Dissecting Causal Relationships Between Plasma Metabolites and Osteoporosis:A Bidirectional Mendelian Randomization Study

Objective To investigate the causal relationships between plasma metabolites and osteoporosis via Mendelian randomization(MR) analysis.Methods Bidirectional MR was used to analyze pooled data from different genome-wide association studies(GWAS). The causal effect of plasma metabolites on osteoporosis was estimated using the inverse variance weighted method, intersections of statistically significant metabolites obtained from different sources of osteoporosis-related GWAS aggregated data was determined, and then sensitivity analysis was performed on these metabolites. Heterogeneity between single nucleotide polymorphisms was evaluated by Cochran's Q test. Horizontal pleiotropy was assessed through the application of the MR-Egger intercept method and the MRPRESSO method. The causal effect of osteoporosis on plasma metabolites was also evaluated using the inverse variance weighted method. Additionally, pathway analysis was conducted to identify potential metabolic pathways involved in the regulation of osteoporosis.Results Primary analysis and sensitivity analysis showed that 77 and 61 plasma metabolites had a causal relationship with osteoporosis from the GWAS data in the GCST90038656 and GCST90044600 datasets, respectively. Five common metabolites were identified via intersection. X-13684 levels and the glucose-to-maltose ratio were negatively associated with osteoporosis, whereas glycoursodeoxycholate levels and arachidoylcarnitine(C20) levels were positively associated with osteoporosis(all P < 0.05). The relationship between X-11299 levels and osteoporosis showed contradictory results(all P < 0.05). Pathway analysis indicated that glycine, serine, and threonine metabolism, valine, leucine, and isoleucine biosynthesis, galactose metabolism, arginine biosynthesis, and starch and sucrose metabolism pathways were participated in the development of osteoporosis.Conclusion We found a causal relationship between plasma metabolites and osteoporosis. These results offer novel perspectives with important implications for targeted metabolite-focused interventions in the management of osteoporosis. 展开更多

The tea plant(Camellia sinensis)is rich in polyphenolic compounds.Particularly,flavan-3-ols and proanthocyanidins(PAs)are essential for the flavor and disease-resistance property of tea leaves.The fifth subgroup of R2R3-MYB transcription factors comprises the primary activators of PA biosynthesis.This study showed that subgroup 5 R2R3-MYBs in tea plants contained at least nine genes belonging to the TT2,MYB5,and MYBPA types.Tannin-rich plants showed an expansion in the number of subgroup 5 R2R3-MYB genes compared with other dicotyledonous and monocot plants.The MYBPA-type genes of tea plant were slightly expanded.qRT–PCR analysis and GUS staining analysis of promoter activity under a series of treatments revealed the differential responses of CsMYB5s to biotic and abiotic stresses.In particular,CsMYB5a,CsMYB5b,and CsMYB5e responded to high-intensity light,high temperature,MeJA,and mechanical wounding,whereas CsMYB5f and CsMYB5g were only induced by wounding.Three genetic transformation systems(C.sinensis,Nicotiana tabacum,and Arabidopsis thaliana)were used to verify the biological function of CsMYB5s.The results show that CsMYB5a,CsMYB5b,and CsMYB5e could promote the gene expression of CsLAR and CsANR.However,CsMYB5f and CsMYB5g could only upregulate the gene expression of CsLAR but not CsANR.A series of site-directed mutation and domain-swapping experiments were used to verify functional domains and key amino acids of CsMYB5s responsible for the regulation of PA biosynthesis.This study aimed to provide insight into the induced expression and functional diversity model of PA biosynthesis regulation in tea plants. 展开更多

High-quality wood products and valuable wood crafts receive everyone’s favor with the rapid development of the economy.In order to improve the cutting surface quality of wood forming parts,the cutting experiment of renewable Fraxinus mandshurica was conducted by waterjet-assisted CO_(2)laser(WACL)technology.A quadratic mathematical model for describing the relationship between surface roughness changes and cutting parameters was established.The effects of cutting speed,flow pressure and laser power on the kerf surface roughness of Fraxinus mandshurica when cutting transversally were discussed by response surface method.The experimental results showed that kerf surface roughness decreased under a lower laser power,higher cutting speed and higher flow pressure.When the cutting speed was 30 mm/s,flow pressure was 1.58 MPa and laser power was 45 W,the actual surface roughness of the optimized Fraxinus mandshurica was 2.41μm,and it was in accord with the theoretically predicted surface roughness value of 2.54μm,so the model fitted the actual situation well.Through the analysis of 3D profile morphology and micromorphology,it was concluded that the optimized kerf surface of Fraxinus mandshurica was smoother,the cell wall was not destroyed and the tracheid was clear.It provides the theoretical basis for wood micromachining. 展开更多

