Effect of Huanglian Jiedu Decoction on the Behavior of Zebrafish with Alzheimer's Disease

[Objectives]To study the effect of Huanglian Jiedu Decoction on the behavior of zebrafish with Alzheimer's disease caused by AlCl 3.[Methods]Each portion of Huanglian Jiedu Decoction was prepared according to the proportion of Coptis chinensis∶Phellodendron chinense Rupr.∶Scutellaria baicalensis Georgi∶Gardenia jasminoides Ellis=63 g∶42 g∶42 g∶63 g.After that,each portion of Huanglian Jiedu Decoction was soaked in 6.3 L water for 30 min and boiled twice at 100℃.The extracts were combined twice and filtered,then concentrated to 308 g/L and put into refrigerator for later use.Before training,zebrafishes were put into T-maze to adapt for 2 d,and then behavioral training was carried out for 7 d.After video recording,the behavior of zebrafish was analyzed by Smart 3.0,and qualified zebrafishes were selected for follow-up experiments.Then 60 successfully trained zebrafishes were randomly divided into control group,model group,positive group,low-dose group,medium-dose group and high-dose group of Huanglian Jiedu Decoction.Except for the control group,all the other groups were exposed to 100μg/L AlCl 3 for 7 d.After that,video was recorded,and behavioral analysis was carried out with behavioral record and analysis software Smart 3.0.And then the zebrafishes in the other four groups except the model group were treated with Huperzine A(4μg/L)and Huanglian Jiedu Decoction(154,308,616 mg/L)for 6 d,respectively.After that,it was recorded and the behavior of each group was analyzed.[Results]There was a significant difference in the time spent in the left area and the percentage of time in the left area between the control group and the model group(P<0.001).The time spent in the left area and the percentage of time in the left area in the model group and positive group,low,medium and high dose groups of Huanglian Jiedu Decoction decreased significantly(P<0.05,P<0.01,P<0.001).The swimming distance in the left area and the percentage of swimming distance in the left area in the model group were significantly higher than those in the control group(P<0.001).There was a significant difference in swimming distance between model group and positive group,low,medium and high dose groups of Huanglian Jiedu Decoction(P<0.01,P<0.001).In the percentage of swimming distance in the left area,there was a significant difference between the model group and the low and high dose groups of Huanglian Jiedu Decoction(P<0.01,P<0.001).[Conclusions]Huanglian Jiedu Decoction can improve the behavior of zebrafish with Alzheimer's disease. 展开更多

Intraocular pressure elevation can induce retinal ganglion cell death and is a clinically reversible risk factor for glaucoma,the leading cause of irreversible blindness.We previously demonstrated that casein kinase-2 inhibition can promote retinal ganglion cell survival and axonal regeneration in rats after optic nerve injury.To investigate the underlying mechanism,in the current study we increased the intraocular pressure of adult rats to 75 mmHg for 2 hours and then administered a casein kinase-2 inhibitor(4,5,6,7-tetrabromo-2-azabenzimidazole or 2-dimethylamino-4,5,6,7-tetrabromo-1H-benzimidazole)by intravitreal injection.We found that intravitreal injection of 4,5,6,7-tetrabromo-2-azabenzimidazole or 2-dimethylamino-4,5,6,7-tetrabromo-1H-benzimidazole promoted retinal ganglion cell survival and reduced the number of infiltrating macrophages.Transcriptomic analysis showed that the mitogen activated protein kinase signaling pathway was involved in the response to intraocular pressure elevation but was not modulated by the casein kinase-2 inhibitors.Furthermore,casein kinase-2 inhibition downregulated the expression of genes(Cck,Htrsa,Nef1,Htrlb,Prph,Chat,Slc18a3,Slc5a7,Scn1b,Crybb2,Tsga10ip,and Vstm21)involved in intraocular pressure elevation.Our data indicate that inhibition of casein kinase-2 can enhance retinal ganglion cell survival in rats after acute intraocular pressure elevation via macrophage inactivation. 展开更多

Melt treatment is well known to have an important influence on the properties of metallic glasses(MGs).However,for the MGs quenched from different melt temperatures with a quartz tube,the underlying physical origin responsible for the variation of properties remains poorly understood.In the present work,we systematically studied the influence of melt treatment on the thermal properties of a Zr50Cu36Al14 glass-forming alloy and unveiled the microscopic origins.Specifically,we quenched the melt at different temperatures ranging from 1.1Tl to 1.5Tl(Tl is the liquidus temperature)to obtain melt-spun MG ribbons and investigated the variation of thermal properties of the MGs upon heating.We found that glass transition temperature,Tg,increases by as much as 36 K,and the supercooled liquid region disappears in the curve of differential scanning calorimetry when the melt is quenched at a high temperature up to 1.5Tl.The careful chemical analyses indicate that the change in glass transition behavior originates from the incorporation of oxygen and silicon in the molten alloys.The incorporated oxygen and silicon can both enhance the interactions between atoms,which renders the cooperative rearrangements of atoms difficult,and thus enhances the kinetic stability of the MGs. 展开更多

