The surface properties of oxidic supports and their interaction with the supported metals play critical roles in governing the catalytic activities of oxide‐supported metal catalysts.When metals are supported on redu...The surface properties of oxidic supports and their interaction with the supported metals play critical roles in governing the catalytic activities of oxide‐supported metal catalysts.When metals are supported on reducible oxides,dynamic surface reconstruction phenomena,including strong metal–support interaction(SMSI)and oxygen vacancy formation,complicate the determination of the structural–functional relationship at the active sites.Here,we performed a systematic investigation of the dynamic behavior of Au nanocatalysts supported on flame‐synthesized TiO_(2),which takes predominantly a rutile phase,using CO oxidation above room temperature as a probe reaction.Our analysis conclusively elucidated a negative correlation between the catalytic activity of Au/TiO_(2) and the oxygen vacancy at the Au/TiO_(2) interface.Although the reversible formation and retracting of SMSI overlayers have been ubiquitously observed on Au/TiO_(2) samples,the catalytic consequence of SMSI remains inconclusive.Density functional theory suggests that the electron transfer from TiO_(2) to Au is correlated to the presence of the interfacial oxygen vacancies,retarding the catalytic activation of CO oxidation.展开更多
Wastewater treatment plants(WWTPs)are important and energy-intensive municipal infrastructures.High energy consumption and relatively low operating performance are major challenges from the perspective of carbon neutr...Wastewater treatment plants(WWTPs)are important and energy-intensive municipal infrastructures.High energy consumption and relatively low operating performance are major challenges from the perspective of carbon neutrality.However,water-energy nexus analysis and models for WWTPs have rarely been reported to date.In this study,a cloud-model-based energy consumption analysis(CMECA)of a WWTP was conducted to explore the relationship between influent and energy consumption by clustering its influent’s parameters.The principal component analysis(PCA)and K-means clustering were applied to classify the influent condition using water quality and volume data.The energy consumption of the WWTP is divided into five standard evaluation levels,and its cloud digital characteristics(CDCs)were extracted according to bilateral constraints and golden ratio methods.Our results showed that the energy consumption distribution gradually dispersed and deviated from the Gaussian distribution with decreased water concentration and quantity.The days with high energy efficiency were extracted via the clustering method from the influent category of excessive energy consumption,represented by a compact-type energy consumption distribution curve to identify the influent conditions that affect the steady distribution of energy consumption.The local WWTP has high energy consumption with 0.3613 kW·h·m^(-3)despite low influent concentration and volumes,across four consumption levels from low(I)to relatively high(IV),showing an unsatisfactory operation and management level.The average oxygenation capacity,internal reflux ratio,and external reflux ratio during high energy efficiency days recognized by further clustering were obtained(0.2924-0.3703 kg O_(2)·m^(-3),1.9576-2.4787,and 0.6603-0.8361,respectively),which could be used as a guide for the days with low energy efficiency.Consequently,this study offers a water-energy nexus analysis method to identify influent conditions with operational management anomalies and can be used as an empirical reference for the optimized operation of WWTPs.展开更多
The object of this study is to prepare iron species-impregnated granular activated carbon as particle electrodes in order to improve their adsorption and electrocatalytic degradation capacity in Benzothiazole removal....The object of this study is to prepare iron species-impregnated granular activated carbon as particle electrodes in order to improve their adsorption and electrocatalytic degradation capacity in Benzothiazole removal.The incorporation of Fe-containing catalysts was performed by Fe(NO_3)_3 impregnation.The obtained samples were characterized by BET,Fourier transform infrared spectroscopy,SEM-EDS,powder X-ray diffraction,X-ray photoelectron spectra and TG.Compared with pure activated carbon,this modified particle electrodes show higher static adsorption capacities and TOC removal,which have respectively increased by25.9% and 54.4%.Both physisorption and chemisorption exist in the process of benzothiazole adsorption,where the latter plays a major role.In this way,the Fe-containing catalysts on modified particle electrodes are demonstrated to make a greater contribution to the improvement of electrocatalytic degradation by decreasing the activated energy by 32%.展开更多
Electrochemical CO2 reduction to chemicals or fuels presents one of the most promising strategies for managing the global carbon balance, which yet poses a significant challenge due to lack of efficient and durable el...Electrochemical CO2 reduction to chemicals or fuels presents one of the most promising strategies for managing the global carbon balance, which yet poses a significant challenge due to lack of efficient and durable electrocatalyst as well as the understanding of the CO2 reduction reaction(CO2RR) mechanism.Benefiting from the large surface area, high electrical conductivity, and tunable structure, carbon-based metal-free materials(CMs) have been extensively studied as cost-effective electrocatalysts for CO2RR.The development of CMs with low cost, high activity and durability for CO2RR has been considered as one of the most active and competitive directions in electrochemistry and material science.In this review article,some up-to-date strategies in improving the CO2RR performance on CMs are summarized.Specifically, the approaches to optimize the adsorption of CO2RR intermediates, such as tuning the physical and electronic structure are introduced, which can enhance the electrocatalytic activity of CMs effectively.Finally, some design strategies are proposed to prepare CMs with high activity and selectivity for CO2RR.展开更多

