A woman who had a uterine malformation was misdiagnosed as uterus didelphys and gave birth to a live, healthy fetus. The correct diagnosis was not made until cesarean section at 37 4/7 weeks. The case suggests that women who are pregnant in a rudimentary horn could obtain a full-term delivery and give birth to a live and healthy baby.展开更多
BACKGROUND Gastric cancer(GC)is a common malignant tumor with high incidence and mortality rates globally,especially in East Asian countries.Helicobacter pylori(H.pylori)infection is a significant and independent risk...BACKGROUND Gastric cancer(GC)is a common malignant tumor with high incidence and mortality rates globally,especially in East Asian countries.Helicobacter pylori(H.pylori)infection is a significant and independent risk factor for GC.However,its underlying mechanism of action is not fully understood.Dickkopf-related protein(DKK)1 is a Wnt signaling antagonist,and cytoskeleton-associated protein(CKAP)4 is a newly identified DKK1 receptor.Recent studies found that the binding of DKK1 to CAKP4 mediated the procancer signaling of DKK1 independent of Wnt signaling.We hypothesize that H.pylori-induced activation of DKK1/CKAP4 signaling contributes to the initiation and progression of GC.AIM To investigate the interaction of H.pylori infection,DKK1 and CAKP4 in GC,as well as the underlying molecular mechanisms.METHODS RNA sequencing was used to identify differentially expressed genes(DEGs)between H.pylori-infected and uninfected primary GC cells.Gain-and loss-offunction experiments were performed to verify the H.pylori-induced upregulation of activator protein-1(AP-1)in GC cells.A dual-luciferase reporter assay and co-immunoprecipitation were used to determine the binding of AP-1 to the DKK1 promoter and DKK1 to CKAP4.Western blotting and immunohistochemistry detected the expression of DKK1,CKAP4,and phosphatidylinositol 3-kinase(PI3K)pathway-related proteins in GC cells and tissues.Functional experiments and tumorigenicity in nude mice detected malignant behavior of GC cells in vitro and in vivo.RESULTS We identified 32 DEGs between primary GC cells with and without H.pylori infection,including JUN,fos-like antigen-1(FOSL1),and DKK1,and confirmed that the three proteins and CKAP4 were highly expressed in H.pylori-infected GC cells,H.pylori-infected gerbil gastric tissues,and human GC tissues.JUN and FOSL1 form AP-1 to transcriptionally activate DKK1 expression by binding to the DKK1 promoter.Activated DKK1 bound to CKAP4,but not the most common Wnt coreceptor low-density lipoprotein receptor-related protein 5/6,to promote GC cell growth,colony formation,migration,invasion,and xenograft tumor growth in nude mice.All these effects were driven by activation of the PI3K/AKT/mammalian target of rapamycin(mTOR)pathway.Targeting the PI3K signaling pathway by LY294002 inhibited DKK1-mediated CKAP4/PI3K signaling activity and the malignant behavior of GC cells.CONCLUSION H.pylori induces JUN and FOSL1 expression to form AP-1,which transcriptionally activates DKK1.Binding of DKK1 to KAKP4 contributes to gastric tumorigenesis via the PI3K/AKT/mTOR pathway.展开更多

Non-pollinated “M3” kiwifruit plant was taken as control. We found that In vitro germination and fertilization ability of different stored pollen sources, and also fruit quality characters including dry matter, total sugar, titratable acid, vitamin C, total polyphenol, total flavonoid, chlorophyll (“a”, “b”, and total) and carotenoid contents except total soluble solid content were significantly different. MH67 and MH45 genotypes were evaluated as the most suitable pollenizers which can be recommended as new candidate cultivars because of long term storage ability and brought about desired fruit quality characters. Confocal Raman microspectroscopy(CRM)is an important tool for analyzing the compositional distribution of cell walls in situ.In this study,we improved the sample preparation method using paraffin-embedded sections com...Confocal Raman microspectroscopy(CRM)is an important tool for analyzing the compositional distribution of cell walls in situ.In this study,we improved the sample preparation method using paraffin-embedded sections combined with hexane dewaxing to obtain high resolution Raman images.We determined that the cell wall components of fiber cells were different from those of ray cells and vessel cells in the xylem of Populus tomentosa.Acetyl bromide and CRM methods produced similar trends when the difference in lignin intensity in the xylem region was compared between transgenic PtrLac4 and wild-type P.tomentosa.However,CRM proved more useful to analyze the lignin distribution in each cell type and distinguished the detailed difference in lignin intensity at the cellular level.Thus,CRM proved to be a useful in situ method to rapidly analyze the spatial variation of lignin content in the xylem of woody plants.展开更多

