Endorepellin downregulation promotes angiogenesis after experimental traumatic brain injury

Endorepellin plays a key role in the regulation of angiogenesis,but its effects on angiogenesis after traumatic brain injury are unclear.This study explored the effects of endorepellin on angiogenesis and neurobehavioral outcomes after traumatic brain injury in mice.Mice were randomly divided into four groups:sham,controlled cortical impact only,adeno-associated virus(AAV)-green fluorescent protein,and AAV-shEndorepellin-green fluorescent protein groups.In the controlled cortical impact model,the transduction of AAV-shEndorepellin-green fluorescent protein downregulated endorepellin while increasing the number of CD31+/Ki-67+proliferating endothelial cells and the functional microvessel density in mouse brain.These changes resulted in improved neurological function compared with controlled cortical impact mice.Western blotting revealed increased expression of vascular endothelial growth factor and angiopoietin-1 in mice treated with AAV-shEndorepellin-green fluorescent protein.Synchrotron radiation angiography showed that endorepellin downregulation promoted angiogenesis and increased cortical neovascularization,which may further improve neurobehavioral outcomes.Furthermore,an in vitro study showed that downregulation of endorepellin increased tube formation by human umbilical vein endothelial cells compared with a control.Mechanistic analysis found that endorepellin downregulation may mediate angiogenesis by activating vascular endothelial growth factor-and angiopoietin-1-related signaling pathways. 展开更多

For it is a n... In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named "2019 novel coronavirus(2019-nCoV)" by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. This rapid advice guideline is suitable for the first frontline doctors and nurses, managers of hospitals and healthcare sections, community residents, public health persons, relevant researchers, and all person who are interested in the 2019-nCoV. 展开更多

The novel severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is the cause of a rapidly spreading illness,coronavirus disease 2019(COVID-19),affecting more than seventeen million people around the world.Diagnosis and treatment guidelines for clinicians caring for patients are needed.In the early stage,we have issued"A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV)infected pneumonia(standard version)";now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline.We formed a working group of clinical experts and methodologists.The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas:chemoprophylaxis,diagnosis,treatments,and discharge management.We searched the literature for direct evidence on the management of COVID-19,and assessed its certainty generated recommendations using the Grading of Recommendations,Assessment,Development and Evaluation(GRADE)approach.Recommendations were either strong or weak,or in the form of ungraded consensus-based statement.Finally,we issued 34 statements.Among them,6 were strong recommendations for,14 were weak recommendations for,3 were weak recommendations against and 11 were ungraded consensus-based statement.They covered topics of chemoprophylaxis(including agents and Traditional Chinese Medicine(TCM)agents),diagnosis(including clinical manifestations,reverse transcription-polymerase chain reaction(RT-PCR),respiratory tract specimens,IgM and IgG antibody tests,chest computed tomography,chest X-ray,and CT features of asymptomatic infections),treatments(including lopinavirritonavir,umifenovir,favipiravir,interferon,remdesivir,combination of antiviral drugs,hydroxychloroquine/chloroquine,interleukin-6 inhibitors,interleukin-1 inhibitors,glucocorticoid,qingfei paidu decoction,lianhua qingwen granules/capsules,convalescent plasma,lung transplantation,invasive or noninvasive ventilation,and extracorporeal membrane oxygenation(ECMO)),and discharge management(including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge).We also created two figures of these recommendations for the implementation purpose.We hope these recommendations can help support healthcare workers caring for COVID-19 patients. 展开更多

Vehicular beaconing plays an important role to facilitate various applications in the paradigm of Internet of Vehicles(IoV). Due to high dynamic and resource limitation in IoV, how to schedule the vehicular beacon broadcast is challenging, especially in dense scenarios. In this paper, we investigate the beacon broadcast scheduling problem considering the Age of Information(AoI). We first propose an algorithm minimizing the expected sum of AoI considering the limited communication resource and vehicle mobility. Then the performance of the proposed algorithm is analyzed. With the proposed algorithm, the optimal solution can be obtained under certain conditions. Extensive simulations are conducted to verify the efficiency, effectiveness and fairness of the proposed solution. 展开更多

