Management Measures for Preventing Postoperative Incision Infections in General Surgery at Primary Hospitals

Objective:To analyze the management measures and effects of preventing postoperative incision infections in the general surgery department of primary hospitals.Methods:Forty-nine surgical patients with 11 healthcare workers who were admitted to the general surgery department of the primary hospital between August 2021 and August 2022 were selected as the routine group for routine incision infection management.Forty-nine surgical patients with 11 healthcare workers admitted to the same department between September 2022 and September 2023 were selected as the prevention group for prophylactic management of postoperative incision infections.The incision infection rate,knowledge,attitude,and practice(KAP)scores,and management satisfaction of the patients as well as the management skill scores of healthcare workers were compared between the two groups.Results:The rate of postoperative incision infection in the prevention group was lower than that in the routine group;after implementing management measures,patients in the prevention group had higher KAP scores than those in the routine group;patients in the prevention group were more satisfied with the management than those in the routine group;and healthcare workers in the prevention group had higher scores than those in the routine group,with P<0.05 for the comparison between the groups.Conclusion:The implementation of preventive management for general surgery patients in primary hospitals can reduce the incidence of postoperative incision infection and improve the KAP of patients,with higher management satisfaction.It can also enhance the management skills of healthcare workers,thus improving their overall management level. 展开更多

The dwarf architecture is an important and valuable agronomic trait in watermelon breeding and has the potential to increase fruit yield and reduce labor cost in crop cultivation.However,the molecular basis for dwarfism in watermelon remains largely unknown.In this study,a recessive dwarf allele(designated as Cldf(Citrullus lanatus dwarfism))was fine mapped in a 32.88 kb region on chromosome 09 using F2 segregation populations derived from reciprocal crossing of a normal line M08 and a dwarf line N21.Gene annotation of the corresponding region revealed that the Cla015407 gene encoding a gibberellin 3β-hydroxylase functions as the best possible candidate gene for Cldf.Sequence analysis showed that the fourth polymorphism site(a G to A point mutation)at the 3′AG splice receptor site of the intron leads to a 13 bp deletion in the coding sequence of Cldf in dwarf line N21 and thus results in a truncated protein lacking the conserved domain for binding 2-oxoglutarate.In addition,the dwarf phenotype of Cldf could be rescued by exogenous GA3 application.Phylogenetic analysis suggested that the small multigene family GA3ox(GA3 oxidase)in cucurbit species may originate from three ancient lineages in Cucurbitaceae.All these data support the conclusion that Cldf is a GA-deficient mutant,which together with the cosegregated marker can be used for breeding new dwarf cultivars. 展开更多

Melatonin is a pleiotropic signaling molecule that regulates plant growth and responses to various abiotic stresses.The last step of melatonin synthesis in plants can be catalyzed by caffeic acid O-methyltransferase(COMT),a multifunctional enzyme reported to have N-acetylserotonin O-methyltransferase(ASMT)activity;however,the ASMT activity of COMT has not yet been characterized in nonmodel plants such as watermelon(Citrullus lanatus).Here,a total of 16 putative O-methyltransferase(ClOMT)genes were identified in watermelon.Among them,ClOMT03(Cla97C07G144540)was considered a potential COMT gene(renamed ClCOMT1)based on its high identities(60.00–74.93%)to known COMT genes involved in melatonin biosynthesis,expression in almost all tissues,and upregulation under abiotic stresses.The ClCOMT1 protein was localized in the cytoplasm.Overexpression of ClCOMT1 significantly increased melatonin contents,while ClCOMT1 knockout using the CRISPR/Cas-9 system decreased melatonin contents in watermelon calli.These results suggest that ClCOMT1 plays an essential role in melatonin biosynthesis in watermelon.In addition,ClCOMT1 expression in watermelon was upregulated by cold,drought,and salt stress,accompanied by increases in melatonin contents.Overexpression of ClCOMT1 enhanced transgenic Arabidopsis tolerance against such abiotic stresses,indicating that ClCOMT1 is a positive regulator of plant tolerance to abiotic stresses. 展开更多

Although male sterility has been identified as a useful trait for hybrid vigor utilization and hybrid seed production,its underlying molecular mechanisms in Cucurbitaceae species are still largely unclear.Here,a spontaneous male-sterile watermelon mutant,Se18,was reported to have abnormal tapetum development,which resulted in completely aborted pollen grains.Map-based cloning demonstrated that the causal gene Citrullus lanatus Abnormal Tapetum 1(ClATM1)encodes a basic helix-loop-helix(bHLH)transcription factor with a 10-bp deletion and produces a truncated protein without the bHLH interaction and functional(BIF)domain in Se18 plants.qRT–PCR and RNA in situ hybridization showed that ClATM1 is specifically expressed in the tapetum layer and in microsporocytes during stages 6–8a of anther development.The genetic function of ClATM1 in regulating anther development was verified by CRISPR/Cas9-mediated mutagenesis.Moreover,ClATM1 was significantly downregulated in the Se18 mutant,displaying a clear dose effect at the transcriptional level.Subsequent dual-luciferase reporter,β-glucuronidase(GUS)activity,and yeast one-hybrid assays indicated that ClATM1 could activate its own transcriptional expression through promoter binding.Collectively,ClATM1 is the first male sterility gene cloned from watermelon,and its self-regulatory activity provides new insights into the molecular mechanism underlying anther development in plants. 展开更多

