Since the complication of monitoring and evaluating the problems about the transgenic expression and its impacts on the receptor in the transgenic crop breeding and other relevant evaluated works,the authors in the pr...Since the complication of monitoring and evaluating the problems about the transgenic expression and its impacts on the receptor in the transgenic crop breeding and other relevant evaluated works,the authors in the present work tried to assess the differences of spectral parameters of the transgenic rice in contrast with its parent group quantitatively and qualitatively,fulfilling the growth monitoring of the transgenic samples.The spectral parameters(spectral morphological characteristics and indices) chosen are highly related to internal or external stresses to the receipts,and thus could be applied as indicators of biophysical or biochemical processes changes of plant.By ASD portable field spectroradiometer with high-density probe,fine foliar spectra of 8 groups were obtained.By analyzing spectral angle and continuum removal,the spectral morphological differences and their locations of sample spectra were found which could be as auxiliary priori knowledge for quantitative analysis.By investigating spectral indices of the samples,the quantitative differences of spectra were revealed about foliar chlorophyll a+b and carotenoid content.In this study both the spectral differences between transgenic and parent groups and among transgenic groups were investigated.The results show that hyperspectral technique is promising and a helpful auxiliary tool in the study of monitoring the transgenic crop and other relevant researches.By this technique,quantitative and qualitative results of sample spectra could be provided as prior knowledge,as certain orientation,for laboratory professional advanced transgenic breeding study.展开更多
A lysozyme gene resistant to rice blast was transferred from the donor transgenic japonica rice Zhonghua 9 (D2-1-2) into a sterile line Pei'ai 64S(PA 64S) and restorer line 9311 of the two-line hybrid rice Liangy...A lysozyme gene resistant to rice blast was transferred from the donor transgenic japonica rice Zhonghua 9 (D2-1-2) into a sterile line Pei'ai 64S(PA 64S) and restorer line 9311 of the two-line hybrid rice Liangyoupeijiu, and the restorer line Minghui 63 (MH 63) of three-line hybrid rice Shanyou 63 by successive backcrossing. The PCR analysis confirmed that foreign lysozyme gene was segregated at ratio of 1 : 1 in backcross generations of B39311, B3MH63 and B2PA64S, and at ratio of 3 : 1 in selfed generations of B2F2 9311, B2F2 MH63 and B1F2 PA64S, indicating that the foreign gene was stably inherited over successive generations as a dominant single copy gene. The resistance against rice blast in backcross or selfed generations and corresponding testcross combinations were investigated in 2003 and 2004. The results showed that the resistance of the transgenic rice to blast had a greater improvement than that of the corresponding recurrent parents or the corresponding check hybrid combinations. The resistance of the advanced backcross and selfed generations to rice blast is much stronger than that of the early generations. The study confirmed that transferring the lysozyme gene into hybrid parents by backcrossing was a simple and effective approach to develop new hybrid rice resistant to rice blast.展开更多
This study aimed to investigate the effects of treatments of rice pollen with different solvents on the seed setting rate.Rice pollen was treated with ultrapure water,0.5%dimethyl sulfoxide,ethanol(1%,5%,and 8%),and 0...This study aimed to investigate the effects of treatments of rice pollen with different solvents on the seed setting rate.Rice pollen was treated with ultrapure water,0.5%dimethyl sulfoxide,ethanol(1%,5%,and 8%),and 0.5%dimethyl sulfoxide+8%ethanol,and the pollen without treatment was taken as the control.Then,crossing was conducted with the treated rice plants as the male parents and plants subjected to emasculation by warm water as the female parents.The seed setting rates of the male and female parents were then recorded.The results showed that the treatments with different solvents significantly decreased the seed setting rates of both male and female parents.The treatment with 1%ethanol resulted in the highest seed setting rates for both male and female parents,while the treatment with 0.5%dimethyl sulfoxide resulted in the lowest seed setting rates for both parents.Although these solvents affected the seed setting rate of rice,they served as intermediate solvents to dissolve chemicals used in the induction of rice haploids.This study layed a technical foundation for subsequent chemical induction of rice haploids.展开更多
