本研究采用网络药理学方法探讨怀膝颗粒治疗骨质疏松症的作用机制。通过中药系统药理学分析平台(Traditional Chinese Medicine System Pharmacology,TCMSP)、中国科学院上海有机化学研究所化学专业数据库等筛选和收集骨碎补和山茱萸等...本研究采用网络药理学方法探讨怀膝颗粒治疗骨质疏松症的作用机制。通过中药系统药理学分析平台(Traditional Chinese Medicine System Pharmacology,TCMSP)、中国科学院上海有机化学研究所化学专业数据库等筛选和收集骨碎补和山茱萸等5味中药的有效成分和作用靶点,并导入Cytoscape软件,构建药材活性成分-靶点网络图。通过TTD数据库、GeneCards数据库和OMIM数据库获得骨质疏松症靶点,再取怀膝颗粒和骨质疏松症的交集靶点,进行蛋白质相互作用网络关系图的构建并通过R语言对其进行GO富集分析和KEGG通路分析。结果收集到活性成分骨碎补15个、山茱萸14个、牛膝17个、川芎6个和鹿茸2个。怀膝颗粒与骨质疏松症相关的交集靶点61个,其中雄激素受体(androgen receptor,AR)、肿瘤坏死因子(tumor necrosis factor,TNF)和白细胞介素-6(Interleukin-6,IL-6)为核心靶点蛋白。GO富集分析和KEGG分析提示怀膝颗粒可能作用于乙型肝炎信号通路(Hepatitis B)、丙型肝炎信号通路(Hepatitis C)和C型凝集素受体信号通路(C-type lectin receptor signaling pathway)并通过在膜区域(membrane region)上抑制类固醇结合(steroidbinding)来治疗骨质疏松症。总之,怀膝颗粒治疗骨质疏松症具有多靶点、多途径和多通路的特点,可为中药治疗骨质疏松症提供新思路。展开更多