Methods Expression of GNAI1 in HCC samples was determined by qRT-PC...Objective To explore the role and regulation of guanine nucleotide-binding protein G(i), a-1 subunit (GNAI1) in hepatocellular carcinoma (HCC). Methods Expression of GNAI1 in HCC samples was determined by qRT-PCR and immunohistochemical (IHC) staining. Huh-7 and SNU-387 cells stably expressing GNAI1 were established by the infection of lentivirus transducing unit containing GNAI1. siRNA against GNAI1 was transfected into SMMC-7721 cells to knock down the GNAI1 expression in HCC cells. Mir-320a/c/d mimics were transfected into SMMC-7721 and SK-Hep-1 cells and the expression of GNAll was determined by Western blot. The migration and invasion of Huh-7, SNU-387, SK-Hep-1 and SMMC-7721 cells were investigated by Transwell assays. Results The GNAI1 protein was significantly downregulated in HCC samples without changes in its mRNA levels. GNAI1 could inhibit the migration and invasion of HCC cells in vitro. Further investigations indicated that GNAI1 was a target of miR-320a/c/d in HCC cells. Transwell assays demonstrated that these microRNAs could promote the migratory ability and invasivesess of HCC cells in vitro. Conclusions GNAII is downregulated in HCC and inhibits the migration and invasion of HCC cells. This study is the first to investigate the role of GNAI1 in cancer. Regulation of GNAI1 by miR-320a/c/d indicates new therapeutic avenues for targeting HCC metastasis.展开更多
The low efficiency of oxygen evolution reaction(OER) is regarded as one of the major roadblocks for metal-air batteries and water electrolysis.Herein,a high-performance OER catalyst of NiFe_(0.2)(oxy)hydroxide(NiFe_(0...The low efficiency of oxygen evolution reaction(OER) is regarded as one of the major roadblocks for metal-air batteries and water electrolysis.Herein,a high-performance OER catalyst of NiFe_(0.2)(oxy)hydroxide(NiFe_(0.2)-O_(x)H_(y)) was developed through topotactic transformation of a Prussian blue analogue in an alkaline solution,which exhibits a low overpotential of only 263 mV to reach a current density of 10 mA cm^(-2) and a small Tafel slope of 35 mV dec-1.Ex-situ/operando Raman spectroscopy results indicated that the phase structure of NiFe_(0.2)-O_(x)H_(y) was irreversibly transformed from the type of α-Ni(OH)_(2) to γ-NiOOH with applying an anodic potential,while ex-situ/operando 57Fe Mossbauer spectroscopic studies evidenced the in-situ production of abundant high-valent iron species under OER conditions,which effectively promoted the OER catalysis.Our work elucidates that the amount of high-valent iron species in-situ produced in the NiFe(oxy)hydroxide has a positive correlation with its water oxidation reaction performance,which further deepens the understanding of the mechanism of NiFe-based electrocatalysts.展开更多
Oxygen is an essential molecule for animal respiration,growth,and survival.Unlike in terrestrial environments,contamination and climate change have led to the frequent occurrence of hypoxia in aquatic environments,thu...Oxygen is an essential molecule for animal respiration,growth,and survival.Unlike in terrestrial environments,contamination and climate change have led to the frequent occurrence of hypoxia in aquatic environments,thus impacting aquatic animal survival.However,the adaptative mechanisms underlying fish responses to environmental hypoxia remain largely unknown.Here,we used large yellow croaker(Larimichthys crocea)and large yellow croaker fry(LYCF)cells to investigate the roles of the Hif-1α/Hsf1/Hsp70 signaling pathway in the regulation of cellular redox homeostasis,and apoptosis.We confirmed that hypoxia induced the expression of Hif-1α,Hsf1,and Hsp70 in vivo and in vitro.Genetic Hsp70 knockdown/overexpression indicated that Hsp70 was required for maintaining redox homeostasis and resisting oxidative stress in LYCF cells under hypoxic stress.Hsp70 inhibited caspase-dependent intrinsic apoptosis by maintaining normal mitochondrial membrane potential,enhancing Bcl-2 mRNA and protein expression,inhibiting Bax and caspase3 mRNA expression,and suppressing caspase-3 and caspase-9 activation.Hsp70 suppressed caspaseindependent intrinsic apoptosis by inhibiting nuclear translocation of apoptosis-inducing factor(AIF)and disturbed extrinsic apoptosis by inactivating caspase-8.Genetic knockdown/overexpression of Hif-1αand dual-luciferase reporter assay indicated that Hif-1αactivated the Hsf1 DNA promoter and enhanced Hsf1 mRNA transcription.Hsf1 enhanced Hsp70 mRNA transcription in a similar manner.In summary,the Hif-1α/Hsf1/Hsp70 signaling pathway plays an important role in regulating redox homeostasis and anti-apoptosis in L.crocea under hypoxic stress.展开更多
