Now,although the identification system of design and operation have been implemented in the evaluation of green building,lacking of appropriate evaluation after use. The actual operation results of many buildings,which have got a green building logo,are not satisfied from the user 's perspective. In this paper,an evaluation method that combines the actual building energy consumption and users' satisfaction has been discussed,based on the post occupancy evaluation( POE) theory and the Big data technology. Through the comparison and analysis of building objective operational metrics and users subjective feelings indicators,the green buildings' POE has been achieved. Finally,the study analyzes the assessed value of green building's POE from the three-time dimensions,short-term,medium-term and long-term. And the outlook of the direction is looking forward to the follow-up study. 展开更多

Objective:To study the inhibition effect of siRNA on the expression of Wisp-1 gene in Hca-F of mouse hepatocellular carcinoma cells strain and also its effect on the proliferation,migration and adhesion of hepatocellular carcinoma cells.Methods:Three expression vectors of siRNA were constructed.Lipo2000 was employed to transfect Hca-F cells and Western blot was used to detect the inhibition effect of siRNA on the expression of Wisp-1 gene.Afterwards,CCK8 was adopted to detect the effect of Wisp-1 siRNA on the proliferation of Hca-F cells;Annexin V-FTTC/PI double staining flow cytometry was used to detect the effect of Wisp-1 siRNA on the apoptosis of Hca-F cells;Transwell was used to detect the effect of Wisp-1 siRNA on the migration of Hca-F cells.The in vitro cell adhesion kit was used to detect of Wisp-1 siRNA on the change in the components of extracellular matrix to which Hca-F cells adhered.Western blot was used to detect the activation of protein kinase B(AKT)/glycogen synthase kinase-3(3pathway and the expression of downstream target protein p53 and matrix metal!oproteinases-2.Results:The siRNA showed interference effect on the expression of Wisp-1 gene.Compared with the control group,after being transfected to cells,Wisp-1 siRNA could significantly inhibit the proliferation,migration and adhesion of Hca-F cells and also promote the cell apoptosis,which was related to the down-regulated phosphorylation of AKT and glycogen synthase kinase-3β and the expression of p53 and matrix metalIoproteinases-2(P<0.05).Conclusions:The inhibition of Wisp-1 expression can reduce the proliferation,migration and adhesion of mouse hepatocellular carcinoma cells,which is related to the AKT/ glycogen synthase kinase-3 β pathway.Wisp-1 gene may be the potential target to cure the hepatocellular carcinoma. 展开更多

AIM:To identify the pathological role of amyloid beta(Aβ) deposition in retinal degeneration,and explore Aβ deposition on the retinal pigment epithelium cells(RPE) layer and the associated structural and functional changes in Alzheimer's disease transgenic mice.METHODS:RPE changes in the eyes of APPswe/PS1 transgenic and none transgenic(NTG) mice over 20 months old were examined.Histological changes were investigated via hematoxylin and eosin(H&E) staining and transmission electron microscopy(TEM) examination,whereas the expression of amyloid precursor protein(APP),Aβ,Zonula occludens-1(ZO-1) and Ionized calcium binding adaptor molecule-1(IBA-1) were investigated using immunohistochemistry and immunofluorescence techniques.All of the obtained results were quantitatively and statistically analyzed.RESULTS:In aged transgenic mice,an APP-positive immunoreaction and Aβ deposition were detected on the RPE layer but were undetectable in NTG mice.The RPE demonstrated some vacuole changes,shortened basal infoldings and basal deposition in histopathological examination and TEM tests,wherein irregular shapes were indicated by ZO-1 disorganization through fluorescence.Furthermore,IBA-1 positive cells were observed to have accumulated and infiltrated into the RPE layer and localized beneath the RPE/Bruch's membrane(Br M) complex,which was accompanied by an increase in BrM thickness in aged transgenic mice in comparison to NTG mice.The IBA-1 positive cells were found to be co-stained with Aβ deposition on the RPE flat mounts.CONCLUSION:The observed Aβ deposition in the RPE layer may cause RPE dysfunction,which is associated with microglia cells infiltration into the retina of aged transgenic mice,suggesting that Aβ deposition probably plays a significant role in RPE-related degenerative disease. 展开更多

