Salsolinol as an RNA m~6A methylation inducer mediates dopaminergic neuronal death by regulating YAP1 and autophagy

Salsolinol(1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline,Sal)is a catechol isoquinoline that causes neurotoxicity and shares structural similarity with 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine,an environmental toxin that causes Parkinson's disease.However,the mechanism by which Sal mediates dopaminergic neuronal death remains unclear.In this study,we found that Sal significantly enhanced the global level of N~6-methyladenosine(m~6A)RNA methylation in PC12 cells,mainly by inducing the downregulation of the expression of m~6A demethylases fat mass and obesity-associated protein(FTO)and alk B homolog 5(ALKBH5).RNA sequencing analysis showed that Sal downregulated the Hippo signaling pathway.The m~6A reader YTH domain-containing family protein 2(YTHDF2)promoted the degradation of m~6A-containing Yes-associated protein 1(YAP1)mRNA,which is a downstream key effector in the Hippo signaling pathway.Additionally,downregulation of YAP1 promoted autophagy,indicating that the mutual regulation between YAP1 and autophagy can lead to neurotoxicity.These findings reveal the role of Sal on m~6A RNA methylation and suggest that Sal may act as an RNA methylation inducer mediating dopaminergic neuronal death through YAP1 and autophagy.Our results provide greater insights into the neurotoxic effects of catechol isoquinolines compared with other studies and may be a reference for assessing the involvement of RNA methylation in the pathogenesis of Parkinson's disease. 展开更多

GULP1是一种含磷酸化酪氨酸结合(phosphotyrosine-binding,PTB)结构域的吞噬衔接蛋白,已有的研究表明它可促进脂肪细胞3T3-L1的糖摄取。为进一步挖掘GULP1下游关键的代谢相关差异基因,本研究对过表达GULP1的脂肪细胞和骨骼肌细胞进行转录组分析,然后对表达异常基因进行生物信息学分析,并通过实时荧光定量PCR(real-time fluorescent quantitative PCR,qRT-PCR)与转录组测序进行相互验证。结果表明:以P<0.05和|Log_(2)Foldchange|≥1为阈值筛选差异表达基因,发现与对照细胞相比,过表达GULP1的脂肪细胞中有278个上调基因和263个下调基因,与代谢相关的GO(Gene Ontology)条目包括胆固醇生物合成过程、胆固醇代谢过程、对脂多糖的反应、脂质代谢过程等,有52个代谢相关差异表达基因富集到10条KEGG(Kyoto Encyclopedia of Genes and Genomes)通路,其中脂质代谢被高度富集;过表达GULP1的骨骼肌细胞有280个上调基因和302个下调基因,与代谢相关的GO条目包括激素代谢过程、对脂多糖的反应、单碳代谢过程等,有86个代谢相关差异表达基因富集到10条KEGG通路,其中氨基酸代谢、脂质代谢、碳水化合物代谢被高度富集。GULP1的生物学功能涉及广泛,包括脂代谢、肿瘤等方面。本研究通过转录组学以及生物信息学分析,筛选出GULP1下游关键的代谢相关差异基因,获得了过表达GULP1后的代谢相关差异基因及信号通路,为今后GULP1下游靶基因的研究提供了重要的理论依据。 展开更多

Impact of comorbid subthreshold depressive symptoms on cancerrelated fatigue and complications in adults with leukemia 被引量:1

