bp presence/absence variant affecting the expression of the CUC2 gene,crucial for leaf serration formation.Finally,we developed a user-friendly web-based tool encompassing the multi-omics resources associated with the Populus super-pangenome(http://www.populus-superpangenome.com).Together,the present pioneering super-pangenome resource in forest trees not only aids in the advancement of breeding efforts of this globally important tree genus but also offers valuable insights into potential avenues for comprehending tree biology.展开更多
Telomeres are specialized structures at the ends of linear chromosomes that protect genome stability.The telomeric repeat-containing RNA(TERRA)that is transcribed from subtelomeric regions can invade into double-stran...Telomeres are specialized structures at the ends of linear chromosomes that protect genome stability.The telomeric repeat-containing RNA(TERRA)that is transcribed from subtelomeric regions can invade into double-stranded DNA regions and form RNA:DNA hybrid-containing structure called R-loop.In tumor cells,R-loop formation is closely linked to gene expression and the alternative lengthening of telomeres(ALT)pathway.Dysregulated R-loops can cause stalled replication forks and telomere instability.However,how R-loops are recognized and regulated,particularly at telomeres,is not well understood.We discovered that ILF3 selectively associates with telomeric R-loops and safeguards telomeres from abnormal homologous recombination.Knocking out ILF3 results in excessive R-loops at telomeres and triggers telomeric DNA damage responses.In addition,ILF3 deficiency disrupts telomere homeostasis and causes abnormalities in the ALT pathway.Using the proximity-dependent biotin identification(BioID)technology,we mapped the ILF3 interactome and discovered that ILF3 could interact with several DNA/RNA helicases,including DHX9.Importantly,ILF3 may aid in the resolution of telomeric R-loops through its interaction with DHX9.Our findings suggest that ILF3 may function as a reader of telomeric R-loops,helping to prevent abnormal homologous recombination and maintain telomere homeostasis.展开更多