The superconducting magnet system of a fusion reactor plays a vital role in plasma confinement,a process that can be dis-rupted by various operational factors.A critical parameter for evaluating the temperature margin of superconducting magnets during normal operation is the nuclear heating caused by D-T neutrons.This study investigates the impact of nuclear heat-ing on a superconducting magnet system by employing an improved analysis method that combines neutronics and thermal hydraulics.In the magnet system,toroidal field(TF)magnets are positioned closest to the plasma and bear the highest nuclear-heat load,making them prime candidates for evaluating the influence of nuclear heating on stability.To enhance the modeling accuracy and facilitate design modifications,a parametric TF model that incorporates heterogeneity is established to expedite the optimization design process and enhance the accuracy of the computations.A comparative analysis with a homogeneous TF model reveals that the heterogeneous model improves accuracy by over 12%.Considering factors such as heat load,magnetic-field strength,and cooling conditions,the cooling circuit facing the most severe conditions is selected to calculate the temperature of the superconductor.This selection streamlines the workload associated with thermal-hydraulic analysis.This approach enables a more efficient and precise evaluation of the temperature margin of TF magnets.Moreover,it offers insights that can guide the optimization of both the structure and cooling strategy of superconducting magnet systems. 展开更多

BACKGROUND According to the degree of intradermal neoplasia in the colorectal exhalation,it can be divided into two grades:Low-grade intraepithelial neoplasia(LGIN)and high-grade intraepithelial neoplasia(HGIN).Currently,it is difficult to accurately diagnose LGIN and HGIN through imaging,and clinical diagnosis depends on postoperative histopathological diagnosis.A more accurate method for evaluating HGIN preoperatively is urgently needed in the surgical treatment and nursing intervention of colorectal polyps.AIM To explore the characteristics and risk factors of HGIN in older patients with colorectal polyps.METHODS We selected 84 older patients diagnosed with HGIN as the HGIN group(n=95 colonic polyps)and 112 older patients diagnosed with LGIN as the LGIN group(n=132 colonic polyps)from Shandong Provincial Hospital Affiliated to Shandong First Medical University.The endoscopic features,demographic characteristics,and clinical manifestations of the two patient groups were compared,and a logistic regression model was used to analyze the risk factors for HGIN in these patients.RESULTS The HGIN group was older and had a higher number of sigmoid colon polyps,rectal polyps,pedunculated polyps,polyps≥1.0 cm in size,polyps with surface congestion,polyps with surface depression,and polyps with villous/tubular adenomas,a higher proportion of patients with diabetes and a family history of colorectal cancer,patients who experienced rectal bleeding or occult blood,patients with elevated carcinoembryonic antigen(CEA)and cancer antigen 199(CA199),and lower nutritional levels and higher frailty levels.The polyp location(in the sigmoid colon or rectum),polyp diameter(≥1.0 cm),pathological diagnosis of(villous/tubular adenoma),family history of colorectal cancer,rectal bleeding or occult blood,elevated serum CEA and CA199 levels,lower nutritional levels and higher frailty levels also are independent risk factors for HGIN.CONCLUSION The occurrence of high-grade neoplastic transformation in colorectal polyps is closely associated with their location,size,villous/tubular characteristics,family history,elevated levels of tumor markers,and lower nutritional levels and higher frailty levels. 展开更多

目的探讨微小RNA(miR)-30a-5p、miR-129-5p在非小细胞肺癌表皮生长因子受体(EGFR)靶向治疗后病人血清中的表达以及与疗效的关系。方法2020年1月~2022年6月行靶向治疗的非小细胞肺癌EGFR基因突变病人186例,根据病人疗效分为有效组(141例... 目的探讨微小RNA(miR)-30a-5p、miR-129-5p在非小细胞肺癌表皮生长因子受体(EGFR)靶向治疗后病人血清中的表达以及与疗效的关系。方法2020年1月~2022年6月行靶向治疗的非小细胞肺癌EGFR基因突变病人186例,根据病人疗效分为有效组(141例)和无效组(45例)。比较两组血清miR-30a-5p、miR-129-5p水平;多因素logistic回归分析疗效的影响因素;ROC曲线分析血清miR-30a-5p、miR-129-5p水平对非小细胞肺癌疗效的预测价值。结果两组吸烟史、TNM分期比较,差异有统计学意义(P<0.05)。无效组血清miR-30a-5p、miR-129-5p的表达水平低于有效组,两组比较差异有统计学意义(P<0.05)。多因素logistic回归分析结果显示,血清miR-30a-5p、miR-129-5p、吸烟史、TNM分期均为非小细胞肺癌疗效的影响因素(P<0.05)。ROC曲线显示,血清miR-30a-5p、miR-129-5p二者联合预测非小细胞肺癌疗效的AUC为0.926,敏感性为91.11%,特异性为83.69%,优于两者各自单独预测(Z二者联合-miR-30a-5P=3.260、Z二者联合-miR-129-5P=3.726,P=0.001、0.000)。结论非小细胞肺癌EGFR靶向治疗后无效组病人血清miR-30a-5p、miR-129-5p水平显著低于有效组,二者联合对非小细胞肺癌EGFR靶向治疗后疗效有较好的预测价值。 展开更多