Purpose: Online reviews on tourism attractions provide important references for potential tourists to choose tourism spots. The main goal of this study is conducting sentiment analysis to facilitate users comprehending the large scale of the reviews, based on the comments about Chinese attractions from Japanese tourism website 4 Travel.Design/methodology/approach: Different statistics-and rule-based methods are used to analyze the sentiment of the reviews. Three groups of novel statistics-based methods combining feature selection functions and the traditional term frequency-inverse document frequency(TF-IDF) method are proposed. We also make seven groups of different rulesbased methods. The macro-average and micro-average values for the best classification results of the methods are calculated respectively and the performance of the methods are shown.Findings: We compare the statistics-based and rule-based methods separately and compare the overall performance of the two method. According to the results, it is concluded that the combination of feature selection functions and weightings can strongly improve the overall performance. The emotional vocabulary in the field of tourism(EVT), kaomojis, negative and transitional words can notably improve the performance in all of three categories. The rule-based methods outperform the statistics-based ones with a narrow advantage.Research limitation: Two limitations can be addressed: 1) the empirical studies to verify the validity of the proposed methods are only conducted on Japanese languages; and 2) the deep learning technology is not been incorporated in the methods.Practical implications: The results help to elucidate the intrinsic characteristics of the Japanese language and the influence on sentiment analysis. These findings also provide practical usage guidelines within the field of sentiment analysis of Japanese online tourism reviews.Originality/value: Our research is of practicability. Currently, there are no studies that focus on the sentiment analysis of Japanese reviews about Chinese attractions. 展开更多

Enhanced Healing of Full-thickness Diabetic Wounds Using Bioactive Glass and Yunnan Baiyao Ointments

In order to accelerate the chronic wounds healing, we investigated the healing effects of bioactive glass and Yuunan baiyao ointments in streptozotocin-induced diabetic rats. The ointments were prepared by mixing 45S5 bioactive glass powder (16% weight) with Vaseline and different weight percentages of Yurman baiyao. Full-thickness defect wounds were created on the back of 130 SD rats and were randomly divided into 8 groups. The wound healing rates were calculated at 4, 7, 10, 14 and 21 days after surgery. The samples were harvested for further observations. Considering the wound closure rate, group 6 (with 5% Yuunan baiyao) has better wound healing performance than other diabetic groups. The lower inflammatory response was observed by gross observation and confirmed by the results of H&E staining and TEM observation. Besides, the proliferation of fibroblasts, the formation of granulation tissue, as well as the vascularization, were improved in group 6 compared to other diabetic groups. All results suggest that bioactive glass and Yunnan baiyao ointments can accelerate the recovery of diabetes-impaired skin wounds, and comparing to other diabetic groups, group 6 (with 5% Yunnan baiyao) has better healing effect. 展开更多

海湾是海洋和陆地相互作用和人类活动最强烈的区域,沉积物中重金属的含量和分布都受到人类活动的影响。为了探究钦州湾沉积物中重金属的含量水平、污染程度和可能来源,本研究分析了钦州湾表层沉积物中Cu、Pb、Zn、Cd、Cr、Hg和As的含量... 海湾是海洋和陆地相互作用和人类活动最强烈的区域,沉积物中重金属的含量和分布都受到人类活动的影响。为了探究钦州湾沉积物中重金属的含量水平、污染程度和可能来源,本研究分析了钦州湾表层沉积物中Cu、Pb、Zn、Cd、Cr、Hg和As的含量及空间分布,采用地质累计指数(Igeo)和潜在生态风险指数(PERI)定量分析了重金属的污染程度和生态风险,并利用自组织映射(SOM)和正定矩阵因子分解法(PMF)模型评估了重金属的可能来源。结果表明:钦州湾表层沉积物重金属中Zn的平均含量最高,Hg平均含量最低;除个别站位的Pb含量符合海洋沉积物质量第二类标准外,其他重金属含量均符合海洋沉积物质量第一类标准;Igeo结果表明钦州湾表层沉积物主要的污染元素为Pb、Hg和Cu;PERI结果表明Hg是钦州湾表层沉积物主要的生态风险元素。SOM和PMF模型结果表明钦州湾表层沉积物重金属主要受自然来源、农业活动、工业生产活动的共同影响,各来源的相对贡献率分别为39.65%、29.15%和31.20%。 展开更多

