Small nutrients detection is challenging due to the high diversity and broad dynamic range of molecules in food samples, and a further issue is to track low abundance toxins. Herein, we developed a novel plasmonic matrix-assisted laser desorption/ionization mass spectrometry(MALDI MS)approach to detect small nutrients and toxins in complex biological emulsion samples. Silver nanoshells(SiO_2@-Ag) with optimized structures were used as matrices andachieved direct analysis of ~ 6 n L of human breast milk without any enrichment or separation. We performed identification and quantitation of small nutrients and toxins with limit-of-detection down to 0.4 pmol(for melamine) and reaction time shortened to minutes, which is superior to the conventional biochemical method currently in use. The developed approach contributes to the near-future application of MALDI MS in a broad field and personalized design of plasmonic materials for real-case bio-analysis.展开更多

When seedlings germinated on sand cultures with liquid MS medium were considered, the relative germination rate, root length, shoot length and fresh seedling weight were significantly correlated with each other (P 0.01), and only the relative shoot lengths were significantly different among the tested genotypes (P 0.05);When both seedlings germinated on MS and MS plus 0.4% NaCl were considered, only the relative shoot length of seedlings germinated on MS was significantly different from that germinated on MS + 0.4% NaCl (P 0.05), and also only the relative shoot lengths were significantly different among the tested genotypes (P 0.01). Raphanus sativa cv. Changfeng, B. juncea cv. JC and Brassica napus cv. Partial correlation analysis indicated that plant height (X1) was significantly correlated with branching height and the number of first branches (P 2) was significantly correlated with pod number of primary inflorescence (P 3) was significantly correlated with number of secondary branches (P -grain weight (P 4) was significantly correlated with seed yield per plant (P 7) was significantly correlated with seed yield per plant (P -grain weight (P -grain weight (X8) was significantly correlated with seed yield per plant (P -grain weight contributed significantly to seed yield per plant at P 7) and 1000-grain weight (X8) to seed yield per plant (Y) is Y = 0.006 X7 + 1.222 X8 - 7.191. The path coefficient of pod number per plant to seed yield per plant was 0.967 and that of 1000-grain weight was 0.194. The determination coefficient of pod number per plant and 1000-grain weight to seed yield per plant was 0.983 and the determination coefficient of other agronomic traits was 0.130. Coefficient of variance indicated that the length of primary inflorescence showed the greatest variation, followed by seed yield per plant, pod number per plant, number of secondary branches, branching height, pod number of primary inflorescence, number of first branches, seed yield per plot, 1000-grain weight and plant height. It was suggested that seed yield per plant in Crambe might be improved by increasing the pod number per plant through selection or cultivation, but the negative correlation between pod number per plant and 1000-grain weight also needs to be considered.展开更多

Recognition of sleep pattern by itself is a challenge issue, especially for elderly-care community, due to both privacy concerns and technical limitations. We propose a novel multi-parametric sensing system called sleep pattern recognition system (SPRS). This system, equipped with a combination of various non-invasive sensors, can mon- itor an elderly user's sleep behavior. It accumulates the de- tecting data from a pressure sensor matrix and ultra wide band (UWB) tags. Based on these two types of complemen- tary sensing data, SPRS can assess the user's sleep pattern automatically via machine learning algorithms. Compared to existing systems, SPRS operates without disrupting the users' sleep. It can be used in normal households with minimal deployment. Results of tests in our real assistive apartment at the Smart Elder-care Lab are also presented in this paper.展开更多

For a product characterized by a single manufacturing cycle and two selling periods, we depict a Two-Stage (TS) ordering strategy with a stochastic dynamic programming model in the view of the whole system, and prove that the expected profit function of the whole system is concave on the first ordering quantity and the remedial ordering quantity, respectively. Then, the optimal ordering decision is developed. Finally, characteristics of the optimal ordering quantities are analyzed with several examples. Our results show that the suggested TS decision model is better than a Quick Response (QR) decision model.展开更多

