Traditional therapies have primarily concentrated on single-target techniques, with a specific emphasis on targeting the vascular endothelial growth factor, but have not reached ideal therapeutic efficacy. In response to this issue, our study introduced a novel nanoparticle system known as CS-siRNA/PEITC&L-cRGD NPs. These chitosan-based nanoparticles have been recognized for their excellent biocompatibility and ability to deliver genes. To enhance their targeted delivery capability, they were combined with a cyclic RGD peptide (cRGD). Targeted co-delivery of gene and chemotherapeutic agents was achieved through the use of a negatively charged lipid shell and cRGD, which possesses high affinity for integrin αvβ3 overexpressed in tumor cells and neovasculature. In this multifaceted approach, co-delivery of VEGF siRNA and phenethyl isothiocyanate (PEITC) was employed to target both tumor vascular endothelial cells and tumor cells simultaneously. The co-delivery of VEGF siRNA and PEITC could achieve precise silencing of VEGF, inhibit the accumulation of HIF-1α under hypoxic conditions, and induce apoptosis in tumor cells. In summary, we have successfully developed a nanoparticle delivery platform that utilizes a dual mechanism of action of anti-tumor angiogenesis and pro-tumor apoptosis, which provides a robust and potent strategy for the delivery of anti-cancer therapeutics.展开更多
Background In the modern sheep production systems,the reproductive performance of ewes determines the economic profitability of farming.Revealing the genetic mechanisms underlying differences in the litter size is imp...Background In the modern sheep production systems,the reproductive performance of ewes determines the economic profitability of farming.Revealing the genetic mechanisms underlying differences in the litter size is important for the selection and breeding of highly prolific ewes.Hu sheep,a high-quality Chinese sheep breed,is known for its high fecundity and is often used as a model to study prolificacy traits.In the current study,animals were divided into two groups according to their delivery rates in three consecutive lambing seasons(namely,the high and low reproductive groups with≥3 lambs and one lamb per season,n=3,respectively).The ewes were slaughtered within 12 h of estrus,and unilateral ovarian tissues were collected and analyzed by 10×Genomics single-cell RNA sequencing.Results A total of 5 types of somatic cells were identified and corresponding expression profiles were mapped in the ovaries of each group.Noticeably,the differences in the ovary somatic cell expression profiles between the high and low reproductive groups were mainly clustered in the granulosa cells.Furthermore,four granulosa cell subtypes were identified.GeneSwitches analysis revealed that the abundance of JPH1 expression and the reduction of LOC101112291 expression could lead to different evolutionary directions of the granulosa cells.Additionally,the expression levels of FTH1 and FTL in mural granulosa cells of the highly reproductive group were significantly higher.These genes inhibit necroptosis and ferroptosis of mural granulosa cells,which helps prevent follicular atresia.Conclusions This study provides insights into the molecular mechanisms underlying the high fecundity of Hu sheep.The differences in gene expression profiles,particularly in the granulosa cells,suggest that these cells play a critical role in female prolificacy.The findings also highlight the importance of genes such as JPH1,LOC101112291,FTH1,and FTL in regulating granulosa cell function and follicular development.展开更多
To explore the green development of automobile enterprises and promote the achievement of the“dual carbon”target,based on the bounded rationality assumptions,this study constructed a tripartite evolutionary game mod...To explore the green development of automobile enterprises and promote the achievement of the“dual carbon”target,based on the bounded rationality assumptions,this study constructed a tripartite evolutionary game model of gov-ernment,commercial banks,and automobile enterprises;introduced a dynamic reward and punishment mechanism;and analyzed the development process of the three parties’strategic behavior under the static and dynamic reward and punish-ment mechanism.Vensim PLE was used for numerical simulation analysis.Our results indicate that the system could not reach a stable state under the static reward and punishment mechanism.A dynamic reward and punishment mechanism can effectively improve the system stability and better fit real situations.Under the dynamic reward and punishment mechan-ism,an increase in the initial probabilities of the three parties can promote the system stability,and the government can im-plement effective supervision by adjusting the upper limit of the reward and punishment intensity.Finally,the implementa-tion of green credit by commercial banks plays a significant role in promoting the green development of automobile enter-prises.展开更多
