Heat shock protein 72 confers protection in retinal ganglion cells and lateral geniculate nucleus neurons via blockade of the SAPK/JNK pathway in a chronic ocular-hypertensive rat model 被引量:10

Optic nerve transection increased the expression of heat shock protein 72 (HSP72) in the lateral geniculate body, indicating that this protein is involved in the prevention of neuronal injury. Zinc sulfate and quercetin induced and inhibited the expression of HSP72, respectively. Intraperitoneal injections of zinc sulfate, SP600125 (c-Jun N-terminal kinase inhibitor), or quercetin were performed on retinal ganglion cells in a Wistar rat model of chronic ocular hypertension. Our results showed that compared with the control group, the expression of HSP72 in retinal ganglion cells and the lateral geniculate body was increased after the injection of zinc sulfate, but was decreased after the injection of quercetin. The expression of phosphorylated c-Jun N-terminal kinases and phosphorylated c-Jun were visible 3 days after injection in the control group, and reached apeak at 7 days. Zinc sulfate and SP600125 significantly decreased the expression of p-c-Jun, whereas quercetin significantly enhanced the expression of this protein. These results suggest that HSP72 protects retinal ganglion cells and lateral geniculate body in a rat model of chronic ocular hypertension from injury by blocking the activation of the stress-activated kinase/c-Jun N-terminal kinase apoptotic pathway. 展开更多

Slope reliability analysis considering inherent spatial variability(ISV)of soil properties is timeconsuming when response surface method(RSM)is used,because of the"curse of dimensionality".This paper proposes an effective method for identification of representative slip surfaces(RSSs)of slopes with spatially varied soils within the framework of limit equilibrium method(LEM),which utilizes an adaptive K-means clustering approach.Then,an improved slope reliability analysis based on the RSSs and RSM considering soil spatial variability,in perspective of computation efficiency,is established.The detailed implementation procedure of the proposed method is well documented,and the ability of the method in identifying RSSs and estimating reliability is investigated via three slope examples.Results show that the proposed method can automatically identify the RSSs of slope with only one evaluation of the conventional deterministic slope stability model.The RSSs are invariant with the statistics of soil properties,which allows parametric studies that are often required in slope reliability analysis to be efficiently achieved with ease.It is also found that the proposed method provides comparable values of factor of safety(FS)and probability of failure(Pf)of slopes with those obtained from direct analysis and lite rature. 展开更多

In this paper,dendritic Bi film electrodes with porous structure had successfully been prepared on glassy carbon electrode using a constant current electrolysis method based on hydrogen bubble dynamic templates.The electrode prepared using a large applied current density showed an increased internal electroactive area and a significantly improved electrochemical performance.The analytical utility of the prepared dendritic Bi film electrodes for the determination of Pb(Ⅱ)and Cd(Ⅱ)in the range of 5–50 μg·L^(-1)were presented in combination with square wave stripping voltammetry in model solution.Compared with non-porous Bi film electrode,the dendritic Bi film electrode exhibited higher sensitivity and lower detection limit.The prepared Bi film electrode with dendritic structure was also successfully applied to real water sample analysis. 展开更多

Morphological and functional abnormalities of vascular endothelial cells(VECs) are risk factors of ischemiareperfusion in skin flaps.Signaling pathway mediated by interleukin-1 receptor(IL-1 R) is essential to hypoxia/reoxygenation(H/R) injury of VECs.While the TIR/BB-loop mimetic(AS-1) disrupts the interaction between IL-1 R and myeloid differentiation primary-response protein 88(MyD88),its role in the VECs dysfunction under H/R is unclear.In this study,we first showed that there was an infiltration of inflammatory cells and the apoptosis of VECs by using a skin flap section from patients who received flap transplantation.We then showed that the H/R treatment induced apoptosis and loss of cell migration of endothelial cell line H926 were attenuated by AS-1.Furthermore,our data suggested that AS-1 inhibits the interaction between IL-1 R and MyD88,and subsequent phosphorylation of IκB and p38 pathway,as well as the nuclear localization of NF-κB subunit p65/p50.Thus,this study indicated that the protective role of AS-1 in H/R induced cellular injury may be due to the AS-1 mediated down-regulation of IL-1 R signaling pathway. 展开更多