BACKGROUND Patients not only experience symptoms caused by cancer but also suffer from the accompanying psychological pain.Therefore,these patients do not have high quality of life.According to the World Health Organization,the incidence of leukemia in China in 2020 was 5.1/100000,the mortality rate was 3.3/100000,and the prevalence rate was 16.7/100000.Therefore,it is important to examine the influence of comorbid subthreshold depressive symptoms on leukemia patients.AIM To determine the impact of comorbid subthreshold depressive symptoms on cancer-related fatigue and complications in leukemia patients,thereby providing a basis for early diagnosis and treatment in clinical practice.METHODS A questionnaire survey was conducted among leukemia patients admitted to a tertiary hospital in Xi'an,Shaanxi Province,China,from August 2022 to December 2023.Patients with a score>16 on the Chinese Classification of Mental Disorders(CCMD-3)and a Hamilton Depression Rating Scale score of 8-17 were classified as the subthreshold depressive group(n=95),while 100 leukemia patients admitted during the same period were classified as the control group.Data were collected using Epidata 3.1 software,and comparisons were made between the two groups regarding general clinical data,the Piper Fatigue Scale(PFS),the Pittsburgh Sleep Quality Index(PSQI),the Numeric Rating Scale for pain assessment,laboratory indicators,and the occurrence of complications.RESULTS In this survey,120 leukemia patients with depression were preliminarily screened,95 patients with subthreshold depression were ultimately selected as the subthreshold depression group,and 100 leukemia patients admitted during the same period were enrolled as the normal group.Comparison of basic clinical data between the two groups revealed no significant differences in age,sex,body mass index,cognitive function,or comorbidity with other chronic diseases.However,there were statistically significant differences in the use of radiotherapy and regular exercise between the two groups(P<0.05).Comparisons of scales and laboratory indicators revealed no significant differences in albumin or PSQI scores between the two groups,but there were statistically significant differences in pain scores,PSQI scores,PFS scores,hemoglobin levels,and C-reactive protein levels(P<0.05).Spearman’s correlation analysis indicated that cancer-related fatigue was correlated with age,hemoglobin levels,C-reactive protein levels,pain,and regular exercise among leukemia patients with subthreshold depression.Multivariate regression analysis revealed that advanced age,combined radiotherapy,pain,and low hemoglobin levels were risk factors for cancer-related fatigue in leukemia patients with comorbid subthreshold depression,while regular exercise was a protective factor against cancer-related fatigue.Follow-up comparisons revealed a significantly lower overall incidence of complications in the control group(4%)than in the depressive group(24.21%;P<0.001).CONCLUSION Leukemia patients with comorbid subthreshold depressive symptoms experience more severe cancer-related fatigue and a higher incidence of complications.These findings may be related to advanced age,combined radiotherapy,pain,and low hemoglobin levels,while regular exercise may effectively alleviate symptoms. 展开更多

In order to improve the performance degradation prediction accuracy of proton exchange membrane fuel cell(PEMFC),a fusion prediction method(CKDG)based on adaptive noise complete ensemble empirical mode decomposition(CEEMDAN),kernel principal component analysis(KPCA)and dual attention mechanism gated recurrent unit neural network(DA-GRU)was proposed.CEEMDAN and KPCA were used to extract the input feature data sequence,reduce the influence of random factors,and capture essential feature components to reduce the model complexity.The DA-GRU network helps to learn the feature mapping relationship of data in long time series and predict the changing trend of performance degradation data more accurately.The actual aging experimental data verify the performance of the CKDG method.The results show that under the steady-state condition of 20%training data prediction,the CKDA method can reduce the root mean square error(RMSE)by 52.7%and 34.6%,respectively,compared with the traditional LSTM and GRU neural networks.Compared with the simple DA-GRU network,RMSE is reduced by 15%,and the degree of over-fitting is reduced,which has higher accuracy.It also shows excellent prediction performance under the dynamic condition data set and has good universality. 展开更多