Assessing benthic habitat quality using biotic indices in the Laizhou Bay, China 被引量:2

As an important part of the Bohai economic rim,the Laizhou Bay has been stressed by serious eco-environmental problems in recent years.In this study,the Shannon-Wiener index(H′),AZTI’s marine biotic index(AMBI)and the multivariate AMBI(M-AMBI)were used to assess the ecological quality status(EQS)of the Laizhou Bay according to macrobenthos data collected annually in August 2011–2014.The results showed that the overall benthic habitat quality in the Laizhou Bay was assessed as“Good”.However,25%of the samples were classified as“Moderate”,“Bad”or“Poor”status under degraded conditions.Ecological group III(EGIII)species which had a certain tolerance to environmental disturbances had a higher proportion in each station,and most of them had appeared the pollution indicator species Capitella capitata.This indicated that the benthic habitat in the Laizhou Bay had been disturbed and polluted to some extent.The comparison of the three indices evaluation result and the RDA analysis showed that the H′and M-AMBI were more suitable when the relative abundance of the single species was high and the macrobenthic community was significantly imbalanced;when the relative abundance of opportunistic species(EGIV and EGV)was high,the AMBI and M-AMBI could reflected the EQS objectively;in an undisturbed and polluted environment,all the three indices could indicate the benthic habitats quality.In summary,the better correlation between the three indices and environmental factors showed that they were well responsive to the tendency of the benthic habitats quality in the Laizhou Bay. 展开更多

Seedlessness is a valuable agronomic trait in watermelon(Citrullus lanatus)breeding.Conventional less seed watermelons are mainly triploid,which has many disadvantages due to unbalanced genome content.Less seed watermelon can be achieved at the diploid level when certain reproductive genes are mutated or by chromosome translocation,which leads to defects during meiosis.However,the formation mechanism of diploid less seed watermelons remains largely unknown.Here,we identified a spontaneous mutant line,watermelon line“148”,which can set seeds normally when self-pollinated.A total of 148×JM F_(1) hybrid plants exhibited seed number reductions to 50.3%and 47.3%of those of the two parental lines,respectively,which are considered to be less seed.Examination of pollen viability and hybridization experiments revealed that F 1 hybrids produce semisterile pollen and ovules.Further cytological observations indicated that semisterility was a result of a reciprocal translocation of chromosomes,which exhibited one quadrivalent ring of four chromosomes at prometaphase I during meiosis.RT-qPCR analysis indirectly confirmed that the semisterile phenotype is caused by chromosome translocation rather than disruption of specific meiotic gene expression.F 2 population genetic analysis indicated that the“148”watermelon line is a homozygous translocation and that the less seed phenotype of the F_(1) hybrid is prompted by one chromosome fragment translocation.The translocated fragment was further fine mapped to a 2.09 Mb region on chromosome 6 by whole-genome resequencing and genetic map cloning procedures.Our work revealed that a 2.09Mb chromosome fragment translocation on chromosome 6,causing meiotic defects at metaphase I during meiosis,leads to diploid less seed watermelon.Our findings provide a new promising method for less seed watermelon breeding at the diploid level,as well as a fragment size reference for breeding less seed watermelon through artificially induced chromosome translocation. 展开更多

Introduction:Sediment and nutrient retention have significant impacts on agricultural productivity and water quality.Obvious changes of climate and land-use have deeply affected freshwater ecosystem services in the last several decades.Nevertheless,it is unclear what the differences are in the impact of climate and land-use changes on freshwater ecosystem services.Outcomes:The impacts of climate and land-use change on sediment and nutrient retention(two essential freshwater ecosystem services)were evaluated with the help of the Integrated Valuation of Environmental Services and Tradeoffs(InVEST)model.The results from the study area suggest a decreasing trend in sediment and nutrient retention from 1995 to 2015 when climate and land-use were changed simultaneously.A decreasing trend in sediment retention and an increasing trend in nutrient retention were presented under scenario A(climate change from 1995 to 2015 with land-use unchanged),while the sediment exported and nutrient retention exhibited a decreasing trend except sediment retained under scenario B(land-use change from 1995 to 2015 with unchanged climate).Rainfall erosivity and soil erodibility were more sensitive to the output of sediment retention,while retention efficiency produced more of an effect on nutrient retention.Discussion and Conclusion:Climate change played a dominant role in the change in sediment retention,while land-use change had a more significant effect on nutrient retention change than climate change.The results of this study provide explicitinformation for land-use managers to reduce the negative effects associated with sediment and nutrient retention. 展开更多

Buildings are responsible for more than forty percent of global energy consump-tion and as much as one third of global greenhouse gas emissions.Meanwhile,the energy conservation and exhaust reduction of a building can be easily understood by accurately calculating a building’s carbon emissions during its operational stage.In the present study,a system dynamics(SD)approach to calculate the energy consumption and carbon emissions from a building during its operational stage is quantitatively developed through a case study on an office building in Nanjing.The obtained results demonstrate that:a)the difference between the results of SD and that of EnergyPlus is so small that a SD approach is acceptable;b)the variation between the real monitored data and that of simulation by SD and EnergyPlus is reasonable;c)the physical meanings of the variables in the SD model are clear;d)the parameters of the SD model and the relationships between the variables can be determined by a qualitative-and-quantitative combined analysis. 展开更多