Objective:We aimed to directly compare the estimated effects of adherence to a healthy lifestyle with those of risk predisposition according to known genetic variants affecting colorectal cancer(CRC)risk,to support ef...Objective:We aimed to directly compare the estimated effects of adherence to a healthy lifestyle with those of risk predisposition according to known genetic variants affecting colorectal cancer(CRC)risk,to support effective risk communication for cancer prevention.Methods:A healthy lifestyle score(HLS)was derived from 5 lifestyle factors:smoking,alcohol consumption,diet,physical activity,and body adiposity.The association of lifestyle and polygenic risk score(PRS)(based on 140 CRC-associated risk loci)with CRC risk was assessed with multiple logistic regression and compared through the genetic risk equivalent(GRE),a novel approach providing an estimate of the effects of adherence to a healthy lifestyle in terms of percentile differences in PRS.Results:A higher HLS was associated with lower CRC risk(4,844 cases,3,964 controls).Those adhering to all 5 healthy lifestyle factors had a 62%(95%CI 54%-68%)lower CRC risk than those adhering to≤2 healthy lifestyle factors.The estimated effect of adherence to all 5 compared with≤2 healthy lifestyle factors was as strong as the effect of having a 79 percentile(GRE 79,95%CI 61-97)lower PRS.The association between a healthy lifestyle and CRC risk was independent of PRS level but was particularly pronounced among those with a family history of CRC in≥1 first-degree relative(P-interaction=0.0013).Conclusions:A healthy lifestyle was strongly inversely associated with CRC risk.The large GRE indicated that CRC risk determined by polygenic risk may be offset to a substantial extent by adherence to a healthy lifestyle.展开更多
Water electrolysis is considered as one most promising technique for hydrogen production.The high efficiency electrocatalyst is the key to accelerating the sluggish kinetics of the hydrogen evolution reaction(HER) in ...Water electrolysis is considered as one most promising technique for hydrogen production.The high efficiency electrocatalyst is the key to accelerating the sluggish kinetics of the hydrogen evolution reaction(HER) in alkaline media.In this work,an efficient HER electrocatalyst with hetero-interfacial metal-metal oxide structure was constructed through a redox solid phase reaction(SPR) strategy.During the annealing process under Ar atmosphere,RuO_(2) and WS_(2)in RuO_(2)/WS_(2)precursor were converted to Ru nanoparticles(NPs) and WO3in situ,where tiny Ru NPs and oxygen vacancies were uniformly distributed onto the newly formed WO3nanosheets.Different characterization techniques were adopted to confirm the successful formation of Ru/WO_(3)electrocatalyst(RWOC).The optimized RWOC sample annealed at 400℃ exhibited the low overpotential value of 13 mV at a current density of 10 mA cm^(-2)and strong durability under the alkaline condition.Density functional theoretical calculations further revealed that the promoted adsorption/desorption rate of reaction intermediates and the accelerated kinetics of HER process were deduced to the synergistic effect between Ru and WO_(3)in electrocatalyst.This work provides a feasible method to fabricate highly efficient HER electrocatalysts.展开更多
The large yellow croaker(Larimichthys crocea),which is an economically important mariculture fish in China,is often exposed to environmental hypoxia.Reactive oxygen species(ROS)homeostasis is essential for the mainten...The large yellow croaker(Larimichthys crocea),which is an economically important mariculture fish in China,is often exposed to environmental hypoxia.Reactive oxygen species(ROS)homeostasis is essential for the maintenance of normal physiological conditions in an organism.Direct evidence that environmental hypoxia leads to ROS overproduction is scarce in marine fish.Furthermore,the sources of ROS overproduction in marine fish under hypoxic stress are poorly known.In this study,we investigated the effects of hypoxia on redox homeostasis in L.crocea and the impact of impaired redox homeostasis on fish.We first confirmed that hypoxia drove ROS production mainly via the mitochondrial electron transport chain and NADPH oxidase complex pathways in L.crocea and its cell line(large yellow croaker fry(LYCF)cells).We subsequently detected a marked increase in the antioxidant systems of the fish.However,imbalance between the pro-oxidation and antioxidation systems ultimately led to excessive ROS and oxidative stress.Cell viability showed a remarkable decrease while oxidative indicators,such as malondialdehyde,proteincarbonylation,and8-hydroxy-2 deoxyguanosine,showed a significant increase after hypoxia,accompanied by tissue damage.Nacetylcysteine(NAC)reduced ROS levels,alleviated oxidative damage,and improved cell viability in vitro.Appropriate uptake of ROS scavengers(e.g.,NAC and elamipretide Szeto-Schiller-31)and inhibitors(e.g.,apocynin,diphenylene iodonium,and 5-hydroxydecanoate)may be effective at overcoming hypoxic toxicity.Our findings highlight previously unstudied strategies of hypoxic toxicity resistance in marine fish.展开更多

