BACKGROUND According to the population statistics in 2023,there were 110000 people aged over 100 years in China,and the experience of using Paxlovid(nirmatrelvir/ritonavir)for centenarians is particularly valuable.Thi...BACKGROUND According to the population statistics in 2023,there were 110000 people aged over 100 years in China,and the experience of using Paxlovid(nirmatrelvir/ritonavir)for centenarians is particularly valuable.This article reports our experience of using Paxlovid in a centenarian with the novel coronavirus disease 2019(COVID-19)infection.CASE SUMMARY A 103-year-old female with mild COVID-19 and renal insufficiency was given sufficient Paxlovid for 2 days and a half dose for 3 days.During treatment,the patient was complicated with lung infection and heart failure,and nucleic acid remained positive.After expert consultation,a full dose of Paxlovid was given again on the 9th day of admission for 2 days and a half dose for 3 days.Meanwhile,anti-heart failure and antibiotics were administered;the heart failure and pulmonary infection were improved.Finally,on the 33^(th) day of admission,nucleic acid turned negative,body temperature returned to normal,cough and sputum,fatigue,poor appetite and other symptoms basically improved.The patient was given Paxlovid via nasal feeding for 2 courses without deterioration of liver and kidney function,diarrhea,nausea and vomiting,myalgia,chest tightness and other side effects,and was discharged from hospital with good recovery.CONCLUSION This case suggests that Paxlovid can be used cautiously in centenarians with renal insufficiency and two courses of treatment can be considered in patients with persistent positive nucleic acid.展开更多
[Objectives] The study aims to improve the ability of inspectors to recognize humoral morphology and provide data reference for the improvement of EQA work of humoral morphology by Guangxi Clinical Laboratory Center. ...[Objectives] The study aims to improve the ability of inspectors to recognize humoral morphology and provide data reference for the improvement of EQA work of humoral morphology by Guangxi Clinical Laboratory Center. [Methods] A retrospective analysis was made on the EQA of humoral morphology by Guangxi Clinical Laboratory Center from 2010 to 2022, and the number of participating laboratories, the rate of return, sample types and maps of EQA of humoral morphology such as urine sediment, vaginal secretions and so on(except feces), and the reasons for the high error rate of the results of the participating laboratories were analyzed. [Results] The number of laboratories participating in the assessment of humoral morphology increased from 98 in 2010 to 371 in 2022. Except for 2011(90.98%), the rate of return was more than 94%, and the highest rate of return was up to 100%. A total of 119 pictures of EQA of humoral morphology were studied, including 101 urine sediment smears, 13 vaginal secretions smears and 5 other smears. The types of specimens were relatively comprehensive. The distribution of the maps was basically reasonable. The urinary sediment was mainly composed of crystalline salts, followed by tubes, and cells and fungi ranked third. The coverage was relatively complete. The error rate of return results from high to low were bacteria(error rate was equal to 40%), tubes and crystalline salts(error rate was close to 29%), and cells and fungi(error rate was about 14%). The main cases with high error rate of return results are as follows: the difference between different tubes was not clear, and the diversity of crystal morphology was not understood enough;the phase division of cells in the same system was wrong. [Conclusions] The analysis and summary of the reasons for the high error rate of the data and results of Guangxi's EQA of humoral morphology is helpful for laboratory physicians to improve their awareness of morphological examination and identify clearly the error type and blurred concept, and lays a foundation for Guangxi Clinical Laboratory Center to carry out EQA of humoral morphology more specifically.展开更多
Heterotopic pancreas is defined as pancreatic tissue found outside the usual anatomical location of the pancreas. It is often an incidental finding and can be found at different sites in the gastrointestinal tract. It...Heterotopic pancreas is defined as pancreatic tissue found outside the usual anatomical location of the pancreas. It is often an incidental finding and can be found at different sites in the gastrointestinal tract. It may become clinically evident when complicated by pathological changes such as inflammation, bleeding, obstruction, and malignant transformation. In this report, a 60-year-old man with carcinoid syndrome caused by heterotopic pancreatic tissue in the duodenum is described, along with a 62-year-old man with abdominal pain caused by heterotopic pancreatic tissue in the gastric antrum. The difficulty of making an accurate diagnosis is highlighted. The patients remain healthy and symptom-free after follow-up of 1 year. Frozen sections may help in deciding the extent of resection intraoperatively. Although heterotopic pancreas is rare, it should be considered in the differential diagnosis of gastrointestinal stromal tumor.展开更多
Neuroendocrine carcinoma(NEC) of the pancreas is rare.We report the case of a 34-year-old man with pancreatic NEC with soft tissue metastasis.The patient presented with right upper abdominal discomfort.Computed tomogr...Neuroendocrine carcinoma(NEC) of the pancreas is rare.We report the case of a 34-year-old man with pancreatic NEC with soft tissue metastasis.The patient presented with right upper abdominal discomfort.Computed tomography revealed a low-density heterogeneous mass in the tail and body of the pancreas that encroached on the greater curvature of the stomach and spleen.We performed exploratory laparotomy and total pancreatectomy with splenectomy and total gastrectomy.Histopathological analysis showed spindleshaped cells with scanty cytoplasm and hyperchromatic nuclei,confirming a primary pancreatic NEC.One month after the surgery,the patient experienced leg swelling.Positron emission tomography-computed tomography revealed high uptake of fludeoxyglucose in the left leg,and the leg was amputated.Histopathological analysis confirmed metastasis of pancreatic NEC.The patient was followed up and received chemotherapy(etoposide and cisplatin).One month after amputation,the level of tumor marker neuron-specific enolase was 142.70 μg/L and computed tomography scan revealed an aggravated metastatic lesion.The patient suffered from unbearable pain and we treated him with odynolysis.Four months postoperatively,the patient died of respiratory failure.展开更多

