为了探究长期干旱胁迫下连翘不同器官的非结构性碳水化合物(NSC)含量与水力特性的协调及响应机制。以连续3年不同水分条件处理后的连翘幼苗为研究对象,设置3个水分处理(适宜供水、中度干旱胁迫和重度干旱胁迫),研究长期干旱胁迫后连翘...为了探究长期干旱胁迫下连翘不同器官的非结构性碳水化合物(NSC)含量与水力特性的协调及响应机制。以连续3年不同水分条件处理后的连翘幼苗为研究对象,设置3个水分处理(适宜供水、中度干旱胁迫和重度干旱胁迫),研究长期干旱胁迫后连翘幼苗的光合特性、生物量的分配、NSC各组分含量、水力特性的变化及其碳水两者之间的相关关系。结果表明:(1)适宜供水、中度干旱、重度干旱胁迫下,枝条的栓塞程度分别为30.7%、41.8%和42.3%,枝条导水率分别为0.95、0.71、0.65 kg m^(-1)s-1MPa^(-1)。(2)重度干旱胁迫显著降低了净光合速率、蒸腾速率、气孔导度、水分利用效率。(3)重度干旱胁迫导致地上和粗根生物量显著降低,细根生物量和根冠比显著增加。此外,各器官的NSC含量显著降低,其根系NSC消耗量最高,根系的可溶性总糖和淀粉含量显著降低,枝条的可溶性总糖、葡萄糖和蔗糖含量增加了12.9%、31.1%和45.7%,而淀粉含量降低了40.7%。(4)枝条栓塞程度和导水率与可溶性总糖、淀粉、蔗糖和葡萄糖含量显著相关,其栓塞程度与可溶性总糖、葡萄糖和蔗糖呈正相关,而与淀粉呈负相关(P<0.01)。综上所述,干旱导致连翘枝条木质部的栓塞程度增加,导水率、光合作用和水分运输效率均显著降低,但连翘通过提高枝条内可溶性总糖、葡萄糖、蔗糖含量和降低淀粉、NSC含量以提高植物在干旱条件下的存活机率及旱后水分恢复能力,研究为半干旱区连翘培育和经营提供理论依据。展开更多
目的探讨免疫调节分子IL-33对绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)模型鼠RANKL/c-Fos/NFATc1信号轴的调控作用及藤黄健骨胶囊(Tenghuang Jiangu capsule,TJC)的干预机制。方法构建PMOP大鼠模型,设置假手术组、PMOP模型...目的探讨免疫调节分子IL-33对绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)模型鼠RANKL/c-Fos/NFATc1信号轴的调控作用及藤黄健骨胶囊(Tenghuang Jiangu capsule,TJC)的干预机制。方法构建PMOP大鼠模型,设置假手术组、PMOP模型组、阳性对照组(0.09 mg/kg)和TJC高、中、低剂量组(0.36、0.18、0.09 g/kg),灌胃给药,每天1次,持续8周。从大鼠末次给药后体质量、骨密度(bone mineral density,BMD)及股骨组织微结构等方面评价TJC的疗效;通过ELISA分析TJC对大鼠血清免疫调节因子IL-33、IL-1、IL-31变化;运用qPCR和Western blotting分析TJC对大鼠股骨OPG、RANKL、RANK、c-Fos、NFATc1表达量的影响。结果与假手术组相比,PMOP模型组大鼠的体质量、骨髓脂肪组织相对面积(relative area of bone marrow adipose tissue,BMAT),IL-1、IL-31变化,RANKL、RANK、c-Fos、NFATc1的表达均出现了明显升高的趋势,而BMD、IL-33变化、OPG的表达则出现了明显下降的趋势(P<0.01);与PMOP模型组相比,TJC高剂量组大鼠体质量出现显著下降趋势、IL-33变化出现显著升高趋势(P<0.01),TJC中、高剂量组大鼠BMAT、RANK的表达出现显著下降趋势、OPG的表达出现显著升高趋势(P<0.05或P<0.01),TJC各剂量组大鼠IL-1、IL-31变化,RANKL、c-Fos、NFATc1出现显著降低趋势,BMD出现显著增加趋势(P<0.05或P<0.01)。结论TJC能够提高PMOP大鼠免疫调控分子IL-33含量,抑制免疫调控分子IL-1、IL-31含量和破骨细胞分化标志分子NFATc1的表达,其机制可能与RANKL/c-Fos/NFATc1抑制骨代谢通路密切相关。展开更多
目的通过对肌萎缩侧索硬化(amyotrophic lateral sclerosis,ALS)、腕管综合征(carpal tunnel syndrome,CTS)及健康对照组正中神经的神经传导及F波指标的对比分析,探讨无创鉴别ALS和CTS的电生理指标。方法回顾性分析作者医院2019年1月至2...目的通过对肌萎缩侧索硬化(amyotrophic lateral sclerosis,ALS)、腕管综合征(carpal tunnel syndrome,CTS)及健康对照组正中神经的神经传导及F波指标的对比分析,探讨无创鉴别ALS和CTS的电生理指标。方法回顾性分析作者医院2019年1月至2020年10月收治的ALS患者(n=45)及CTS患者(n=53),并选择同期健康体检者(n=58)作为健康对照,对三组正中神经的神经传导及F波指标进行分析,并对ALS组临床资料(包括ALS的病程、起病部位、病情进展方式、近或远端起病方式等)与正中神经F波指标进行相关性分析。结果远端运动潜伏期(distal motor latency,DML)、复合肌肉动作电位(compound muscle action potential,CMAP)、感觉神经传导速度(sensory nerve conduction velocity,SCV)三组间差异有统计学意义(均P<0.05),且三组间两两比较差异均有统计学意义(均P<0.05)。F波出波率(F-wave persistent,FP)、重复F波百分比(the percentage of repeated F waves,RFP)、巨大F波(giant F waves,GF)出现与不出现受试者比值、重复巨大F波(repeated giant F waves,RGF)出现与不出现受试者比值在三组间差异有统计学意义(均P<0.05)。与CTS组比较,ALS组正中神经DML〔(4.51±0.91)ms比(4.02±0.76)ms,P<0.05〕更短、CMAP波幅下降更明显〔(10.76±3.98)mV比(5.15±3.86)mV,P<0.05〕、SCV更快〔(42.54±12.07)m/s比(58.81±6.21)m/s,P<0.05〕、F波最短潜伏期与M波潜伏期差(difference between F-wave minimal latency and M-wave latency,Fmin-M)更长〔(20.05±3.44)ms比(22.05±2.47)ms,P<0.05〕、FP降低〔95.0(80.0,100.0)%比50.0(25.0,85.0)%,U=2046.50,P<0.05〕、RFP更高〔0(0,10.0)%比15.0(0,27.5)%,U=885.00,P<0.05〕、GF出现与不出现受试者比值〔3/50比14/31,χ^(2)=98.00,P<0.05〕及RGF出现与不出现受试者比值更高〔0/53比7/38,χ^(2)=98.00,P<0.05〕。ALS组FP与病程(r=-0.49,P=0.003)、病情进展方式(r=-0.43,P=0.020)及起病部位(r=-0.50,P=0.003)呈负相关,RFP与病情进展方式(r=-0.42,P=0.022)及起病部位(r=-0.45,P=0.008)呈负相关。结论正中神经感觉传导相对保留而CMAP波幅降低是ALS鉴别于CTS的重要电生理特点。出现正中神经RGF是ALS与CTS鉴别的重要指标;ALS在腕以远和腕以近均有运动传导障碍,不能将远端运动潜伏期延长作为鉴别ALS与CTS的指标。展开更多