AIM To evaluate diagnostic yields of capsule endoscopy(CE) and/or single-balloon enteroscopy(SBE) in patients with suspected small bowel diseases.METHODS Were trospectively analyzed 700 patients with suspected small bowel diseases from September 2010 to March 2016. CE, SBE, or SBE with prior CE was performed in 401, 353, and 47 patients, respectively. Data from clinical and endoscopy records were collected for analysis. Indications, procedure times, diagnostic yields, and complications were summarized and evaluated.RESULTS The overall diagnostic yield for the CE group was 57.6%. The diagnostic yield of CE in patients with obscure gastroin testinal bleeding(OGIB) was significantly greater than that in patients with no bleeding(70.5% vs 43.8%, P < 0.01). The overall diagnostic yield of SBE was 69.7%. There was no difference in the diagnostic yield of SBE between patients with OGIB and those with no bleeding(72.5% vs 68.9%, P = 0.534). Forty-seven patients underwent CE prior to SBE. Among them, the diagnostic yield of SBE with positive findings on prior CE was 93.3%. In addition, SBE detected two cases with superficial ulcer and erosive lesions in the small bowel, which were missed by CE. However, one case with lymphoma and two with Crohn's disease were not confirmed by SBE. The rate of capsule retention was 2.0%. There were no significant complications during or after SBE examinations.CONCLUSION SBE is a safe and effective technique for diagnosing small bowel diseases. SBE with prior CE seemed to improve the diagnostic yield of small bowel diseases. 展开更多

As a natural and environmentally friendly renewable material,Northeast China ash wood(NCAW)(Fraxinus mandshurica Rupr.)was cut by water-jet assisted CO_(2) laser(WACL),the surface quality was evaluated by surface roughness of cut section.The surface roughness was measured by three-dimensional(3D)profilometry.Furthermore,the micromorphology of machined surface was observed by scanning electronic microscopy(SEM).Carbon content changes of machined surface were measured by energy dispersive spectrometer(EDS).A relationship between surface roughness and cutting parameters was established using response surface methodology(RSM).It is concluded that the cutting speed,laser power and water pressure played an important role in surface roughness of cut section.The surface roughness increased as an increase in laser power.It decreased caused by increasing of cutting speed and water pressure.Measurements revealed that the surface quality of NCAW part was improved using the optimized combination of cutting parameters.The established quadratic mathematical model of a good prediction is helpful for matching suitable cutting parameters to obtain expected surface quality. 展开更多

Tea(Camellia sinensis)is a well-known beverage crop rich in polyphenols with health benefits for humans.Understanding how tea polyphenols participate in plant resistance is beneficial to breeding resistant varieties and uncovering the resistance mechanisms.Here,we report that a Colletotrichum infection-induced‘pink ring’symptom appeared outside the lesion,which is highly likely to occur in resistant cultivars.By identifying morphological feature-specific metabolites in the pink ring and their association with disease resistance,and analysis of the association between metabolite and gene expression,the study revealed that the accumulation of anthocyanin-3-O-galactosides,red phytotoxin compounds resistant to anthracnose,plays a pivotal role in the hypersensitive response surrounding infection sites in tea plants.The results of genetic manipulation showed that the expression of CsF3Ha,CsANSa,CsUGT78A15,CsUGT75L43,and CsMYB113,which are involved in anthocyanin biosynthesis,is positively correlated with anthracnose-resistance and the formation of the pink ring.Further phosphorus quantification and fertilization experiments confirmed that phosphate deficiency caused by anthracnose is involved in the occurrence of the pink ring.Genetic manipulation studies indicated that altering the expression levels of Pi transporter proteins(CsPHT2-1,CsPHT4;4)and phosphate deprivation response transcription factors(CsWRKY75-1,CsWRKY75-2,CsMYB62-1)enhances resistance to anthracnose and the formation of the pink ring symptom in tea plants.This article provides the first evidence that anthocyanin-3-O-galactosides are the anthracnose-resistant phytoalexins among various polyphenols in tea plants,and this presents an approach for identifying resistance genes in tea plants,where genetic transformation is challenging. 展开更多

