Recent studies have proposed three lymphatic drainage systems in the brain,that is,the glymphatic system,the intramural periarterial drainage pathway,and meningeal lymphatic vessels,whose roles in various neurological diseases have been widely explored.The glymphatic system is a fluid drainage and waste clearance pathway that utilizes perivascular space and aquaporin-4 protein located in the astrocyte endfeet to provide a space for exchange of cerebrospinal fluid and interstitial fluid.The intramural periarterial drainage pathway drives the flow of interstitial fluid through the capillary basement membrane and the arterial tunica media.Meningeal lymphatic vessels within the dura mater are involved in the removal of cerebral macromolecules and immune responses.After ischemic stroke,impairment of these systems could lead to cerebral edema,accumulation of toxic factors,and activation of neuroinflammation,while restoration of their normal functions can improve neurological outcomes.In this review,we summarize the basic concepts of these drainage systems,including drainage routes,physiological functions,regulatory mechanisms,and detection technologies.We also focus on the roles of lymphatic drainage systems in brain injury after ischemic stroke,as well as recent advances in therapeutic strategies targeting these drainage systems.These findings provide information for potential novel strategies for treatment of stroke. 展开更多

Spinal cord injury causes accumulation of a large number of leukocytes at the lesion site where they contribute to excessive inflammation.Overproduced chemokines are responsible for the migratory process of the leukocytes,but the regulatory mechanism underlying the production of chemokines from resident cells of the spinal cord has not been fully elucidated.We examined the protein levels of macrophage migration inhibitory factor and chemokine C-C motif chemokine ligand 2 in a spinal cord contusion model at different time points following spinal cord injury.The elevation of macrophage migration inhibitory factor at the lesion site coincided with the increase of chemokine C-C motif chemokine ligand 2 abundance in astrocytes.Stimulation of primary cultured astrocytes with different concentrations of macrophage migration inhibitory factor recombinant protein induced chemokine C-C motif chemokine ligand 2 production from the cells,and the macrophage migration inhibitory factor inhibitor 4-iodo-6-phenylpyrimidine attenuated the stimulatory effect.Further investigation into the underlying mechanism on macrophage migration inhibitory factor-mediated astrocytic production of chemokine C-C motif chemokine ligand 2 revealed that macrophage migration inhibitory factor activated intracellular JNK signaling through binding with CD74 receptor.Administration of the macrophage migration inhibitory factor inhibitor 4-iodo-6-phenylpyrimidine following spinal cord injury resulted in the reduction of chemokine C-C motif chemokine ligand 2-recruited microglia/macrophages at the lesion site and remarkably improved the hindlimb locomotor function of rats.Our results have provided insights into the functions of astrocyte-activated chemokines in the recruitment of leukocytes and may be beneficial to develop interventions targeting chemokine C-C motif chemokine ligand 2 for neuroinflammation after spinal cord injury. 展开更多

The low intrinsic growth capacity of neurons and an injury-induced inhibitory milieu are major contributo rs to the failure of sensory and motor functional recovery following spinal cord injury.Heat shock transcription factor 1(HSF1),a master regulator of the heat shock response,plays neurogenetic and neuroprotective roles in the damaged or diseased central nervous system.However,the underlying mechanism has not been fully elucidated.In the present study,we used a gecko model of spontaneous nerve regeneration to investigate the potential roles of gecko HSF1(gHSF1) in the regulation of neurite outgrowth and inflammatory inhibition of macrophages following spinal cord injury.gHSF1 expression in neurons and microglia at the lesion site increased dramatically immediately after tail amputation.gHSF1 ove rexpression in gecko primary neuro ns significantly promoted axonal growth by suppressing the expression of suppressor of cytokine signaling-3,and fa cilitated neuro nal survival via activation of the mitogen-activated extracellular signal-regulated kinase/extracellular regulated protein kinases and phosphatidylinositol 3-kinase/protein kinase B pathways.Furthermore,gHSF1 efficiently inhibited the macrophagemediated inflammatory response by inactivating 1kappa B-alpha/NF-kappaB signaling.Our findings show that HSF1 plays dual roles in promoting axonal regrowth and inhibiting leukocyte inflammation,and provide new avenues of investigation for promoting spinal co rd injury repair in mammals. 展开更多

