Low-affinity nitrate transporter genes have been identified in subfamilies 4-8 of the rice nitrate transporter 1(NRT1)/peptide transporter family(NPF),but the OsNPF3 subfamily responsible for nitrate and phytohormone transport and rice growth and development remains unknown.In this study,we described OsNPF3.1 as an essential nitrate and phytohormone transporter gene for rice tillering and nitrogen utilization efficiency(NUtE).OsNPF3.1 possesses four major haplotypes of its promoter sequence in 517 cultivars,and its expression is positively associated with tiller number.Its expression was higher in the basal part,culm,and leaf blade than in other parts of the plant,and was strongly induced by nitrate,abscisic acid(ABA)and gibberellin 3(GA_3)in the root and shoot of rice.Electrophysiological experiments demonstrated that OsNPF3.1 is a pH-dependent low-affinity nitrate transporter,with rice protoplast uptake assays showing it to be an ABA and GA_3 transporter.OsNPF3.1 overexpression significantly promoted ABA accumulation in the roots and GA accumulation in the basal part of the plant which inhibited axillary bud outgrowth and rice tillering,especially at high nitrate concentrations.The NUtE of OsNPF3.1-overexpressing plants was enhanced under low and medium nitrate concentrations,whereas the NUtE of OsNPF3.1 clustered regularly interspaced short palindromic repeats(CRISPR)plants was increased under high nitrate concentrations.The results indicate that OsNPF3.1 transports nitrate and phytohormones in different rice tissues under different nitrate concentrations.The altered OsNPF3.1 expression improves NUtE in the OsNPF3.1-overexpressing and CRISPR lines at low and high nitrate concentrations,respectively. 展开更多

Photosynthesis affects crop growth and yield.The roles of microRNAs(miRNAs)in photosynthesis are little known.In the present study,the role of the OsNF-YB7–OsMIR5810–OsMRLP6 regulatory module in photosynthesis was investigated.The malectin-like protein gene OsMRLP6 was identified as a target gene of osa-miR5810(miR5810).Overexpression in rice of miR5810 or down-expression of OsMRLP6 resulted in reduced expression of genes involved in chloroplast development and photosynthesis and decreased net photosynthetic rate,finally leading to lower shoot biomass and grain yield.Down-expression of miR5810 and overexpression of OsMRLP6 showed the opposite effect.Overexpression of transcription factor OsNF-YB7 elevated expression of OsMIR5810 in rice seedlings by binding to its promoter.The OsNFYB7–OsMIR5810–OsMRLP6 regulatory module affects photosynthesis to mediate growth and grain yield. 展开更多

Comparison of DAA and PLA Approaches for Total Hip Replacement in the Treatment of Femoral Neck Fractures

Objectives: Femoral neck fractures are becoming more common within nowadays. This research is to explore the clinical effect of primary total hip replacement (THA) via direct anterior approach (DAA) and posterolateral approach (PLA) in the treatment of femoral neck fracture. Methods: Retrospective analysis of 100 cases of elderly patients with femoral neck fracture who underwent total hip arthroplasty admitted to Tianyou Hospital affiliated to Wuhan University of Science and Technology from January 2019 to January 2022. 50 patients treated with DAA approach were included in the observation group, and 50 patients treated with PLA approach were included in the control group. The operation indexes, postoperative acetabular abduction angle and anteversion angle, hip joint function, Harris score and complications were compared between the two groups. Result: The length of incision in the observation group was shorter than that in the control group, and the amount of intraoperative bleeding and postoperative hospital stay were shorter than those in the control group (P < 0.05);There was a statistically significant difference between the two groups in the ratio of acetabular abduction angle and its safe zone, and the length difference of both lower limbs (P < 0.05), while there was no statistically significant difference between the two groups in the ratio of acetabular anteversion angle and its safe zone, eccentricity, and its recovery rate (P > 0.05);Harris score of hip joint: 6 months after operation, the anterior approach group was significantly higher than the posterolateral approach group (P < 0.05), and there was no statistical difference between the two groups 12 months after operation (P > 0.05);The total incidence of complications in the observation group was lower than that in the control group, with a statistically significant difference (P Conclusion: DAA and PLA approaches for total hip replacement can restore the hip joint structure of patients with femoral neck fractures and achieve good results, but DAA approach has greater advantages in early postoperative recovery, improvement of hip joint function, small surgical injury, high application value, so it is recommended. 展开更多

MicroRNAs(miRNAs)are small single-stranded,non-coding RNA molecules involved in the pathogenesis and progression of cancer,including osteosarcoma.We aimed to clarify the pathways involving miRNAs using new bioinformatics tools.We applied WikiPathways and PathVisio,two open-source platforms,to analyze miRNAs in osteosarcoma using miRTar and ONCO.IO as integration tools.We found 1298 records of osteosarcoma papers associated with the word“miRNA”.In osteosarcoma patients with good response to chemotherapy,miR-92a,miR-99b,miR-193a-5p,and miR-422a expression is increased,while miR-132 is decreased.All identified miRNAs seem to be centered on the TP53 network.This is the first application of PathVisio to determine miRNA pathways in osteosarcoma.MiRNAs have the potential to become a useful diagnostic and prognostic tool in the management of osteosarcoma.PathVisio is a full pathway editor with the potentiality to illustrate the biological events,augment graphical elements,and elucidate all the physical structures and interactions with standard external database identifiers. 展开更多

A Research on the Early Effect of Unicompartmental Knee Arthroplasty for Unicompartment Osteoarthritis of the Knee

Objective: To evaluate the early clinical effect of unicompartmental knee arthroplasty (UKA) for medial unicompartment osteoarthritis of knee in the old people, providing the evidence for the treatment of clinic. Methods: From January of 2016 to January of 2017, 20 cases of knee osteoarthritis in medial unicompartment were treated by unicompartmental knee arthroplasty. The knee joint range of motion (ROM), visual analogue scale (VAS) and knee functional Hospital for Special Surgery (HSS) scores were preoperatively recorded. The loss of blood and time of operation were intraoperative recorded. The satisfaction of patient, knee joint range of motion (ROM) and knee functional special surgical hospital score (HSS score) were postoperatively recorded at one year. Results: All patients have been followed up at an average of twelve months. The operation time was 94 - 135 min, mean 105.6 min. The blood loss was 150 - 500 ml, mean 315 ml. The postoperative HSS and ROM were both reduced. The last follow-up, there were no meniscus subluxation, loose prosthesis, and lateral ventricular osteoarthritis or other complications. Conclusion: Unicompartmental knee arthroplasty has the advantages of small operation trauma, early postoperative effect and quick recovery. 展开更多