Here, we identi...As a sensor of cytosolic DNA, the role of cyclic GMP-AMP synthase (cGAS) in innate immune response is well established, yet how its functions in different biological conditions remain to be elucidated. Here, we identify cGAS as an essential regulator in inhibiting mitotic DNA double-strand break (DSB) repair and protecting short telomeres from end-to-end fusion independent of the canonical cGAS-STING pathway. cGAS associates with telomeric/subtelomeric DNA during mitosis when TRF1/TRF2/POT1 are deficient on telomeres. Depletion of cGAS leads to mitotic chromosome end-to-end fusions predominantly occurring between short telomeres. Mechanistically, cGAS interacts with CDK1 and positions them to chromosome ends. Thus, CDK1 inhibits mitotic non-homologous end joining (NHEJ) by blocking the recruitment of RNF8. cGAS-deficient human primary cells are defective in entering replicative senescence and display chromosome end-to-end fusions, genome instability and prolonged growth arrest. Altogether, cGAS safeguards genome stability by controlling mitotic DSB repair to inhibit mitotic chromosome end-to-end fusions, thus facilitating replicative senescence.展开更多

It was suggested that seed yield per plant in Crambe might be improved by increasing the pod number per plant through selection or cultivation, but the negative correlation between pod number per plant and 1000-grain weight also needs to be considered.

文摘As a foodborne pathogen of global importance,Salmonella enterica serovar Enteritidis(S.Enteritidis)is a threat to public health that is mainly spread by poultry products.Intestinal Enterobacteriaceae can inhibit the colonization of S.Enteritidis and are regarded as a potential antibiotic substitute. We investigated,in chicks, the anti-S.Enteritidis effects of Escherichia coli(E.coli)Nissle 1917,the most wellknown probiotic member of Enterobacteriaceae.Eighty 1-d-old healthy female AA broilers were randomly divided into 4 groups, with 20 in each group, namely the negative control(group P),the E.coli Nissle 1917-treated group(group N),the S.Enteritidis-infected group(group S)and the E.coli Nissle1917-treated and S.Enteritidis-infected group(group NS).From d 5 to 7,chicks in groups N and NS were orally gavaged once a day with E.coli Nissle 1917 and in groups P and S were administered the same volume of sterile PBS.At d 8,the chicks in groups S and NS were orally gavaged with S.Enteritidis and in groups P and N were administered the same volume of sterile PBS.Sampling was conducted 24 h after challenge. Results showed that gavage of E.coli Nissle 1917 reduced the spleen index,Salmonella loads, and inflammation(P<0.05).It improved intestinal morphology and intestinal barrier function(P<0.05).S.Enteritidis infection significantly reduced mRNA expression of angiotensin-converting enzyme 2(ACE2)and solute carrier family 6-member 19(SLC6A19)in the cecum and the content of Gly,Ser,Gln,and Trp in the serum(P<0.05).Pretreatment with E.coli Nissle 1917 yielded mRNA expression of ACE2and SLC6A19 in the cecum and levels of Gly,Ser,Gln,and Trp in the serum similar to that of uninfected chicks(P<0.05).Additionally,E.coli Nissle 1917 altered cecum microbiota composition and enriched the abundance of E.coli,Lactobacillales, and Lachnospiraceae. These findings reveal that the probiotic E.coli Nissle 1917 reduced S.Enteritidis infection and shows enormous potential as an alternative to antibiotics.