However, in the PLLA/PDLA blends with higher molecular weights, the phase separation occurs and the SC exhibits weak memory after melting, which lead to a small amount of SC together with a large amount of homochiral crystallites (HC) develop during crystallization from the melt. In this study, a small content of graphite oxide was blended with PLLA and PDLA to form ternary blends, and it was exciting to find that the formation of SC was enhanced gradually with the content of graphite oxide. The SC exclusively developed when 2 wt% graphite oxide was incorporated into the PLLA/PDLA, and the crystallinity with ∼50% was received even during fast cooling from the melt (−50 ℃/min). The acceleration formation of SC was speculated due to the interaction between PLA molecular chains and the hydroxyl groups on the surface of graphite oxide and the obstruction of proliferation of graphite oxide.展开更多

However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of "Darmor-bzh"and "Tapidor,"respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. Our study provides important insights into the genomic footprints of rapeseed evolution and flowering-time divergence among three ecotype groups,and will facilitate screening of molecular markers for accelerating rapeseed breeding.展开更多

The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigat...The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.展开更多

Ferulic acid inclusion complexes(FA-HP-β-CD)effects on tyramine accumulation and gene expression of Enterobacter hormaechei MW386398 in traditional smoked horsemeat sausages were studied.Reverse transcription-quantit...Ferulic acid inclusion complexes(FA-HP-β-CD)effects on tyramine accumulation and gene expression of Enterobacter hormaechei MW386398 in traditional smoked horsemeat sausages were studied.Reverse transcription-quantitative polymerase chain reaction(RT-qPCR)was used to identify the expression of tyrosine decarboxylase(TDC)clusters in the tyramine producing pathway.In addition,relevant parameters such as microbial growth,moisture content,pH,and tyramine accumulation in pure cultures and during fermentation of smoked horsemeat sausages were evaluated.Ferulic acid(FA)and FA-HP-β-CD inclusion complexes both significantly decreased tyramine accumulation,as well as significantly inhibited tyrosine decarboxylase(tdcA)and tyrosine/tyramine antiporter(tyrP)genes expression(p<0.05).Furthermore,FA and FA-HP-β-CD inclusion complexes both reduced the number of Enterobacteriaceae in smoked horsemeat sausages,but did not alter the main microbiota.However,FA-HP-β-CD inclusion complexes exhibited stronger ability in hindering TDC related gene expression,reducing tyramine accumulation and antibacterial properties.These results indicate that FA-HP-β-CD inclusion complexes are able to reduce tyramine accumulation in smoked horsemeat sausages by inhibiting Enterobacteriaceae growth and gene expression.展开更多

Herein we reported 11 sulfur-containing coumarins as cell imaging probes. Different sulfur speciation of the 4 representative coumarins SC1-SC4 renders them significantly different subcellular localizations and cellular uptake pathways: SC1 containing thioether group located in lysosomes, while sulfoxide and sulfone compounds SC2 and SC3 distributed in the whole cell. Furthermore, the cationic sulfonium containing compound SC4 was internalized by clathrin-mediated endocytosis and localized at mitochondria. By analyzing the molecular parameters of all 11 coumarins, we found that different sulfur speciation affected their lipophilicity and electrostatic surface potential. These two key factors play roles in altering biological behaviors of the coumarins. The results revealed the importance of sulfur speciation on the physicochemical properties and thus subcellular localization of bioprobes. This is useful for designing new functional bioprobes.展开更多

