Objective:To explore the effect of Hupo powder(HP)on autophagy in menstrual blood-derived stem cells(MenSCs)with endometriosis(EMT).Methods:EMT MenSCs(E-MenSCs)and healthy MenSCs(H-MenSCs)were isolated from the menstrual blood of patients with EMT and healthy female participants,respectively.We identified their stem cells’characteristics via adipogenic and osteogenic differentiation.Twelve male SpragueeDawley rats received 0.9% NaCl and HP-dispensing granules by gastric irrigation to prepare blank serum and medicated serum,respectively.We used serum concentrations of 5%,10%,and 20%,each at administered times of 12,24,and 48 h to select the best condition.These cells were divided into three groups:blank serum of the control group,blank serum of the model group,and medicated serum of the HP group.H-MenSCs were used in the control group,while E-MenSCs were used in the model and HP groups.We analyzed cell viability using a cell counting kit-8 assay,observed cell morphology,evaluated the amounts of auto-phagosomes and autolysosomes by transmission electron microscopy,and detected the protein expression of autophagy markers(LC3-II and Beclin1)by Western blot.Results:E-MenSCs and H-MenSCs became long fusiform with a diffuse radial pattern,forming lipid droplets and calcium nodules after adipogenic and osteogenic differentiation.We then used the best conditiond 20% serum and 48 hdfor the subsequent experiments.In contrast to the model group,the HP group exhibited lower cell viability(=0.007),larger amounts of autophagosomes and autolysosomes(P<0.001 and P=0.001,respectively),and higher expression of LC3-II and Beclin1(P=0.021 and P=0.019,respectively).Conclusion:Hupo powder can promote autophagy in E-MenSCs,which might be one of the mechanisms underlying its therapeutic effects. 展开更多

A series of barium-tungstate-based phosphors doped with different concentrations of Dy<sup>3+</sup> were synthesized by solid-state reaction method. Photoluminescence properties and decay lifetime of Dy<sup>3+</sup>-doped BaWO<sub>4</sub> samples were studied. The results indicated that luminescent properties of BaWO<sub>4</sub>:Dy<sup>3+</sup> depended on the Dy<sup>3+</sup> concentration, and the inner energy could transfer from  to Dy<sup>3+</sup>. The quality of the light was checked by estimating CIE parameters, and the results showed that BaWO<sub>4</sub>:Dy<sup>3+</sup> was a potential candidate as blue-green luminescent materials in white LED because of its excellent emission spectrum excited by UV light. 展开更多

Microarray analysis showed that 24 hours after cerebral ischemia, there were nine up-regulated and 27 down-regulated microRNA genes in cortical tissue. Bioinformatic analysis showed that bcl-2 was the target gene of microRNA-384-5p and microRNA-494, and caspase-3 was the target gene of microRNA-129, microRNA-320 and microRNA-326. Real-time PCR and western blot analyses showed that 24 hours after cerebral ischemia, bcl-2 mRNA and protein levels in brain tissue were significantly decreased, while caspase-3 mRNA and protein levels were significantly increased. This suggests that following cerebral ischemia, differentially expressed microRNA-384-5p, microRNA-494, microRNA-320, microRNA-129 and microRNA-326 can regulate bcl-2 and caspase-3 expression in brain tissue. 展开更多

目的探讨产前瑜伽干预对产后抑郁症发生情况的影响。方法选取2013年6月-2017年6月在本院待产的惠州地区孕妇作为试验研究样本,梳理分析病例资料,挑选出200例符合试验要求的孕妇进行入组试验,瑜伽组95例进行瑜伽运动干预,瑜伽练习自孕期1... 目的探讨产前瑜伽干预对产后抑郁症发生情况的影响。方法选取2013年6月-2017年6月在本院待产的惠州地区孕妇作为试验研究样本,梳理分析病例资料,挑选出200例符合试验要求的孕妇进行入组试验,瑜伽组95例进行瑜伽运动干预,瑜伽练习自孕期16周开始至分娩结束,对照组105例则仅作常规护理。产后分三个阶段(产后3天、42天、3个月)分别对受试个体的抑郁情况进行量化评分,以爱丁堡产后抑郁量表(EPDS)为参考标准进行抑郁评估。结果瑜伽组在产后三个评价阶段,评价得分情况均低于对照组(P<0.05);瑜伽组的抑郁发生率明显低于对照组(P<0.05)。结论对待产孕妇产前行瑜伽干预,可有效将降低产后抑郁症发生率,临床应用前景好。 展开更多

