BACKGROUND Thrombocytopenia 2,an autosomal dominant inherited disease characterized by moderate thrombocytopenia,predisposition to myeloid malignancies and normal platelet size and function,can be caused by 5’-untranslated region(UTR)point mutations in ankyrin repeat domain containing 26(ANKRD26).Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1)have been identified as negative regulators of ANKRD26.However,the positive regulators of ANKRD26 are still unknown.AIM To prove the positive regulatory effect of GATA binding protein 2(GATA2)on ANKRD26 transcription.METHODS Human induced pluripotent stem cells derived from bone marrow(hiPSC-BM)INTRODUCTION Ankyrin repeat domain containing protein 26(ANKRD26)acts as a regulator of adipogenesis and is involved in the regulation of feeding behavior[1-3].The ANKRD26 gene is located on chromosome 10 and shares regions of homology with the primate-specific gene family POTE.According to the Human Protein Atlas database,the ANKRD26 protein is localized to the Golgi apparatus and vesicles,and its expression can be detected in nearly all human tissues[4].Moreover,UniProt annotation revealed that ANKRD26 is localized in the centrosome and contains coiled-coil domains formed by spectrin helices and ankyrin repeats[5,6].The most common disease related to ANKRD26 is thrombocytopenia 2(THC2),which is a rare autosomal dominant inherited disease characterized by lifelong mild-to-moderate thrombocytopenia and mild bleeding[7-9].Caused by the variants in the 5’-untranslated region(UTR)of ANKRD26,THC2 is defined by a decrease in the number of platelets in circulating blood and results in increased bleeding and decreased clotting ability[8,10].Due to the point mutations that occur in the 5’-UTR of ANKRD26,its negative transcription factors(TFs),Runt related transcription factor 1(RUNX1)and friend leukemia integration 1(FLI1),lose their repression effect[11].The persistent expression of ANKRD26 increases the activity of the mitogen activated protein kinase and extracellular signal regulated kinase 1/2 signaling pathways,which are potentially involved in the regulation of thrombopoietin-dependent signaling and further impair proplatelet formation by megakaryocytes(MKs)[11].However,the positive regulators of ANKRD26,which might be associated with THC2 pathology,are still unknown. 展开更多

The pyrolysis of cyclohexane,methylcyclohexane,and ethylcyclohexane have been studied behind reflected shock waves at pressures of 5 and10 bar and at temperatures of 930-1550 K for 0.05%fuel diluted by Argon.A single-pulse shock tube(SPST)is used to perform the pyrolysis experiments at reaction times varying from 1.65 to 1.74 ms.Major products are obtained and quantified using gas chromatography analysis.A flame ionization detector and a thermal conductivity detector are used for species identification and quantification.Kinetic modeling has been performed using several detailed and lumped chemical kinetic mechanisms.Differences in modeling results among the kinetic models are described.Reaction path analysis and sensitivity analysis are performed to determine the important reactions controlling fuel pyrolysis and their influence on the predicted concentrations of reactant and product species profiles.The present work provides new fundamental knowledge in understating pyrolysis characteristics of cyclohexane compounds and additional data set for detailed kinetic mechanism development. 展开更多

Chinese soft-shelled turtles (Trionyx sinens) in culture farms using an artificial warming system in Zhejiang, China, often show typical signs of white-spot disease such as white spots on their bodies, skin lesions, anorexia and eventually death. The sick turtles were mostly 5-80 g in weight. A suspected fungal pathogen was isolated from the sick turtles and verified as Paecilomyces lilacinus by sequence analysis of the internal transcribed spacer (ITS) of its ribosomal DNA (rDNA). Detailed morphological examinations were also conducted to confirm the white-spot disease. 展开更多

The energy and trajectory of the electron, which is irradiated by a high-power laser pulse in a cylindrical plasma channel with a uniform positive charge and a uniform negative current, have been analyzed in terms of a single-electron model of direct laser acceleration. We find that the energy and trajectory of the electron strongly depend on the positive charge density, the negative current density, and the intensity of the laser pulse. The electron can be accelerated significantly only when the positive charge density, the negative current density, and the intensity of the laser pulse are in suitable ranges due to the dephasing rate between the wave and electron motion. Particularly, when their values satisfy a critical condition. the electron can stay in phase with the laser and gain the largest energy from the laser. With the enhancement of the electron energy, strong modulations of the relativistic factor cause a considerable enhancement of the electron transverse oscillations across the channel, which makes the electron trajectory become essentially three-dimensional, even if it is flat at the early stage of the acceleration. 展开更多

