血清4型禽腺病毒强毒株GY株的分离鉴定和致病性研究 被引量:6

从山东某疑似感染禽腺病毒的发病鸡群中采集病料并分离到一株禽腺病毒,克隆纯化后进行PCR检测和序列分析,鉴定该毒株为血清4型禽腺病毒,命名为GY株。用LMH细胞繁殖的GY株病毒滴度可以达到107.5 TCID_(50)/0.1mL,并建立了纯净稳定的种子... 从山东某疑似感染禽腺病毒的发病鸡群中采集病料并分离到一株禽腺病毒,克隆纯化后进行PCR检测和序列分析,鉴定该毒株为血清4型禽腺病毒,命名为GY株。用LMH细胞繁殖的GY株病毒滴度可以达到107.5 TCID_(50)/0.1mL,并建立了纯净稳定的种子批。动物致病性试验结果显示,攻毒鸡只8/10死亡,剖检均出现心包积液,肝脏肿大、出血或坏死,表明该毒株为强毒株。就攻毒途径、攻毒剂量等方面研究发现,GY株经胸部肌肉注射后发病率和发病时间相对颈部皮下注射途径更有优势,确定攻毒途径为肌肉注射;通过比较不同剂量(5×10^(3.0)~5×10^(6.0) TCID50)GY株病毒攻击SPF鸡的致病性,将攻毒剂量定为5×10^(5.0) TCID_(50)。研究表明,GY株可作为禽腺病毒4型攻毒用强毒株。 展开更多

The ultrastructure of porcine kidney(PK)-15 cells was examined after lipofectamine-aided transfection of the molecular clone of the P1 agent.PK-15 cells transfected with the tandem dimer of the P1molecular DNA clone had numbers of intracytoplasmic inclusions,and a few cells had intranuclear inclusions.Intracytoplasmic inclusions were round to oval and 0.1-0.3μm in diameter,and intranuclear inclusions,which were more electron dense,were of two general types:the first were round and small(0.1μm approximately)and the second were hexagonal and larger(0.4-0.8μm in diameter).Cells transfected with the tandem dimer of the P1 molecular DNA clone tested positive for P1 DNA at passage 5.This is the first report that the P1 molecular clone has infectivity in vitro and it will provide fundamental materials for further study of the biological characterization of P1. 展开更多

Porcine reproductive and respiratory syndrome virus.(PRRSV) actively induces cell apoptosis both in vitro and in vivo, which can contribute critically to viral pathogenesis. Previous studies have shown that the PRRSV nonstructural protein 4 (nsp4) is an important mediator of this process, but the underlying molecular details remain poorly understood. In this study, we found that the PRRSV nsp4 interacted with the mitochondrial inner membrane protein cytochrome cl (cyto.cl) and induced its proteolytic cleavage. Interestingly, the cleaved N-terminal fragment of cyto.cl was found to exert apoptotic activity, which could cause mitochondrial fragmentation, resulting in apoptotic cell death. And RNA interference (RNAi) silencing experiments further confirmed the crucial role which cyto.cl played in nsp4- and PRRSV-induced cell apoptosis. Thus, our data provide an important piece of mechanistic clues for PRRSV-induced cell apoptosis and also elucidate a novel mechanism for the 3C-like proteases in this finding. 展开更多

目的:探讨地西他滨联合低剂量CAG(阿糖胞苷+阿柔比星+粒细胞刺激因子)对老年急性髓系白血病(AML)的影响。方法:采用前瞻性研究方法,以我院2017年10月–2020年3月收治的100例老年AML患者为研究对象,按照随机数字表法分为对照组(50例)和... 目的:探讨地西他滨联合低剂量CAG(阿糖胞苷+阿柔比星+粒细胞刺激因子)对老年急性髓系白血病(AML)的影响。方法:采用前瞻性研究方法,以我院2017年10月–2020年3月收治的100例老年AML患者为研究对象,按照随机数字表法分为对照组(50例)和观察组(50例)。对照组给予地西他滨联合标准剂量CAG治疗,观察组给予地西他滨联合低剂量CAG治疗。对比分析两组血液指标、血清免疫指标、治疗效果、不良反应及预后情况。结果:治疗后,观察组患者血红蛋白含量显著高于对照组(P<0.05);观察组患者血清CD4^(+)和CD8^(+)细胞数量显著低于对照组(P<0.05);观察组患者各项不良反应发生率均低于对照组,其中血小板减少发生率与对照组相比差异具有统计学意义(P<0.05)。随访18个月后,观察组患者的总生存率高于对照组(P<0.05)。结论:地西他滨联合低剂量CAG治疗老年AML疗效较好,不良反应发生率低,患者预后较好。 展开更多