Background:Most bone-related injuries to grassroots troops are caused by training or accidental injuries.To establish preventive measures to reduce all kinds of trauma and improve the combat effectiveness of grassroots troops,it is imperative to develop new strategies and scafolds to promote bone regeneration.Methods:In this study,a porous piezoelectric hydrogel bone scafold was fabricated by incorporating polydopamine(PDA)-modified ceramic hydroxyapatite(PDA-hydroxyapatite,PHA)and PDA-modified barium titanate(PDABaTiO_(3),PBT)nanoparticles into a chitosan/gelatin(Cs/Gel)matrix.The physical and chemical properties of the Cs/Gel/PHA scafold with 0–10 wt%PBT were analyzed.Cell and animal experiments were performed to characterize the immunomodulatory,angiogenic,and osteogenic capabilities of the piezoelectric hydrogel scafold in vitro and in vivo.Results:The incorporation of BaTiO_(3) into the scafold improved its mechanical properties and increased self-generated electricity.Due to their endogenous piezoelectric stimulation and bioactive constituents,the prepared Cs/Gel/PHA/PBT hydrogels exhibited cytocompatibility as well as immunomodulatory,angiogenic,and osteogenic capabilities;they not only effectively induced macrophage polarization to M2 phenotype but also promoted the migration,tube formation,and angiogenic differentiation of human umbilical vein endothelial cells(HUVECs)and facilitated the migration,osteodifferentiation,and extracellular matrix(ECM)mineralization of MC3T3-E1 cells.The in vivo evaluations showed that these piezoelectric hydrogels with versatile capabilities significantly facilitated new bone formation in a rat large-sized cranial injury model.The underlying molecular mechanism can be partly attributed to the immunomodulation of the Cs/Gel/PHA/PBT hydrogels as shown via transcriptome sequencing analysis,and the PI3K/Akt signaling axis plays an important role in regulating macrophage M2 polarization.Conclusion:The piezoelectric Cs/Gel/PHA/PBT hydrogels developed here with favorable immunomodulation,angiogenesis,and osteogenesis functions may be used as a substitute in periosteum injuries,thereby offering the novel strategy of applying piezoelectric stimulation in bone tissue engineering for the enhancement of combat efectiveness in grassroots troops. 展开更多

目的:探讨舒适化医疗技术在牙科恐惧症(child dental fear,DF)儿童口腔治疗中的临床应用。方法:将纳入的94例CDF患儿根据治疗方法分为47例对照组(Ctrl)和47例实验组(Exp),Ctrl组采用传统束缚治疗,Exp组采用口腔舒适化治疗。比较两组治... 目的:探讨舒适化医疗技术在牙科恐惧症(child dental fear,DF)儿童口腔治疗中的临床应用。方法:将纳入的94例CDF患儿根据治疗方法分为47例对照组(Ctrl)和47例实验组(Exp),Ctrl组采用传统束缚治疗,Exp组采用口腔舒适化治疗。比较两组治疗前后的恐惧程度[采用儿童畏惧调查表-牙科分量表(children's fear survey schedule-dental subscale,CFSS-DS)评估]、口腔相关生活质量[采用口腔健康相关生活质量量表(the early childhood oral health impact scale,ECOHIS)评估]和疼痛程度(采用Wong-Baker面部表情量表评估)及接诊时、治疗前、治疗后的心率变异性[相关指标心率(heart rate,HR)、低频功率(low frequency power,LF)、高频功率(high frequency power,HF)、LF/HF]。结果:Ctrl组患儿治疗后的CFSS-DS评分较治疗前明显升高,ECOHIS评分较治疗前明显降低(P<0.05);而Exp组治疗后的CFSS-DS评分和ECOHIS评分均明显低于治疗前和Ctrl组(P<0.05);与Ctrl组比较,Exp组患儿疼痛程度明显降低(P<0.05)。两组患儿治疗前的HR、LF和LF/HF水平均明显高于接诊时,而HF水平明显低于接诊时,且Exp组中上述指标变化幅度小于Ctrl组(P<0.05);同时,Exp组治疗后HR、LF和LF/HF水平较治疗前和Ctrl组降低;而HF水平较治疗前和Ctrl组明显升高(P<0.05)。结论:舒适化医疗技术不仅可降低CDF患儿恐惧与疼痛程度,提高口腔相关生活质量,还可减轻患儿心率变异性的改变,提升舒适度。 展开更多