The applications of laser-induced breakdown spectroscopy(LIBS) on classifying complex natural organics are relatively limited and their accuracy still requires improvement.In this work,to study the methods on classifi...The applications of laser-induced breakdown spectroscopy(LIBS) on classifying complex natural organics are relatively limited and their accuracy still requires improvement.In this work,to study the methods on classification of complex organics,three kinds of fresh leaves were measured by LIBS.100 spectra from 100 samples of each kind of leaves were measured and then they were divided into a training set and a test set in a ratio of 7:3.Two algorithms of chemometric methods including the partial least squares discriminant analysis(PLS-DA) and principal component analysis Mahalanobis distance(PCA-MD) were used to identify these leaves.By using 23 lines from 16 elements or molecules as input data,these two methods can both classify these three kinds of leaves successfully.The classification accuracies of training sets are both up to 100% by PCA-MD and PLS-DA.The classification accuracies of the test set are 93.3% by PCA-MD and 97.8% by PLS-DA.It means that PLS-DA is better than PCA-MD in classifying plant leaves.Because the components in PLS-DA process are more suitable for classification than those in PCA-MD process.We think that this work can provide a reference for plant traceability using LIBS.展开更多

T...In this paper, the outage perfor- mance of a cognitive relaying network over Nakagami-m fading channels, employing simultaneous wireless information and power transfer (SWIPT) technology is analyzed and evaluated. The operation of this network is considered in conjunction with the convention- al decode-and-forward (DF) and incremental DF (IDF) protocols. For the conventional DF protocol, it is assumed that there is no direct link between the secondary transmitter (S) and the secondary destination (D), while (for both protocols) after harvesting energy, the relay node (R) always helps to forward the resulting signal to D. However, for the IDF protocol, R assists in relaying S's information to D only when the direct communication between S and D has failed. Furthermore, for both DF and IDF protocols, we assume there is no power supply for R, and R harvests energy from the transmitted signal of S. We derive exact ana- lytical expressions for the outage probability at D in DF and IDF protocols, respectively, in terms of the bivariate Meijer's G-function. Performance evaluation results obtained by means of Monte-Carlo simulations are also provided and have validated the correctness of the oroDosed analysis.展开更多

[ Method ] Gene fragment BC encoding the hybrid antimicrobial peptide Buforin II-Cecropin B was synthesized by SOE-PCR with six primers designed according to the published amino acid sequences. Then, the BC gene fragment was ligated into pET32a vector and expressed in BI21. Antimierobial ability of the crude hybrid antimicrobial peptide was detected with the Oxford cup method. [ Result] The BC gene fragment was correctly amplified by SOE-PCR and ligated into pET32a vector, to construct the expression vector pET32-BC. The hybrid antimicrobial peptide buforin II-cecropin B was expressed in BI21 after induction by IPTG. The optimal induction time was 2 h. Antimicrobial activity assay suggested that the hybrid antimicrobial peptide possessed antimicrobial activity to Escherichia coli and Bacillus subtilis. [ Conclusion] The hybrid antimicrobial peptide buforin II-cecropin B exhibited antimicrobial activity to Gram-negative E. coli and Gram-positive B. subtilis strains.展开更多
Selective hydrogenation of hydroxyaldehydes to polyalcohols is challenging due to the competitive hydrogenation of C=O and CAO.This study develops heterogeneous Cu catalysts for the selective synthesis of ethylene gly...Selective hydrogenation of hydroxyaldehydes to polyalcohols is challenging due to the competitive hydrogenation of C=O and CAO.This study develops heterogeneous Cu catalysts for the selective synthesis of ethylene glycol via batch liquid-phase hydrogenation of glycolaldehyde.SiO_(2)supported Cu,fabricated by ammonia evaporation,enables to catalyze the C=O bond hydrogenation with retaining the CAO bond intact,yielding higher selective hydrogenation activity with ethylene glycol selectivity up to 99.8%relative to MgO,Al_(2)O_(3),CeO_(2),and TiO_(2)supports and Cu/SiO_(2)synthesized by deposition–precipitation and impregnation.Characterizations confirm that highly efficient 20Cu/SiO_(2)-AE-623 K catalyst fabricated by ammonia evaporation is featured with larger Cu^(0)and Cu^(+)surface areas,of which the Cu^(+)species created from reducing copper phyllosilicate exhibit higher reactivity.A synergistic effect between Cu^(+)and Cu^(0)facilitates the selective adsorption/activation of glycolaldehyde on Cu^(+)sites and the dissociation of H_(2)on Cu^(0)sites,bringing a remarkable improvement in the selective hydrogenation performance.展开更多