In this study, we present a catalog of molecular clumps extracted from ^(13)CO(J=1-0) emission data of the Milky Way Imaging Scroll Painting(MWISP) project. The data covers the inner Milky Way within the longitude range10°≤l≤20° and the latitude strip of |b|≤ 5°. 25. The workflow for the extraction of clumps, namely Facet-SS-3DClump, consists of two parts: the identification of clump candidates and their verification. First, Facet-SS-3DClump employs FacetClumps to identify clump candidates. Subsequently, high-confidence clumps are obtained by cross-matching with the clumps detected by other algorithms, such as dendrogram. Second, these high-confidence clumps are used as prior knowledge to train a semi-supervised deep clustering approach, SS-3D-Clump, which is applied to verify clump candidates detected by FacetClumps, providing confidence levels for the molecular clumps. Finally, the catalog comprising 18,757 molecular clumps was obtained using Facet-SS-3D-Clump, and the catalog is 90% complete above 37 K km s^(-1). We observe a significant deviation of the mean Galactic latitude for clumps within |b|≤ 2° from the midplane, with b=-0°.110. We found that 82.3% of the dust clumps correspond to ^(13)CO clumps by matching with Herschel infrared dust clumps. In the future, Facet-SS-3D-Clump will be applied to detect ^(13)CO clumps in the entire MWISP data. 展开更多

Influence of liquid film shape on evaporation performance of agitated thin film evaporator

The agitated thin film evaporator(ATFE),which is known for its high efficiency,force the material to form a film through the scraping process of a scraper,followed by evaporation and purification.The complex shape of the liquid film inside the evaporator can significantly affect its evaporation capability.This work explores how change in shape of the liquid films affect the evaporation of the materials with non-Newtonian characteristics,achieved by changing the structure of the scraper.Examining the distribution of circumferential temperature,viscosity,and mass transfer of the flat liquid film shows that the film evaporates rapidly in shear-thinning region.Various wavy liquid films are developed by using shear-thinning theory,emphasizing the flow condition in the thinning area and the factors contributing to the exceptional evaporation capability.Further exploration is conducted on the spread patterns of the wavy liquid film and flat liquid film on the evaporation wall throughout the process.It is noted that breaking the wavy liquid film on the evaporating wall during evaporation is challenging due to its film-forming condition.For which the fundamental causes are demonstrated by acquiring the data regarding the flow rate and temperature of the liquid film.The definitive findings of the analysis reveal a significant improvement in the evaporation capability of the wavy liquid film.This enhancement is attributed to increasing the shear-thinning areas and maintaining the overall shape of the film throughout the entire evaporation process. 展开更多

Plasma equilibrium reconstruction provides essential information for tokamak operation and physical analysis.An extensive and reliable set of magnetic diagnostics is required to obtain accurate plasma equilibrium.This study designs and optimizes the magnetic diagnostics layout for the reconstruction of the equilibrium of the plasma according to the scientific objectives,engineering design parameters,and limitations of the Chinese Fusion Engineering Test Reactor(CFETR).Based on the CFETR discharge simulation,magnetic measurement data are employed to reconstruct consistent plasma equilibrium parameters,and magnetic diagnostics'number and position are optimized by truncated Singular value decomposition,verifying the redundancy reliability of the magnetic diagnostics layout design.This provides a design solution for the layout of the magnetic diagnostics system required to control the plasma equilibrium of CFETR,and the developed design and optimization method can provide effective support to design magnetic diagnostics systems for future magnetic confinement fusion devices. 展开更多

[目的]地铁盾构隧道因上方河道开挖卸荷产生隆起变形,可能会影响地铁隧道的正常运行。为此,应就软土地区河道开挖工程对下方地铁隧道的变形影响进行研究,并制定地铁隧道变形控制措施。[方法]介绍了案例工程的概况,采用有限元方法对其实测数据进行分析,验证了土体本构模型及参数的合理性。基于具有典型软土特征的宁波地区地层剖面和土层参数,建立了土体简化模型,依次分析了河道开挖宽度变化对下方地铁隧道变形的影响、河道开挖深度变化对下方地铁隧道变形的影响、软土层埋深对地铁隧道变形的影响。介绍了门架式土体加固措施及门架式梁板加固措施两种变形控制措施,将考虑了控制变形措施的计算模型与基准模型的有限元计算结果进行了比较分析。[结果及结论]在土体加固和河道开挖阶段,下方地铁隧道变形分别表现为沉降和隆起;河道开挖断面面积相同时,减小河道的开挖深度、增加河道开挖宽度,有利于减少下方地铁隧道变形;地铁隧道竖向位移主要由其下方土体回弹变形产生;门架式梁板加固措施对于地铁隧道隆起变形的控制效果明显。 展开更多