为揭示厦门海域水体富营养化程度的时空变化特征,本研究根据2003年、2013年和2020年秋季在厦门海域开展的3个航次的调查数据,对水体中的溶解无机氮(DIN)和活性磷酸盐(DIP)含量及氮磷比的时空分布进行了研究,并通过第三代潜在性富营养化... 为揭示厦门海域水体富营养化程度的时空变化特征,本研究根据2003年、2013年和2020年秋季在厦门海域开展的3个航次的调查数据,对水体中的溶解无机氮(DIN)和活性磷酸盐(DIP)含量及氮磷比的时空分布进行了研究,并通过第三代潜在性富营养化评价模型(potential eutrophication assessment model,PE3)评价了厦门海域富营养化程度的变化。结果表明,厦门海域营养盐平均含量及氮磷摩尔比平均值整体上呈现出湾内向湾外递减的空间分布趋势,高值主要集中在厦门西海域和九龙江口临近海域,低值主要出现在大嶝海域。厦门海域DIN平均含量表现为2013年>2020年>2003年,其值分别为(0.789±0.527)、(0.539±0.234)和(0.345±0.167)mg/L;DIP平均含量表现为分别2013年>2003年>2020年,其值分别为(0.070±0.082)、(0.040±0.021)和(0.032±0.009)mg/L;氮磷摩尔比平均比值表现为2020年>2013年>2003年,其值分别为36.2±9.0、28.1±12.7和23.0±19.0。厦门海域的富营养化不仅受沿岸人为活动和陆源径流输入的影响,还受到海域中的海水养殖活动和水体交换能力等的影响。随着近年来厦门市不断加强海洋空间规划的管控,构建陆海统筹的污染防治体系,开展海洋生态综合修复,相较于2003年和2013年,2020年厦门海域富营养化程度明显降低,厦门市海洋生态修复工作取得了显著成效。 展开更多

This article utilizes daily precipitation data from 28 national meteorological stations in northern Shanxi Province spanning from 1972 to 2020,and the US NCEP/NCAR monthly average reanalysis and ERA5 monthly average reanalysis data.The study employs techniques such as empirical orthogonal function(EOF)decomposition,MannKendall mutation and other methods to investigate the spatiotemporal distribution of extreme precipitation index in northern Shanxi and their correlation with atmospheric circulation.The research results show that:the absolute index,relative index,intensity index and sustained dry period index(CDD)in the continuous index appear from southwest to northeast.The spatial distribution characteristics of the central region decrease,while the continuous wet period(CWD)decreases from the central to the east and west.The three indices Rx1day,Rx5day,and CWD mutated in 1978,1975,and 1983 respectively,and other extreme precipitation indices all appeared in a sudden change from a low-value period to a high-value period occurred around 2010.In the high-value years of the summer extreme precipitation index,there is a significant negative anomaly in the height field in the mid-high latitude regions of Eurasia.Northern Shanxi is controlled by a broad low-pressure trough in the Lake Baikal area.Water vapor transported via the east,west,and south routes converges in the northern Shanxi region and encounters cold air from the north.There is a strong upward motion anomaly at 500 hPa in the troposphere,and the dynamic conditions of upper-level divergence and lower-level convergence lead to more summer extreme precipitation in the northern Shanxi region.Conversely,in the low-value years of the summer extreme precipitation index,northern Shanxi is affected by a strong high-pressure ridge north of Lake Baikal.There is a downward motion anomaly at 500 hPa,and the northern Shanxi region lacks water vapor.The cold and warm air cannot converge,and both the water vapor conditions and dynamic conditions are poor,which is not conducive to the production of extreme precipitation in northern Shanxi. 展开更多

