Objective:To investigate whether acupoint penetration acupuncture(APA)could regulate chondrocyte autophagy and apoptosis via the PI3K/Akt-mTOR signaling pathway to reduce cartilage degeneration in knee osteoarthritis(...Objective:To investigate whether acupoint penetration acupuncture(APA)could regulate chondrocyte autophagy and apoptosis via the PI3K/Akt-mTOR signaling pathway to reduce cartilage degeneration in knee osteoarthritis(KOA)rats.Methods: KOA was induced in rats via intra-articular injection of sodium iodoacetate resolution.Forty male Sprague-Dawley rats were randomly assigned to blank control,model,APA,electro-acupuncture(EA),and sham model groups(n=8)and those in the APA and EA groups received their respective therapies.Following completion of the treatment course,histological examinations of cartilage and muscle were conducted.Levels of apoptosis-and autophagy-related factors,including Bax,Bcl-2,mTOR,ULK-1,and Beclin-1 protein,and mRNAs were assessed.Additionally,β-endorphin(β-EP)concentrations in the brain and serum were measured.Results: Histological analysis revealed that APA alleviated cartilage and muscle damage compared with the model group.APA inhibited cartilage degeneration by modulating the expression of apoptosis-and autophagy-related proteins and mRNA,thus preventing chondrocyte apoptosis.In the APA group,Bax and mTOR protein levels were significantly lower than those in the model group(both P=.024).Conversely,the Bcl-2 expression level was significantly higher than that in the EA group(P=.035).Additionally,ULK-1 expression was significantly lower than that in the EA group(P=.045).The mRNA level of Bax was significantly higher than that in the blank control group(P<.001).However,Beclin-1 levels were significantly higher than those in both the model and EA groups(both P<.001).ELISA results showed a significant decrease in the concentration ofβ-EP in the brains of the rats in the APA group compared with those in the model group(P=.032).Conclusions: APA reduced osteoarthritis-related pain and alleviated cartilage damage by upregulating chondrocyte autophagy and down-regulating apoptosis via signaling pathways involving PI3K/Akt-mTOR in KOA rats.展开更多
Marine shale gas in South China is widely distributed and demonstrates an enriched resource. Compared with the North American commercial shale gas field, the shale formation in South China is featured by old age, mult...Marine shale gas in South China is widely distributed and demonstrates an enriched resource. Compared with the North American commercial shale gas field, the shale formation in South China is featured by old age, multiple stages tectonic movements, and high thermal evolution degree and complex reservoir forming conditions. As a result, the existing theories and technical methods of exploration in North America cannot be simply applied to South China. Since 2007, based on the in-depth study on a large quantity of analytical test data, we have conducted the theory and technology research for the southern marine shale gas, found Wufeng-Longmaxi Group deep water shelf high-quality shale gas critical parameters coupling laws, proposed the “Binary Enrichment” theory for highly evolved marine shale gas in southern complex tectonic zone, and established a zone selection and evaluation criteria, which are based on the quality of shale, for the key of preservation condition, and for the purpose of economy. Taking this as a guide, we selected Longmaxi formation Lower Silurian of Fuling area in southeastern Sichuan Province as the preferred breakthrough field for the shale gas exploration. The Jiaoye 1 well has the daily gas production of 20.3 × 10<sup>4</sup> m<sup>3</sup>, which embarked the found of China’s first large-scale business development gas field, the Fuling shale gas field. And we submit the first domestic shale gas proven reserves of 1067.5 × 10<sup>8</sup> m<sup>3</sup>. The Fuling shale gas reservoirs are deep water shelf marine high-quality shale, with favorable thickness and even distribution, without dissection in the middle. They are typical self-generation and self-storage shale gas reservoirs. The gas fields have high production gas well;high pressure gas reservoir, good gas components and good exploit results, and there for the Fuling shale gas field is characterized by mid-depth, high pressure, high reservoir pressure, and high quality gas accumulation. Up to March 24, 2014, all of the 101 fracture-gas testing wells have obtained middle and upper shale airflow, with the average single well test production of 32.6 × 10<sup>4</sup> m<sup>3</sup>/d. In the exploration and development process, technology series in geological evaluation, horizontal well, drilling, well completion, piecewise fracturing techniques have been gradually formed. It is of great importance to commercially develop the domestic shale gas and promote the restructuring of China’s energy structure.展开更多