文摘Background We aimed to explore predictive measures for intravenous immunoglobulin(IVIG)resistance in children with Kawasaki disease(KD).Methods Patients diagnosed with KD were enrolled in this study.Univariate analysis and multiple logistic regression were utilized to analyze the clinical features and laboratory results prior to IVIG-treatment of the two groups.Independent predictors of IVIG resistance were analyzed, and a predictive model for KD children with IVIG resistance was constructed. Results A total of 277 children with KD,180 boys and 97 girls, aged 2-128(median 23)months,were enrolled in the study.Compared with the IVIG-responsive group, the IVIG-resistant group had higher levels of the peripheral neutrophil count, mean platelet volume, mean platelet volume-to-lymphocyte ratio and C-reactive protein, and total serum bilirubin, but lower levels of peripheral lymphocyte count, serum albumin and serum prealbumin.Age(in months),peripheral neutrophil count, lymphocyte count and mean platelet volume and serum albumin were independent indicators for IVIG resistance by multivariate logistic regression analysis.A logistic regression model and a scoring system were set up,where cut-off values of—0.46 and 6.5 points yielded sensitivities of 83.9%and 77.4%,and specificities of 74.8%and 61.0%,respectively.The areas under the curve(AUC)were 0.808 in the logistic regression model, and 0.750 in the scoring system. Conclusion Our model for predicting IVIG-resistant children with KD,involving age(months),peripheral neutrophil count, lymphocyte count and mean platelet volume and serum albumin prior to IVIG-treatment,is helpful for clinical prediction of children with IVIG-resistant KD.
文摘The quality of sleep may be a reflection of an el- derly individual's health state, and sleep pattern is an im- portant measurement. Results of tests in our real assistive apartment at the Smart Elder-care Lab are also presented in this paper.

文摘This paper investigates the ordering decision problem for a short-life-cycle product under Bayesian updating. Our results show that the suggested TS decision model is better than a Quick Response (QR) decision model.

文摘While the majority of all human cancers court teract telomere shortening by expressing telomerase,-15%of all cancers maintain telomere length by a telomerase?independent mechanism known as alternative lengthening of telomeres(ALT).Here,we show that high load of intrinsic DNA damage is present in ALT cancer cells, leading to apoptosis stress by activating p53-independent,but JNK/c-IVIyc-dependent apoptotic pathway.Notably,ALT cells expressing wild-type p53 show much lower apoptosis than p53-deficient ALT cells.Mechanistically,we find that intrinsic DNA damage in ALT cells induces low level of p53 that is insufficient to initiate the transcription of apoptosis-related genes, but is sufficient to stimulate the expression of key components of mTORC2(mTOR and Rictor),which in turn leads to phosphorylation of AKT.Activated AKT(p-AKT)thereby stimulates downstream anti-apoptotic events.Therefore,p53 and AKT are the key factors that suppress sponta?neous apoptosis in ALT cells.Indeed, inhibition of p53 or AKT selectively induces rapid death of ALT cells in vitro, and p53 inhibitor severely suppresses the growth of ALT-cell xenograft tumors in mice. These findings reveal a previously unrecognized function of p53 in antiapoptosis and identify that the inhibition of p53 or AKT has a potential as therapeutics for specifically targeting ALT cancers.
文摘As a sensor of cytosolic DNA, the role of cyclic GMP-AMP synthase (cGAS) in innate immune response is well established, yet how its functions in different biological conditions remain to be elucidated. Altogether, cGAS safeguards genome stability by controlling mitotic DSB repair to inhibit mitotic chromosome end-to-end fusions, thus facilitating replicative senescence.

文摘Permission delegation has become a new way for data sharing by delegating the authorized permission to other users.A flexible authorization model with strict access control policies is promising for electronic health record(EHR)sharing with security. In this paper,a blockchain-based multi-hop permission delegation scheme with controllable delegation depth for EHR sharing has been presented. We use the interplanetary file system(IPFS)for storing the original EHRs.Smart contracts and proxy re-encryption technology are implemented for permission delegation. In order to ensure data security,we use attribute-based encryption to provide fine-grained access control.Additionally, blockchain is used to achieve traceability and immutability. We deploy smart contracts so that the delegation depth can be set by delegators.Security analysis of the proposed protocol shows that our solution meets the designed goals. Finally,we evaluate the proposed algorithm and implement the scheme on the Ethereum test chain. Our scheme outperforms the competition in terms of performance, according to the results of our experiments.