The preexistence of immune cells in the tumor microenvironment substantiates the efficacy of immunotherapy in cancer patients.Although the complex intratumoral immune heterogeneity has been extensively studied in sing...The preexistence of immune cells in the tumor microenvironment substantiates the efficacy of immunotherapy in cancer patients.Although the complex intratumoral immune heterogeneity has been extensively studied in single cell resolution,hi-res spatial investigations are limited.In this study,we performed a spatial transcriptome analysis of 4 colorectal adenocarcinoma specimens and 2 paired distant normal specimens to identify the molecular pattern involved in a discontinuous inflammatory response in pathologically annotated cancer regions.Based on the location of spatially varied gene expression,we unmasked the spatially-varied immune ecosystem and identified the locoregional“warmed-up”immune response in predefined“cold”tumor with substantial infiltration of immune components.This“warmed-up”immune profile was found to be associated with the in-situ copy number variance and the tissue remodeling process.Further,“warmed-up”signature genes indicated improved overall survival in CRC patients obtained from TCGA database.展开更多

The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigat...The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.展开更多
Improving the performance and reducing the manufacturing costs are the main directions for the development of organic solar cells in the future.Here,the strategy that uses chemical structure modification to optimize t...Improving the performance and reducing the manufacturing costs are the main directions for the development of organic solar cells in the future.Here,the strategy that uses chemical structure modification to optimize the photoelectric properties is reported.A new narrow bandgap(1.30 eV)chlorinated non-fullerene electron acceptor(Y15),based on benzo[d][1,2,3] triazole with two 3-undecylthieno[2’,3’:4,5] thieno[3,2-b] pyrrole fused-7-heterocyclic ring,with absorption edge extending to the near-infrared(NIR) region,namely A-DA’D-A type structure,is designed and synthesized.Its electrochemical and optoelectronic properties are systematically investigated.Benefitting from its NIR light harvesting,the fabricated photovoltaic devices based on Y15 deliver a high power conversion efficiency(PCE) of 14.13%,when blending with a wide bandgap polymer donor PM6.Our results show that the A-DA’D-A type molecular design and application of near-infrared electron acceptors have the potential to further improve the PCE of polymer solar cells(PSCs).展开更多
In plant pathology,the correct naming of a species is essential for determining the causal agents of disease.Species names not only serve the general purpose of concise communication,but also are critical for effectiv...In plant pathology,the correct naming of a species is essential for determining the causal agents of disease.Species names not only serve the general purpose of concise communication,but also are critical for effective plant quarantine,prevent-ing the introduction of new pathogens into a territory.Many phytopathogenic genera have multiple species and,in several genera,disagreements between the multiple prevailing species concept definitions result in numerous cryptic species.Some of these species were previously called by various names;forma speciales(specialised forms),subspecies,or pathotypes.However,based on new molecular evidence they are being assigned into new species.The frequent name changes and lack of consistent criteria to delineate cryptic species,species,subspecies,forms,and races create increasing confusion,often making communication among biologists arduous.Furthermore,such ambiguous information can convey misleading evo-lutionary concepts and species boundaries.The aim of this paper is to review these concepts,clarify their use,and evaluate them by referring to existing examples.We specifically address the question,“Do plant pathogens require a different ranking system?”We conclude that it is necessary to identify phytopathogens to species level based on data from multiple approaches.Furthermore,this identification must go beyond species level to clearly classify hitherto known subspecies,forms and races.In addition,when naming phytopathogenic genera,plant pathologists should provide more information about geographic locations and host ranges as well as host specificities for individual species,cryptic species,forms or races.When describing a new phytopathogen,we suggest that authors provide at least three representative strains together with pathogenicity test results.If Koch’s postulates cannot be fulfilled,it is necessary to provide complementary data such as associated disease severity on the host plant.Moreover,more sequenced collections of species causing diseases should be published in order to stabilise the boundaries of cryptic species,species,subspecies,forms,and races.展开更多