Lithium sulfur(Li-S)battery is a kind of burgeoning energy storage system with high energy density.However,the electrolyte-soluble intermediate lithium polysulfides(Li PSs)undergo notorious shuttle effect,which seriously hinders the commercialization of Li-S batteries.Herein,a unique VSe_(2)/V_(2)C heterostructure with local built-in electric field was rationally engineered from V_(2)C parent via a facile thermal selenization process.It exquisitely synergizes the strong affinity of V_(2)C with the effective electrocatalytic activity of VSe_(2).More importantly,the local built-in electric field at the heterointerface can sufficiently promote the electron/ion transport ability and eventually boost the conversion kinetics of sulfur species.The Li-S battery equipped with VSe_(2)/V_(2)C-CNTs-PP separator achieved an outstanding initial specific capacity of 1439.1 m A h g^(-1)with a high capacity retention of 73%after 100 cycles at0.1 C.More impressively,a wonderful capacity of 571.6 mA h g^(-1)was effectively maintained after 600cycles at 2 C with a capacity decay rate of 0.07%.Even under a sulfur loading of 4.8 mg cm^(-2),areal capacity still can be up to 5.6 m A h cm^(-2).In-situ Raman tests explicitly illustrate the effectiveness of VSe_(2)/V_(2)C-CNTs modifier in restricting Li PSs shuttle.Combined with density functional theory calculations,the underlying mechanism of VSe_(2)/V_(2)C heterostructure for remedying Li PSs shuttling and conversion kinetics was deciphered.The strategy of constructing VSe_(2)/V_(2)C heterocatalyst in this work proposes a universal protocol to design metal selenide-based separator modifier for Li-S battery.Besides,it opens an efficient avenue for the separator engineering of Li-S batteries. 展开更多

Dual-atom catalysts(DACs) afford promising potential for oxygen reduction electrocatalysis due to their high atomic efficiency and high intrinsic activity.However,precise construction of dual-atom sites remains a challenge.In this work,a post-modification strategy is proposed to precisely fabricate DACs for oxygen reduction electrocatalysis.Concretely,a secondary metal precursor is introduced to the primary single-atom sites to introduce direct metal-metal interaction,which ensures the formation of desired atom pair structure during the subsequent pyrolysis process and allows for successful construction of DACs.The as-prepared FeCo-NC DAC exhibits superior oxygen reduction electrocatalytic activity with a half-wave potential of 0,91 V vs.reversible hydrogen electrode.Zn-air batteries equipped with the FeCo-NC DAC demonstrate higher peak power density than those with the Pt/C benchmark.More importantly,this post-modification strategy is demonstrated universal to achieve a variety of dual-atom sites.This work presents an effective synthesis methodology for precise construction of catalytic materials and propels their applications in energy-related devices. 展开更多

The exploitation of electrocatalysts with high activity and durability for HER is desirable for future energy systems,but it is still a challenge.NMPs have attracted increasing attentions,but the preparation process often needs toxic regents or dangerous reaction conditions.Herein,we develop a general green method to fabricate metal-rich NMPs anchored on NPG through pyrolyzing DNA cross-linked complexes.The obtained Ru_(2) P-NPG exhibits an ultrasmall overpotential of 7 mV at 10 mA cm^(2) and ultralow Tafel slope of 33 mV dec^(-1) in 1.0 mol L?1 KOH,even better than that of commercial Pt/C.In addition,Ru 2 P-NPG also shows low overpotentials of 29 and 78 mV in 0.5 mol L^(-1) H_(2)SO_(4) and 1.0 mol L^(-1) PBS,respectively.The superior activity can be attributed to the ultrafine dispersion of Ru 2 P nanoparticles for more accessible sites,more defects formed for abundant active sites,the two-dimensional plane structure for accelerated electron transfer and mass transport,as well as the regulation of electron distribution of the catalyst.Moreover,the synthetic method can also be applied to prepare other metal-rich noble metal phosphides(Pd_(3)P-NPG and Rh_(2)P-NPG),which also exhibits high activity for HER.This work provides an effective strategy for designing NMP-based electrocatalysts. 展开更多