Objective:Based on the quality analysis of Nauclea officinalis in different growth years and different months in the same year,the most suitable harvest year and the specific month in the same year were determined according to the evaluation of the active components in the stem of medicinal parts of Nauclea officinalis.Methods:LC-MS/MS method was used to investigate the quality of Nauclea officinalis from different growth years and different months in the same year,mainly including the content determination of the active ingredients in the medicine.Results:With the prolongation of production life,the content of active ingredients in stem of Nauclea officinalis increased continuously.After reaching a certain number of years,the content of active ingredients increased little;In the middle of the same year,from mid-October to mid-June of the next year,the content of iso-vincin lactam was relatively high,especially between the end of March and the end of May.The content was high and stable,and the highest point was the stem sample collected in May.Conclusion:The content of each component in the stem of Nauclea officinalis with a growth life of more than 5 years has reached a relatively high and stable level.Nauclea officinalis with a growth life of more than 5 years should be collected as raw materials for medicinal materials.The month with high and stable content of isocyanoside lactam,namely May of each year,was taken as the best harvest time of gallbladder in one year.The best harvest time was from mid-October to mid-June of the next year,or from March to May of each year. 展开更多

Colloidal quantum dots(CQDs)are of great interest to photovoltaic(PV)technologies as they possess the benefits of solution-processability,size-tunability,and roll-to-roll manufacturability,as well as unique capabilities to harvest near-infrared(NIR)radiation.During the last decade,lab-scale CQD solar cells have achieved rapid improvement in the power conversion efficiency(PCE)from~1%to 18%,which will potentially exceed 20%in the next few years and approach the performance of other PV technologies,such as perovskite solar cells and organic solar cells.In the meanwhile,CQD solar cells exhibit long lifetimes either under shelf storage or continuous operation,making them highly attractive to industry.However,in order to meet the industrial requirements,mass production techniques are necessary to scale up the fabrication of those lab devices into large-area PV modules,such as roll-to-toll coating.This paper reviews the recent developments of large-area CQD solar cells with a focus on various fabrication methods and their principles.It covers the progress of typical large-area coating techniques,including spray coating,blade coating,dip coating,and slot-die coating.It also discusses next steps and new strategies to accomplish the ultimate goal of the low-cost large-area fabrication of CQD solar cells and emphasizes how artificial intelligence or machine learning could facilitate the developments of CQD solar cell research. 展开更多

Porcine circovirus 3(PCV3)has been detected in major pig-producing countries around the world since its first report in the US in 2016.Most current studies have focused on epidemiological investigations and detection methods of PCV3 because of lack of live virus strains for research on its pathogenesis in porcine cells or even in pigs.We constructed a recombinant plasmid pCMV-Cap carrying the PCV3 orf2 gene to investigate the effects of capsid(Cap)protein expression on autophagic response in human embryonic kidney cell line 293 T(HEK293 T).We demonstrate that PCV3 Cap protein induced complete autophagy shown as formation of autophagosomes and autophagosome-like vesicles as well as LC3-II conversion from LC3-I via inhibiting phosphorylation of the mammalian target of rapamycin(mTOR)in HEK293 T cells.The ubiquitin–proteasome pathway is also involved in the autophagy process.These findings provide insight for further exploration of PCV3 pathogenetic mechanisms in porcine cells. 展开更多