文摘This article is the 15th contribution in the Fungal Diversity Notes series,wherein 115 taxa from three phyla,nine classes,28 orders,48 families,and 64 genera are treated.Fungal taxa described and illustrated in the present study include a new family,five new genera,61 new species,five new combinations,one synonym,one new variety and 31 records on new hosts or new geographical distributions.Ageratinicolaceae fam.nov.is introduced and accommodated in Pleosporales.The new genera introduced in this study are Ageratinicola,Kevinia,Pseudomultiseptospora(Parabambusicolaceae),Marasmiellomycena,and Vizzinia(Porotheleaceae).Newly described species are Abrothallus altoandinus,Ageratinicola kunmingensis,Allocryptovalsa aceris,Allophoma yuccae,Apiospora cannae,A.elliptica,A.pallidesporae,Boeremia wisteriae,Calycina papaeana,Clypeo-coccum lichenostigmoides,Coniochaeta riskali-shoyakubovii,Cryphonectria kunmingensis,Diaporthe angustiapiculata,D.campylandrae,D.longipapillata,Diatrypella guangdongense,Dothiorella franceschinii,Endocalyx phoenicis,Epicoc-cum terminosporum,Fulvifomes karaiensis,F.pannaensis,Ganoderma ghatensis,Hysterobrevium baoshanense,Inocybe avellaneorosea,I.lucida,Jahnula oblonga,Kevinia lignicola,Kirschsteiniothelia guangdongensis,Laboulbenia caprina,L.clavulata,L.cobiae,L.cosmodisci,L.nilotica,L.omalii,L.robusta,L.similis,L.stigmatophora,Laccaria rubriporus,Lasiodiplodia morindae,Lyophyllum agnijum,Marasmiellomycena pseudoomphaliiformis,Melomastia beihaiensis,Nemania guangdongensis,Nigrograna thailandica,Nigrospora ficuum,Oxydothis chinensis,O.yunnanensis,Petriella thailandica,Phaeoacremonium chinensis,Phialocephala chinensis,Phytophthora debattistii,Polyplosphaeria nigrospora,Pronectria loweniae,Seriascoma acutispora,Setoseptoria bambusae,Stictis anomianthi,Tarzetta tibetensis,Tarzetta urceolata,Tetraploa obpyriformis,Trichoglossum beninense,and Tricoderma pyrrosiae.We provide an emendation for Urnula ailaoshanensis Agaricus duplocingulatoides var.brevisporus introduced as a new variety based on morphology and phylogeny.

文摘The preexistence of immune cells in the tumor microenvironment substantiates the efficacy of immunotherapy in cancer patients.Although the complex intratumoral immune heterogeneity has been extensively studied in single cell resolution,hi-res spatial investigations are limited.In this study,we performed a spatial transcriptome analysis of 4 colorectal adenocarcinoma specimens and 2 paired distant normal specimens to identify the molecular pattern involved in a discontinuous inflammatory response in pathologically annotated cancer regions.Based on the location of spatially varied gene expression,we unmasked the spatially-varied immune ecosystem and identified the locoregional“warmed-up”immune response in predefined“cold”tumor with substantial infiltration of immune components.This“warmed-up”immune profile was found to be associated with the in-situ copy number variance and the tissue remodeling process.Further,“warmed-up”signature genes indicated improved overall survival in CRC patients obtained from TCGA database.