Purpose: Nanophthalmos is a rare congenital disorder associated with a high incidence of angle-closure glaucoma..We followed the clinical manifestations of three siblings to evaluate their responses to various treatments.Methods:.Three sisters with nanophthalmos were followed from 2000 to 2013..Glaucoma and cataract treatments were performed whenever indicated.Results: The oldest sister had chronic elevation of intraocular pressure(IOP) and underwent laser peripheral iridotomy(LPI)on both eyes,.followed by uneventful phacoemulsification with intraocular lens(IOL) implantation on the left eye and phacotrabeculectomy with IOL implantation on the right eye.The middle sister had acute elevation of IOP and initially underwent phacoemulsification combined with implantation of two IOLs on her left eye and LPI on her right eye. Severe uveal effusion occurred when phacoemulsification was performed on her right eye 6 years later, but ultimately was completely resolved. In both sisters, stable IOP and visual results were achieved after lensectomy. The youngest sister, who had suspected angle-closure, achieved a stable IOP and visual results with prophylactic LPI alone.Conclusion: In nanophthalmic eyes, the severity of the disease may foreshadow the severity of surgical complications and responses to therapy. 展开更多

With advantages of strong drive capability,nested-loop secondary linear machine(NLS-LM)has great potentiality in linear metro.For its secondary structure with multiple loops,it is difficult to calculate the electromagnetic thrust of NLS-LM reasonably.Hence,in this paper,one thrust calculation method is proposed considering variable loop inductance and transient loop current.Firstly,to establish the secondary winding function,the modeling domain is confined to a limited range,and the equivalent loop span is employed by analyzing the coupling relationship between primary and secondary.Then,in order to obtain the secondary flux density,the transient secondary current is solved based on the loop impedance and induced voltage.Finally,the electromagnetic thrust can be calculated reasonably by the given primary current sheet and the calculated secondary flux density.Comprehensive simulations and experiments have demonstrated the effectiveness of the proposed method. 展开更多

Estrogen is imperative to mammalian reproductivity,metabolism,and aging.However,the hormone activating estrogen receptor(ERs)αcan cause major safety concerns due to the enrichment of ERαin female tissues and certain malignancies.In contrast,ERβis more broadly expressed in metabolic tissues and the skin.Thus,it is desirable to generate selective ERβagonist conjugates for maximizing the therapeutic effects of ERs while minimizing the risks of ERαactivation.Here,we report the design and production of small molecule conjugates containing selective non-steroid ERβagonists Gtx878 or genistein.Treatment of aged mice with our synthesized conjugates improved aging-associated declines in insulin sensitivity,visceral adipose integrity,skeletal muscle function,and skin health,with validation in vitro.We further uncovered the benefits of ERβconjugates in the skin using two inducible skin injury mouse models,showing increased skin basal cell proliferation,epidermal thickness,and wound healing.Therefore,our ERβ-selective agonist conjugates offer novel therapeutic potential to improve aging-associated conditions and aid in rejuvenating skin health. 展开更多