Anderson localization of phonons is a kind of phonon wave effect,which has been proved to occur in many structures with disorders.In this work,we introduced aperiodicity to boron nitride/carbon nanotube superlattices(...Anderson localization of phonons is a kind of phonon wave effect,which has been proved to occur in many structures with disorders.In this work,we introduced aperiodicity to boron nitride/carbon nanotube superlattices(BN/C NT SLs),and used molecular dynamics to calculate the thermal conductivity and the phonon transmission spectrum of the models.The existence of phonon Anderson localization was proved in this quasi one-dimensional structure by analyzing the phonon transmission spectra.Moreover,we introduced interfacial mixing to the aperiodic BN/C NT SLs and found that the coexistence of the two disorder entities(aperiodicity and interfacial mixing)can further decrease the thermal conductivity.In addition,we also showed that anharmonicity can destroy phonon localization at high temperatures.This work provides a reference for designing thermoelectric materials with low thermal conductivity by taking advantage of phonon localization.展开更多
<strong>Objective: </strong>To investigate the concentration and clinical significance of serum ferritin (SF) in patients diagnosed with advanced gastric cancer before and after treatment. <strong>Me...<strong>Objective: </strong>To investigate the concentration and clinical significance of serum ferritin (SF) in patients diagnosed with advanced gastric cancer before and after treatment. <strong>Methods:</strong> Forty patients with advanced gastric cancer diagnosed by cytology or pathology in our hospital were selected, including 25 males and 15 females, aged from 48 to 85 years, and the median age was 61.0 years. 40 healthy volunteers matched with age and education were selected as the control group. In order to study the changes of SF level in the treatment of advanced gastric cancer, we divided the patients into effective group (efficacy evaluation as partial remission or complete remission), ineffective group (efficacy evaluation as no remission) and recurrence group according to the efficacy after treatment. Then the difference of SF level between different groups and the relationship between SF level and curative effect were analyzed. There was no significant difference in gender and age among all groups. <strong>Results</strong>: The SF levels in the newly diagnosed group, stage III patient group and stage IV patient group were significantly higher than those in the control group. The level of SF in stage IV patients was significantly higher than that in stage III patients. There were significant differences in SF level between the effective treatment group, the newly diagnosed group and the ineffective treatment group, but there was no significant difference in SF level between the newly diagnosed group and the ineffective treatment group. <strong>Conclusion: </strong>SF level has a certain value in the auxiliary diagnosis of gastric cancer, and it also has a certain guiding significance for the evaluation of curative effect and prognosis after treatment.展开更多

OBJECTIVE: To determine the effects of continuous peripheral nerve block by tetrodotoxin before and after nerve injury on GAP-43 expression in the dorsal root ganglion during the development of neuropathic pain. DESIGN: A randomized controlled animal experiment. SETTINGS: Department of Anesthesiology, the Second Hospital of Xiamen City; Department of Anesthesiology, the Second Affiliated Hospital of Shantou University Medical College. MATERIALS: Thirty-five Spragne Dawley (SD) rats, weighing 200 - 250 g, were randomly divided into four groups: control group (n =5), simple sciatic nerve transection group (n =10), peripheral nerve block before and after sciatic nerve transection groups (n =10). All the sciatic nerve transection groups were divided into two subgroups according to the different postoperative survival periods: 3 and 7 days (n =5) respectively. Mouse anti-GAP-43 monoclonal antibody (Sigma Co., Ltd.), supervision TM anti-mouse reagent (HRP, Changdao antibody diagnosis reagent Co., Ltd., Shanghai), and HMIAS-100 image analysis system (Qianping Image Engineering Company, Tongji Medical University) were employed in this study. METHODS: This experiment was carried out in the Department of Surgery and Pathological Laboratory, the Second Affiliated Hospital of Shantou University Medical College from April 2005 to April 2006. ①The animals were anesthetized and the right sciatic nerve was exposed and transected at 1 cm distal to sciatic notch. ② Tetrodotoxin 10 μg/kg was injected percutaneously between the greater trochanter and the posterior superior iliac spine of fight hind limb to block the sciatic nerve proximally at 1 hour before or 4 hours after nerve injury respectively, the injection was repeated in all the rats every 12 hours.③ At 3 or 7 days after nerve injury, immunohistochemistry and image analysis were used to evaluate the expression of GAP-43 in the dorsal root ganglions of L5 to the transected sciatic nerve, and quantitative analysis was also performed. ④ Statistical analysis was performed using one way analysis of variance followed by t test. MAIN OUTCOME MEASURE: Expression of GAP-43 in the fight dorsal root ganglions of L5. RESULTS: All the 35 SD rats were involved in the final analysis of results. In normal rats, there were very low expressions of GAP-43 in the dorsal root ganglions. In simple sciatic nerve transection rats 3 and 7 days after sciatic nerve transection, the average absorbance value of GAP-43 immunopositive neurons were significantly different from that in normal rats (t =8.806, 6.771, P 〈 0.01). Whereas 3 and 7 days after sciatic nerve transection in rats with peripheral nerve block before and after nerve injury, the average absorbance value of GAP-43 immunopositive neurons were not significantly different from that in normal rats (P 〉 0.05). CONCLUSION: Local anesthetic continuous peripheral nerve block before or after nerve injury can suppress nerve injury induced high expression of GAP-43 during the development of neuropathic pain.展开更多