Tetramethylpyrazine(TMP) is a traditional Chinese herbal medicine with strong antiinflammatory and cartilage protection activities, and thus a promising candidate for treating osteoarthritis. However, TMP is rapidly cleared from the joint cavity after intra-articular injection and requires multiple injections to maintain efficacy. The aim of this study was to encapsulate TMP into poly(lactic-co-glycolic acid)(PLGA) microspheres to enhance the TMP retention in the joint, reducing injection frequencies and decreasing dosage. TMP microspheres were prepared by emulsion/solvent evaporation method. The intra-articular retention of the drug was assessed by detecting the drug concentration distributed in the joint tissue at different time points. The therapeutic effect of TMP microspheres was evaluated by the swelling of knee joints and histologic analysis in papain-induced OA rat model. The prepared freezedried microspheres with a particle size of about 10 μm can effectively prolong the retention time of the drug in the articular cavity to 30 d, which is 4.7 times that of the TMP solution.Intra-articular injection of TMP microspheres efficiently relieved inflammatory symptoms,improved joint lesions and decreased the depletion of proteoglycan. In conclusion, intraarticular injection of TMP loaded microspheres was a promising therapeutic method in the treatment of OA. 展开更多

Background:Metabolic diseases pose considerable burden on the healthcare system worldwide,indicating the significance of prevention and treatment.In constitution theory of traditional Chinese medicine,phlegm-dampness constitution(PDC)is the common basis of metabolic diseases.In clinical practice,Huatan Qushi(HTQS)decoction targeting on PDC can effectively improve metabolic indicators.However,its underlying biochemical mechanism still remains unclear.Methods:Eight PDC participants received HTQS decoction for three months.Their blood was collected at baseline and 1 and 3 months after intervention started.Related biomedical indicators were detected.High-throughput sequencing and RT-qPCR were used for validation.Due to the missing data,repeated measures with missing values in mixed models were used.Results:After 3-month treatment,HDL-C level increased(P<.001)and FBG,FINS,and HbA1c all showed decreasing trend at different time points(all P<.05).After miRNA high-throughput sequencing,compared with the baseline,differential miRNAs at 1 and 3 months were screened,and target gene prediction and KEGG pathway enrichment analysis were performed.The results displayed that metabolic disease-related pathways mainly included pathways in cancer,PI3K-Akt signaling pathway,etc.Further,RT-qPCR showed that hsa-miR-1237e3p differed statistically(P=.008).Then we validated the target genes of hsa-miR-1237e3p in the“Pathways in Cancer”pathway including SDF1,AC,CRK,and HGF,also known as upstream target genes of PI3K/AKT pathway.The results showed that two indicators of CRK and HGF were in statistical significance(P=.045 and P=.036,respectively).Conclusion:PDC serves as a common basis for various metabolic diseases.Through adjusting PDC,HTQS decoction can improve biomedical indicators including blood glucose,HbA1c,insulin,and HDL-C.The target pathway is“Pathways in cancer”.Specifically,HTQS decoction acts on targets of CRK and HGF by regulating hsa-miR-1237e3p,and probably exerts effects on their downstream PI3K/AKT pathway. 展开更多

Background:Endometriosis is a hormone-and organ-dependent disease with unclear pathogenesis.Danggui Shaoyao powder(DSP)has been used for the treatment of endometriosis for many years and has been proven to be effective,but its underlying molecular mechanisms remain unclear.In this study,we explored the molecular targets and pathways of DSP in the treatment of endometriosis mainly from the perspective of network pharmacology,and provided some novel ideas for managing this disease.Methods:In the present study,we focused on the underlying mechanisms between DPS and endometriosis,via oral bioavailability screening,drug similarity assessment,target identification,network analysis,drug relocation,and molecular docking using network pharmacology.Results:By bioinformatics,54 active components of DSP were predicted from the 534 components provided by the TCMSP database.We then found 85 validated targets and 6059 predicted targets for herbal components,and 2241 targets for endometriosis.After mapping,there were 255 targets in common.After that,network analysis was performed to screen key networks and core targets.Ninetynine indirect targets were collected from 14 direct ones.GO and KEGG analyses identified 63 enriched functions and 10 pathways(P<.01).According to the disease target,dipyridamole could undergo drug repositioning for endometriosis(score?1).The mechanism of action of dipyridamole on endometriosis was compared with that of DSP.Finally,eight targets,namely,AR,ESR1,HMGCR,NOS2,NR3C1,PPARG,PTGES,and PTGS2,were validated by molecular docking,suggesting that the probable molecular mechanisms by which DSP treats endometriosis are through seven validated pathways:hsa05200,hsa04915,hsa01100,hsa00900,hsa04920,hsa01130,and hsa00590.Conclusion:In this study,we uncovered the mechanism by which DSP can treat endometriosis using a drugecomponentetarget interaction network.This approach can provide some novel ideas for the development of Chinese medicine and the clinical management of endometriosis. 展开更多