应用遥感技术监测丹江口水库氨氮分布研究

丹江口水库作为南水北调中线工程的水源地,其水质状况直接影响到受水区经济发展和居民用水安全。本研究针对丹江口水库现状水质监测存在的耗时耗力、成本高、监测点有限等现象,基于遥感技术和智能算法的基本理论,利用高分辨率遥感数据... 丹江口水库作为南水北调中线工程的水源地,其水质状况直接影响到受水区经济发展和居民用水安全。本研究针对丹江口水库现状水质监测存在的耗时耗力、成本高、监测点有限等现象,基于遥感技术和智能算法的基本理论,利用高分辨率遥感数据和实测水质数据研究典型水质参数氨氮的遥感反演技术。将构建的人工神经网络模型应用于丹江口水库,获得2007~2009年丹江口水库氨氮浓度时空分布图。结果表明,人工神经网络模型能有效地反演丹江口水库氨氮的时空分布情况,率定期和验证期的平均相对误差分别为3.59%和19.28%,相较于多元回归模型的8.3%和22.4%而言,人工神经网络模型反演效果更为精准,更能模拟实际情况;研究时段内,丹江口水库水质总体上趋好,氨氮浓度均低于地表水环境质量II类标准限值,丰水期明显高于枯水期。水质遥感反演作为一种非传统性的监测手段,具有空间全覆盖、快速和成本低等传统水质监测难以比拟的优点,能够作为实测数据的有益补充,极具有深入研究和推广的价值和必要。 展开更多

AIM: To compare the effectiveness of needle-free incision suture closure with butterfly tape and traditional secondary suturing techniques in treating incision infection. METHODS: Two hundred and twenty-three patients with incision infection following hepatobiliary surgery at a tertiary hospital were randomly divided into three groups: 90 patients were closed by needle-free incision suture closure, which gradually closed the incision wound when drainage from incision infection was visibly decreased and healthy granulation tissues had grown; 79 patients were closed by butterfly bandage; another 54 patients were closed by traditional secondary suturing technique. Healing time of incision infection was calculated from the beginning of dressing change to the healing of the incision. RESULTS: Healing time in the needle-free incision suture closure group (24.2 +/- 7.2 d) was significantly shorter than that in the butterfly bandage group (33.3 +/- 11.2 d) and the traditional secondary suturing group (36.2 +/- 15.3 d) (P < 0.05). Healing time in the butterfly bandage group appeared to be slightly shorter than that in the secondary suture group, but the difference was not statistically significant (P > 0.05). CONCLUSION: Needle-free incision suture closure could gradually close the infection wound at the same time of drainage and dressing change, thereby shortening the healing time. BACKGROUND The accurate classification of focal liver lesions(FLLs)is essential to properly guide treatment options and predict prognosis.Dynamic contrast-enhanced computed tomography(DCE-CT)is still the cornerstone in the exact classification of FLLs due to its noninvasive nature,high scanning speed,and high-density resolution.Since their recent development,convolutional neural network-based deep learning techniques has been recognized to have high potential for image recognition tasks.AIM To develop and evaluate an automated multiphase convolutional dense network(MP-CDN)to classify FLLs on multiphase CT.METHODS A total of 517 FLLs scanned on a 320-detector CT scanner using a four-phase DCECT imaging protocol(including precontrast phase,arterial phase,portal venous phase,and delayed phase)from 2012 to 2017 were retrospectively enrolled.FLLs were classified into four categories:Category A,hepatocellular carcinoma(HCC);category B,liver metastases;category C,benign non-inflammatory FLLs including hemangiomas,focal nodular hyperplasias and adenomas;and category D,hepatic abscesses.Each category was split into a training set and test set in an approximate 8:2 ratio.An MP-CDN classifier with a sequential input of the fourphase CT images was developed to automatically classify FLLs.The classification performance of the model was evaluated on the test set;the accuracy and specificity were calculated from the confusion matrix,and the area under the receiver operating characteristic curve(AUC)was calculated from the SoftMax probability outputted from the last layer of the MP-CDN.RESULTS A total of 410 FLLs were used for training and 107 FLLs were used for testing.The mean classification accuracy of the test set was 81.3%(87/107).The accuracy/specificity of distinguishing each category from the others were 0.916/0.964,0.925/0.905,0.860/0.918,and 0.925/0.963 for HCC,metastases,benign non-inflammatory FLLs,and abscesses on the test set,respectively.The AUC(95%confidence interval)for differentiating each category from the others was 0.92(0.837-0.992),0.99(0.967-1.00),0.88(0.795-0.955)and 0.96(0.914-0.996)for HCC,metastases,benign non-inflammatory FLLs,and abscesses on the test set,respectively.CONCLUSION MP-CDN accurately classified FLLs detected on four-phase CT as HCC,metastases,benign non-inflammatory FLLs and hepatic abscesses and may assist radiologists in identifying the different types of FLLs. 展开更多