China has the largest population in the world and has a huge medical needs.Research on China’s health workforce indicate that China encounter problems of shortages,imbalances and maldistribution in medical resources.Until 2014,China’s total health workforce are 10.2 million,of which licensed doctors are 2.4 million and licensed assistant doctors are 0.5 million(1).Most of these doctors are distributed in state-owned health institutions in relatively developed areas,working as civil servants,rather than independent medical professionals.Taking eye care as an example,there was only about 26 ophthalmologists per million population in China according to the latest domestic survey in 2016(2).And it is reported that cataract surgery rate in China is lagging behind.Only 1,425 people per million received cataract surgery in 2014,while about 5,000 people per million receive such surgery in India and 8,000 people in the US every year(3).Additionally,China has been suffering a high attrition rate among physicians and medical graduates over the past 10 years,going with low physician production,and rapid population growth(4).Based on its national conditions,China institute double reforms of its health-care systems and medical education. 展开更多

Firstly, the mechanism of static and dynamic end effects is analyzed in aspect of direct coupling, winding asymmetry and transient secondary current. Furthermore, based on the winding theory for short primary linear machines, the machine parameters are established qualitatively considering pulsating magnetic field of NLS-LDFM. Skin is the largest organ of the human body and a possible source of stem cells for research and cell-based therapy. We have isolated a population of mesenchymal stem cell-like pluripotent cells from human epidermis, termed human (h) EMSCPCs. This preliminary study tested if these hEMSCPCs can be induced to differentiate into neural-like cells. Human EMSCPCs were first cultured for four to seven days in a serum-free neural stem cell (NSC) medium for pre-induction. During pre-induction, hEMSCPCs coalesced into dense spheres that resembled neural rosettes. In the presence of a conditioned differentiation medium, pre-induced cells took on the morphological characteristics of neural cells, including slender projections with inflated or claw-like ends that contacted the soma or projections of other cells as revealed by confocal microscopy. Moreover, these differentiating cells expressed the neural-specific markers β-III tubulin, MAP2, GFAP, and synapsin I as evidenced by immunocytochemistry. Both pre-induced hEMSCPCs and uninduced hEMSCPCs were labeled with CM-DiI and transplanted into the vitreous cavities of nude mice. Transplanted cells were examined four weeks later in frozen eyeball sections by immunofluorescence staining, which demonstrated superior retinal migration and neural differentiation of pre-induced cells. Our study is the first to demonstrate that hEMSCPCs possess the capacity to differentiate into neural-like cells, suggesting potential uses for the treatment of retinal diseases such as age-related macular degeneration. 展开更多

In the middle of May to September 2014, the research team conducted survey and field measurement on rural residential energy consumption and environment condition in China’s local city in order to grasp the lifestyle pattern and its correlation on energy use in the demand side. In this paper, the analyses of people lifestyle and summer energy consumption will be presented through statistical analyses based on data which collected from questionnaire survey. The results of questionnaire were presented and discussed. Based on the questionnaire results, we classified the housing types into several groups then compared the energy use of each type. The major influential factors on housing energy consumption were analyzed and the results were discussed. The results of field measurement during summer which conducted to several types of houses were also discussed. Finally, the results will propose some strategies on lowering housing energy use in the village. Further, the results will give conceptual layout to find the correlation between environmental quality, thermal condition, and energy consumption. 展开更多

There is growing evidence of the high transmission potential of COVID-19 through virus-laden aerosols.Because aerosols are inhaled in various concentrations,an overall assessment of transmission risks at different indoor scales is crucial.However,a comprehensive risk assessment method that evaluates the direct link between short-range and room-scale zones is stl lacking.In this paper,a risk assessment model combining both short-range and room-scale effects is developed to obtain effective reproduction number in confined indoor environments,called Turbulent Jet Wells Riley(TJWR)model.Combined with the viral load data and aerosol generation data of different human respiratory activities,the Monte Carlo simulation method is applied to calculate the quanta emission rate,which further provides the input parameters of the TJWR model.Three known outbreaks(Hangzhou banquet hall X,Guangzhou restaurant Y,and Hong Kong restaurant Z,China)are chosen to validate the TJWR model.Results show that the TJWR model is more efficient than the original Wells-Riley model.The average relative error of the TJWR model ranges between 9%and 44%,while for the Wells-Riley model,it ranges between 57%and 78%.The TJWR model also proves that infection risk assessments using the well-mixed assumption can systematically underestimate the transmission risk for those close to the source.Additionally,there is a significant positive linear correlation between the total number of exposed individuals at the short-range and the effective reproduction number.This newly developed TJWR model has great potential for rapid and real-time overall airborne transmission risk assessment in buildings and cities. 展开更多