Congenital bilateral absence of the vas deferens(CBAVD)is observed in 1%–2%of males presenting with infertility and is clearly associated with cystic fibrosis transmembrane conductance regulator(CFTR)mutations.CFTR is one of the most well-known genes related to male fertility.The frequency of CFTR mutations or impaired CFTR expression is increased in men with nonobstructive azoospermia(NOA).CFTR mutations are highly polymorphic and have established ethnic specificity.Compared with F508Del in Caucasians,the p.G970D mutation is reported to be the most frequent CFTR mutation in Chinese patients with cystic fibrosis.However,whether p.G970D participates in male infertility remains unknown.Herein,a loss-of-function CFTR p.G970D missense mutation was identified in a patient with CBAVD and NOA.Subsequent retrospective analysis of 122 Chinese patients with CBAVD showed that the mutation is a common pathogenic mutation(4.1%,5/122),excluding polymorphic sites.Furthermore,we generated model cell lines derived from mouse testes harboring the homozygous Cftr p.G965D mutation equivalent to the CFTR variant in patients.The Cftr p.G965D mutation may be lethal in spermatogonial stem cells and spermatogonia and affect the proliferation of spermatocytes and Sertoli cells.In spermatocyte GC-2(spd)ts(GC2)Cftr p.G965D cells,RNA splicing variants were detected and CFTR expression decreased,which may contribute to the phenotypes associated with impaired spermatogenesis.Thus,this study indicated that the CFTR p.G970D missense mutation might be a pathogenic mutation for CBAVD in Chinese males and associated with impaired spermatogenesis by affecting the proliferation of germ cells. 展开更多

A one-step dual flow immunochromatographic assay (DICGA), based on a competitive format, was de- veloped for simultaneous quantification of ochratoxin A (OTA) and zearalenone (ZEN) in corn, wheat, and feed sam- ples. The limit of detection for OTA was 0.32 ng/ml with a detection range of 0.53-12.16 ng/ml, while for ZEN it was 0.58 ng/ml with a detection range of 1.06-39.72 ng/ml. The recovery rates in corn, wheat, and feed samples ranged from 77.3% to 106.3% with the coefficient of variation lower than 15%. Naturally contaminated corn, wheat, and feed samples were analyzed using both DICGA and liquid chromatography-tandem mass spectrometry (LC-MS/MS) and the correlation between the two methods was evaluated using a regression analysis. The DICGA method shows great potential for simple, rapid, sensitive, and cost-effective quantitative detection of OTA and ZEN in food safety control. 展开更多

Porcine epidemic diarrhea virus(PEDV) is a highly infectious pathogen that can cause severe diseases in pigs and result in enormous economic losses in the worldwide swine industry. Previous studies revealed that PEDV exhibits an obvious capacity for modulating interferon(IFN) signaling or expression. The newly discovered type III IFN, which plays a crucial role in antiviral immunity, has strong antiviral activity against PEDV proliferation in IPEC-J2 cells. In this study, we aimed to investigate the effect of PEDV nucleocapsid(N) protein on type III IFN-λ. We found that the N proteins of ten PEDV strains isolated between 2013 and 2017 from different local farms shared high nucleotide identities, while the N protein of the CV777 vaccine strain formed a monophyletic branch in the phylogenetic tree. The N protein of the epidemic strain could antagonize type III IFN, but not type I or type II IFN expression induced by polyinosinic-polycytidylic acid(poly(I:C)) in IPEC-J2 cells. Subsequently, we demonstrated that the inhibition of poly(I:C)-induced IFN-λ3 production by PEDV N protein was dependent on the blocking of nuclear factor-κB(NF-κB) nuclear translocation. These findings might help increase understanding of the pathogenesis of PEDV and its mechanisms for evading the host immune response. 展开更多

Porcine circovirus type 2(PCV2)has recently been reported to elicit the unfolded protein response(UPR)via activation of the PERK/e IF2α(RNA-activated protein kinase-like endoplasmic reticulum(ER)kinase/eukaryotic initiation factor 2α)pathway.This study attempted to examine which viral protein might be involved in inducing UPR and whether this cellular event would lead to apoptosis of the cells expressing the viral protein.By transient expression,we found that both replicase(Rep)and capsid(Cap)proteins of PCV2 could induce ER stress as shown by increased phosphorylation of PERK with subsequent activation of the eI F2α-ATF4(activating transcription factor 4)-CHOP(CCAAT/enhancer-binding protein homologous protein)axis.Cap expression,but not Rep,significantly reduced antiapoptotic B-cell lymphoma-2(Bcl-2)and increased caspase-3 cleavage,possibly due to increased expression of CHOP.Since knockdown of PERK by RNA interference clearly reduced Cap-induced CHOP expression,caspase-3cleavage,and apoptotic cell death possibly by partially rescuing Bcl-2 expression,we propose that there is connection between Cap-induced UPR and apoptosis via the PERK/eI F2α/ATF4/CHOP/Bcl-2 pathway.This study,together with our earlier studies,provides insight into the mechanisms underlying PCV2 pathogenesis. 展开更多