Objective:To investigate the therapeutic efficacy and safety of Sijunzi Decoction (SJZD) for peptic ulcers (PUs) through the assessment of randomized controlled tHals (RCTs).Methods:Five English and four Chinese datab...Objective:To investigate the therapeutic efficacy and safety of Sijunzi Decoction (SJZD) for peptic ulcers (PUs) through the assessment of randomized controlled tHals (RCTs).Methods:Five English and four Chinese databases were systematically searched to identify eligible RCTs that investigated the effect of SJZD for PUs.SJZD was used either independently or concomitantly with routine anti-ulcer treatments and compared with no intervention,placebo,or conventional therapy.Outcomes evaluated included ulcer healing,symptom improvement,Helicobacter pylori (Hp) eradication,ulcer recurrence,and adverse reactions.Statistical analysis and risk of bias was evaluated by Revman 5.3.Results:Fourteen trials involving 1476 participants fulfilled the selection criteria.All trials were presented with a high risk of bias.Analyses showed that modified SJZD had significant benefit on ulcer healing compared with ranitidine and sucralfate and triple therapy with omeprazole,amoxicillin and clarithromycin.Compared with this omeprazole triple therapy and therapy using pantoprazole,the addition of modified SJZD was superior.Regarding symptom improvement,modified SJZD was significantly better than ranitidine and sucralfate and had an added benefit when supplemented with either of two omeprazole triple therapies (omeprazole,amoxicillin and clarithromycin,and omeprazole,levofloxacin and furazolidone) or pantoprazole.Furthermore,modified SJZD had a favorable effect on Hp eradication compared with cimetidine and additional benefit was noted when complemented with either of the two omeprazole triple therapies or therapy consisting of omeprazole and amoxicillin.Modified SJZD was superior at reducing ulcer recurrence among healed patients compared with cimetidine.Modified SJZD with triple-drug regimen comprising omeprazole,amoxicillin and clarithromycin was also found to be more effective at reducing ulcer relapse compared with the triple therapy alone.No serious adverse events were reported.Conclusion:Modified SJZD,used independently or as a complementary treatment to conventional ulcer-healing treatments,promoted PU healing,alleviated symptoms,eradicated Hp,and reduced ulcer relapse.Modified SJZD was relatively safe.However,rigorous randomized controlled trials are required to provide further support for these effects because of the high risk of bias and limited sample size of the included trials.展开更多
[Objectives]To observe the effect of aqueous extract of Houttuynia cordata on the content of serum TNF-αand IL-6 in mice with abdominal infection caused by Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginos...[Objectives]To observe the effect of aqueous extract of Houttuynia cordata on the content of serum TNF-αand IL-6 in mice with abdominal infection caused by Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Proteus vulgaris and Klebsiella pneumoniae,so as to investigate the antibacterial effect of aqueous extract of Houttuynia cordata in vivo.[Methods]A total of 72 KM mice were randomly divided into six groups:model group,blank group,cefdinir dispersible tablet group(0.25 g/kg),high,medium and low dose groups of aqueous extract of Houttuynia cordata(40,20,10 g/kg crude drug).There were 12 mice in each group,half male and half female,continuously given intragastric administration for three days,once a day.The blank group and the model group were intragastrically infused with normal saline,and the other groups were given corresponding drugs with a volume of 0.02 mL/g.After 30 min of administration on the third day,the mice were injected intraperitoneally with 0.5 mL of bacterial suspension to make bacterial infection.The mice in the blank group were intraperitoneally injected with the same amount of normal saline,followed by intragastric administration 6 h after intraperitoneal injection and intragastric administration again after 24 h.The mice were observed for three days,and the changes of body weight before and after infection were recorded.On the fourth day of infection,eyeball blood was taken,serum was separated,and the concentration of inflammatory factors TNF-αand IL-6 in serum was determined.[Results]Compared with the model group,the high,medium and low dose groups of H.cordata aqueous extract had protective effects on mice with abdominal infection caused by S.aureus,E.coli,P.aeruginosa,P.vulgaris and K.pneumoniae,and decreased the content of TNF-αand IL-6 in serum.[Conclusions]The aqueous extract of H.cordata had a certain antibacterial effect on acute abdominal infection caused by S.aureus,E.coli,P.aeruginosa,P.vulgaris and K.pneumoniae,and its mechanism was related to TNF-αand IL-6.展开更多