文摘Wastewater treatment plants(WWTPs)are important and energy-intensive municipal infrastructures.High energy consumption and relatively low operating performance are major challenges from the perspective of carbon neutrality.However,water-energy nexus analysis and models for WWTPs have rarely been reported to date.In this study,a cloud-model-based energy consumption analysis(CMECA)of a WWTP was conducted to explore the relationship between influent and energy consumption by clustering its influent’s parameters.The principal component analysis(PCA)and K-means clustering were applied to classify the influent condition using water quality and volume data.The energy consumption of the WWTP is divided into five standard evaluation levels,and its cloud digital characteristics(CDCs)were extracted according to bilateral constraints and golden ratio methods.Our results showed that the energy consumption distribution gradually dispersed and deviated from the Gaussian distribution with decreased water concentration and quantity.The days with high energy efficiency were extracted via the clustering method from the influent category of excessive energy consumption,represented by a compact-type energy consumption distribution curve to identify the influent conditions that affect the steady distribution of energy consumption.The local WWTP has high energy consumption with 0.3613 kW·h·m^(-3)despite low influent concentration and volumes,across four consumption levels from low(I)to relatively high(IV),showing an unsatisfactory operation and management level.The average oxygenation capacity,internal reflux ratio,and external reflux ratio during high energy efficiency days recognized by further clustering were obtained(0.2924-0.3703 kg O_(2)·m^(-3),1.9576-2.4787,and 0.6603-0.8361,respectively),which could be used as a guide for the days with low energy efficiency.Consequently,this study offers a water-energy nexus analysis method to identify influent conditions with operational management anomalies and can be used as an empirical reference for the optimized operation of WWTPs.

文摘The object of this study is to prepare iron species-impregnated granular activated carbon as particle electrodes in order to improve their adsorption and electrocatalytic degradation capacity in Benzothiazole removal.The incorporation of Fe-containing catalysts was performed by Fe(NO_3)_3 impregnation.The obtained samples were characterized by BET,Fourier transform infrared spectroscopy,SEM-EDS,powder X-ray diffraction,X-ray photoelectron spectra and TG.Compared with pure activated carbon,this modified particle electrodes show higher static adsorption capacities and TOC removal,which have respectively increased by25.9% and 54.4%.Both physisorption and chemisorption exist in the process of benzothiazole adsorption,where the latter plays a major role.In this way,the Fe-containing catalysts on modified particle electrodes are demonstrated to make a greater contribution to the improvement of electrocatalytic degradation by decreasing the activated energy by 32%.

文摘Electrochemical CO2 reduction to chemicals or fuels presents one of the most promising strategies for managing the global carbon balance, which yet poses a significant challenge due to lack of efficient and durable electrocatalyst as well as the understanding of the CO2 reduction reaction(CO2RR) mechanism.Benefiting from the large surface area, high electrical conductivity, and tunable structure, carbon-based metal-free materials(CMs) have been extensively studied as cost-effective electrocatalysts for CO2RR.The development of CMs with low cost, high activity and durability for CO2RR has been considered as one of the most active and competitive directions in electrochemistry and material science.In this review article,some up-to-date strategies in improving the CO2RR performance on CMs are summarized.Specifically, the approaches to optimize the adsorption of CO2RR intermediates, such as tuning the physical and electronic structure are introduced, which can enhance the electrocatalytic activity of CMs effectively.Finally, some design strategies are proposed to prepare CMs with high activity and selectivity for CO2RR.

