The association between PTPRD/PTPRT mutations and immunotherapeutic efficacy was analyzed. Then, the association between PTPRD/PTPRT mutation and immune profiles was analyzed using The Cancer Genome Atlas(TCGA) cohort. Results: A total of 2,392 patients across 20 cancer types were included in this study. Our results showed that patients harboring PTPRD/PTPRT mutation, especially co-mutations, had a significantly elevated response rate to immunotherapy in multiple cancers. Patients with PTPRD/PTPRT mutation had a higher objective response rate(ORR)(P=0.002), longer overall survival(OS)(P=0.005) and progression-free survival(PFS)(P=0.038).Importantly, the above findings were further verified in validation cohorts. In addition, we found that the PTPRD/PTPRT co-mutations(co-mut) subgroup exhibited an immune-activated phenotype, the wild-type subgroup tended to have an immune-desert phenotype, and the uni-mutation(uni-mut) subgroup might have an immune-mixed phenotype. Our further analyses suggested that combining programmed cell death ligand 1(PDL1) expression and PTPRD/PTPRT mutation can be used to screen patients who may benefit from immunotherapy. Conclusions: PTPRD/PTPRT mutation could serve as a potential predictive biomarker for cancer immunotherapy.展开更多

The aim of this study was to evaluate the role and indications of diagnostic laparoscopy in the detection of intra-abdominal metastasis. Methods: Standard diagnostic laparoscopy with peritoneal cytology examination was performed prospectively on patients who were clinically diagnosed with primary local advanced gastric cancer (CT≥2M0). We calculated the rate of intra-abdominal metastases identified by diagnostic laparoscopy, and examined the relationship between peritoneal dissemination (P) and cytology results (CY). Split-sample method was applied to find clinical risk factors for intra-abdominal metastasis. Multivariate logistic regression analysis and receiver-operator characteristic (ROC) analysis were performed in training set to find out risk factors ofintra-abdominal metastasis, and then validate it in testing set. Results: Out of 249 cM0 patients, 51 (20.5%) patients with intra-abdominal metastasis were identified by diagnostic laparoscopy, including 20 (8.0%) P1CY1, 17 (6.8%) POCY1 and 14 (5.6%) P1CY0 patients. In the training set, multivariate logistic regression analysis and ROC analysis showed that the depth of tumor invasion on computer tomography (CT) scan ≥21 mm and tumor-occupied 〉2 portions of stomach are predictive factors of metastasis. In the testing set, when diagnostic laparoscopy was performed on patients who had one or two of these risk factors, the sensitivity and positive predictive value for detecting intra-abdominal metastasis were 90.0% and 32.1%, respectively. Conclusions: According to our results, depth of tumor invasion and tumor-occupied portions of stomach are predictive factors ofintra-abdominal metastasis.展开更多

Detuning parameters between frequencies of atomic transitions and those of the corresponding driving lasers are carefully chosen to offset the blockade effect of two Rydberg atoms, so that an effective Hamiltonian, representing a single-photon detuning L-type three-level system and concluding the quantum state of two Rydberg atoms excited simultaneously, is obtained. The adiabatic-passage technique, based on the effective Hamiltonian, is adopted to implement a two-atom phase gate by using two time-dependent Rabi frequencies. Numerical simulations indicate that a high-fidelity two-qubit p-phase gate is constructed and its operation time does not have to be controlled accurately. Besides, owing to the long coherence time of the Rydberg state, the phase gate is robust against atomic spontaneous emission.展开更多

文摘Optical gain and loss of microcavity greatly affect the quality of lasing, how to improve optical gain and decrease optical loss is of great significance for the preparation of laser. In this study, four types standard microdisks with different thicknesses of 2.2μm,1.9μm,1.7μm,and 1.45μm were fabricated by micromachining technology process to modulate optical gain and loss of microdisk lasing. The whispering gallery mode lasing in the ultraviolet range of Ga N microdisk devices was investigated for these devices in order to clarify the effect of microdisk thickness on device characteristics. The quality factor Q and lasing mode number for different thicknesses are calculated from the stimulated spectra. The lifetimes of the exciton combination properties of the devices were observed using time-resolved PL spectroscopy. The lasing modes are modulated, and the lifetime decreases, while the Q factor of the devices first increases and then decreases with decreasing thickness. All these results are induced by optical gain and loss competition.
