[Methods]The AD model rat was rep...[Objectives] To study the effects and mechanism of notoginsenoside Rg1 on the spatial learning and memory and phosphorylated tau protein in the AD( Alzheimer's Disease) model rat. [Methods]The AD model rat was replicated by injection of Aβ_(25-35) in the left lateral ventricles of SD rats. The low dose( 25 mg/kg),middle dose( 50 mg/kg) and high dose( 100 mg/kg) notoginsenoside Rg1 was used for intragastric administration,respectively,two times every day. After 4 weeks,the Morris water maze test was done to detect the learning and memory capacity,and the immunoblotting,immunohistochemical methods were used to detect the changes in the phosphorylation level and distribution of tau protein in hippocampus of the rats. [Results] After the intracerebroventricular injection of Aβ_(25-35),the learning and memory capacity of the model rats was significantly lower than the learning and memory capacity of the normal control rats. The immunoblotting test results showed that the phosphorylation level of tau protein threonine 231 site( Thr231) in hippocampus was significantly increased,and the nonphosphorylation level was significantly decreased. The morphological testing results showed that the phosphorylation level of tau protein Thr231 of AD model rats was increased markedly in region of DG,CA1 and CA3 of the hippocampus. The intervention of the middle dose notoginsenoside Rg1 could significantly improve the learning and memory capacity of the model rats in Morris water maze. The notoginsenoside Rg1 in three different doses could all reduce the phosphorylation level of tau protein Thr231 in the hippocampal DG,CA1,CA3 regions,and there were no significant differences among the three doses. [Conclusions]The notoginsenoside Rg1 could improve Aβ_(25-35)-induced spatial learning and memory impairment of the AD model rats,and decreased the phosphorylation level of tau protein in hippocampus.展开更多

Methods: DAC and AS2O3 monotherapy, combination treatment...Purpose: Our study was to detect the effect of Decitabine (DAC) combined with arsenic trioxide (AS2O3) on DAPK gene and HL-60 cell proliferation and apoptosis. Methods: DAC and AS2O3 monotherapy, combination treatment and DAC pretreatment were used in this study after incubating with HL-60 cell for 24 h, 48 h, 72 h. CCK8 was used to detect the cell proliferation of HL-60 cell. Flow cytometry was used to detect the cell apoptosis. Then, we used RT-PCR to obtain the gene expression level of DAPK. Results: HL-60 cells were treated with different concentrations of DAC (20 μmol/L, 40 μmol/L, 80 μmol/L), AS2O3 (1 μmol/L, 2.5 μmol/L, 5 μmol/L) monotherapy for 24 h, 48 h, 72 h;along with the extension of the drug concentration and time, proliferation inhibition rate had gradually increased. Monotherapy of DAC, AS2O3 could inhibit the proliferation and induce apoptosis of HL-60 cells, and was time- and dose-dependent. DAC (80 μmol/L) was firstly used for pretreatment, and then, different concentrations of AS2O3 (1 μmol/L, 2.5 μmol/L, 5 μmol/L) were used for 24 h, 48 h, 72 h. It was found that cell proliferation inhibition rate and apoptosis rate had increased significantly. When the two drugs were used together, the increasing proliferation inhibition rate, apoptosis rate and DAPK had become more obvious. Conclusion: DAC and AS2O3 had a synergetic effect for the HL-60 cell proliferation inhibition, apoptosis and expression of DAPK.展开更多

Analysis of expression data of the family of DUF1644- containing genes indicated that they may regulate responses to abiotic stress in rice. Here we present our discovery of the role of Os.SIDP366, a member of the DUF1644 gene family, in response to drought and salinity stresses in rice. Transgenic rice plants overexpressing OsSIDP366 showed enhanced drought and salinity tolerance and reduced water loss as compared to that in the control, whereas plants with downregulated OsSIDP366 expression levels using RNA interference (RNAi) were more sensitive to salinity and drought treatments. The sensitivity to abscisic acid (ABA) treatment was not changed in OsStDP366-overexpressing plants, and OsSIDP366 expression was not affected in ABA- deficient mutants. Subcellular localization analysis revealed that OsSIDP366 is presented in the cytoplasmic foci that colocalized with protein markers for both processing bodies (PBs) and stress granules (SGs) in rice protoplasts. Digital gene expression (DGE) profile analysis indicated that stress-related genes such as SNACl, OsHAK5 and PRs were upregulated in OsSIDP366-overexpressing plants. These results suggest that OsSIDP366 may function as a regulator of the PBs/SGs and positively regulate salt and drought resistance in rice.展开更多

