Objective: To observe the antitumor effect and mechanism of recombinant human endostatin(Endostar) injection in tumor combined with intraperitoneal injection of cisplatin on subcutaneous transplanted Lewis lung cancer...Objective: To observe the antitumor effect and mechanism of recombinant human endostatin(Endostar) injection in tumor combined with intraperitoneal injection of cisplatin on subcutaneous transplanted Lewis lung cancer in rats. Methods: A total of 30 C57 rats were selected, and the monoplast suspension of Lewis lung cancer was injected into the left axilla to prepare the subcutaneous transplanted tumor models in the axilla of right upper limb. The models were randomly divided into Groups A, B, and C. Medication was conducted when the tumor grew to 400 mm3. Group A was the control group without any interventional treatment. Group B was injected with Endostar 5 mg.kg-1.d for 10 d. Group C was given the injection of Endostar 5 mg.kg-1.d combined with intraperitoneal injection of cisplatin 5 mg.kg-1.d for 10 d. All the rats in three groups were executed the day after the 10-d medication and the tumor was taken off for measurement of volume and mass changes and calculation of antitumor rate, after which the vascular endothelial growth factor(VEGF) concentration in rats' plasma was determined by ELISA. The tumor tissues were cut for the preparation of conventional biopsies. After hematoxylin-eosin staining, the pathologic histology was examined to observe the structures of tumor tissues, VEGF score and microvessel density(MVD) in each group. Results: The volume and mass of tumor in Groups B and C were significantly lower than Group A(P < 0.05) while the tumor volume and mass in Group C were significantly lower than Group B(P < 0.05). The antitumor rate in Group C was significantly higher than Group B(P < 0.05), but the tumor VEGF score, MVD and plasma VEGF level in Group C were significantly lower than Groups A and B(P < 0.05). In Group B, the tumor VEGF score, MVD and plasma VEGF level were significantly lower than Group A(P < 0.05). The microscopic image of Group C showed that its number of active tumor cells and the blood capillary around tumor was significantly smaller than that of Groups A and B, and meanwhile atrophy and liquefactive necrosis were seen in local tumor. Conclusions: Endostar injection combined with intraperitoneal injection of cisplatin is effective in reducing tumor VEGF score and MVD of transplanted tumor tissues in rats with Lewis lung cancer to obstruct the nutrient supply of tumor cells and kill tumor cells, so that the inhibition of tumor cell proliferation and metastasis can be achieved with a remarkable effect.展开更多

Methods:.A total of 56 patients with TED and 30 controls were recruited in this prospective observational cohort study. TED patients were divided into active TED and inactive TED types according to a seven-point modified formulation of the Clinical Activity Score (CAS). All participants underwent full eye examinations including Ocular Surface Disease Index (OSDI) score, tear film break-up time (TBUT), fluorescein staining and Schirmer I test. Thirty nine patients with thyroid-associated orbitopathy.(TAO) received Nelson's grade with conjunctival impression cytology. Proptosis, palpebral fissure width and lagophthalmos were assessed. Results:.Ocular surface parameters including proptosis, palpebral fissure width and lagophthalmos did not differ between active and inactive TED patients(P>0.05). Both active and inactive TED patients obtained higher fluorescein staining scores, lower TBUT scores and significantly lower Schirmer test scores than those of controls (P<0.001 for all). Additionally, the TBUT score was significantly lower and the OSDI score significantly higher in the active TED group compared with those in the inactive TED group (P<0.001 for both). Impression cytology revealed a higher proportion of grade 2-3 changes in the active TED group compared with the inactive TED group(P<0.001). Conclusion:.Orbital inflammation in TED patients may lead to decreased tear film stability and ocular surface squamous metaplasia.展开更多