BACKGROUND: Studies have demonstrated iron deposition in Alzheimer's disease (AD) patients. Therefore, quantitative measurements and tracing of iron deposition are important for early detection and treatment.OBJECTIVE: To quantitatively measure iron deposition in the brain and evaluate the relationship between iron deposition and AD using magnetic susceptibility-weighted imaging. DESIGN, TIME AND SETTING: A case-control study was performed at the Department of Radiology, Huashan Hospital of Fudan University from February to July 2008. PARTICIPANTS: A total of 20 AD patients, comprising 11 males and 9 females, with a mean age of 72.5 years (range, 51-80 years), and 20 healthy volunteers, comprising 10 males and 10 females, with a mean age of 69.9 years (range, 55-78 years), were selected. AD was diagnosed according to the National Institute of Neurological and Communicative Disorders and Stroke-Alzheimer Disease and Related Disorders Association criteria.METHODS: Sagittal T2-weighted images were acquired to locate precise positions of the anterior and posterior commissures. The susceptibility-weighted magnetic resonance images were parallel to the anterior-posterior commissural line through the use of a three-dimensional gradient-echo sequence. All participants underwent measurement of corrected phase (CP) value and Mini-Mental State Examination. Pearson correlation coefficients were used to assess the association between CP values and Mini-Mental State Examination results.MAIN OUTCOME MEASURES: CP values of regions of interest in the hippocampal regions were measured on CP images.RESULTS: CP values were significantly reduced in bilateral hippocampal regions of AD patients compared with normal controls (P〈0.01), but there were no significant differences between left and right CP values (P〉0.05). The hippocampal mean CP value positively correlated with the Mini-Mental State Examination score in AD patients and normal controls (r= 0.57, P〈0.01). CONCLUSION: Susceptibility-weighted imaging can be used to measure CP values to determine iron deposition in the brains of AD patients and could serve as a useful diagnostic tool for AD. 展开更多

Background:Melanocytic nevus is mainly treated by complete or partial removal.However,predicting the risk of malignant transformation of melanocytic nevi and which treatment patients should receive,surgical or nonsurgical management,to gain the best results and aesthetic outcomes is controversial.Methods:Global literature on melanocytic nevus treatment,published between 1997 and 2022,was scanned using the Web of Science Core Collection database.Microsoft Office Excel,CiteSpace V,VOSviewer,Scimago Graphica,Bibliometrix,and Biblioshiny packages in R were used for the bibliometric analysis to summarize the leading countries,institutions,professors,and research trends in this field.Results:This study included 1723 articles.Publications and citations exhibited positive trends over the past 20 years.The United States had the most productive organizations and publications in the comprehensive worldwide cooperation network,and China was recently one of the most active major participants.Professor Giovanni Pellacani,whose H-index,G-index,and M-index ranked first in this field,founded a virtual biopsy using reflectance confocal microscopy.In addition,Krengel and Kinsler contributed significantly to diagnosing and treating melanocytic nevi.The top 25 keywords in recent years were mostly about the mechanisms and risk factors for the malignant transformation of nevi.Conclusion:The future trend for melanocytic nevi treatment is to specify genotype-phenotype and genotypeoutcome correlations,choose proper therapy to reduce the risk of malignant transformation,and simultaneously achieve the best aesthetic outcomes. 展开更多