The model incorporates the factors of project divisibility and interdependenc...In this paper, we develop an extended model for the project portfolio selection problem over a planning horizon with multiple time periods. The model incorporates the factors of project divisibility and interdependency at the same time for real-life applications. The project divisibility is considered as a strategy, not an unfortunate event as in the literature, in choosing the best execution schedule for the projects, and the classical concept of"project interdependencies" among fully executed projects is then extended to the portions of executed projects. Additional constraints of reinvestment consideration, setup cost, cardinality restriction, precedence relationship and scheduling are also included in the model. For efficient computations, an equivalent mixed integer linear programming representation of the proposed model is derived. Numerical examples under four scenarios are presented to highlight the characteristics of the proposed model. In particular, the positive effects of project divisibility are shown for the first time.展开更多
OBJECTIVE:To investigate biological indicators of sub-optimal health status and provide means of objective assessment of sub-optimal health status.METHODS:We set the unified standards for diagnosing a SHS.We tested va...OBJECTIVE:To investigate biological indicators of sub-optimal health status and provide means of objective assessment of sub-optimal health status.METHODS:We set the unified standards for diagnosing a SHS.We tested various laboratory indicators in 407 cases that we selected randomly from2807 subjects and collected 15 mL of fasting venous blood from each case.We measured serum immunoglobulin A(IgA)and immunoglobulin G(IgG)concentrations,serum beta endorphins(β-EP),cortisol(C),testosterone(T),plasma adrenocorticotropic hormone(ACTH)and serum T lymphocyte subsets CD3+and CD4+.RESULTS:Mean serum testosterone concentrations and their ratio to cortisol(C)concentrations weresignificantly higher in the healthy group than in those with sub-optimal health status(P<0.01).Mean serum CD3+concentrations were significantly higher in those with sub-optimal health status than in the healthy group(P<0.05).CONCLUSION:Decreased serum testosterone/cortisol ratio may be an objective indication of sub-optimal health status.Changes in neuroendocrine and immunological indicators may explain some of the symptoms,including malaise and poor work performance,attributable to persistent or relapsing fatigue in subjects with sub-optimal health status.展开更多
Purpose The high energy photon source(HEPS)is the fourth-generation synchrotron photon source.Compared with the third-generation synchrotron photon source,the brightness is 100-1000 times higher,and the electron emitt...Purpose The high energy photon source(HEPS)is the fourth-generation synchrotron photon source.Compared with the third-generation synchrotron photon source,the brightness is 100-1000 times higher,and the electron emittance of the storage ring is low to the diffraction limit of light.Through physical calculations,it is required that the stability of the storage ring quadrupole magnet power supply be better than 10 ppm,and the accuracy of output current be better than 80 ppm.This high demand for technical parameter poses a challenge to the development of high precision and stability power supplies.Methods The main circuit topology of the power supply adopts a phase-shifted full bridge soft switching scheme,which avoids interference caused by switching noise and improves power stability and efficiency.The high-precision digital power supply controller based on FPGA improves the sampling speed and control accuracy of the power supply,and the constant temperature control circuit ensures that the output current of the power supply meets the requirements of HEPS for power supply performance.Results In the batch testing section,a testing facility was built to test the stability,accuracy,repeatability,voltage ripple,and other parameter of high precision and stability power supplies.After a year and a half of testing,the performance tests of 1066 power supplies,including linear accelerators power supplies,booster power supplies,storage rings power supplies,dipole and quadrupole combined power supplies,dipole and quadrupole power supplies,were completed.The results all met and exceeded the design specifications.Conclusions The HEPS high precision and stability power supply meets the design requirements in terms of current stability,accuracy,repeatability,voltage ripple,and other aspects.The batch test results show that the power supply performance using the full bridge phase shifting soft switching technology combined with high-precision digital controller scheme is excellent,and the power supply consistency is good,providing a guarantee for the successful operation of HEPS in the future.展开更多
Purpose The high-energy photon source(HEPS)is the first fourth-generation synchrotron photon source in China(Jiang et al.in Sci Sin Phys Mech Astron 44:1075-1094,2014).The strip electrode and its power supply provide ...Purpose The high-energy photon source(HEPS)is the first fourth-generation synchrotron photon source in China(Jiang et al.in Sci Sin Phys Mech Astron 44:1075-1094,2014).The strip electrode and its power supply provide necessary hardware support for correcting the magnetic field of the wiggler as reported by Zhao and Yin(Particle accelerator technology,Higher Education Press,Beijing,2006).The design of the conventional power supply for accelerators,with a large output current and high output voltage,making it easy to achieve high stability of the output current.The strip electrode power supply poses difficulties in achieving high stability due to its small output current and voltage.Because of size limitations,as well as the requirements of the power architecture,there are currently no commercial products for this type of power supply.Methods Designed and developed 32 strip electrode power supplies.Considering the large number of power supplies and limited space,a rack structure design was adopted,placing all power supplies in one rack.The main circuit of the power supply adopts a full bridge transformation structure(Chao et al.in Radiat Detect Technol Methods 6:470-478,2022),and the control scheme adopts full digital control based on FPGA(Long and Cheng in At Energy Sci Technol 43:780-784,2009).The power supply has two control modes:local operation and remote operation as reported by Lu(Particle accelerator technology,Hunan University,Changsha,2010).Results The test shows that all power supplies meet the indicator requirements,with a stability of less than 100 ppm.Conclusions The strip electrode power supply can effectively correct the multipole integral field error and improve the integral field performance of the wiggler.展开更多
