Although several approaches for the enrichment of EpSCs have been established, enriching a pure population of viable EpSCs is still a challenging task. An improved approach is worth developing to enhance the purity and viability of EpSCs. Here we report that cell size combined with collagen type IV adhesiveness can be used in an improved approach to enrich pure and viable human EpSCs. We separated the rap- idly adherent keratinocytes into three populations that range in size from 5-7 μm (population A), to 7-9 μm (population B), to ≥9μm (population C) in diameter, and found that human putative EpSCs could be further enriched in population A with the smallest size. Among the three populations, population A displayed the highest density of plintegrin receptor, contained the highest percentage of cells in G0/G1 phase, showed the highest nucleus to cytoplasm ratio, and possessed the highest colony formation efficiency (CFE). When injected into murine blastocysts, these cells participated in multi-tissue formation. More significantly, compared with a previous approach that sorted putative EpSCs according to pl-integrin antibody staining, the viability of the EpSCs enriched by the improved approach was significantly enhanced. Our results provide a putative strategy for the enrichment of human EpSCs, and encourage further study into the role of cell size in stem cell biology.展开更多

The radial and axial displacements are assumed to have quadratic and cubic variations through the thickness, respectively. It is important that the radial stress is approximated by a cubic expansion satisfying the boundary conditions at the inner and outer surfaces, and the corresponding strain should be least-squares compatible with the strain derived from the strain-displacement relation. The equations of motion are derived from the integration of the equilibrium equations of stresses, which are solved by precise integration method (PIM). Numerical results are.obtained, and compared with FE simulations and dynamic thermo-elasticity solutions, which indicates that the high order shell theory is capable of predicting the transient thermal response of an orthotropic (or isotropic) thick hollow cylinder efficiently, and for the detonation tube of actual pulse detonation engines (PDE) heated continuously, the thermal stresses will become too large to be neglected, which are not like those in the one time experiments with very short time.展开更多

This kind of deposits have simple mineralogy dominated by galena and sphalerite commonly associated with calcite and other gangue minerals. Both galena and sphalerite have low and high variable Re concentrations and thus Re-Os dating of these minerals have been less promising. In addition, the recovery of Re is extremely low for galena when conventional method was applied, lending additional difficulty in precisely dating galena. In this study, we investigate the recovery of Re using different media for anion exchange separation and reporte a revised preparation method for Re-Os dating of galena and sphalerite. By using the new protocol, two reliable Re-Os isochron ages of galena and sphalerite from the Fule(20.4±3.2 Ma) and Laochang(308±25 Ma) Pb-Zn deposits in Yunnan Province, SW China, are achieved.展开更多

文摘The transient thermal response of a thick orthotropic hollow cylinder with finite length is studied by a high order shell theory. Numerical results are.obtained, and compared with FE simulations and dynamic thermo-elasticity solutions, which indicates that the high order shell theory is capable of predicting the transient thermal response of an orthotropic (or isotropic) thick hollow cylinder efficiently, and for the detonation tube of actual pulse detonation engines (PDE) heated continuously, the thermal stresses will become too large to be neglected, which are not like those in the one time experiments with very short time.

文摘For polar codes, the performance of successive cancellation list(SCL)decoding is capable of approaching that of maximum likelihood decoding. However, the existing hardware architectures for the SCL decoding suffer from high hardware complexity due to calculating L decoding paths simultaneously, which are unfriendly to the devices with limited logical resources, such as field programmable gate arrays(FPGAs).In this paper,we propose a list-serial pipelined hardware architecture with low complexity for the SCL decoding, where the serial calculation and the pipelined operation are elegantly combined to strike a balance between the complexity and the latency. Moreover,we employ only one successive cancellation(SC)decoder core without L×L crossbars, and reduce the number of inputs of the metric sorter from 2L to L+2.Finally,the FPGA implementations show that the hardware resource consumption is significantly reduced with negligible decoding performance loss.

文摘Elastic diodes with nonreciprocity have the potential to enable unidirectional modulation of elastic waves. However,it is a challenge to achieve nonreciprocity at low frequencies(<100 Hz)using existing elastic diodes. This paper proposes a quasizero-stiffness(QZS)elastic diode to resolve such a tough issue and fulfill high-quality low-frequency nonreciprocity. The proposed elastic diode is invented by combining a QZS locally resonant metamaterial with a linear one, where the beneficial nonlinearity of the QZS metamaterial facilitates opening an amplitude-dependent band gap at very low frequencies.Firstly, the dispersion relation of the QZS metamaterial is derived theoretically based on the harmonic balance method(HBM).Then, the transmissibility of the QZS elastic diode in both the forward and backward directions is calculated through theoretical analyses and numerical simulations.Additionally, the influences of system parameters on the low-frequency nonreciprocal effect are discussed. The results indicate that considerable nonreciprocity is observed at a quite low frequency(e.g.,9 Hz),which is achieved by amplitude-dependent local resonance combined with interface reflection. Finally,a machine learning-based design optimization is introduced to evaluate and enhance the nonreciprocal effect of the QZS elastic diode. With the aid of machine learning(ML),the computational cost of predicting nonreciprocal effects during design optimization can be significantly reduced.Through design optimization, the nonreciprocal frequency bandwidth can be broadened while maintaining considerable isolation quality at low frequencies.