目的 探讨基于微环境理论的延续性护理对截瘫患者皮瓣修复术后再发压力性损伤(pressure injury,PI)的影响,以期为提高延续性护理质量提供依据。方法 2016年1月至2020年3月,便利抽样法选取截瘫伴PI患者132例为研究对象,按随机数字表法将... 目的 探讨基于微环境理论的延续性护理对截瘫患者皮瓣修复术后再发压力性损伤(pressure injury,PI)的影响,以期为提高延续性护理质量提供依据。方法 2016年1月至2020年3月,便利抽样法选取截瘫伴PI患者132例为研究对象,按随机数字表法将其分为观察组和对照组,对照组给予常规术后护理和随访,观察组在对照组的基础上给予基于微环境理论的延续性护理,主要包括居室和皮肤局部的温湿度调控。两组患者均进行前瞻性随访记录PI首次再发情况,随访间隙3个月。结果 对照组有9例再发PI,无PI生存时间M(P_(0)~P_(100))为7(1~29)个月;观察组有3例再发PI,无PI生存时间M(P_(0)~P_(100))为32(13~36)个月。两组无PI复发生存曲线差异有统计学意义(χ^(2)=3.958,P=0.047)。结论 在微环境理论指导下的延续性护理能减少深度PI截瘫患者皮瓣修复术后PI的再发,值得推广。 展开更多

目的观察急诊绿色通道在抢救急性重症颅脑外伤患者中的应用价值。方法选取2012年1月1日-2014年10月30日医院收治的急性重症颅脑外伤患者23例作为对照组,2014年11月1日-2017年10月31日医院开展急诊绿色通道收治的急性重症颅脑外伤患者45... 目的观察急诊绿色通道在抢救急性重症颅脑外伤患者中的应用价值。方法选取2012年1月1日-2014年10月30日医院收治的急性重症颅脑外伤患者23例作为对照组,2014年11月1日-2017年10月31日医院开展急诊绿色通道收治的急性重症颅脑外伤患者45例作为观察组,统计所有入组患者检查及检验开具时间、检查检验完成时间、相关科室会诊时间、到院时间、送手术时间、手术开始时间、患者平均住院时间,以及术前、术后5 d及术后2周格拉斯哥昏迷评分(GCS)。分析观察组阶段开展急诊绿色通道的临床价值。结果观察组检查及检验开具时间、检查检验完成时间、相关科室会诊时间、到院时间均短于对照组,差异均有统计学意义(P<0.05)。2组送达手术室时间比较差异不显著(P>0.05);观察组受伤到手术开始时间、患者平均住院时间均短于对照组,差异均有统计学意义(P<0.05)。2组术前GCS评分差异不显著(P>0.05),观察组术后5 d、术后2周GCS评分均高于对照组,差异有统计学意义(P<0.05)。结论针对急性重症颅脑外伤患者通过开展急诊绿色通道可缩短救治时间,减少患者平均住院时间,具有较高临床应用价值。 展开更多

目的探讨血清淀粉酶(AMY)及脂肪酶(LPS)升高在重症儿童中的发生及其与胰腺损伤的关系。方法选取2016年5月至2017年12月在我院儿科重症监护病房(PICU)治疗的重症患儿168例,检测患儿血清AMY和LPS水平,根据AMY和LPS水平将患儿分为三组,A组... 目的探讨血清淀粉酶(AMY)及脂肪酶(LPS)升高在重症儿童中的发生及其与胰腺损伤的关系。方法选取2016年5月至2017年12月在我院儿科重症监护病房(PICU)治疗的重症患儿168例,检测患儿血清AMY和LPS水平,根据AMY和LPS水平将患儿分为三组,A组111例(AMY和LPS正常),B组40例(AMY或LPS升高>1倍正常值,而<3倍正常值),C组17例(AMY或LPS升高≥3倍正常值),比较各组小儿危重病例评分(PCIS)、胰岛素内分泌指标、死亡率等。结果 AMY或LPS升高的总发生率为33. 93%; A组患儿年龄0. 94(0. 20,3. 20)岁,低于B组和C组,差异有统计学意义(P<0. 05); C组PCIS评分为(70. 46±5. 06)分,低于A组和B组,差异有统计学意义(P<0. 05); B组PCIS评分为(79. 16±4. 54)分,低于A组,差异有统计学意义(P<0. 05); C组胰岛素和C肽分别为(24. 06±5. 24) m U/L和(3. 32±0. 62) ng/m L,高于A组和B组,而HOMA-IR指数为(4. 21±20. 15),明显低于A组和B组(P<0. 05); B组胰岛素和C肽分别为(16. 11±4. 80) m U/L和(2. 60±0. 51) ng/m L,明显高于A组(P<0. 05),而HOMA-IR指数为(90. 42±32. 15),明显低于A组,差异有统计学意义(P<0. 05); C组和B组胰腺厚度增大率分别为60. 00%和76. 47%,明显高于A组,差异有统计学意义(P<0. 05); C组和B组患儿死亡率分别为29. 41%和22. 50%,明显高于A组,差异有统计学意义(P<0. 05)。结论有部分重症儿童可发生血清AMY或LPS升高,其与胰腺损伤以及患儿预后有一定关系,可作为重症儿童病情的预警指标。 展开更多