ODVs are released from occlusion bodies in the midgut where they initiate a primary infection. Due to the lack of an in vitro system, the molecular mechanism of ODV infection is still unclear. Here we present data demonstrating that Helicoverpa armigera nucleopolyhedrovirus (HearNPV) ODV infected cultured Hz-AM1 cells in a pH dependent manner. The optimal pH for ODV infection was 8.5, which is same to that in the microvilli of midgut epithelial cells, the ODV native infection sites. Antibodies neutralization analysis indicated that four HearNPV oral infection essential genes p74, pif-l, pif-2 and pif-3 are also essential for HearNPV ODV infection in vitro. Thus, HearNPV-HzAM1 system can be used to analyze the mechanism of ODV entry. 展开更多

Renal aspergillosis(RAsp)is a rare complication in liver transplant(LT)recipients.Here we report RAsp in two LT recipients.In both patients,RAsp occurred more than 90 d after allogenetic orthotropic LT,and all the clinical findings were unspecific.RAsp involved unilateral kidney in Case one and bilateral kidneys in Case two.Both computed tomography(CT)and magnetic resonance imaging(MRI)revealed renal abscesses,with progressively enhanced walls and separationsand unenhanced alveolate areas after contrast agent administration.On unenhanced CT images they showed inhomogeneous hypo-attenuation.On fat-suppressed T2-weighted images(T2WIs),the walls and separations of the abscesses showed slightly low signal intensity and the central parts of the lesions showed slightly high signal intensity.Both on CT and MRI,there were some hints of renal infarction or chronic ischemia.Both cases were treated by radical nephrectomy followed by adjuvant antifungal treatment.They all recovered well. 展开更多

BACKGROUND: The high recurrence rate of hepatolithiasis and the high operative risk of right posterior, caudate or multiple lobe hepatectomy are the unsettled problems in hepatobiliary surgery. The present study was to investigate the efficacy of chemical hepatectomy performed via applying sequential embolization of the branches of the bile duct and portal vein to the targeted hepatic lobe. METHODS: The bile duct and portal vein branches of the median hepatic lobe of rats were treated with: 1) bile duct embolization followed by portal vein ligation(BDE+PVL) and 2) portal vein ligation followed by bile duct embolization(PVL+BDE). The efficacy of chemical hepatectomy in BDE+PVL and PVL+BDE groups was compared with that of sole BDE by histology and Western blotting analysis of collagen I expression. RESULTS: After six weeks of the chemical hepatectomy, rats in the BDE group showed hepatocyte damages, fibrosis and 'selfcut' only in the periphery of the embolized lobe. In contrast, rats in the PVL+BDE and BDE+PVL groups exhibited complete necrosis of hepatocytes and replacement with proliferative ductules and collagen fibers, leading to complete fibrosis and 'self-cut' phenomenon in the whole targeted lobe. Collagen I expression in the PVL+BDE group was slightly higher than that in the BDE+PVL group; however, no statistically significant difference was noted. CONCLUSION: The sequential embolization of the bile duct and portal vein branches to the targeted hepatic lobe may bea feasible and effective approach to acheive the ideal effect of chemical hepatectomy in a short period of time. 展开更多

In order to enhance the strength of sustainable walnut shell/Co-PES(WSPC)sintered parts,wax-filtrated posttreatment was carried out.The effects of treating fluid temperature,preheating time and immersion time on the bending strength of WSPC wax-filtrated parts were analyzed by single factor analysis method.To obtain an accurate model for predicting the bending strength of the WSPC wax-filtrated part,the experiments were involved by using Box-Behnken design(BBD).Main parameters,such as treating fluid temperature,preheating time and immersion time,and their interactive effects were analyzed through analysis of variance(ANOVA)and graphical contours.The results demonstrated that all parameters’direct effects were significant to bending strength of the WSPC wax-filtrated part.Its optimum value was 5.0 MPa when the treating fluid temperature of 70°C,preheating time of 50 min,and immersion time of 20 s.The predicted models effectively validated had good predicting accuracy.The WSPC wax-filtrated part using optimal processing parameters was processed by investment casting,and then the metal casting of dimensional stability and smooth surface was obtained.Investment casting was done using WSPC wax-filtrated parts under optimal process parameters and then metal parts with stable structure size and smooth surface can be obtained,which indicates that WSPC material can be used for investment casting. 展开更多