目的考察S100β蛋白在新生大鼠脑损伤中的作用及重组人红细胞生成素(rhEPO)干预机制的研究。方法将2日龄SD大鼠分成3组:假手术组、缺氧缺血脑损伤(HIBD)模型组(HIBD组)和rhEPO治疗组(rhEPO组),每组40只。HIBD组和rhEPO组大鼠组均存在HIB... 目的考察S100β蛋白在新生大鼠脑损伤中的作用及重组人红细胞生成素(rhEPO)干预机制的研究。方法将2日龄SD大鼠分成3组:假手术组、缺氧缺血脑损伤(HIBD)模型组(HIBD组)和rhEPO治疗组(rhEPO组),每组40只。HIBD组和rhEPO组大鼠组均存在HIBD,rhEPO组腹腔注射rhEPO 5 000 u/(kg·次)。采用ELISA法检测大鼠出生后第3、5和7天及治疗后6和24 h,第3和7天外周血的S100β蛋白表达水平;选取2015年2月—2017年10月在大连医科大学附属二院妇产科或新生儿科确诊为脑损伤的新生儿25例,选择同期出生的健康新生儿29例为对照。脑损伤的新生儿均给予皮下注射rhEPO。采用ELISA法检测rhEPO治疗前和治疗后第3、7和14天外周血的S100β蛋白表达水平。结果 3组大鼠出生后第3、5和7天外周血S100β蛋白表达水平比较:①不同时间点的S100β蛋白表达无差异(P>0.05);②3组S100β蛋白表达无差异(P>0.05);③3组的S100β蛋白表达变化趋势无差异(P>0.05)。3组大鼠治疗后6、24 h,第3和7天外周血S100β蛋白表达比较:①不同时间点的S100β蛋白表达有差异(P <0.05);②3组的S100β蛋白表达有差异(P <0.05),rhEPO组和HIBD组较假手术组高;③3组的S100β蛋白表达变化趋势有差异(P <0.05)。两组新生儿治疗前、治疗后第3、7和14天外周血S100β蛋白表达比较:①不同时间点的S100β蛋白表达有差异(P <0.05);②两组S100β蛋白表达有差异(P <0.05),脑损伤组较对照组高;③两组的S100β蛋白表达变化趋势有差异(P <0.05)。脑损伤组外周血的S100β蛋白表达呈双峰变化。对照组,轻度和重度脑损伤新生儿组治疗前、治疗后第3、7和14天外周血S100β蛋白表达比较:①不同时间点的S100β蛋白表达有差异(P <0.05);②3组的S100β蛋白表达有差异(P <0.05),重度脑损伤组较其他两组高;③3组的S100β蛋白表达变化趋势有差异(P <0.05)。轻度和重度脑损伤新生儿组外周血的S100β蛋白表达呈双峰变化。结论 S100β蛋白在新生大鼠及新生儿脑损伤可呈双峰变化,其中轻度新生儿脑损伤的S100β蛋白无双峰变化,rhEPO可用于治疗新生大鼠及新生儿脑损伤。 展开更多

AIM: To establish a method detecting porcine endogenous retrovirus (PERV) in China experimental minipigs and to evaluate the safety of PERV in three individuals treated with bioartificial liver support systems based on porcine hepatocytes. METHODS: Porcine hepatocytes were isolated with two-stage perfusion method, then cultured in the bioreactor, which is separated by a semipermeable membrane (0.2μm) from the lumen through which the patients' blood plasma was circulated. After posthemoperfusion, patients' blood was obtained for screening. Additionally, samples of medium collected from both intraluminal and extraluminal compartments of the laboratory bioreactor and culture supernate in vitro was analyzed. The presence of viral sequences was estimated by polymerase chain reaction (PCR) and reverse transcriptase-polymerase chain reaction (RTPCR). Finally, the infection of virus in the supernate of common culture was ascertained by exposure to the fetal liver cells. RESULTS: PERV-specific gag sequences were found in the porcine hepatocytes using RT-PCR. and were detected in all samples from the intraluminal, extraluminal samples and culture supernate. However, culture supernatant from primary porcine hepatocytes (cleared of cellular debris) failed to infect human fetal liver cells. Finally, RT-PCR detected no PERV infection was found in the blood samples obtained from three patients at various times post-hemoperfusion. CONCLUSION: The assays used are specific and sensitive, identified by second PCR. PERVs could be released from hepatocytes cultured in bioreactor without the stimulation of mitogen and could not be prevented by the hollow fiber semipermeable membrane, indicating the existence of PERV safety in extracorporeal bioartificial liver support system (EBLSS). 展开更多