文摘Dear Editor,As an illuminating cancer hallmark,polymorphic microbiomes profoundly impact cancer phenotypes by promoting or repressing cancer initiation and progression.1 Diversity and composition in the gut microbiome are significantly associated with the response rate of anti-PD1 immunotherapy in melanoma.2 In addition to the gut microbiome,a large number of microbiomes colonizing in human tumors have been shown to play significant roles in cancer development.3 However,a comprehensive understanding of intratumor microbiomes in cancer immunotherapy is lacking,largely due to the challenge of investigating intratumor microbiomes in anti-cancer immunotherapy.

文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.

文摘Improving the performance and reducing the manufacturing costs are the main directions for the development of organic solar cells in the future.Here,the strategy that uses chemical structure modification to optimize the photoelectric properties is reported.A new narrow bandgap(1.30 eV)chlorinated non-fullerene electron acceptor(Y15),based on benzo[d][1,2,3] triazole with two 3-undecylthieno[2’,3’:4,5] thieno[3,2-b] pyrrole fused-7-heterocyclic ring,with absorption edge extending to the near-infrared(NIR) region,namely A-DA’D-A type structure,is designed and synthesized.Its electrochemical and optoelectronic properties are systematically investigated.Benefitting from its NIR light harvesting,the fabricated photovoltaic devices based on Y15 deliver a high power conversion efficiency(PCE) of 14.13%,when blending with a wide bandgap polymer donor PM6.Our results show that the A-DA’D-A type molecular design and application of near-infrared electron acceptors have the potential to further improve the PCE of polymer solar cells(PSCs).

文摘In plant pathology,the correct naming of a species is essential for determining the causal agents of disease.Species names not only serve the general purpose of concise communication,but also are critical for effective plant quarantine,prevent-ing the introduction of new pathogens into a territory.Many phytopathogenic genera have multiple species and,in several genera,disagreements between the multiple prevailing species concept definitions result in numerous cryptic species.Some of these species were previously called by various names;forma speciales(specialised forms),subspecies,or pathotypes.However,based on new molecular evidence they are being assigned into new species.The frequent name changes and lack of consistent criteria to delineate cryptic species,species,subspecies,forms,and races create increasing confusion,often making communication among biologists arduous.Furthermore,such ambiguous information can convey misleading evo-lutionary concepts and species boundaries.The aim of this paper is to review these concepts,clarify their use,and evaluate them by referring to existing examples.We specifically address the question,“Do plant pathogens require a different ranking system?”We conclude that it is necessary to identify phytopathogens to species level based on data from multiple approaches.Furthermore,this identification must go beyond species level to clearly classify hitherto known subspecies,forms and races.In addition,when naming phytopathogenic genera,plant pathologists should provide more information about geographic locations and host ranges as well as host specificities for individual species,cryptic species,forms or races.When describing a new phytopathogen,we suggest that authors provide at least three representative strains together with pathogenicity test results.If Koch’s postulates cannot be fulfilled,it is necessary to provide complementary data such as associated disease severity on the host plant.Moreover,more sequenced collections of species causing diseases should be published in order to stabilise the boundaries of cryptic species,species,subspecies,forms,and races.

文摘To the Editor:Chlamydia trachomatis(C.trachomatis)is obligate intracellular pathogens,often resulting in development of chronic disease and tissue damage as a consequence of the host inflammatory immune response as well as persistent infections,[1]which is associated with the activation of mitogen-activated protein kinase(MAPK)and extracellular regulated protein kinases(ERK)mediated by Toll-like receptors(TLRs)and myeloid differentiation primary response gene 88(MyD88).[2]Furthermore,C.trachomatis has evolved the ability to block the apoptosis of its host cells involving the interaction of autophagy and apoptosis.[3]Novel strategies to prevent and treat C.trachomatis infection effectively are needed.
