Candidate genes conferring ethylene-response in cultivated peanuts determined by BSA-seq and fine-mapping

Ethylene plays essential roles in plant growth,development and stress responses.The ethylene signaling pathway and molecular mechanism have been studied extensively in Arabidopsis and rice but limited in peanuts.Here,we established a sand-culture method to screen pingyangmycin mutagenized peanut lines based on their specific response to ethylene(“triple response”).An ethylene-insensitive mutant,inhibition of peanut hypocotyl elongation 1(iph1),was identified that showed reduced sensitivity to ethylene in both hypocotyl elongation and root growth.Through bulked segregant analysis sequencing,a major gene related to iph1,named AhIPH1,was preliminarily mapped at the chromosome Arahy.01,and further narrowed to a 450-kb genomic region through substitution mapping strategy.A total of 7014 genes were differentially expressed among the ACC treatment through RNA-seq analysis,of which only the Arahy.5BLU0Q gene in the candidate mapping interval was differentially expressed between WT and mutant iph1.Integrating sequence variations,functional annotation and transcriptome analysis revealed that a predicated gene,Arahy.5BLU0Q,encoding SNF1 protein kinase,may be the candidate gene for AhIPH1.This gene contained two single-nucleotide polymorphisms at promoter region and was more highly expressed in iph1 than WT.Our findings reveal a novel ethylene-responsive gene,which provides a theoretical foundation and new genetic resources for the mechanism of ethylene signaling in peanuts. 展开更多

Soy isoflavones are natural tyrosine kinase inhibitors closely associated with decreased morbidity and mortality of various tumors.The activation of tyrosine kinases such as ERBB2 is the mechanism by which cholecystitis transforms into gallbladder cancer(GBC),therefore,it is important to investigate the relationship between long-term exposure to soy isoflavones and the occurrence and progression of GBC.This case-control study(n=85 pairs)found that the high level of plasma soy isoflavoneDgenistein(GEN)was associated with a lower risk of gallbladder cancer(≥326.00 ng/m L compared to≤19.30ng/m L,crude odds ratio 0.15,95%CI 0.04–0.59;P for trend=0.016),and that the level of GEN exposure negatively correlated with Ki67 expression in GBC tissue(n=85).Consistent with these results,the proliferation of GBC cells was inhibited in the long-term exposure models of GEN in vitro and in vivo.The long-term exposure to GEN reduced the tyrosine kinase activity of ERBB2 and impaired the function of the PTK6-AKT-GSK3βaxis,leading to downregulation of the MCM complex in GBC cells.In summary,long-term exposure to GEN associated with soy products intake might play a certain role in preventing GBC and even inhibiting the proliferation of GBC cells. 展开更多

Metasurfaces composed of spatially arranged ultrathin subwavelength elements are promising photonic devices for manipulating optical wavefronts,with potential applications in holography,metalens,and multiplexing communications.Finding microstructures that meet light modulation requirements is always a challenge in designing metasurfaces,where parameter sweep,gradient-based inverse design,and topology optimization are the most commonly used design methods in which the massive electromagnetic iterations require the design computational cost and are sometimes prohibitive.Herein,we propose a fast inverse design method that combines a physicsbased neural network surrogate model(NNSM)with an optimization algorithm.The NNSM,which can generate an accurate electromagnetic response from the geometric topologies of the meta-atoms,is constructed for electromagnetic iterations,and the optimization algorithm is used to search for the on-demand meta-atoms from the phase library established by the NNSM to realize an inverse design.This method addresses two important problems in metasurface design:fast and accurate electromagnetic wave phase prediction and inverse design through a single phase-shift value.As a proof-of-concept,we designed an orbital angular momentum(de)multiplexer based on a phase-type metasurface,and 200 Gbit/s quadrature-phase shift-keying signals were successfully transmitted with a bit error rate approaching 1.67×10^(-6).Because the design is mainly based on an optimization algorithm,it can address the“one-to-many”inverse problem in other micro/nano devices such as integrated photonic circuits,waveguides,and nano-antennas. 展开更多