Engineering transition metal compounds(TMCs)catalysts with excellent adsorption-catalytic ability has been one of the most effec-tive strategies to accelerate the redox kinetics of sulfur cathodes.Herein,this review focuses on engineering TMCs catalysts by cation doping/anion doping/dual doping,bimetallic/bi-anionic TMCs,and TMCs-based heterostructure composites.It is obvious that introducing cations/anions to TMCs or constructing heterostructure can boost adsorption-catalytic capacity by regulating the electronic structure including energy band,d/p-band center,electron filling,and valence state.Moreover,the elec-tronic structure of doped/dual-ionic TMCs are adjusted by inducing ions with different electronegativity,electron filling,and ion radius,resulting in electron redistribution,bonds reconstruction,induced vacancies due to the electronic interaction and changed crystal structure such as lat-tice spacing and lattice distortion.Different from the aforementioned two strategies,heterostructures are constructed by two types of TMCs with different Fermi energy levels,which causes built-in electric field and electrons transfer through the interface,and induces electron redistribution and arranged local atoms to regulate the electronic structure.Additionally,the lacking studies of the three strategies to comprehensively regulate electronic structure for improving catalytic performance are pointed out.It is believed that this review can guide the design of advanced TMCs catalysts for boosting redox of lithium sulfur batteries. 展开更多

Peanut(Arachis hypogaea L.)is a globally important oil crop.Web blotch is one of the most important foliar diseases affecting peanut,which results in serious yield losses worldwide.Breeding web blotch-resistant peanut varieties is the most effective and economically viable method for minimizing yield losses due to web blotch.In the current study,a bulked segregant analysis with next-generation sequencing was used to analyze an F2:3 segregating population and identify candidate loci related to web blotch resistance.Based on the fine-mapping of the candidate genomic interval using kompetitive allele-specific PCR(KASP)markers,we identified a novel web blotch resistance-related locus spanning approximately 169 kb on chromosome 16.This region included four annotated genes,of which only Arahy.35VVQ3 had a non-synonymous single nucleotide polymorphism in the coding region between the two parents.Two markers(Chr.16.12872635 and Chr.16.12966357)linked to this gene were shown to be co-segregated with the resistance of peanut web blotch by 72 randomly selected recombinant inbred lines(RIL),which could be used in marker-assisted breeding of resistant peanut varieties. 展开更多

Ni^(2+)and Cd^(2+)in wastewater accumulated through the ecological chain and could jeopardize human health.Adsorption of Ni^(2+)and Cd^(2+)from wastewater using recovered perlite was an important way to solve the problem of resource utilization of solid waste from agar production.Our previous study confirmed that recovered perlite from agar extraction residue had better pore size and specific surface area than commercial perlite.However,the adsorption efficiency and adsorption mechanism of recovered perlite were the main factors limiting its adsorption application.The adsorption process of Ni^(2+)and Cd^(2+)by recovered perlite in aqueous solution was described by the pseudo-second-order kinetic equation,and the relevant adsorption mechanism was mainly chemisorption.Compared with commercial perlite,the adsorption removal rate of Ni^(2+)and Cd^(2+)by enzymatic recovered perlite could reach 92.9%and 89.2%,respectively,and were improved by 12.63%and 13.03%.Langmuir isothermal adsorption model could better describe the isothermal adsorption process of recovered perlite on heavy metal Ni^(2+)and Cd^(2+),and the relevant adsorption mechanism was mainly monolayer adsorption.The X-ray photoelectron spectroscopy(XPS)results indicated that the decrease of Si—O Si^(2+)hydroxyl coordination bond and the increase of C—Si bond might make the binding effect of recovered perlite with heavy metals stronger.The competitive adsorption of Ni^(2+)and Cd^(2+)by recovered perlite was still dominated by chemisorption and monolayer adsorption.This study was expected to provide a theoretical basis and technical support for the removal of Ni^(2+)and Cd^(2+)from wastewater using recovered perlite from seaweed residue. 展开更多