文摘Objective To explore the role and regulation of guanine nucleotide-binding protein G(i), a-1 subunit (GNAI1) in hepatocellular carcinoma (HCC). Methods Expression of GNAI1 in HCC samples was determined by qRT-PCR and immunohistochemical (IHC) staining. Huh-7 and SNU-387 cells stably expressing GNAI1 were established by the infection of lentivirus transducing unit containing GNAI1. siRNA against GNAI1 was transfected into SMMC-7721 cells to knock down the GNAI1 expression in HCC cells. Mir-320a/c/d mimics were transfected into SMMC-7721 and SK-Hep-1 cells and the expression of GNAll was determined by Western blot. The migration and invasion of Huh-7, SNU-387, SK-Hep-1 and SMMC-7721 cells were investigated by Transwell assays. Results The GNAI1 protein was significantly downregulated in HCC samples without changes in its mRNA levels. GNAI1 could inhibit the migration and invasion of HCC cells in vitro. Further investigations indicated that GNAI1 was a target of miR-320a/c/d in HCC cells. Transwell assays demonstrated that these microRNAs could promote the migratory ability and invasivesess of HCC cells in vitro. Conclusions GNAII is downregulated in HCC and inhibits the migration and invasion of HCC cells. This study is the first to investigate the role of GNAI1 in cancer. Regulation of GNAI1 by miR-320a/c/d indicates new therapeutic avenues for targeting HCC metastasis.

文摘The low efficiency of oxygen evolution reaction(OER) is regarded as one of the major roadblocks for metal-air batteries and water electrolysis.Herein,a high-performance OER catalyst of NiFe_(0.2)(oxy)hydroxide(NiFe_(0.2)-O_(x)H_(y)) was developed through topotactic transformation of a Prussian blue analogue in an alkaline solution,which exhibits a low overpotential of only 263 mV to reach a current density of 10 mA cm^(-2) and a small Tafel slope of 35 mV dec-1.Ex-situ/operando Raman spectroscopy results indicated that the phase structure of NiFe_(0.2)-O_(x)H_(y) was irreversibly transformed from the type of α-Ni(OH)_(2) to γ-NiOOH with applying an anodic potential,while ex-situ/operando 57Fe Mossbauer spectroscopic studies evidenced the in-situ production of abundant high-valent iron species under OER conditions,which effectively promoted the OER catalysis.Our work elucidates that the amount of high-valent iron species in-situ produced in the NiFe(oxy)hydroxide has a positive correlation with its water oxidation reaction performance,which further deepens the understanding of the mechanism of NiFe-based electrocatalysts.

文摘Oxygen is an essential molecule for animal respiration,growth,and survival.Unlike in terrestrial environments,contamination and climate change have led to the frequent occurrence of hypoxia in aquatic environments,thus impacting aquatic animal survival.However,the adaptative mechanisms underlying fish responses to environmental hypoxia remain largely unknown.Here,we used large yellow croaker(Larimichthys crocea)and large yellow croaker fry(LYCF)cells to investigate the roles of the Hif-1α/Hsf1/Hsp70 signaling pathway in the regulation of cellular redox homeostasis,and apoptosis.We confirmed that hypoxia induced the expression of Hif-1α,Hsf1,and Hsp70 in vivo and in vitro.Genetic Hsp70 knockdown/overexpression indicated that Hsp70 was required for maintaining redox homeostasis and resisting oxidative stress in LYCF cells under hypoxic stress.Hsp70 inhibited caspase-dependent intrinsic apoptosis by maintaining normal mitochondrial membrane potential,enhancing Bcl-2 mRNA and protein expression,inhibiting Bax and caspase3 mRNA expression,and suppressing caspase-3 and caspase-9 activation.Hsp70 suppressed caspaseindependent intrinsic apoptosis by inhibiting nuclear translocation of apoptosis-inducing factor(AIF)and disturbed extrinsic apoptosis by inactivating caspase-8.Genetic knockdown/overexpression of Hif-1αand dual-luciferase reporter assay indicated that Hif-1αactivated the Hsf1 DNA promoter and enhanced Hsf1 mRNA transcription.Hsf1 enhanced Hsp70 mRNA transcription in a similar manner.In summary,the Hif-1α/Hsf1/Hsp70 signaling pathway plays an important role in regulating redox homeostasis and anti-apoptosis in L.crocea under hypoxic stress.