The heat shock cognate protein 70 (Hsc70) is a member of a 70-kDa heat shock protein (HSP70) family that functions as molecular chaperones.In this study,a novel Hsc70 gene from Chinese soft-shelled turtle (Pelodiscus sinensis) (tHsc70) was identified.The tHsc70 full-length complementary DNA (cDNA) is 2 272 bp long with a 1 941-bp open reading frame (ORF) encoding 646 amino acids.Three characteristic signature regions of the HSP70 family,two major domains of an adenosine triphosphate (ATP)/guanosine triphosphate (GTP) binding domain (ABD),and a substrate-binding domain (SBD) were present in the predicted tHsc70 amino acid sequence.The tHsc70 gene was expressed in Escherichia coli BL21 and the expression product reacted with the anti-Hsc70 mouse monoclonal antibody by Western blotting.Homology analysis revealed that tHsc70 shared identity from 53.9% to 87.7% at the nucleotide level,and 49.1% to 99.5% at the amino acid level with the known Hsc70s.Phylogenetic analysis showed that tHsc70 was clustered together with the Hsc70 gene of another reptile species (Alligator mississippiensis).The tHsc70 was expressed in the liver,lung,heart,and skeletal muscle.The expression patterns of tHsc70 messenger RNA (mRNA) differed among different tissues under different durations of heat stress at 40 °C.Adaptation at 25 °C for 1 h after heat stress was also different among tissues and length of heat stress.Irrespective of different profiles of expression under heat stress,tHsc70 may play roles in protecting turtles from thermal stress. 展开更多

Objective:To investigate the relationship between the concentration of L-carnitine in semen and sperm parameters and investigate the epigenetic profile in sperm cell after L-carnitine usage.Methods:From February 2017 to February 2018,46 semen samples from asthenospermic males and 41 semen samples from healthy donors were acquired.Motility parameters were assessed using computer-assisted sperm analysis(CASA,n=78)and the DNA fragmentation index(DFI)was evaluated through flow cytometry(n=86),%DFI=%cells outside main population.Other oxidative stress markers,such as reactive oxygen species(ROS)levels(n=86)and the mitochondria DNA copy numbers,were detected(n=78).The concentration of L-carnitine and acetyl-L-carnitine was detected(n=82),and methylation was analyzed(n=30).After that,we collected 13 fresh semen samples from asthenospermic males and 23 fresh semen samples from healthy donors.These samples were used in a freeze-thaw model that was used to determine whether adding L-carnitine could change sperm progressive motility(n=23),apoptosis index(n=9),and methylation analysis(n=7).In total,we have done 13 asthenospermia samples for Western blot,and except for the poor Western result,we analyzed 6 samples for H3K9ac detection,7 samples for H3K9m3 and H3K27m3 detection,and immunofluorescence(n=3).Finally,we had recruited 30 volunteers,and they were given oral administration of L-carnitine for 3 months and then collected semen samples at different time points for methylation analysis.Results:The concentration of acetyl-L-carnitine is negatively correlated with the%DFI value(r^2=0.1090;P=0.0026),and the concentration of acetyl-L-carnitine is positively correlated with sperm forward motility(r^2=0.0543;P=0.0458)and ROS(r^2=0.1854;P<0.0001),and the acetyl-L-carnitine level is negatively correlated with%DFI in asthenospermia(r^2=0.1701;P=0.0066),and the level of acetyl-L-carnitine in asthenospermic semen is significantly lower than the normal group(P=0.0419).In addition,this study indicates that adding L-carnitine significantly improved sperm motility(P=0.0325)and reduced sperm apoptosis(P=0.0032).Importantly,Western blotting(P=0.0429)and immunofluorescence staining results showed that the addition of L-carnitine reduced H3K9Me3 methylation level in sperm,respectively.Furthermore,semen samples from asthenospermic patients had reduced methylation levels in a specific region(16^thP=0.0003;17^thP=0.0016)of the brain-derived neurotrophic factor(BDNF)promoter.The 16^th methylation decreased with age(r^2=0.1564;P=0.0306),and the 17^th methylation was decreased after treatment with L-carnitine for 28 days(P=0.0341).Conclusion:L-carnitine can reduce the%DFI and also affect the methylation of the histone modification marker in sperm as a possible epigenetic regulator. 展开更多