High-voltage transmission lines play a crucial role in facilitating the utilization of renewable energy in regions prone to desertification. The accumulation of atmospheric particles on the surface of these lines can significantly impact corona discharge and wind-induced conductor displacement. Accurately quantifying the force exerted by particles adhering to conductor surfaces is essential for evaluating fouling conditions and making informed decisions. Therefore, this study investigates the changes in electric field intensity along branched conductors caused by various fouling layers and their resulting influence on the adhesion of dust particles. The findings indicate that as individual particle size increases, the field strength at the top of the particle gradually decreases and eventually stabilizes at approximately 49.22 k V/cm, which corresponds to a field strength approximately 1.96 times higher than that of an unpolluted transmission line. Furthermore,when particle spacing exceeds 15 times the particle size, the field strength around the transmission line gradually decreases and approaches the level observed on non-adhering surface. The electric field remains relatively stable. In a triangular arrangement of three particles, the maximum field strength at the tip of the fouling layer is approximately 1.44 times higher than that of double particles and 1.5 times higher compared to single particles. These results suggest that particles adhering to the transmission line have a greater affinity for adsorbing charged particles. Additionally, relevant numerical calculations demonstrate that in dry environments, the primary adhesion forces between particles and transmission lines follow an order of electrostatic force and van der Waals force. Selenopeptides may be a valuable bioactive compound to promote gut microbiota-targeted therapeutic methods for intestinal disease and hepatopathy.However,limited information is available on the utilization of selenopeptides by gut microbiota,especially Selenium(Se)function.For this purpose,the present study aimed to investigate the protective effect of selenopeptide(RYNA(Se)MNDYT,Se-P2,purity of≥95%)and its original peptide(RYNAMNDYT,P2,purity of≥95%)in vivo by the microbiota-metabolite axis and further analyze the potential contribution of Se biofortification to Se-P2 bioactivity.The results showed that Se-P2 exhibits a higher protective effect on lipopolysaccharide(LPS)-induced inflammation than P2,including pathology of the colon and liver,which suggested that the bioactivity of P2 was promoted by the organic combination of Se.Notably,gut microbiota composition tended to be a healthy structure by Se-P2 pretreatment in LPS-injured mice,which had a positive effect on LPS-induced gut microbiota dysbacteriosis.Additionally,only Se-P2 promoted an increase in the relative abundance of Lactobacillus,Alistipes,and Roseburia and a decrease in the relative abundance of Akkermansia,Erysipelatoclostridium,and Bacteroides in LPS-injured mice.The changes in gut microbiota were obviously correlated with the changes in metabolites and affected the metabolic pathways of valine,leucine,isoleucine,phenylalanine,tyrosine,and tryptophan biosynthesis and phenylalanine metabolism.This may be one of the key reasons for Se-P2 to exert bioactivity through the microbiota-metabolite axis.Furthermore,Se-biofortification in Se-enriched Cordyceps militaris affected the parental proteins of Se-P2 to modulate mitogen-activated protein kinase,GPI anchored protein,and carbohydrate metabolism,translation,folding,sorting and degradation,which may contribute to the bioactivity of Se-P2.Our study provides information on the effect of Se on selenopeptides in vivo,which further promotes the prospective applications of selenopeptides as dietary supplements. 展开更多

目的:探讨基于食管癌增强动脉期CT图像的深度学习及影像组学特征联合临床资料预测其T2、T3分期。方法:回顾性收集2015年5月至2024年4月皖南医学院第一附属医院经病理学确诊的388例食管癌患者的临床及CT资料,按照7:3比例随机分为训练集(... 目的:探讨基于食管癌增强动脉期CT图像的深度学习及影像组学特征联合临床资料预测其T2、T3分期。方法:回顾性收集2015年5月至2024年4月皖南医学院第一附属医院经病理学确诊的388例食管癌患者的临床及CT资料,按照7:3比例随机分为训练集(271例)和验证集(117例)。提取食管癌增强CT动脉期图像的影像组学及深度学习特征,使用最小绝对收缩和选择算子算法进行降维和筛选特征,并分别建立组学标签得分(Radscore)和深度学习标签得分(Deepscore)。采用单因素及多因素Logistic回归分析筛选独立危险因素,构建临床、影像组学、深度学习和联合模型,并绘制联合模型列线图。使用受试者工作特征曲线下面积(AUC)评价模型的诊断效能并用DeLong检验比较其差异,用决策曲线评价模型的临床净收益,校正曲线评价模型的校准度。结果:经降维后筛选出9个影像组学特征和12个深度学习特征。多因素Logistic回归分析显示肿瘤长度、边界、Radscore及Deepscore为鉴别食管癌T2、T3分期的独立危险因素。联合模型的AUC在训练集为0.867,与临床模型(0.774,P<0.001)、影像组学模型(0.795,P<0.001)和深度学习模型(0.821,P=0.001)之间的差异均有统计学意义;在验证集为0.810,与临床模型(0.653,P=0.002)、影像组学模型(0.719,P=0.033)、深度学习模型(0.750,P=0.009)之间的差异均有统计学意义。决策曲线显示联合模型在训练集及验证集的临床获益均最高,校正曲线显示联合模型在训练集及验证集均拟合良好(P=0.084、0.053)。结论:基于食管癌增强动脉期的CT图像的深度学习和影像组学特征,结合临床特征能较准确地预测其术前的T2、T3分期,可辅助临床制定治疗方案。 展开更多