High mobility group box 1(HMGB1) interacts with pattern-recognition receptors of immune cells to activate the inflammatory response. Astrocytes play a positive role in the inflammatory response of the central nervous system by expressing a broad range of patternrecognition receptors. However, the underlying relationship between HMGB1 and the inflammatory reaction of astrocytes remains unclear. In this study, we established rat models of spinal cord injury via laminectomy at the T8–10 level, and the injured spinal cord was subjected to transcriptome sequencing. Our results showed that the HMGB1/Toll-like receptor 4(TLR4) axis was involved in the activation of astrocyte inflammatory response through regulation of cyclooxygenase 2(COX2)/prostaglandin E2(PGE2) signaling. Both TLR4 and COX2 were distributed in astrocytes and showed elevated protein levels following spinal cord injury. Stimulation of primary astrocytes with recombinant HMGB1 showed that COX2 and microsomal PGE synthase(mPGES)-1, rather than COX1, mPGES-2, or cytosolic PGE synthase, were significantly upregulated. Accordingly, PGE2 production in astrocytes was remarkably increased in response to recombinant HMGB1 challenges. Pharmacologic blockade of TLR2/4 attenuated HMGB1-mediated activation of the COX2/PGE2 pathway. Interestingly, HMGB1 did not impact the production of tumor necrosis factor-α or interleukin-1β in astrocytes. Our results suggest that HMGB1 mediates the astrocyte inflammatory response through regulating the COX2/PGE2 signaling pathway. A new signal readout method for position-sensitive multi-output detectors,such as those in high-energy spectroscopy measurement and nuclear imaging,was developed by combining the charge division circuit,summing circuit and charge-to-time conversion(QTC) circuit.The 64 outputs of a Hamamatsu H8500 position-sensitive photomultiplier tube were processed,and three digital pulses were generated.The widths of digital pulses were determined using the time-to-digital converter in an field programmable gate array.The energy and position information of incident y-rays is estimated based on the proportionality between the width of digital pulses and input charge created by y-photons.A prototype was built using discrete components and tested,and the energy and position resolutions were improved compared with that obtained with standard ADCs.This method greatly simplifies the front-end electronics and the digital interface.It enables a compact electronics system and an easy integration into an ASIC. 展开更多

Schwann cells are not only myelinating cells, but also function as immune cells and express numerous innate pattern recognition receptors, including the Toll-like receptors. Injury to peripheral nerves activates an inflammatory response in Schwann cells. However, it is unclear whether specific endogenous damage-associated molecular pattern molecules are involved in the inflammatory response following nerve injury. In the present study, we demonstrate that a key damage-associated molecular pattern molecule, high mobility group box 1(HMGB1), is upregulated following rat sciatic nerve axotomy, and we show colocalization of the protein with Schwann cells. HMGB1 alone could not enhance expression of Toll-like receptors or the receptor for advanced glycation end products(RAGE), but was able to facilitate migration of Schwann cells. When Schwann cells were treated with HMGB1 together with lipopolysaccharide, the expression levels of Toll-like receptors and RAGE, as well as inflammatory cytokines were upregulated. Our novel findings demonstrate that the HMGB1 pathway activates the inflammatory response in Schwann cells following peripheral nerve injury. 展开更多

A three-dimensional fluid model is developed to investigate the radio-frequency inductively coupled H2 plasma in a reactor with a rectangular expansion chamber and a cylindrical driver chamber,for neutral beam injection system in CFETR.In this model,the electron effective collision frequency and the ion mobility at high E-fields are employed,for accurate simulation of discharges at low pressures(0.3 Pa-2 Pa)and high powers(40 kW-100 kW).The results indicate that when the high E-field ion mobility is taken into account,the electron density is about four times higher than the value in the low E-field case.In addition,the influences of the magnetic field,pressure and power on the electron density and electron temperature are demonstrated.It is found that the electron density and electron temperature in the xz-plane along permanent magnet side become much more asymmetric when magnetic field enhances.However,the plasma parameters in the yz-plane without permanent magnet side are symmetric no matter the magnetic field is applied or not.Besides,the maximum of the electron density first increases and then decreases with magnetic field,while the electron temperature at the bottom of the expansion region first decreases and then almost keeps constant.As the pressure increases from 0.3 Pa to 2 Pa,the electron density becomes higher,with the maximum moving upwards to the driver region,and the symmetry of the electron temperature in the xz-plane becomes much better.As power increases,the electron density rises,whereas the spatial distribution is similar.It can be summarized that the magnetic field and gas pressure have great influence on the symmetry of the plasma parameters,while the power only has little effect. 展开更多