We evaluated the influence of five mutations (3321 delA, 441delA, 1249 insG, E1795X and S3296X) in a cohort of 65 IV Chinese patients and in 100 control individuals using the Sequenom? MassARRAY? system. The null allele frequency of 3321delA was 52.31%(34/65). FLG mutation 441delA was only found in one IV patients. FLG mutations 1249insG, E1795X and S3296X were not found in these patients. These findings show that the mutation 3321delA represent the most frequent genetic cause in Southern Chinese IV patients. Our findings confirm and extend the knowledge of the influence of FLG mutations in IV.展开更多

Associ...Dyskeratosis follicularis (Darier’s disease, DD) is rare autosomal dominant disease characterized by hyperkeratotic papules that coalesce into plaques and occur primarily in seborrheic or intertriginous areas. Associated findings include nail abnormalities. A 3-year-old boy presented with porcupine-like appearance for 2 years. The lesion from the back was taken for light microscopy and electron microscopy. He was treated with acitretin (0.31 mg/d to 0.66 mg/d) for 8 years. Light microscopy and electron microscopy showed that the typical features of DD. The patient show good respond to the treatment. During 8 years treatment, the patient had dry mouth and pruritus. The skeletal abnormalities didn’t happen in the patient. The serum lipid profile, liver function and renal function within normal lever after treatment. Our findings showed that porcupine-like appearance is a unique pattern of DD. Acitretin may be a useful therapeutic agent in children with DD and less likely to cause skeletal problems.展开更多

Grape skin,one of the by-products of grape processing,is rich in oxyresveratrol and other bioactive compounds.Oxyresveratrol is a polyphenolic non-flavonoid with anti-inflammatory and anti-aging properties among other...Grape skin,one of the by-products of grape processing,is rich in oxyresveratrol and other bioactive compounds.Oxyresveratrol is a polyphenolic non-flavonoid with anti-inflammatory and anti-aging properties among other biological activities,but little research has been done on its antioxidant mechanism.This study aims to evaluate the regulation of oxyresveratrol on Keap1-Nrf2-ARE signaling pathway.Firstly,oxyresveratrol in grape skin was detected by HPLC-MS/MS,and then its interaction mechanism with Keap1 was investigated by molecular docking.The result of Western blot further confirmed that oxyresveratrol can bind with Nrf2 to induce its nuclear translocation.The Luciferase reporter gene analysis revealed the agonistic effects of oxyresveratrol on ARE.Oxyresveratrol increased not only the mRNA expression levels of ARE-mediated genes NQO1,HO-1,and GCLM,but also their protein expression levels,suggesting that oxyresveratrol inhibited H_(2)O_(2)-induced oxidative damage.Therefore,oxyresveratrol can be applied as a potential antioxidant by activating the Keap-Nrf2-ARE signaling pathway.展开更多
Precision-targeted detection of cancer in a rapid and sensitive fashion remains a significant challenge in prevention,management,and in particular,treatment.Herein,we have designed and synthesized a unique cyclooxygen...Precision-targeted detection of cancer in a rapid and sensitive fashion remains a significant challenge in prevention,management,and in particular,treatment.Herein,we have designed and synthesized a unique cyclooxygenase-2(COX-2)fluorescence probe,dimethylamine-9,10-distyrylanthracene-indomethacin(NDSA-IMC),that could visualize the site of highly expressed COX-2 in the Golgi apparatus of cancer cells,using 9,10-diatyrylanthracene derivative as the luminous unit and a selective inhibitor,indomethacin(IMC),as the recognizing moiety for COX-2.In an aqueous solution,the free state NDSA-IMC showed a weak emission in the absence of COX-2 but enhanced emission in the presence of the enzyme due to the restriction of intramolecular motion of aggregation-induced emission-active NDSA-IMC when bound to COX-2.Cell imaging and flow cytometry experiments indicated that NDSA-IMC could discriminate between cancer and normal cells and visualize the Golgi apparatus of cancer cells via specific targeting of COX-2.Therefore,NDSA-IMC might potentially detect early cancer lesions and ultimately mitigate the population of cancer burden in society.展开更多