文摘The embryonic mesoderm comprises heterogeneous cell subpopulations with distinct lineage biases. It is unclear whether a bias for the human hematopoietic lineage emerges at this early developmental stage. In this study,we integrated single-cell transcriptomic analyses of human mesoderm cells from embryonic stem cells and embryos, enabling us to identify and define the molecular features of human hematopoietic mesoderm(HM)cells biased towards hematopoietic lineages. We discovered that BMP4 plays an essential role in HM specification and can serve as a marker for HM cells.Mechanistically,BMP4 acts as a downstream target of HDAC1,which modulates the expression of BMP4 by deacetylating its enhancer.Inhibition of HDAC significantly enhances HM specification and promotes subsequent hematopoietic cell differentiation. In conclusion, our study identifies human HM cells and describes new mechanisms for human hematopoietic development.

文摘Dating of lead-zinc deposits is of critical importance for better understanding of ore genesis, but has long been a big challenge due to the lack of suitable minerals that can be unequivocally linked to the ore genesis and that can be used for tradition radiometric methods. By using the new protocol, two reliable Re-Os isochron ages of galena and sphalerite from the Fule(20.4±3.2 Ma) and Laochang(308±25 Ma) Pb-Zn deposits in Yunnan Province, SW China, are achieved.

文摘Vibration reduction has always been one of hot and important topics in mechanical engineering, especially for the special measurement instrument. In this paper,a novel limb-inspired bionic structure is proposed to generate negative stiffness and design a new quasi-zero stiffness isolator via torsion springs, distinguishing from the existing tension spring structures in the literature. The nonlinear mathematical model of the proposed structure is developed and the corresponding dynamic properties are further investigated by using the Harmonic Balance method and ADAMS verification. To evaluate the vibration isolation performance, typical three-springs quasi-zero stiffness(TS QZS)system is selected to compare with the proposed bionic structure. And the graphical processing unit(GPU)parallel technology is applied to perform necessary two-parameter analyses, providing more insights into the effects of parameters on the transmissibility. It is shown that the proposed structure can show advantages over the typical TS QZS system in a wider vibration isolation range for harmonic excitation case and shorter decay time for the impact excitation case.

文摘In this paper,a tunable locally resonant metamaterial is proposed for low-frequency band gaps. The local resonator composed of two pairs of folded slender beams and a proof mass is designed based on the theory of compliant mechanism. The design optimization on geometric parameters is carried out to fulfil the quasi-zero-stiffness property. The locally resonant metamaterial is formed by periodically arranged unit cells, and the transmittance of longitudinal wave is studied through three aspects:numerical predictions, finite element simulations and experimental tests. The variation trends revealed by these three methods match well with one another:the band gap moves to lower frequency and both its depth and width get smaller and smaller with the increase of pre-compression(Δ).The band gap overlays the frequency range of 73.10–92.38 Hz and 16.78–19.49 Hz atΔ=0mm andΔ=10mm,respectively,providing a wide range of tunability.Besides, the ultralow-frequency band gap can be achieved asΔapproaches 10 mm.This study may provide an avenue for achieving the tunable ultralow-frequency locally resonant band gap.

文摘Hair follicles(HFs)undergo cycles of degeneration(catagen),rest(telogen),and regeneration(anagen)phases.Anagen begins when the hair follicle stem cells(HFSCs)obtain sufficient activation cues to overcome suppressive signals, mainly the BMP pathway, from their niche cells. Here,we unveil that mTOR complex 1(mTORC1)signaling is activated in HFSCs,which coincides with the HFSC activation at the telogen-to-anagen transition.By using both an inducible conditional gene targeting strategy and a pharmacological inhibition method to ablate or inhibit mTOR signaling in adult skin epithelium before anagen initiation,we demonstrate that HFs that cannot respond to mTOR signaling display significantly delayed HFSC activation and extended telogen.Unexpectedly,BMP signaling activity is dramatically prolonged in mTOR signaling-deficient HFs.Through both gain-and loss-of-function studies in vitro,we show that mTORC1 signaling negatively affects BMP signaling, which serves as a main mechanism whereby mTORC1 signaling facilitates HFSC activation.Indeed,in vivo suppression of BMP by its antagonist Noggin rescues the HFSC activation defect in mTORC1-null skin. Our findings reveal a critical role for mTOR signaling in regulating stem cell activation through counterbalancing BMP-mediated repression during hair regeneration.

文摘Friction plays a pivotal role in the sliding-mode triboelectric energy harvester(TEH),which not only enables the charge transfer between two dielectrics, but also influences the energy harvesting performance by affecting the dynamic response of the TEH.How to evaluate the effects of the friction on TEHs is important for optimizing TEHs in engineering practices. In order to analyze the effects of the friction on the dynamic response and evaluate the energy harvesting performance ofTEHs,the paper models the friction of a devised non-linear TEH based on the Coulomb friction model and the Macro-slip friction theory. The TEH equips a pair of magnets, rendering a switching between the bistability and the monostability by tuning the distance between two magnets. The dynamic model of the non-linear TEH is established by the extended Hamilton principle. The effects of friction in slidingmode TEH are dissected in detail. The influences of parameters on both the mechanical and electrical responses are also systemically studied to explore an optimal energy harvesting performance in the low-frequency range. This work provides a guideline for designing and accurately analyzing a sliding-mode TEH.