A feedback control of fuel recycling via real-time boron powder injection,addressing the issue of continuously increasing recycling in long-pulse plasma discharges,has been successfully developed and implemented on EAST tokamak.The feedback control system includes four main parts:the impurity powder dropper(IPD),a diagnostic system measuring fuel recycling level represented by D_(α)emission,a plasma control system(PCS)implementing the Proportional Integral Derivative(PID)algorithm,and a signal converter connecting the IPD and PCS.Based on this control system,both active control and feedback control experiments have recently been performed on EAST with a full metal wall.The experimental results show that the fuel recycling can be gradually reduced to lower level as PCS control voltage increases.In the feedback control experiments,it is also observed that the D_(α)emission is reduced to the level below the target D_(α)value by adjusting boron injection flow rate,indicating successful implementation of the fuel recycling feedback control on EAST.This technique provides a new method for fuel recycling control of long pulse and high parameter plasma operations in future fusion devices. 展开更多

We have experimentally achieved the all-optical trapping of a ^(40)Ca^(+)ion.An optical dipole trap was established using a high-power,far-detuned,tightly focused laser with a wavelength of 532 nm.The single ^(40)Ca^(+)ion was trapped without any RF fields and demonstrated a long lifetime of over 3 s.In this experiment,we implemented several measures to improve the optical trapping probability,including focusing the dipole beam waist near the diffraction limit,precisely compensating for stray electric fields,and mitigating electron shelving in metastable states.The optical trapping of a ^(40)Ca^(+)ion eliminates the influence of micromotion induced by RF fields,potentially paving the way for development of all-optical trapping ion optical clocks. 展开更多

为了应对国产雷达所有元器件均国产化需求,设计了一款工作频点在1.2GHZ~1.4 GHz的国产氮化镓(gallium nitride,GaN)固态雷达发射机。使用国产GaN功率模块对发射系统最大限度地进行小型化设计,降低组件的重量,加强设备的高可靠性及维护... 为了应对国产雷达所有元器件均国产化需求,设计了一款工作频点在1.2GHZ~1.4 GHz的国产氮化镓(gallium nitride,GaN)固态雷达发射机。使用国产GaN功率模块对发射系统最大限度地进行小型化设计,降低组件的重量,加强设备的高可靠性及维护方便性设计,进一步完善GaN功率模块在线测试功能,使产品具有高可靠性和完善的雷达故障诊断功能。增加系统的监控、检测及显示功能,并将各种工作状态信号都传输到主监控上,使系统的各种工作状态都能够在终端上显示,使操作人员通过终端界面便可以检查发射系统的各种工作参数,判断发射机的工作状态。在电磁兼容方面通过谐波滤波器对二次谐波的抑制,测试结果表明,发射系统输出信号的二次谐波小于等于-66dB。 展开更多

新型水凝胶微针具有安全包裹多相药物、药物持续、可控释放等优点,在微针透皮给药领域极具应用潜力。文中重点研究了交联透明质酸微针贴片的优化工艺,并对微针贴片进行了性能评价。在碱性条件下使用1,4-丁二醇二缩水甘油醚(BDDE)为交联... 新型水凝胶微针具有安全包裹多相药物、药物持续、可控释放等优点,在微针透皮给药领域极具应用潜力。文中重点研究了交联透明质酸微针贴片的优化工艺,并对微针贴片进行了性能评价。在碱性条件下使用1,4-丁二醇二缩水甘油醚(BDDE)为交联剂对透明质酸进行交联,利用微浇注法制备交联透明质酸微针(cHA-MN),并优化其工艺。对所制备的微针贴片进行了外观形貌、力学性能、体外刺入性能及溶胀性能表征,验证其能否满足使用要求。实验结果表明,使用最优工艺制备的交联透明质酸微针贴片力学性能优良,可以顺利刺破皮肤,并实现溶胀功能。使用制备的微针搭载2.5%的右旋布洛芬药物,体外透皮扩散结果表明,微针在0~6 h释放出约38%药物,随后以一定速度缓慢释放至42 h,累计释放率约90%。 展开更多

The present work investigates an analysis of Eco-Tourism Climate Resources in Xingwe<span>n, China based on the Comfort Index and Negative Air (Oxygen) Ion (“NAI” for short in this article) concentration. The daily temperature, precipitation, wind speed, sunshine hours, and relative humidity data of the Xingwen National Meteorological Station from 1999 to 2018, as well as the NAI data of Feiwu Cave station and Shoushan Lake station in Xingwen of 2018 were used in this study. Based on the analysis of the characteristics of basic meteorological elements, the index of climate comfort of human living environment and the NAI variation in Xingwen, the eco-tourism climate resources in this area were evaluated comprehensively. The results show that: The climate is mild, the precipitation is abundant and the seasonal variation trend of precipitation and heat is similar in the study area. The annual average temperature of this area is 17.8<span style="color:#4F4F4F;font-family:-apple-system, "font-size:16px;white-space:normal;background-color:#FFFFFF;">&#176;</span>C, the annual average precipitation is 1096 mm, the annual average precipitation days are 186 days, the annual average relative humidity is 81%, the annual average wind speed is 1.3 m/s, and the annual average sunshine </span><span>hours are 999 h. The living environment comfort level reaches the “comfort” level for 6 months per year. In addition, the concentration of NAI in Xingwen remains at the highest level 7 throughout the year, and the rate of good air quality is extremely high. Overall, the study area has high-quality ecological climate resources that are conducive to the development of forest recuperation, leisure and vacation and other ecological tourism activities.</span> 展开更多