The objective of the present investigation was to estimate the prevalence of Toxoplasma gondii infection and co-infection with porcine reproductive and respiratory syndrome virus(PRRSV), classical swine fever virus(CS... The objective of the present investigation was to estimate the prevalence of Toxoplasma gondii infection and co-infection with porcine reproductive and respiratory syndrome virus(PRRSV), classical swine fever virus(CSFV) and porcine circovirus type 2(PCV-2) in pigs in China. A total of 372 tissues or serum samples collected from pigs distributed in 9 provinces/municipalities of China during the period from February 2011 to November 2012 were assayed for T. gondii antigens and antibodies using enzyme linked immunosorbent assay(ELISA) technique, while the PCR was designed for the detection of the PRRSV, CSFV and PCV-2, respectively. The total positive rate of T. gondii, PRSSV, CSFV and PCV-2 was 9.14%(34/372), 50.00%(186/372), 37.10%(138/372) and 3.23%(12/372), respectively. Among the 34 T. gondii positive samples, 26 samples were simultaneously infected with T. gondii and viruses, while the remaining eight samples were infected with T. gondii alone. In addition, the co-infection rate of T. gondii with PRSSV, T. gondii with PRSSV and CSFV, T. gondii with PRSSV and PCV-2, T. gondii with CSFV and PCV-2, T. gondii with PRSSV, CSFV and PCV-2 was 1.61%(6/372), 4.03%(15/372), 0.27%(1/372), 0.27%(1/372) and 0.81%(3/372), respectively. The results of the present survey revealed that PRRSV and CSFV were the common pathogens co-existing with porcine toxoplasmosis in China, and both of them could increase the chances of T. gondii infection in pig. This is the first report of T. gondii co-infections with viruses in pigs. It is very important to understand the interactions of parasite and virus, and can be used as reference data for the control and prevention of co-infections of T. gondii and viruses in pigs. 展开更多

The nonstructural protein 10(nsp10) of porcine reproductive and respiratory syndrome virus(PRRSV) encodes for helicase which plays a vital role in viral replication. In the present study, a truncated form of nsp10, termed nsp10 a, was found in PRRSV-infected cells and the production of nsp10 a was strain-specific. Mass spectrometric analysis and deletion mutagenesis indicated that nsp10 a may be short of about 70 amino acids in the N terminus of nsp10. Further studies by rescuing recombinant viruses showed that the Glu-69 in nsp10 was the key amino acid for nsp10 a production. Finally, we demonstrated that nsp10 a exerted little influence on the growth kinetics of PRRSV in vitro. 展开更多

The objective of this study was to construct the infectious clone of encephalomyocarditis virus (EMCV) BJC3 strain.The genomic cDNA of the virus was amplified by three overlapping segments using RT-PCR,and cloned into low-copy plasmid pWSK29 to construct the full-length cDNA clone pWSKBJC3/ w.The pWSKBJC3/w was in vitro transcribed and transfected into BHK-21 cells to rescue the virus.The results showed that the full-length cDNA clone was infectious and the virus could be rescued in BHK-21 cells.The rescued virus designated RvBJC3W was identified by RT-PCR and indirect immunofluorescence assay(IFA).The rescued virus had similar growth characteristics to its parental virus BJC3 and retained pathogenicity for mice.Our results indicate that the first infectious cDNA clone of EMCV in China has been successfully established and provides an essential tool for investigating the molecular basis of pathogenicity of EMCV. 展开更多
