Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort wit...Objective:To test the diagnostic performance of percent free prostate-specific antigen(%fPSA)in predicting any prostate cancer(PCa)and high-grade prostate cancer(HGPCa)in a retrospective multi-center biopsy cohort with a PSA level of 4.0e10.0 ng/mL in China.Methods:Consecutive patients with a PSA of 4.0-10.0 ng/mL who underwent transrectal ultrasound-guided biopsy were enrolled at 16 Chinese medical centers from January 1st,2010 to December 31st,2013.Total and free serum PSA determinations were performed using three types of electro-chemiluminescence immunoassays recalibrated to the World Health Organization(WHO)standard.The diagnostic accuracy of PSA,%fPSA,and %fPSA in combination with PSA(%fPSA t PSA)was determined using the area under the receiver operating characteristic(ROC)curve(AUC).Results:A total of 2310 consecutive men with PSA levels between 4.0 and 10.0 ng/mL were included,and the detection rate of PCa was 25.1%.The AUC of%fPSA and %fPSA t PSA in predicting any PCa was superior to PSA alone in men aged≥60 years(0.623 vs.0.534,p<0.0001)but not in men aged 40e59 years(0.517 vs.0.518,p=0.939).Similar result was yield in predicting HGPCa.Conclusion:In a clinical setting of Chinese men with 4.0e10.0 ng/mL PSA undergoing initial prostate biopsy,adding %fPSA to PSA can moderately improve the diagnostic accuracy for any PCa and HGPCa compared with PSA alone in patients≥60 but not in patients aged 40-59 years.展开更多

Background:Ovarian cancer is one of the most widespread malignant diseases of the female reproductive system worldwide.The plurality of ovarian cancer is diagnosed with metastasis in the abdominal cavity.Epithelial-me...Background:Ovarian cancer is one of the most widespread malignant diseases of the female reproductive system worldwide.The plurality of ovarian cancer is diagnosed with metastasis in the abdominal cavity.Epithelial-mesenchymal transition(EMT)exerts a vital role in tumor cell metastasis.However,it remains unclear whether long non-coding RNA(lncRNA)are implicated in EMT and influence ovarian cancer cell invasion and metastasis.This study was designed to investigate the impacts of lncRNA AC005224.4 on ovarian cancer.Methods:LncRNA AC005224.4,miR-140-3p,and snail family transcriptional repressor 2(SNAI2)expression levels in ovarian cancer and normal ovarian tissues were determined using real-time quantitative polymerase chain reaction(qRT-PCR).Cell Counting Kit-8(CCK-8)and Transwell(migration and invasion)assays were conducted to measure SKOV3 and CAOV-3 cell proliferation and metastasis.E-cadherin,N-cadherin,Snail,and Vimentin contents were detected using Western blot.Nude mouse xenograft assay was utilized to validate AC005224.4 effects in vivo.Dual-luciferase reporter gene assay confirmed the targeted relationship between miR-140-3p and AC005224.4 or SNAI2.Results:AC005224.4 and SNAI2 upregulation and miR-140-3p downregulation were observed in ovarian cancer tissues and cells.Silencing of AC005224.4 observably moderated SKOV3 and CAOV-3 cell proliferation,migration,invasion,and EMT process in vitro and impaired the tumorigenesis in vivo.miR-140-3p was a target of AC005224.4 and its reduced expression level was mediated by AC005224.4.miR-140-3p mimics decreased the proliferation,migration,and invasion of ovarian cancer cells.SNAI2 was identified as a novel target of miR-140-3p and its expression level was promoted by either AC005224.4 overexpression or miR-140-3p knockdown.Overexpression of SNAI2 also facilitated ovarian cancer cell viability and metastasis.Conclusion:AC005224.4 was confirmed as an oncogene via sponging miR-140-3p and promoted SNAI2 expression,contributing to better understanding of ovarian cancer pathogenesis and shedding light on exploiting the novel lncRNA-directed therapy against ovarian cancer.展开更多
Dear Editor,Aberrant DNA methylation gets involved in cancer initiation,progression,and recurrence,which in turn makes it an ideal cancer biomarker.Various methylation markers or their panels have been developed in di...Dear Editor,Aberrant DNA methylation gets involved in cancer initiation,progression,and recurrence,which in turn makes it an ideal cancer biomarker.Various methylation markers or their panels have been developed in diverse cancer types.However,the model-constructing based marker mining strategy and incompatibility of application have greatly impeded their ways to clinic.Thus,single methylation marker applicable to all/most cancer types and multiple clinical scenarios is desperately needed.The hope came from the unexpected observation that HIST1H4F was universally hypermethylated in all 17 cancer types;thus,we raised the concept of“Universal Cancer Only Marker(UCOM)”and established a paradigm for discovery and clinical application of UCOM.1 Recently,a novel UCOM,hypermethylated PCDHGB7,was identified and found to advance cervical cancer(CC)screening to the precancerous stage.2 During the screening of UCOM,we discerned a bunch of cancer cell-differentially methylated regions.1 Among them,sine oculis(SIX)homeobox family of transcription factors,which were found to function as tumorigenesis regulator by promoting epithelial-to-mesenchymal transition and metastasis recently in addition to their traditional roles in tissue formation and organogenesis,3 sparked our special attention.Herein,we interrogate whether SIX6 methylation could serve as a novel UCOM and its potential applications.展开更多