Among these constitutions,the main feature of the yang-deficiency constitution (YADC) is intolerance of the cold,which has been proven to reduce quality of life and confer susceptibility to specific diseases.Previous studies explored the genetic and transcriptional bases of YADC.In this experiment,we explored the potential mechanism of YADC using protein microarray,to deepen our understanding of its biological mechanism.Methods:Subjects identified with a YADC (n =12) or a balance constitution (BC;n =12) in accordance with the Classification and Determination Standards of Constitutions in Traditional Chinese Medicine were selected.Blood was collected to separate serum and protein microarray technology was used to analyze serum protein expression.Results:The clustering of subjects' constitutions based on protein expression profiling largely coincided with the TCM classification.Based on false discovery rate correction (P <.01) and fold change ≥ 5 or ≤ 0.2,a total of 85 proteins differentially expressed in YADC compared with their status in BC were selected,including 64 upregulated and 21 downregulated ones.Enrichment analysis suggested that subjects with YADC are susceptible to endocrine and energy metabolism disorders,as well as decline in immune function.Conclusion:This study revealed that YADC exhibits systematic differences in its protein expression profile.Moreover,we can potentially explain the characteristics of YADC partly via differentially expressed proteins. 展开更多

In order to solve this problem,the technology of ridge-mulching and side-sowing of spring corn is put forward,the supporting compound operation seeder is developed,and the effect of different speed on the quality of sowing is tested and analyzed. Under the test conditions described in this paper,the seeding operation with a high speed( up to 6 km/h) can be realized,and the quality of the seeding operation can meet the requirements of the national standards. The application of this machine can solve the problem of " drought damage at the booting stage" for spring corn,thereby realizing the deep fusion of farming machine and agronomy in dry farming of northern China,and achieving the integration of farming machine and agronomy. 展开更多

与主胰管毗邻的良性肿瘤,通常需要扩大切除范围,甚至实施胰腺-消化道吻合来恢复胰腺的外分泌功能,增加了术后严重并发症的风险.因此,如何维护主胰管的连续性和生理功能成为胰腺外科亟须解决的问题.为了减少手术对良性肿瘤患者的创伤,我... 与主胰管毗邻的良性肿瘤,通常需要扩大切除范围,甚至实施胰腺-消化道吻合来恢复胰腺的外分泌功能,增加了术后严重并发症的风险.因此,如何维护主胰管的连续性和生理功能成为胰腺外科亟须解决的问题.为了减少手术对良性肿瘤患者的创伤,我们借鉴了桥梁合龙的原理,提出了胰管(修复)外科的概念,即对胰管进行整形或修复,恢复及维护其连续性和正常生理功能的一系列胰腺外科术式.在临床中,根据不同的肿瘤部位和切除范围,胰管(修复)外科可以分为主胰管修复术、胰腺端-端重建术、主胰管替代术和分支胰管型导管内乳头状黏液肿瘤(branch duct intraductal papillary mucinous neoplasm,BD-IPMN)局部切除等术式.为了探究胰腺端-端重建在生物力学上的可行性,我们建立了间断缝合术、连续缝合术和“荣式”手术的3种有限元模型,并进行拉伸模拟实验.“荣式”手术能够使拉伸时的应力分布更加均匀,拉伸受力更强,并且减少了缝线对胰腺组织的切割力,从而能够使得胰腺端-端对拢时耐受更大的拉力.胰管(修复)外科的提出明确了保护主胰管连续性和生理功能的重要性,避免扩大性切除手术和消化道重建,有助于促进患者术后恢复和提高远期生活质量,有望改变传统的胰腺良性疾病的手术治疗理念,通过创伤最小化和功能正常化的有机结合,使患者获得最佳的治疗结果. 展开更多

主动脉夹层是由于各种原因导致的主动脉内膜、中膜撕裂,主动脉内膜与中膜分离,血液流入,致使主动脉腔被分割为真腔和假腔。主动脉夹层是死亡风险最高的心血管急症之一,若前48 h内未行手术治疗,其病死率高达50%。近年来随着诊断及外科手... 主动脉夹层是由于各种原因导致的主动脉内膜、中膜撕裂,主动脉内膜与中膜分离,血液流入,致使主动脉腔被分割为真腔和假腔。主动脉夹层是死亡风险最高的心血管急症之一,若前48 h内未行手术治疗,其病死率高达50%。近年来随着诊断及外科手术技术的进步,主动脉夹层病死率显著下降。随着生存率的提高,相关研究发现患者预后与夹层术后并发症密切相关,术后并发症严重影响患者术后康复并降低其术后生活质量。神经系统并发症,尤其是缺血性脑卒中、脊髓缺血,是主动脉夹层术后最常见、最严重的并发症,如何预防术后神经系统并发症并降低其发生率已成为目前的研究热点,如何诊断与治疗术后神经系统并发症也受到神经科医师的广泛关注。本文报道2例主动脉夹层术后并发神经系统并发症患者的诊疗过程,并结合文献进行学习,以提高神经科医师对主动脉夹层术后并发神经系统并发症的警惕性与关注度。 展开更多