More importantly, encapsulated by PLGA nanoparticles, cryptand tri Znsalen 1 displayed visible intracellular fluorescence whereas monomeric Znsalen 5 could not. 文摘Rapeseed (Brassica napus),an important oilseed crop,has adapted to diverse climate zones and latitudes by forming three main ecotype groups,namely winter,semiwinter,and spring types. However,genetic variations underlying the divergence of these ecotypes are largely unknown. Here,we report the global pattern of genetic polymorphisms in rapeseed determined by resequencing a worldwide collection of 991 germplasm accessions.A total of 5.56 and 5.53 million singlenucleotide polymorphisms (SNPs)as Well as 1.86 and 1.92 million InDels were identified by mapping reads to the reference genomes of "Darmor-bzh"and "Tapidor,"respectively.We generated a map of allelic drift paths that shows splits and mixtures of the main populations,and revealed an asymmetric evolution of the two subgenomes of B.napus by calculating the genetic diversity and linkage disequilibrium parameters.Selective-sweep analysis revealed genetic changes in genes orthologous to those regulating various aspects of plant development and response to stresses.A genome-wide association study identified SNPs in the promoter regions of FLOWERING LOCUS T and FLOWERING LOCUS C orthologs that corresponded to the different rapeseed ecotype groups. Our study provides important insights into the genomic footprints of rapeseed evolution and flowering-time divergence among three ecotype groups,and will facilitate screening of molecular markers for accelerating rapeseed breeding.

文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.
文摘Copper sulfide (Cu7S4) nanoparticles coated with an ultra-high payload (N5.0 ~ 107 fluorine atoms per particle) of fluorinated ligands (oleylamine functionalized 3,5-bis(trifluoromethyl)benzaldehyde, 19FOAm) exhibited a single intense 19F magnetic resonance (MR) signal and efficient near infrared photothermal performance in water medium. In vivo assessment revealed strong 19F MR signals at cancerous lesions and effective inhibition of tumor growth after photothermal treatment, indicating the great potential of these fabricated nanoprobes for simultaneous 19F MR imaging and photothermal therapy.

文摘Ferulic acid inclusion complexes(FA-HP-β-CD)effects on tyramine accumulation and gene expression of Enterobacter hormaechei MW386398 in traditional smoked horsemeat sausages were studied.Reverse transcription-quantitative polymerase chain reaction(RT-qPCR)was used to identify the expression of tyrosine decarboxylase(TDC)clusters in the tyramine producing pathway.In addition,relevant parameters such as microbial growth,moisture content,pH,and tyramine accumulation in pure cultures and during fermentation of smoked horsemeat sausages were evaluated.Ferulic acid(FA)and FA-HP-β-CD inclusion complexes both significantly decreased tyramine accumulation,as well as significantly inhibited tyrosine decarboxylase(tdcA)and tyrosine/tyramine antiporter(tyrP)genes expression(p<0.05).Furthermore,FA and FA-HP-β-CD inclusion complexes both reduced the number of Enterobacteriaceae in smoked horsemeat sausages,but did not alter the main microbiota.However,FA-HP-β-CD inclusion complexes exhibited stronger ability in hindering TDC related gene expression,reducing tyramine accumulation and antibacterial properties.These results indicate that FA-HP-β-CD inclusion complexes are able to reduce tyramine accumulation in smoked horsemeat sausages by inhibiting Enterobacteriaceae growth and gene expression.

文摘Targeted-delivery is of great importance to molecular probes and drugs for cell biology study. Herein we reported 11 sulfur-containing coumarins as cell imaging probes. Different sulfur speciation of the 4 representative coumarins SC1-SC4 renders them significantly different subcellular localizations and cellular uptake pathways: SC1 containing thioether group located in lysosomes, while sulfoxide and sulfone compounds SC2 and SC3 distributed in the whole cell. Furthermore, the cationic sulfonium containing compound SC4 was internalized by clathrin-mediated endocytosis and localized at mitochondria. By analyzing the molecular parameters of all 11 coumarins, we found that different sulfur speciation affected their lipophilicity and electrostatic surface potential. These two key factors play roles in altering biological behaviors of the coumarins. The results revealed the importance of sulfur speciation on the physicochemical properties and thus subcellular localization of bioprobes. This is useful for designing new functional bioprobes.

文摘Exploring the factors to control Znsalen aggregation is of importance to design functional materials in catalysis, optical materials and biological imaging. In this work, we synthesized and characterized four cryptand type tri Znsalen complexes and found that cryptand structure could efficiently minimize intermolecular Zn…O interaction. More importantly, encapsulated by PLGA nanoparticles, cryptand tri Znsalen 1 displayed visible intracellular fluorescence whereas monomeric Znsalen 5 could not. These results provide a new access to design new luminescent materials with the potential application in optics and biological studies.