目的分析视频回顾对严重多发伤患者规范化接诊与院内救治流程改进效果。方法选入惠州市第一人民医院于2017年1月-2018年12月期间,出车接诊120例多发伤患者数据,即损伤严重评分(ISS分数)多25分的患者,对就诊时间,将前60例患者予以基础方... 目的分析视频回顾对严重多发伤患者规范化接诊与院内救治流程改进效果。方法选入惠州市第一人民医院于2017年1月-2018年12月期间,出车接诊120例多发伤患者数据,即损伤严重评分(ISS分数)多25分的患者,对就诊时间,将前60例患者予以基础方式急救,而后通过视频录像对关键操作指标的时间进行记录,设定为对照组,将后60例患者设定为研究组,抢救后通过视频录像予以回顾,总结同时学习急救流程,强调时间点的有效掌控,提升创伤患者急救时效的观念。对比两组视频回顾的9个时间关键点,同时对视频回顾学习法前后的指标时间进行分析。结果两组除多功能监护时间对比无差异性(P>0.05),其余指标对比有统计学意义(P<0.05);研究组急诊救治用时、抢救成功率、患者转运缺陷、抢救物品单元缺失率优于对照组。结论将视频回顾方法应用于诊疗中,有利于创伤急救时效性的提高。 展开更多

目的探究骨盆骨折院前急救和院内救治的损伤控制效果。方法择取2016年11月至2019年12月我院收治的经急诊CT检查确诊的101例骨盆骨折患者为观察组,另回顾性分析2012年1月至2016年10月157例骨盆骨折患者的临床资料,并将其设为对照组。观... 目的探究骨盆骨折院前急救和院内救治的损伤控制效果。方法择取2016年11月至2019年12月我院收治的经急诊CT检查确诊的101例骨盆骨折患者为观察组,另回顾性分析2012年1月至2016年10月157例骨盆骨折患者的临床资料,并将其设为对照组。观察组根据骨科损伤控制(DCO)理论开展对应治疗,对照组行常规院前急救和院内救治。比较两组死亡率、并发症发生情况、实验室指标、骨折复位效果及远期疗效。结果观察组患者死亡率低于对照组,骨折复位优良率和Cole评分高于对照组,差异具有统计学意义(P<0.05)。观察组感染、DIC及MODS发生率均低于对照组,差异具有统计学意义(P<0.05)。复苏4 h后,观察组PT短于对照组,HCT、PLT高于对照组,血乳酸含量低于对照组,差异具有统计学意义(P<0.05)。结论在骨盆骨折患者的院前急救、院内救治中开展DCO理论能有效提升救治效果,降低患者并发症发生率及死亡率,值得在临床中推广。 展开更多

PCR技术从黑曲霉(Aspergillus niger)基因组扩增获得1个全长1 790 bp的α-L-阿拉伯呋喃糖苷酶基因,基因含有1个50 bp的内含子序列,其对应的蛋白质序列含有5个O糖基化位点和9个N糖基化位点,N端含有18个氨基酸的信号肽序列。将目的基因与表达载体pPICZαA连接并在毕赤酵母X-33诱导表达,获得重组α-L-阿拉伯呋喃糖苷酶。重组酶的相对分子质量为70 kDa,最适反应pH和温度分别为pH 5.5和50℃;金属离子Cu^(2+)、Zn^(2+)和Fe^(2+)对重组酶的酶活有抑制作用,Fe^(3+)对重组酶的酶活有促进作用;以4-Nitrophenylα-L-arabinofuranoside为底物测得酶的Km值和Vmax值分别为0.78 mmol/L和2.57μmol/(min·mg)。在大麦麦芽协定糖化的初始阶段添加31.2 mU/g重组酶,麦汁的过滤速度提高了12.8%。以大麦麦芽阿拉伯木聚糖为底物,α-L-阿拉伯呋喃糖苷酶与木聚糖酶有较好的协同作用。 展开更多