Background Inflammatory linear verrucous epidermal nevus(ILVEN)is a rare type of epidermal nevus that occurs in infancy and develops along Blaschko’s lines.Due to its rarity and clinical/histological similarities with other cutaneous disorders,correct diagnosis is sometimes difficult.In the present case,the whole exome sequencing of blood found no germline mutation,but the sequencing of the biopsy specimen revealed a novel potential pathogenic somatic mutation,C>T at rs200881715(p.R698X)in the cspp1 gene,which may provide new insight into the mechanism of ILVEN.Patient concerns A 10-year-old girl presented with linear,pruritic,erythematous,scaly,and exophytic verrucous skin lesions on her left thigh,buttock,and hand unilaterally along Blaschko’s lines from the third day after birth.Diagnosis Biopsy revealed psoriasiform acanthosis,papillary hyperplasia with orthohyperkeratosis,and parakeratosis of squamous epithelium,which are histological characteristics of ILVEN.Interventions Owing to the large lesion size and significant refractory response to previous treatments,multiple-stage surgical resections with a tissue expander were performed.Outcomes After surgical resection and defect reconstruction with a tissue expander,the outcome was satisfactory for the patient and her family.No recurrence was detected during the follow-up visits.Conclusion ILVEN is a rare type of cutaneous hyperplastic disease,and treatment is extremely challenging.Biopsy should be performed at the diagnostician’s discretion or if refractory to conservative treatments.Timely diagnosis and surgical intervention could result in satisfactory outcomes. 展开更多

The scarcity of wettability,insufficient active sites,and low surface area of graphite felt(GF)have long been suppressing the performance of vanadium redox flow batteries(VRFBs).Herein,an ultra-homogeneous multipledimensioned defect,including nano-scale etching and atomic-scale N,O codoping,was used to modify GF by the molten salt system.NH_(4)Cl and KClO_(3) were added simultaneously to the system to obtain porous N/O co-doped electrode(GF/ON),where KClO_(3) was used to ultra-homogeneously etch,and O-functionalize electrode,and NH4Cl was used as N dopant,respectively.GF/ON presents better electrochemical catalysis for VO_(2)+/VO_(2)+ and V3+/V2+ reactions than only O-functionalized electrodes(GF/O)and GF.The enhanced electrochemical properties are attributed to an increase in active sites,surface area,and wettability,as well as the synergistic effect of N and O,which is also supported by the density functional theory calculations.Further,the cell using GF/ON shows higher discharge capacity,energy efficiency,and stability for cycling performance than the pristine cell at 140 mA cm^(−2) for 200 cycles.Moreover,the energy efficiency of the modified cell is increased by 9.7% from 55.2% for the pristine cell at 260 mA cm^(−2).Such an ultra-homogeneous etching with N and O co-doping through“boiling”molten salt medium provides an effective and practical application potential way to prepare superior electrodes for VRFB. 展开更多

In this paper,carbon microsphere prepared by hydrothermal treatment was activated by KHCO_3 at high temperature,and employed as the catalyst for VO^(2+)/VO_2^+redox reaction for vanadium redox flow battery(VRFB).Carbon microsphere can be etched by KHCO_3 due to the reaction between the pyrolysis products of KHCO_3 and carbon atoms.Moreover,KHCO_3 activation can bring many oxygen functional groups on carbon microsphere,further improving the wettability of catalyst and increasing the active sites.The electrocatalytic properties of carbon microsphere from hydrothermal treatment are improved by high temperature carbonization,and can further be enhanced by KHCO_3 activation.Among carbon microsphere samples,the VO^(2+)/VO_2^+redox reaction exhibits the highest electrochemical kinetics on KHCO_3 activated sample.The cell using KHCO_3 activated carbon microsphere as the positive catalyst demonstrates higher energy efficiency and larger discharge capacity,especially at high current density.The results reveal that KHCO_3 activated carbon microsphere is an efficient,low-cost carbon-based catalyst for VO^(2+)/VO_2^+redox reaction for VRFB system. 展开更多