BACKGROUND Anti-contactin-associated protein-like 2(CASPR2)antibody encephalitis is an autoimmune disorder characterized by the presence of antibodies against the voltage-gated potassium channel.This leads to neurological symptoms,such as seizures,cognitive decline,and neuropathic pain,primarily affecting the limbic system.The prognosis of this disorder varies among individuals.CASE SUMMARY The patient,a girl aged nine years and nine months,underwent treatment for 14 to 21 d.The main clinical manifestations were vomiting and unclear consciousness,positive pathological signs,normal cranial computed tomography and magnetic resonance imaging,and abnormal electroencephalogram.The child was discharged after receiving immunoglobulin and hormone treatment.Subsequent follow-up over a period of 15 months after discharge,conducted through telephone and outpatient visits,showed no recurrence of symptoms.CONCLUSION Anti-CASPR2 antibody autoimmune encephalitis in children is rare,mainly manifested as convulsions,mental abnormalities,cognitive impairment,and neuropathic pain,among others.Timely evaluation for autoimmune encephalitis antibodies is crucial,especially in cases of recurrent central nervous system involvement in children. 展开更多

Called island-crossing tunnels,some specific underwater tunneling projects face constraints imposed by geological and water conditions,necessitating their passage through artificial or natural islands.The longitudinal of the tunnel follows aW-shaped distribution.The congestion situation does not allowfor immediate longitudinal smoke exhaust at the early stage of the fire,and the natural spread of smoke is complicated.An exhaustive investigation was carried out to analyze the smoke behaviors during a fire incident,employing the fire dynamics software FDS,considering five slopes and four fire locations.The simulation results reveal that the layer of high-temperature smoke becomes thicker as one gets closer to the fire source.The thermal pressure difference significantly impacts the temperature distribution within the tunnel and the distance of smoke spread.The value of the thermal pressure difference is significantly affected by changes in slope.It reaches a maximum of 157 Pa at a 5%slope,while it is only 41 Pa at a 1%slope when the fire occurs at the V-point.Fire hazards vary across locations within the W-shaped tunnel,necessitating separate consideration of the V-point and inverted V-point fire characteristics.The mass flow rate in small and large slope tunnels shows different decay rates due to variations in the main forces acting on the movement.Hence,two equations have been developed to predict the smoke mass flow rate,indicating a nonlinear relationship with the tunnel slope and the distance fromthe fire source.The tunnel slope inversely affects the smoke mass flowrate at the same location.The results can be utilized as a reference for conducting evacuation operations and aiding rescues during aW-shaped tunnel fire. 展开更多

Frequency can realize remote control when staff uses a table to give instruction and set disinfection mode, and then the intelligent disinfection robot returns automatically to the charging pile to charge when the instruction is completed. It can also autonomously move to sterilize without human participation, which makes man-machine separation and accurate disinfection come true. The chance of contact infection and exposure is decreased when an intelligent disinfection robot is used to sterilize the environment and object surface in an isolation ward, which can also reduce occupational exposure, achieve occupational protection of medical workers and ensure there is no hospital infection. 展开更多
