Glioma is one of the lethal central nervous system tumors.The infiltrative and invasive growth nature makes it difficult to identify the boundary between glioma and the normal tissues,resulting in inevitable recurrenc...Glioma is one of the lethal central nervous system tumors.The infiltrative and invasive growth nature makes it difficult to identify the boundary between glioma and the normal tissues,resulting in inevitable recurrence after surgery operation.Gliomas do not metastasize,so to prevent the residual tumor from proliferating or invading is a key challenge.Previous report indicated that hypotaurine could facilitate glioma invasion and suppress demethylases’activities.Using a hypotaurine synthesis deficient U251 cell line,we proved that the cells invasion ability was impaired.Gene expression profile analysis exhibited that knocking down one of the key enzymes of hypotaurine synthesis,2-aminoethanethiol dioxygenase(ADO),significantly affected the extracellular matrix-receptor process.Of that process,Wnt5a expression was severely upregulated by decreased intracellular ADO expression.Cells cultured at the presence of hypotaurine showed a decrease in intracellular Wnt5a protein and mRNA levels.This phenotype was due to hypermethylation of Wnt5a promoter,which was most likely the result of hypotaurine’s inhibiting demethylases activities.Collectively,this study demonstrated that hypotaurine synthesis deficient U251 cells were prone to epigenetic modification and Wnt5a seemed to be a tumor suppressor under that circumstance.This tumor suppression effect is warranted to be reevaluated in real tumor samples and the relevant evidence might contribute to develop new glioma interference strategies.展开更多

However, the characteristics of scalp and hair have not yet been studied in Chinese, the largest population in the world, especially in terms of ageing. The purpose of this study is to investigate ageing features in Chinese women’s hair and scalp. Methods: In total, 1343 women in China aged 20 to 65 participated in this research. Hair density and diameter were measured and analysed with Folliscope®. Hair color and scalp conditions such as dandruff and erythema were evaluated through microscopic images. Measuring equipment was also used to detect scalp water content, barrier function and sebum secretion. Results: Hair density decreased gradually with ageing, while hair diameter peaked at 40s. The color of hair became bright with ageing because of gray hair and color fading. The degree of scalp sebum and dandruff was higher at younger ages. Interestingly, scalp erythema became more severe with ageing. Conclusion: This study shows the physical and physiological changes of Chinese women’s hair and scalp. Most aging features (hair diameter, scalpsebum contents, dandruff and erythema) were noticeable in their 40s, but hair density began to decline in their 30s. This first study of aging hair and scalp in Chinese women would be helpful for understanding the ageing phenomenon and development of hair and scalp care products.展开更多

Background:This study aimed to develop a comprehensive instrument for evaluating and ranking clinical practice guidelines,named Scientific,Transparent and Applicable Rankings tool(STAR),and test its reliability,validi...Background:This study aimed to develop a comprehensive instrument for evaluating and ranking clinical practice guidelines,named Scientific,Transparent and Applicable Rankings tool(STAR),and test its reliability,validity,and usability.Methods:This study set up a multidisciplinary working group including guideline methodologists,statisticians,journal editors,clinicians,and other experts.Scoping review,Delphi methods,and hierarchical analysis were used to develop the STAR tool.We evaluated the instrument’s intrinsic and interrater reliability,content and criterion validity,and usability.Results:STAR contained 39 items grouped into 11 domains.The mean intrinsic reliability of the domains,indicated by Cronbach’sαcoefficient,was 0.588(95%confidence interval[CI]:0.414,0.762).Interrater reliability as assessed with Cohen’s kappa coefficient was 0.774(95%CI:0.740,0.807)for methodological evaluators and 0.618(95%CI:0.587,0.648)for clinical evaluators.The overall content validity index was 0.905.Pearson’s r correlation for criterion validity was 0.885(95%CI:0.804,0.932).The mean usability score of the items was 4.6 and the median time spent to evaluate each guideline was 20 min.Conclusion:The instrument performed well in terms of reliability,validity,and efficiency,and can be used for comprehensively evaluating and ranking guidelines.展开更多

To address the challenge of highly sensitive and accurate detection of biomarkers in complex environments,a rational engineering strategy for designing electrochemical immunosensing platform is proposed.Herein,we deve...To address the challenge of highly sensitive and accurate detection of biomarkers in complex environments,a rational engineering strategy for designing electrochemical immunosensing platform is proposed.Herein,we develop a microsensor chip through the combination of multiplexed electrodes and microfluidic channels for the parallel detection of human interleukins(IL-6 and IL-8).For the construction of an efficient sensing interface,the conductive silver nanowires(Ag NWs)wrapped with zeolitic imidazolate framework(ZIF-8)thin film(denoted as ZIF-8@Ag NWs)are prepared,and then employed for the multi-functionalization of electrodes.The immunodetection of ILs is based on the direct signal transduction ability of Ag NWs and specific interaction of periodically arranged Zn2+ions in ZIF-8 films with biomolecules,which offer the high assay sensitivity and good specificity.The immunosensor chip achieves a wide detection range from pg/mL to ng/mL and possesses the ability to resist non-specific proteins adsorption in biological complex media.Further clinical serum samples assay verifies that the combination of IL-6 and IL-8 levels yields high diagnostic accuracy.Principal component analysis(PCA)reveals that 18 patient samples could be fully separated from healthy control samples.The low-cost,easily fabricated electrochemical immunosensing platform provides a rapid serum test for diagnosis and personalized therapy of inflammatory diseases,and can also be generalized to other immunoreaction-based biomarkers detections.展开更多

Dihydronicotinamide adenine dinucleotide(NADH)is an important enzyme in all living cells,which is found to be abnormally expressed in cancer cells.Since it is redox-active,an electrochemical detection method would be ...Dihydronicotinamide adenine dinucleotide(NADH)is an important enzyme in all living cells,which is found to be abnormally expressed in cancer cells.Since it is redox-active,an electrochemical detection method would be suitable for monitoring its concentration in biological fluids.Here we present a strategy for specific determination of NADH in real human serum by using RhIr@MoS2 nanohybrids based microsensor.To implement the protocol,RhIr nanocrysrals are in-situ grown onto MoS2 interlayers forming a nanohybrid structure(RhIr@MoS2).After being locally deposited on an electrochemical microsensor,it could be used for the analysis of NADH.The developed RhIr@MoS2 nanohybrids based microsensor possesses the ability for analyzing NADH at the applied potential of 0.07 V(much lower than most reported values).The detection limit is evaluated as low as 1 nmol/L even in bovine serum albumin(BSA)media.In addition,the sampling analysis of human serum from cancer patients and health controls shows that the microsensor displays good diagnostic sensitivity and specificity,illustrating that this developed detection technique is a relatively accurate method for measuring NADH in biological fluids.The proposed electrochemical microsensor assay also owns the benefits of convenience,disposable and easy processing,which make it a great possibility for future point-of-care cancer diagnosis.展开更多