Vanadium redox flow battery(VRFB)is one of the most promising large-scale energy storage systems,which ranges from kilowatt to megawatt.Nevertheless,poor electrochemical activity of electrode for two redox couples still restricts the extensive applications of VRFB.Compared with V^(2+)/V^(3+)redox reaction,V^(2+)/V^(3+)reaction plays a more significant role in voltage loss of VRFB owing to slow heterogeneous electron transfer rate.Herein,N-doped carbon materials derived from scaphium scaphigerum have been developed as negative electrocatalyst by hydrothermal carbonization and high-temperature nitridation treatments.The undoped carbon material hardly has electrocatalytic ability for V^(2+)/V^(3+)reaction.Based on this,N-doped carbon materials with urea as nitrogen source exhibit excellent electrocatalytic properties.And the material nitrided at 850°C(SSC/N-850)exhibits the best performance among those from700 to 1000℃.SSC/N-850 can accelerate the electrode process including V^(2+)/V^(3+)reaction and mass transfer of active ions due to the large reaction place,more active sites,and good hydrophilicity.The effect of catalyst on comprehensive performance of cell was evaluated.SSC/N-850 can improve the charge-discharge performance greatly.Utilization of SSC/N-850 can lessen the electrochemical polarization of cell,further resulting in increased discharge capacity and energy efficiency.Discharge capacity and energy efficiency increase by 81.5%and 9.8%by using SSC/N-850 as negative catalyst at 150 m A cm^(-2),respectively.Our study reveals that the developed biomass-derived carbon materials are the low-cost and efficient negative electrocatalyst for VRFB system. 展开更多

We have previously reported that angiopoietin-1 significantly redu... Doxorubicin (Dox) is a major anticancer chemotherapeutic agent. However, it causes cardiomyopathy due to the side effect of cardiomyocyte apoptosis. We have previously reported that angiopoietin-1 significantly reduced myocardial infarction after ischemic injury and protected cardiomyocytes from oxidative stress-induced apoptosis. It is hypothesized that angiopoietin-1 may protect cardiomyocytes from Dox-induced apoptosis. Cardiomyocytes H9C2 were transfected with adenovirus expressing angiopoietin-1 (Ad5-Ang-1) 24 h before the cells were chal- lenged with Dox at a concentration of 2 ~tmol/L. Ad5-GFP served as the vector control. Cardiomyocyte apoptosis was evaluated using Annexin V-FITC staining and caspase-3 and caspase-8 activity was determined by Western blotting. The results showed that Dox treatment significantly induced cardiomyocyte apoptosis as evidenced by the greater number of Annexin V-FITC stained cells and increases in caspase-3 and caspase-8 activity. In contrast, overexpression of angiopoietin-1 significantly prevented Dox-induced cardiomyocyte apoptosis. To elucidate the mechanisms by which angiopoietin-1 protected cells from Dox-induced apoptosis, we analyzed both extrinsic and intrinsic apoptotic signaling pathways. We observed that angiopoietin-1 prevented Dox-induced activation of both extrinsic and intrinsic apoptotic signaling pathways. Specifically, angiopoietin-1 prevented DOX-induced in- creases in FasL and Bax levels and cleaved caspase-3 and caspase-8 levels in H9C2 cells. In addition, overexpres- sion of angiopoietin-1 also activated the pro-survival phosphoinositide-3 kinase (PI3K)/Akt signaling pathway and decreased Dox-induced nuclear factor-kappaB (NF-~:B) activation. Our data suggest that promoting the expression of angiopoietin-1 could be a potential approach for reducing Dox-induced cardiomyocyte cytoxicity. 展开更多

Background Human interferon-α1b(hIFN-α1b)is the first genetically engineered drug approved by the National Medical Product Administration for cancer therapy in China.hIFN-α1b is a relatively safe therapy and can improve survival in advanced melanoma and its long-term efficacy against resection of stage Ⅲ melanoma is unknown.Objective The objective of this study is to assess the safety and efficacy of hIFN-α1b in patients with resected stage Ⅲ melanoma.Methods The clinical data were collected from patients who underwent resection of stage ⅢB or ⅢC melanoma and received subcutaneous injection of hIFN-α1b every other day.The efficacy of hIFN-α1b was determined by rate of recurrence-free survival(RFS),distant metastasis-free survival(DMFS)and overall survival(OS).Safety data were acquired by review of adverse events at follow-ups.Results The study included a total of 61 patients.The median follow-up duration was 45 months.The RFS at 12,24,and 36 months were observed to be 75.4%,47.4%,and 37.2%respectively,while the DMFS rates were found to be 83.6%,65.5%,and 62.2%.Furthermore,the OS rates at these time points were recorded as being 100%,81.9%,and 71.5%.The overall tolerability of hIFN-α1b was satisfactory,with only 8.2%of the patients experiencing grade 3/4 toxicity and no treatment-related deaths observed.Conclusion Our findings suggest that adjuvant hIFN-α1b treatment is relatively safe and may potentially enhance the rate of RFS,DMFS and OS in patients who have undergone resection for stage ⅢB or ⅢC melanoma. 展开更多