文摘Objective:We aimed to directly compare the estimated effects of adherence to a healthy lifestyle with those of risk predisposition according to known genetic variants affecting colorectal cancer(CRC)risk,to support effective risk communication for cancer prevention.Methods:A healthy lifestyle score(HLS)was derived from 5 lifestyle factors:smoking,alcohol consumption,diet,physical activity,and body adiposity.The association of lifestyle and polygenic risk score(PRS)(based on 140 CRC-associated risk loci)with CRC risk was assessed with multiple logistic regression and compared through the genetic risk equivalent(GRE),a novel approach providing an estimate of the effects of adherence to a healthy lifestyle in terms of percentile differences in PRS.Results:A higher HLS was associated with lower CRC risk(4,844 cases,3,964 controls).Those adhering to all 5 healthy lifestyle factors had a 62%(95%CI 54%-68%)lower CRC risk than those adhering to≤2 healthy lifestyle factors.The estimated effect of adherence to all 5 compared with≤2 healthy lifestyle factors was as strong as the effect of having a 79 percentile(GRE 79,95%CI 61-97)lower PRS.The association between a healthy lifestyle and CRC risk was independent of PRS level but was particularly pronounced among those with a family history of CRC in≥1 first-degree relative(P-interaction=0.0013).Conclusions:A healthy lifestyle was strongly inversely associated with CRC risk.The large GRE indicated that CRC risk determined by polygenic risk may be offset to a substantial extent by adherence to a healthy lifestyle.

文摘Water electrolysis is considered as one most promising technique for hydrogen production.The high efficiency electrocatalyst is the key to accelerating the sluggish kinetics of the hydrogen evolution reaction(HER) in alkaline media.In this work,an efficient HER electrocatalyst with hetero-interfacial metal-metal oxide structure was constructed through a redox solid phase reaction(SPR) strategy.During the annealing process under Ar atmosphere,RuO_(2) and WS_(2)in RuO_(2)/WS_(2)precursor were converted to Ru nanoparticles(NPs) and WO3in situ,where tiny Ru NPs and oxygen vacancies were uniformly distributed onto the newly formed WO3nanosheets.Different characterization techniques were adopted to confirm the successful formation of Ru/WO_(3)electrocatalyst(RWOC).The optimized RWOC sample annealed at 400℃ exhibited the low overpotential value of 13 mV at a current density of 10 mA cm^(-2)and strong durability under the alkaline condition.Density functional theoretical calculations further revealed that the promoted adsorption/desorption rate of reaction intermediates and the accelerated kinetics of HER process were deduced to the synergistic effect between Ru and WO_(3)in electrocatalyst.This work provides a feasible method to fabricate highly efficient HER electrocatalysts.

文摘The large yellow croaker(Larimichthys crocea),which is an economically important mariculture fish in China,is often exposed to environmental hypoxia.Reactive oxygen species(ROS)homeostasis is essential for the maintenance of normal physiological conditions in an organism.Direct evidence that environmental hypoxia leads to ROS overproduction is scarce in marine fish.Furthermore,the sources of ROS overproduction in marine fish under hypoxic stress are poorly known.In this study,we investigated the effects of hypoxia on redox homeostasis in L.crocea and the impact of impaired redox homeostasis on fish.We first confirmed that hypoxia drove ROS production mainly via the mitochondrial electron transport chain and NADPH oxidase complex pathways in L.crocea and its cell line(large yellow croaker fry(LYCF)cells).We subsequently detected a marked increase in the antioxidant systems of the fish.However,imbalance between the pro-oxidation and antioxidation systems ultimately led to excessive ROS and oxidative stress.Cell viability showed a remarkable decrease while oxidative indicators,such as malondialdehyde,proteincarbonylation,and8-hydroxy-2 deoxyguanosine,showed a significant increase after hypoxia,accompanied by tissue damage.Nacetylcysteine(NAC)reduced ROS levels,alleviated oxidative damage,and improved cell viability in vitro.Appropriate uptake of ROS scavengers(e.g.,NAC and elamipretide Szeto-Schiller-31)and inhibitors(e.g.,apocynin,diphenylene iodonium,and 5-hydroxydecanoate)may be effective at overcoming hypoxic toxicity.Our findings highlight previously unstudied strategies of hypoxic toxicity resistance in marine fish.

文摘The effect of AlGaN interlayer in quantum barrier on the electroluminescence characteristics of GaN-based green light emitting diodes(LEDs)grown on silicon substrate was investigated.The results show that AlGaN interlayer is beneficial to improve the luminous efficiency of LED devices and restrain the phase separation of In GaN.The former is ascribed to the inserted AlGaN layers can play a key role in determining the carrier distribution and screening dislocations in the active region,and the latter is attributed to the increased compressive stress in the quantum well.However,when the electrical stress aging tests were performed at a current density of 100 A/cm^2,LED devices with AlGaN interlayers are more likely to induce the generation/proliferation of defects in the active region under the effect of electrical stress,resulting in the reduced light output power at low current density.