Genetic control of the timing of flowering in woody plants is complex and has yet to be adequately investigated due to their long life-cycle and difficulties in genetic modification.Studies in Populus,one of the best ...Genetic control of the timing of flowering in woody plants is complex and has yet to be adequately investigated due to their long life-cycle and difficulties in genetic modification.Studies in Populus,one of the best woody plant models,have revealed a highly conserved genetic network for flowering timing in annuals.However,traits like continuous flowering cannot be addressed with Populus.Roses and strawberries have relatively small,diploid genomes and feature enormous natural variation.With the development of new genetic populations and genomic tools,roses and strawberries have become good models for studying the molecular mechanisms underpinning the regulation of flowering in woody plants.Here,we review findings on the molecular and genetic factors controlling continuous flowering in roses and woodland strawberries.Natural variation at TFL1 orthologous genes in both roses and strawberries seems be the key plausible factor that regulates continuous flowering.However,recent efforts suggest that a two-recessive-loci model may explain the controlling of continuous flowering in roses.We propose that epigenetic factors,including non-coding RNAs or chromatin-related factors,might also play a role.Insights into the genetic control of flowering time variation in roses should benefit the development of new germplasm for woody crops and shed light on the molecular genetic bases for the production and maintenance of plant biodiversity.展开更多

Thirty pregnant Landrace sows were as...This experiment was conducted to investigate the effects of dietary ratios of n-6:n-3 polyunsaturated fatty acids (PUFA) on the performance of lactating sows and their piglets. Thirty pregnant Landrace sows were assigned to one of three treatments from d 108 of gestation until weaning (26-29 d) and were fed diets containing different ratios of n-6:n-3 PUFA including 3:1,9:1 and 13:1. The effects on sow and litter production traits were examined together with an assessment of sow body condition. No differences were detected among the treatments for the daily feed intake of sows or changes in sow weight and back-fat levels during lactation (P 〉 0.05). Litter size at d 14 and d 21 were tended to increase in 3:1 treatment compared with 9:1 and 13:1 treatments (P 〈 0.10). Litter weight gain (1.77 kg/d) from d 0 to d 14 was tended to increase in 9:1 groups compared with the other two treatments (P 〈 0.10). A significant difference was observed for the content of a -linolenic acid, total n-3 PUFA, and the ratio of n-6:n-3 PUFA in the colostrum, milk, and piglets plasma (P 〈 0.01). The effects of different ratios of n-6:n-3 PUFA in sow diets on colostrum, milk and piglet plasma immunoglobulin concentrations are studied. No difference was observed among treatments in the concentrations of IgM, and IgA in colostrum (P 〉 0.05). A great significant difference for IgG concentration was observed among 3 group in colostrum. A great significant difference for IgA, and IgM (P〈 0.01) concentrations in piglet plasma at d14 and a significant difference for IgG(P 〈 0.05) was observed at d14. Furthermore, at d 21 of lactation, piglet plasma IgG and IgA concentration were greater in 3:1 compared with 13:1 group (P 〈 0.01). In summary, the current study demonstrated that altering the ratio of n-6:n-3 PUFA in lactating sow diet had an effect on the immune component including immunoglobulin and cytokines, and it tended to increase the litter average daily gain and improve the immune status of piglets when dietary ratio of n-6:n-3 PUFA was 9:1.展开更多
In this paper,a method for rapid detection of cordycepin in food was established based on surface-enhanced Raman spectroscopic analysis method.The surface-enhanced Raman substrates were screened and the Raman detectio...In this paper,a method for rapid detection of cordycepin in food was established based on surface-enhanced Raman spectroscopic analysis method.The surface-enhanced Raman substrates were screened and the Raman detection conditions and sample pretreatment methods were optimized.The optimal substrate was gold nanocolloid,and the optimal detection conditions were as follows:the addition amount of gold nanocolloid was 200μL of gold nanocolloid and the sample addition amount was 5μL.Under the optimal conditions,the detection limit of cordycepin was 1 mg/L,and the sample pretreatment was performed by methanol extraction.Based on the established surface-enhanced Raman spectroscopy(SERS)method,cordycepin in two Cordyceps militaris was detected,which confirmed the practical application of this method.展开更多