Living crystallization-driven self-assembly(CDSA)provides robust access to uniformπ-conjugated nanostructures(CNSs)from block copolymers(BCPs)containing a crystallineπ-conjugated segment with controlled dimension,mo...Living crystallization-driven self-assembly(CDSA)provides robust access to uniformπ-conjugated nanostructures(CNSs)from block copolymers(BCPs)containing a crystallineπ-conjugated segment with controlled dimension,morphology and composition,which show appealing applications in biomedicine,photocatalysis and microelectronics.To further expand the application spectrum of these CNSs,the development of facile strategies toward diverse CNSs with varying structures/functionalities is highly desired.Herein,BCPs consisting of oligo(p-phenylene ethynylene)-b-poly(polypropyl-3-methanethiol acrylate)(OPE_(9)-bPMTPA35and OPE_(9)-b-PMTPA_(58);the subscript represents the number of repeat unit of each block)consisting of a crystallineπ-conjugated core-forming OPE_(9)segment and a corona-forming PMTPA block are synthesized.By efficient“click-type”alkylation of methylthio groups,OPE_(9)-b-PMTPA with varying contents of sulfonium unit is obtained.Uniform ribbon-like micelles with different widths and lengths can then be generated in a controlled manner via the self-seeding approach of living CDSA.Additionally,negatively charged polymeric and Ag nanoparticles can be immobilized on sulfonium/methylthio-containing shells by taking advantage of electrostatic attraction and coordination interaction,respectively.Interestingly,the ribbon-like micelles with positively charged shells exhibit antibacterial activity against E.coli.Given the ease of modification of PMTPA-based shell and attractive opto-electronic/photocatalytic properties ofπ-conjugated units,the combination of methylthio-chemistry and living CDSA opens a new avenue to generate multi-functional CNSs for widespread applications from biomedicine to photocatalysis.展开更多

MAIN OUTCOME MEASURE: Expression of GAP-43 in the fight dorsal root ganglions of L5. RESULTS: All the 35 SD rats were involved in the final analysis of results. In normal rats, there were very low expressions of GAP-43 in the dorsal root ganglions. In simple sciatic nerve transection rats 3 and 7 days after sciatic nerve transection, the average absorbance value of GAP-43 immunopositive neurons were significantly different from that in normal rats (t =8.806, 6.771, P 〈 0.01). Whereas 3 and 7 days after sciatic nerve transection in rats with peripheral nerve block before and after nerve injury, the average absorbance value of GAP-43 immunopositive neurons were not significantly different from that in normal rats (P 〉 0.05). CONCLUSION: Local anesthetic continuous peripheral nerve block before or after nerve injury can suppress nerve injury induced high expression of GAP-43 during the development of neuropathic pain.

文摘Objective:To investigate the therapeutic efficacy and safety of Sijunzi Decoction (SJZD) for peptic ulcers (PUs) through the assessment of randomized controlled tHals (RCTs).Methods:Five English and four Chinese databases were systematically searched to identify eligible RCTs that investigated the effect of SJZD for PUs.SJZD was used either independently or concomitantly with routine anti-ulcer treatments and compared with no intervention,placebo,or conventional therapy.Outcomes evaluated included ulcer healing,symptom improvement,Helicobacter pylori (Hp) eradication,ulcer recurrence,and adverse reactions.Statistical analysis and risk of bias was evaluated by Revman 5.3.Results:Fourteen trials involving 1476 participants fulfilled the selection criteria.All trials were presented with a high risk of bias.Analyses showed that modified SJZD had significant benefit on ulcer healing compared with ranitidine and sucralfate and triple therapy with omeprazole,amoxicillin and clarithromycin.Compared with this omeprazole triple therapy and therapy using pantoprazole,the addition of modified SJZD was superior.Regarding symptom improvement,modified SJZD was significantly better than ranitidine and sucralfate and had an added benefit when supplemented with either of two omeprazole triple therapies (omeprazole,amoxicillin and clarithromycin,and omeprazole,levofloxacin and furazolidone) or pantoprazole.Furthermore,modified SJZD had a favorable effect on Hp eradication compared with cimetidine and additional benefit was noted when complemented with either of the two omeprazole triple therapies or therapy consisting of omeprazole and amoxicillin.Modified SJZD was superior at reducing ulcer recurrence among healed patients compared with cimetidine.Modified SJZD with triple-drug regimen comprising omeprazole,amoxicillin and clarithromycin was also found to be more effective at reducing ulcer relapse compared with the triple therapy alone.No serious adverse events were reported.Conclusion:Modified SJZD,used independently or as a complementary treatment to conventional ulcer-healing treatments,promoted PU healing,alleviated symptoms,eradicated Hp,and reduced ulcer relapse.Modified SJZD was relatively safe.However,rigorous randomized controlled trials are required to provide further support for these effects because of the high risk of bias and limited sample size of the included trials.