文摘The applications of laser-induced breakdown spectroscopy(LIBS) on classifying complex natural organics are relatively limited and their accuracy still requires improvement.In this work,to study the methods on classification of complex organics,three kinds of fresh leaves were measured by LIBS.100 spectra from 100 samples of each kind of leaves were measured and then they were divided into a training set and a test set in a ratio of 7:3.Two algorithms of chemometric methods including the partial least squares discriminant analysis(PLS-DA) and principal component analysis Mahalanobis distance(PCA-MD) were used to identify these leaves.By using 23 lines from 16 elements or molecules as input data,these two methods can both classify these three kinds of leaves successfully.The classification accuracies of training sets are both up to 100% by PCA-MD and PLS-DA.The classification accuracies of the test set are 93.3% by PCA-MD and 97.8% by PLS-DA.It means that PLS-DA is better than PCA-MD in classifying plant leaves.Because the components in PLS-DA process are more suitable for classification than those in PCA-MD process.We think that this work can provide a reference for plant traceability using LIBS.

文摘In this paper, the outage perfor- mance of a cognitive relaying network over Nakagami-m fading channels, employing simultaneous wireless information and power transfer (SWIPT) technology is analyzed and evaluated. The operation of this network is considered in conjunction with the convention- al decode-and-forward (DF) and incremental DF (IDF) protocols. For the conventional DF protocol, it is assumed that there is no direct link between the secondary transmitter (S) and the secondary destination (D), while (for both protocols) after harvesting energy, the relay node (R) always helps to forward the resulting signal to D. However, for the IDF protocol, R assists in relaying S's information to D only when the direct communication between S and D has failed. Furthermore, for both DF and IDF protocols, we assume there is no power supply for R, and R harvests energy from the transmitted signal of S. We derive exact ana- lytical expressions for the outage probability at D in DF and IDF protocols, respectively, in terms of the bivariate Meijer's G-function. Performance evaluation results obtained by means of Monte-Carlo simulations are also provided and have validated the correctness of the oroDosed analysis.

文摘[ Objective ] This study aimed to investigate the antimicrobial activity of hybrid antimicrobial peptide buforin II-cecropin B. [ Method ] Gene fragment BC encoding the hybrid antimicrobial peptide Buforin II-Cecropin B was synthesized by SOE-PCR with six primers designed according to the published amino acid sequences. Then, the BC gene fragment was ligated into pET32a vector and expressed in BI21. Antimierobial ability of the crude hybrid antimicrobial peptide was detected with the Oxford cup method. [ Result] The BC gene fragment was correctly amplified by SOE-PCR and ligated into pET32a vector, to construct the expression vector pET32-BC. The hybrid antimicrobial peptide buforin II-cecropin B was expressed in BI21 after induction by IPTG. The optimal induction time was 2 h. Antimicrobial activity assay suggested that the hybrid antimicrobial peptide possessed antimicrobial activity to Escherichia coli and Bacillus subtilis. [ Conclusion] The hybrid antimicrobial peptide buforin II-cecropin B exhibited antimicrobial activity to Gram-negative E. coli and Gram-positive B. subtilis strains.

文摘Selective hydrogenation of hydroxyaldehydes to polyalcohols is challenging due to the competitive hydrogenation of C=O and CAO.This study develops heterogeneous Cu catalysts for the selective synthesis of ethylene glycol via batch liquid-phase hydrogenation of glycolaldehyde.SiO_(2)supported Cu,fabricated by ammonia evaporation,enables to catalyze the C=O bond hydrogenation with retaining the CAO bond intact,yielding higher selective hydrogenation activity with ethylene glycol selectivity up to 99.8%relative to MgO,Al_(2)O_(3),CeO_(2),and TiO_(2)supports and Cu/SiO_(2)synthesized by deposition–precipitation and impregnation.Characterizations confirm that highly efficient 20Cu/SiO_(2)-AE-623 K catalyst fabricated by ammonia evaporation is featured with larger Cu^(0)and Cu^(+)surface areas,of which the Cu^(+)species created from reducing copper phyllosilicate exhibit higher reactivity.A synergistic effect between Cu^(+)and Cu^(0)facilitates the selective adsorption/activation of glycolaldehyde on Cu^(+)sites and the dissociation of H_(2)on Cu^(0)sites,bringing a remarkable improvement in the selective hydrogenation performance.
