Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family Fabaceae.It is a close relative of cultivated Vigna species such as adzuki bean(V.angularis),cowpea(V.unguiculata),mung bean(V.radiata),and blackgram(V.mungo),and is distributed throughout the tropics.With its ability to tolerate salt stress,beach pea has great potential to contribute salt-tolerance genes for developing salt-tolerant cultivars in cultivated Vigna species.However,it is still underutilized in Vigna breeding programs.A draft genome sequence of beach pea was generated using a high-throughput next-generation sequencing platform,yielding 23.7 Gb of sequence from 79,929,868 filtered reads.A de novo genome assembly containing 68,731 scaffolds gave an N50 length of 10,272 bp and the assembled sequences totaled 365.6 Mb.A total of 35,448 SSRs,including 3574 compound SSRs,were identified and primer pairs for most of these SSRs were designed.Genome analysis identified 50,670 genes with mean coding sequence length 1042 bp.Phylogenetic analysis revealed highest sequence similarity with V.angularis,followed by V.radiata.Comparison with the V.angularis genome revealed 16,699 SNPs and 2253 InDels and comparison with the V.radiata genome revealed 17,538 SNPs and 2300 InDels.To our knowledge this is the first draft genome sequence of beach pea derived from an accession(ANBp-14-03)adapted locally in the Andaman and Nicobar Islands of India.The draft genome sequence may facilitate the genetic enhancement in cultivated Vigna species. 展开更多

Diacerein protects against iodoacetate-induced osteoarthritis in the femorotibial joints of rats 被引量:1

The present study was undertaken to investigate the effect of diacerein on the histopathology of articular cartilage and subchondral bone of the femorotibial joint in rats. Osteoartbritis was induced in rats after single intra-articular injection of sodium iodoacetate. Rats were sacrificed 1, 2, 4, and 8 weeks post intra-articular injection to evaluate the progression of histopathogenesis of osteoarthritis. Diacerein was orally administered (15 mg/kg) once daily post 1 and 2 weeks of iodoacetate injection in two groups, respectively, for up to 12 weeks. Articular cartilage and sub- chondral bone of the rats of both groups were examined after 8 and 12 weeks, respectively. Quantitative histological analyses were performed by scoring these sections as per the OARSI system. Chondroitin sulfate was also estimated in articular cartilage by decrease in absorbance of methylene blue on complexation with chondroitin sulfate using a spectrophotometer. Intra-articular injection of iodoacetate induced loss of articular cartilage with progressive sub- chondral bone sclerosis and degeneration. Based on histopathological and biochemical findings, diacerein treatment showed chondroprotective effect. Furthermore, the chondroprotective effect of diacerein was found to be more pro- nounced after 12 weeks as compared to 8 weeks in both cases (i.e., post 1 and 2 weeks of iodoacetate injection). Similar results were observed by investigation of chondroitin sulfate during biochemical study, showing the chon- droprotective effect. In conclusion, diacerein exhibits chondroprotective effect in rats with late onset of action. 展开更多

Post-surgical evaluation of dry socket formation after surgical removal of impacted mandibular third molar—A prospective study 被引量:1

Aims & Objectives: This study was performed to evaluate the incidence, risk factors (age, sex, infection, radiographic difficulty of the extraction, tobacco use) contributing to the development of dry socket and clinical features following surgical removal of impacted mandibular third molar. Materials and Methods: The study included 63 patients during period of September 2009 to September 2011 in the age group of 18 to 53 years with impacted mandibular third molars all of which were surgically removed under local anesthesia by the same operator. Following surgery all patients received a single dose of 4 mg corticosteroid intravenously and similar prescriptions for analgesics, antibiotics and postoperative instructions. All patients were reviewed at 3rd and 7th day postoperatively. Results: In this study incidence of dry socket was 6.3%. We also noticed onset of symptom mostly appeared within 48 hours. Postoperatively pain, bare bone and halitosis were the most significant clinical feature at 3rd postoperative day and pain, empty socket and bare bone were most significant clinical feature at 7th postoperative day. Variables like right 3rd molar impaction, patients mean age between 19 - 32 years, female, preoperative infection, radiographically difficult impaction and habitual tobacco users showed a higher incidence of dry socket formation. Conclusion: Incidence of dry socket formation is multifactorial and therefore, there is need to evaluate all factors, with special attentions in handling these patients to reduce the incidence of dry socket formation. 展开更多

BACKGROUND:Gallbladder carcinoma is a common malignancy in the Indian subcontinent.It commonly metastasizes through lymphatics,direct invasion,and hematogenous spread.A common extra-abdominal site of metastasis is the lungs.Simultaneous metastasis to breast and ovary is extremely rare.METHOD:This report describes an unusual case of carcinoma gallbladder metastasizing to the breast and ovary at the same time.RESULTS:A 45-year-old woman came to us with complaints of flatulent dyspepsia associated with weight loss and anorexia.Ultrasound of the abdomen revealed hepatomegaly with thick-walled gallbladder with multiple stones and a mass at the fundus,but normal uterus and ovary.Contrast-enhanced computer tomography of the abdomen showed a gallbladder mass infiltrating the liver parenchyma.The patient underwent radical cholecystectomy.Histopathological examination revealed a poorly-differentiated adenocarcinoma with margins free from tumor infiltration.One month after surgery she developed a breast lump.Ultrasound of the abdomen for metastatic workup revealed an ovary mass.Simple mastectomy and salphingo-opherectomy were performed,and histopathological examination revealed a metastatic adenocarcinoma.The patient is now on chemotherapy with gemcitabin.CONCLUSION:This is an unusual case of carcinoma of the gallbladder with metastasis to the breast and ovary,which has not been documented before. 展开更多