Objective:To research the effect of conventional treatment(Gingko biloba extract(GBE)+continuous positive airway pressure(CPAP)ventilation)combined with acupuncture on the changes of blood pressure,respiratory efficiency,oxidative index,and clinical effect in patients with OSAHS combined with hypertension.Method Selecting 80 patients with OSAHS combined with hypertension,then they were randomly divided into observation group(n=40)and control group(n=40).The patients in the control group were given conventional drug therapy(GBE)and rehabilitation and CPAP,and the observation group were given acupuncture on this basic,they were treated 14 days.The primary endpoints were the changes from baseline to day 14 in blood pressure;the secondary endpoints were changes from baseline to day 14 in the respiratory efficiency(PSG);the tertiary endpoints were changes from baseline to day 14 in the oxidative related substances(SOD,GSH-Px,ROS,and MDA).Results After 14-day treatment,the respiratory efficiency index were better than pretherapy(p<0.05),and the observation group were better than control group(p<0.05);the blood pressure were lower than pretherapy(p<0.05),and the observation group were lower than control group(p<0.05);the ROS and MDA were lower than pretherapy(p<0.05),and the observation group were lower than control group(p<0.05);the SOD,GSH-Px were higher than pretherapy(p<0.05),and the observation group were higher than control group(p<0.05);the clinical effect of hypotensive in the observation group were better control group(p<0.05).Conclusion 14-day treatment of conventional treatment combined with acupuncture can improve the imbalance of oxidation and antioxidation and blood pressure. 展开更多

Case studies have reported on the application of bevacizumab in the treatment of severe brain edema. Waves occurring in cold-rolled plates or sheets can be divided into longitudinal and transverse waves. Classical leveling theories merely solve the problem of longitudinal waves, while no well accepted method can be employed for transverse waves. In order to investigate the essential deformation law of leveling for plates with transverse waves, a 2.5-dimensional (2.5- D) analytical approach was proposed. In this model, the plate was transversely divided into some strips with equal width; the strips are considered to be in the state of plane strain and each group of adjacent strips are assumed to be deformation compatible under stress. After calculation, the bending deformation of each strip and the leveling effect of overall plate were obtained by comprehensNe consideration of various strips along with the width. Bending of roller is a main approach to eliminate the transverse waves, which is widely accepted by the industry, but the essential effect of bending of roller on the deformation of plates and the calculation of bending of roller are unknown. According to the 2.5-D analytical model, it can be found that, for plates, it is neutral plane offsetting and middle plane elongation or contraction under inner stress that can effectively improve plate shape. Taking double side waves as an example, the appropriate values of bending of roller were obtained by the 2.5-D analytical model related to different initial unevenness, which was applicable to the current on-line adjusting of bending of roller in rolling industry. 展开更多

AMCheX: Accurate Analysis of Missing-Check Bugs for Linux Kernel

The Linux kernel adopts a large number of security checks to prevent security-sensitive operations from being executed under unsafe conditions.If a security-sensitive operation is unchecked,a missing-check issue arises.Missing check is a class of severe bugs in software programs especially in operating system kernels,which may cause a variety of security issues,such as out-of-bound accesses,permission bypasses,and privilege escalations.Due to the lack of security specifications,how to automatically identify security-sensitive operations and their required security checks in the Linux kernel becomes a challenge for missing-check analysis.In this paper,we present an accurate missing-check analysis method for Linux kernel,which can automatically infer possible security-sensitive operations.Particularly,we first automatically identify all possible security check functions of Linux.Then according to their callsites,a two-direction analysis method is leveraged to identify possible security-sensitive operations.A missing-check bug is reported when the security-sensitive operation is not protected by its corresponding security check.We have implemented our method as a tool,named AMCheX,on top of the LLVM(Low Level Virtual Machine)framework and evaluated it on the Linux kernel.AMCheX reported 12 new missing-check bugs which can cause security issues.Five of them have been confirmed by Linux maintainers. 展开更多