Aiming at the problem of music noise introduced by classical spectral subtraction,a shorttime modulation domain(STM)spectral subtraction method has been successfully applied for singlechannel speech enhancement.However,due to the inaccurate voice activity detection(VAD),the residual music noise and enhanced performance still need to be further improved,especially in the low signal to noise ratio(SNR)scenarios.To address this issue,an improved frame iterative spectral subtraction in the STM domain(IMModSSub)is proposed.More specifically,with the inter-frame correlation,the noise subtraction is directly applied to handle the noisy signal for each frame in the STM domain.Then,the noisy signal is classified into speech or silence frames based on a predefined threshold of segmented SNR.With these classification results,a corresponding mask function is developed for noisy speech after noise subtraction.Finally,exploiting the increased sparsity of speech signal in the modulation domain,the orthogonal matching pursuit(OMP)technique is employed to the speech frames for improving the speech quality and intelligibility.The effectiveness of the proposed method is evaluated with three types of noise,including white noise,pink noise,and hfchannel noise.The obtained results show that the proposed method outperforms some established baselines at lower SNRs(-5 to +5 dB). 展开更多

To overcome the limitations of conventional speech enhancement methods, such as inaccurate voice activity detector(VAD) and noise estimation, a novel speech enhancement algorithm based on the approximate message passing(AMP) is adopted. AMP exploits the difference between speech and noise sparsity to remove or mute the noise from the corrupted speech. The AMP algorithm is adopted to reconstruct the clean speech efficiently for speech enhancement. More specifically, the prior probability distribution of speech sparsity coefficient is characterized by Gaussian-model, and the hyper-parameters of the prior model are excellently learned by expectation maximization(EM) algorithm. We utilize the k-nearest neighbor(k-NN) algorithm to learn the sparsity with the fact that the speech coefficients between adjacent frames are correlated. In addition, computational simulations are used to validate the proposed algorithm, which achieves better speech enhancement performance than other four baseline methods-Wiener filtering, subspace pursuit(SP), distributed sparsity adaptive matching pursuit(DSAMP), and expectation-maximization Gaussian-model approximate message passing(EM-GAMP) under different compression ratios and a wide range of signal to noise ratios(SNRs). 展开更多

BACKGROUND Although several trials have shown that the addition of antifoaming agents to polyethylene glycol(PEG)can improve bowel preparation,whether PEG plus antifoaming agents have a beneficial role in the detection of benign tumors during colonoscopy has yet to be confirmed.Our aim was to clarify whether adding simethicone to PEG solution could improve the detection of benign colorectal tumors.AIM To clarify whether adding simethicone to PEG solution could improve the detection of benign colorectal tumors.METHODS The PubMed,EMBASE,and Cochrane Library databases were searched for articles published prior to September 2019.The outcomes included the detection rates of colorectal adenomas and polyps.RESULT Twenty studies were eligible.Although there was no difference in the colorectal adenoma detection rate(ADR),a significant effect of simethicone for diminutive adenomas(<10 mm)was revealed in the group taking simethicone.We also found that simethicone could significantly improve the ADR in the proximal colon but did not affect the colorectal polyp detection rate.Furthermore,the subgroup analyses revealed a beneficial effect of simethicone on the ADR among Asians(P=0.005)and those with an ADR<25%(P=0.003).Moreover,it was a significant finding that the low dose simethicone was as effective as the high dose one with respect to the detection of benign colorectal tumors.CONCLUSION In summary,the addition of simethicone to PEG might improve the detection of diminutive adenomas in the right colon by colonoscopy in Asia.Low-dose simethicone was recommended for the detection of benign colorectal tumors.However,large clinical trials are necessary to validate our results and determine the ideal dose of simethicone. 展开更多

In this study, DERB1A transcription factor and stress-induced promoter rd29A were isolated respectively and amplified from Arabidopsis thaliana, se- quenced and analyzed by DNAsis. In addition, the stress-induced promoter rd29A was utilized to construct the plant expression vector of DERB1A, which was transformed into Agrobacterium tumefaciens. Furthermore, the transgenic regeneration system of fresh-cut chrysanthemum from callus to plantlets was established successfully. On this basis, chrysanthemum leaf-disc explants were genetically transformed with Agrobacterium-mediated method. Two positive transgenie plantlets were obtained in vitro. Based on PCR detection, DREB1A transcription factor was integrated into chrysanthemum genome, which laid the foundation for breeding new transgenie cultivars of fresh-cut chrysanthemum with high comprehensive stress resistance, good cmalitv and high field. 展开更多