Characterization is absolutely necessary and is a must in order to understand and estimate different silver nanoparticle (nm) size in specific group wise manner which corresponds to group wise in number & sizes, and their importance and effect on biological tissue and organs with agglomeration for nano toxicological studies in environments, the acute toxicity of colloidal silver nano particles (AgNps) were studied in fresh dissected tissues of Swiss Albino mice and their fetuses. In this manuscript, an attempt is made to demonstrate the synthesis and characterization of silver nano particles with a wide range of sizes (from 2.75 nm up to 1908.2 nm in radius) by reducing silver nitrate powder with polyvinyl pyrollidone in aqueous solutions in the presence of a sodium borohydride stabilizer. The resulting particles were found spherical aggregates with a rough surface and poly dispersity index below 18.26% (>0.783 PDI). The particle optical, cumulative, diluents and electrical conductivity properties were examined by dynamic light scattering and zeta potential but morphology was evaluated after examination by transmission electron microscopy & image-j. Silver nanoparticles were directly coated with polyvinyl pyrollidone with a sodium borohydride stabilizer. Optical properties on a single-particle level were studied by means of auto correlation function measurements. The effective poly dispersity index of the charged silver nanoparticles was low enough to form a colloidal crystal at low ionic strength. Colloidal form is found more toxic than suspended particles in 1.5 molar sodium chloride solution;this shows increase of silver nanoparticles size due to agglomeration, will reduce the toxicity but increase teratogenicity. 展开更多

CAD tools (Agilent’s Advance Design System software) have been used for simulation and optimization of the circuits. 180 degree balanced mixer configuration has been adopted due to better RF to LO isolation requirement. The circuit is designed on microstrip line using RT Duroid (Dielectric Constant = 2.22). Biomimetic cell culture substrates are developed as an alternative to the conventional substrates.They provide necessary biochemical and biophysical cues to the cells from their surrounding environment for their optimal growth,behaviour and physiology.Changes in physiology of cells growing on biomimetic substrate can essentially affect results of in vitro biological experiments such as drug cytotoxicity,nanoparticle internalization or signalling pathways.As majority of ECM proteins are fibrous in nature,nanofibrous scaffolds have more biomimicking properties.Therefore,in this study,we developed ECM mimicking polycaprolactone-chitosan nanofiber substrate and evaluated its effect on cell morphology,proliferation,cell cycle and ECM production.Further,cellular uptake of BSA-AuNCs has been assessed on conventional and biomimetic substrate in order to demonstrate the effect of these events on cellular properties.It was observed that the cells that were grown for 15 days on the nanofibers,had majority of cells in the proliferative phase of cell cycle compared to TCPS.Moreover,these cells showed extensive collagen and fibronectin production.Due to these conditions C3H10T1/2 cells displayed higher cell internalization of BSA-AuNCs.Overall,this study indicates that the nano-topographical and biochemical environment could alter the cell proliferative behaviour and ECM production,which affects the cell internalization of BSA-AuNCs.Also,PCL-chitosan nanofibrous substrate could be a better alternative to TCPS for cell culture studies. 展开更多

Skin wound healing involves a coordinated cellular response to achieve complete reepithelialisation.Elevated levels of reactive oxygen species(ROS)in the wound environment often pose a hindrance in wound healing resulting in impaired wound healing process.Cerium oxide nanoparticles(CeNPs)have the ability to protect the cells from oxidative damage by actively scavenging the ROS.Furthermore,matrices like nanofibers have also been explored for enhancing wound healing.In the current study CeNP functionalised polycaprolactone(PCL)-gelatin nanofiber(PGNPNF)mesh was fabricated by electrospinning and evaluated for its antioxidative potential.Wide angle XRD analysis of randomly oriented nanofibers revealed^2.6 times reduced crystallinity than pristine PCL which aided in rapid degradation of nanofibers and release of CeNP.However,bioactive composite made between nanoparticles and PCLgelatin maintained the fibrous morphology of PGNPNF upto 14 days.The PGNPNF mesh exhibited a superoxide dismutase(SOD)mimetic activity due to the incorporated CeNPs.The PGNPNF mesh enhanced proliferation of 3T3-L1 cells by^48%as confirmed by alamar blue assay and SEM micrographs of cells grown on the nanofibrous mesh.Furthermore,the PGNPNF mesh scavenged ROS,which was measured by relative DCF intensity and fluorescence microscopy;and subsequently increased the viability and proliferation of cells by three folds as it alleviated the oxidative stress.Overall,the results of this study suggest the potential of CeNP functionalised PCL-gelatin nanofibrous mesh for wound healing applications. 展开更多