OBJECTIVE To explore the protective effects of total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja on STZ-stimulated INS-1 cells through regulating of autophagy and apoptosis.METHODS INS-1cells wer...OBJECTIVE To explore the protective effects of total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja on STZ-stimulated INS-1 cells through regulating of autophagy and apoptosis.METHODS INS-1cells were cultured in media containing 3n M STZ and different doses of total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja.The proliferation of cells was examined by MTT assay,ROS content were detected by fluorescence enzyme label.The levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),hydrogen peroxidase(CAT),malondialdehyde(MDA)were also measured by colorimetry.The activities of caspase-3,9 were also observed by Caspase colorimetric assay kit in each group.The expressions of Bcl-2 m RNA and Bax m RNA were detected by Real time PCR,protein expression of LC3-Ⅱand PARP were detected by Western blotting.RESULTS Compared with the control group,total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja could promote the proliferation of INS-1 cells no matter with STZ or not when its concentration lower than 25μg·m L-1;but when its concentration higher than 100μg·m L-(1use individually)or 50μg·m L-1(combined use),total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja might significantly inhibite the growth of the cells whether STZ existed or not.Total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja(6.25,12.5,25μg·m L-1)might inhibit INS-1cell apoptosis,decrease intra-cellular ROS contents,improve the s upernatant liquid SOD,GSH-PX,CAT activities,decrease MDA level,promote INS-1 cell secreting insulin,decrease the protein expressions of autophagy protein LC3Ⅱand apoptosis regulating protein cleaved PARP protein,up-regulate the anti-apoptotic protein Bcl-2 m RNA expression,down-regulate the pro-apoptotic protein Bax m RNA expression and Bcl-2and Bax,reduce the activity of caspase-9 and caspase-3(P<0.05 or P<0.01).CONCLUSION Total triterpenoids extracts from Cyclocarya paliurus(Batal.)Iljinskaja has good protective effect on STZ-stimulated INS-1cells.It can inhibit STZ injured INS-1 cells to overproduce ROS production,enhance endogenous antioxidant enzymes(GSH-Px,SOD,CAT activities),reduce the expression of autophagy protein LC3Ⅱand apoptosisregulating protein cleaved PARP,up-reglute the antiapoptosis protein Bcl-2 expression,down-regulate the pro-apoptotic protein Bax expression,decrease the caspase-9 and caspase-3 activities,and improve the INS-1cell survival rate,and then play a protective effect on damaged INS-1 cells.展开更多