The electric unmanned aerial vehicles (UAVs) are rapidly growing due to their abilities to perform some difficult or dangerous tasks as well as many public services including real-time monitoring, wireless coverage, search and rescue, wildlife surveys, and precision agriculture. However, the electrochemical power supply system of UAV is a critical issue in terms of its energy/power densities and lifetime for service endurance. In this paper, the current power supply systems used in UAVs are comprehensively reviewed and analyzed on the existing power configurations and the energy management systems. It is identified that a single type of electrochemical power source is not enough to support a UAV to achieve a long-haul flight;hence, a hybrid power system architecture is necessary. To make use of the advantages of each type of power source to increase the endurance and achieve good performance of the UAVs, the hybrid systems containing two or three types of power sources (fuel cell,battery, solar cell, and supercapacitor,) have to be developed. In this regard, the selection of an appropriate hybrid power structure with the optimized energy management system is critical for the efficient operation of a UAV. It is found that the data-driven models with artificial intelligence (AI) are promising in intelligent energy management. This paper can provide insights and guidelines for future research and development into the design and fabrication of the advanced UAV power systems. 展开更多

目的了解中国健康定量CT(QCT)大数据项目2018—2019年入组人群的基线特征。方法以中国健康定量CT大数据项目2018年1月至2019年12月符合标准的入组人群为研究对象, 骨密度和腹内脂肪的测量采用Mindways的定量CT Pro Model 4系统。描述性... 目的了解中国健康定量CT(QCT)大数据项目2018—2019年入组人群的基线特征。方法以中国健康定量CT大数据项目2018年1月至2019年12月符合标准的入组人群为研究对象, 骨密度和腹内脂肪的测量采用Mindways的定量CT Pro Model 4系统。描述性分析入组人群的年龄、性别、地区分布以及身高、体重、腹围、血压、血常规及血生化等健康指标的基线情况, 并采用单因素方差分析分性别对骨密度和腹内脂肪年龄变化趋势进行趋势检验。结果经对主要指标的纳入排除标准及异常值筛选后, 最终86 113例纳入本研究, 入组率为92.47%, 其中女性35 431例(41.1%), 男性50 682例(58.9%), 男女比值为1.43。总体年龄为(50.3±12.7)岁, 男性为(50.2±12.8)岁, 女性为(50.4±12.5)岁, 性别差异无统计学意义(P>0.05)。按地区划分, 华东地区入组最多, 共43 833例(50.90%), 其次为华中地区16 434例(19.08%), 东北地区最低2 914例(3.38%)。与主要研究结局相关的健康信息指标入组率基本在70%以上, 且两性差异均有统计学意义(均P<0.05)。女性骨密度为(139.33±46.76)mg/cm3, 男性为(135.90±36.48)mg/cm3, 不同性别骨密度整体表现为随年龄递减趋势(均P<0.001);女性腹内脂肪面积为(116.39±56.23)cm2, 男性为(191.67±77.07)cm2, 不同性别整体表现为随年龄递增趋势(均P<0.001)。结论基于QCT测量的基线骨密度和腹内脂肪均存在性别差异且年龄趋势不同, 健康信息指标也有性别差异;此外, 由于纳入数据的地区差异, 也需考虑地区因素。 展开更多