文摘[Objectives]To observe the effect of aqueous extract of Houttuynia cordata on the content of serum TNF-αand IL-6 in mice with abdominal infection caused by Staphylococcus aureus,Escherichia coli,Pseudomonas aeruginosa,Proteus vulgaris and Klebsiella pneumoniae,so as to investigate the antibacterial effect of aqueous extract of Houttuynia cordata in vivo.[Methods]A total of 72 KM mice were randomly divided into six groups:model group,blank group,cefdinir dispersible tablet group(0.25 g/kg),high,medium and low dose groups of aqueous extract of Houttuynia cordata(40,20,10 g/kg crude drug).There were 12 mice in each group,half male and half female,continuously given intragastric administration for three days,once a day.The blank group and the model group were intragastrically infused with normal saline,and the other groups were given corresponding drugs with a volume of 0.02 mL/g.After 30 min of administration on the third day,the mice were injected intraperitoneally with 0.5 mL of bacterial suspension to make bacterial infection.The mice in the blank group were intraperitoneally injected with the same amount of normal saline,followed by intragastric administration 6 h after intraperitoneal injection and intragastric administration again after 24 h.The mice were observed for three days,and the changes of body weight before and after infection were recorded.On the fourth day of infection,eyeball blood was taken,serum was separated,and the concentration of inflammatory factors TNF-αand IL-6 in serum was determined.[Results]Compared with the model group,the high,medium and low dose groups of H.cordata aqueous extract had protective effects on mice with abdominal infection caused by S.aureus,E.coli,P.aeruginosa,P.vulgaris and K.pneumoniae,and decreased the content of TNF-αand IL-6 in serum.[Conclusions]The aqueous extract of H.cordata had a certain antibacterial effect on acute abdominal infection caused by S.aureus,E.coli,P.aeruginosa,P.vulgaris and K.pneumoniae,and its mechanism was related to TNF-αand IL-6.

文摘Living crystallization-driven self-assembly(CDSA)provides robust access to uniformπ-conjugated nanostructures(CNSs)from block copolymers(BCPs)containing a crystallineπ-conjugated segment with controlled dimension,morphology and composition,which show appealing applications in biomedicine,photocatalysis and microelectronics.To further expand the application spectrum of these CNSs,the development of facile strategies toward diverse CNSs with varying structures/functionalities is highly desired.Herein,BCPs consisting of oligo(p-phenylene ethynylene)-b-poly(polypropyl-3-methanethiol acrylate)(OPE_(9)-bPMTPA35and OPE_(9)-b-PMTPA_(58);the subscript represents the number of repeat unit of each block)consisting of a crystallineπ-conjugated core-forming OPE_(9)segment and a corona-forming PMTPA block are synthesized.By efficient“click-type”alkylation of methylthio groups,OPE_(9)-b-PMTPA with varying contents of sulfonium unit is obtained.Uniform ribbon-like micelles with different widths and lengths can then be generated in a controlled manner via the self-seeding approach of living CDSA.Additionally,negatively charged polymeric and Ag nanoparticles can be immobilized on sulfonium/methylthio-containing shells by taking advantage of electrostatic attraction and coordination interaction,respectively.Interestingly,the ribbon-like micelles with positively charged shells exhibit antibacterial activity against E.coli.Given the ease of modification of PMTPA-based shell and attractive opto-electronic/photocatalytic properties ofπ-conjugated units,the combination of methylthio-chemistry and living CDSA opens a new avenue to generate multi-functional CNSs for widespread applications from biomedicine to photocatalysis.