目的建立中国健康成年女性人群定量CT(QCT)腰椎骨密度(BMD)正常参考值, 并探究其地区差异。方法收集中国健康定量CT大数据项目符合纳入标准的健康女性人群35 431例。采用美国 Mindways 的定量CT系统, 测量L1、L2椎体中心层面BMD, 取平... 目的建立中国健康成年女性人群定量CT(QCT)腰椎骨密度(BMD)正常参考值, 并探究其地区差异。方法收集中国健康定量CT大数据项目符合纳入标准的健康女性人群35 431例。采用美国 Mindways 的定量CT系统, 测量L1、L2椎体中心层面BMD, 取平均值。单因素方差分析比较不同年龄段之间和不同地区之间的女性腰椎BMD差异。按10岁年龄间隔, 同年龄段不同地区之间两两比较。结果中国健康成年女性人群峰值骨密度出现在20~29岁年龄段[东北(183.01±24.58)mg/cm^(3)、华北(188.93±24.80)mg/cm^(3)、华东(187.54±27.71)mg/cm^(3)、华南(186.22±33.72)mg/cm^(3)、华中(176.33±24.91)mg/cm^(3)、西南(182.25±28.00)mg/cm^(3)], 随后随年龄增加呈下降趋势。各地区BMD水平随着年龄增长而降低, 70岁以前, 华中和西南地区BMD[(176.23±24.91)至(90.38±28.12)mg/cm^(3), (182.25±28.00)至(88.55±25.68)mg/cm^(3)]始终处于较低水平, 低于东北[(183.01±24.58)至(99.69±27.85)mg/cm^(3))、华北(188.93±24.80)至(95.89±26.12)mg/cm^(3)]、华东[(187.54±27.71)至(95.65±27.86)mg/cm^(3)]地区(P<0.01);≥70岁各地区BMD趋于一致(P>0.05)。华中和西南地区在40~60岁年龄段BMD值接近(P>0.05), ≥60岁各地区BMD均低于骨量异常值(均P<0.05)。≥50岁女性西南地区骨质疏松检出率最高(25.65%), 华北地区最低(17.30%)。结论建立了中国健康女性人群的QCT骨密度参考值数据库, 西南地区BMD值最低, 华南地区BMD值最高。 展开更多

目的探究中国成年男性人群定量CT(QCT)脊柱骨密度正常参考值和中国不同地区骨密度差异。方法选取2018年1月至2019年12月在中国东北、华北、华东、华南、华中和西南6个地区接受低剂量胸部CT肺癌筛查的男性人群,采用Mindways公司的QCT系... 目的探究中国成年男性人群定量CT(QCT)脊柱骨密度正常参考值和中国不同地区骨密度差异。方法选取2018年1月至2019年12月在中国东北、华北、华东、华南、华中和西南6个地区接受低剂量胸部CT肺癌筛查的男性人群,采用Mindways公司的QCT系统测量腰椎椎体的骨密度。以10岁为1个年龄段,计算骨密度均值和下降率,按美国放射学院脊柱QCT骨质疏松诊断标准统计骨质疏松患病率。结果纳入男性50682例,年龄为(50.2±12.8)岁(20~98岁)。中国男性总体脊柱QCT峰值骨密度(173.11±28.56)mg/cm3,出现在20~29岁,然后随年龄增长呈下降趋势。在70岁之前,华南地区男性骨密度相对较高,华中和西南地区较低,东北、华北和华东地区居中,差异具有统计学意义(均P<0.001),70~79岁和≥80岁2个年龄段各地区男性骨密度差异均无统计学意义(均P>0.05)。中国男性脊柱QCT骨密度总体下降约46.92%,40~49岁年龄段下降明显加速。≥50岁男性骨质疏松总体患病率约为11.42%,其中西南地区和华中地区患病率较高(分别为14.72%和13.87%),华北和华南地区较低(分别为8.53%和7.71%)。结论建立了中国成年男性人群的脊柱QCT骨密度范围,华南地区骨密度值最高,华中地区骨密度值最低。 展开更多
