Methods: The breast...Objective: The purpose of this study was to evaluate the impact of the negative lymph node (LN) count on the survival of the breast cancer patients in early stage after the axillary dissection. Methods: The breast cancer patients with T1-2N0-1M0 stage between January 2001 and December 2005 in Jiangsu Cancer Hospital, who underwent the axillary LNs dissection, were enrolled in this study. We analyzed the data of these patients including information of follow-up and postop- erative pathological results. All patients were divided into two groups according to the axillary LN status and each group was divided into four subgroups according to the negative LN count. Cox regression analysis was performed to screen the patho- logical factor including the negative LN count on the survival and to compare the different negative LN count on the survival. Results: COX proportional hazard regression model showed that the survival of the breast cancer was significantly associ- ated with the negative LN count. In T1 2N0 group, when the negative LN count was 3 or less, 4 to 5, 6 to 9 and 10 or more, the median survival time was (82.6 ±4.1) months, (101.5 ± 1.3) months, (104.7 ±1.0) months, and (110.5 ±0.9) months respectively (P 〈 0.05). In T1-2N0 group, when the negative LN count was 6 or less, 7 to 8, 9 to 10 and 11 or more, the median survival time was (95.4 ± 1.9) months, (101.8 ± 1.1) months, (104.9 ± 1.0) months, and (106.5 ± 0.9) months respectively (P 〈 0.05). Conclusion: The negative LN count can reflect the adequacy of the axillary dissection. Increasing negative LN count is independently associated with improved survival in pT1-2N0M0 or pT1-2N0M0 staging breast cancer patients. The negative LN count should be considered for incorporation into staging for breast cancer with the axillary LN dissection.展开更多

A formalized reasoning method—CPNI (Colored Petri Net for Planning in incomplete environment) based on two kinds of actions is proposed, and the reasoning rule as Fluent Calculus in incomplete environment is applied. Robot planning experiment is modeled and simulated by using the tool CPNTools and the result shows the state knowledge of the door and the action sequence to reach the goal can be generated automatically in the CPNI net system.展开更多
Background: Brucellosis poses a serious threat to human and animal health,particularly in developing countries such as China.The Inner Mongolia Autonomous Region is one of the most severely brucellosis-endemic provinc...Background: Brucellosis poses a serious threat to human and animal health,particularly in developing countries such as China.The Inner Mongolia Autonomous Region is one of the most severely brucellosis-endemic provinces in China.Currently,the host immune responses functioning to control Brucella infection and development remain poorly understood.The aim of this study is to further clarify the key immunity characteristics of diverse stages of brucellosis in Inner Mongolia.Methods: We collected a total of 733 blood samples from acute(n=137),chronic(n=316),inapparent(n=35),recovery(n=99),and healthy(n=146)groups from the rural community of Inner Mongolia between 2014 and 2015.The proportions of CD4^(+),CD8^(+),Th1,Th2,and Th17 T cells in peripheral blood and the expression of TLR2 and TLR4 in lymphocytes,monocytes and granulocytes were examined using flow cytometry analysis.The differences among the five groups were compared using one-way ANOVA and the Kruskal–Wallis method,respectively.Results: Our results revealed that the proportions of CD4^(+) and CD8^(+) T cells were significantly different among the acute,chronic,recovery,and healthy control groups(P<0.05),with lower proportions of CD4^(+) T cells and a higher proportion of CD8^(+) T cells in the acute,chronic,and recovery groups.The proportion of Th1 cells in the acute,chronic,and inapparent groups was higher than that in the healthy and recovery groups;however,there was no significant difference between patients and healthy individuals(P>0.05).The proportion of Th2 lymphocytes was significantly higher in the acute and healthy groups than in the inapparent group(P<0.05).The proportion of Th17 cells in the acute group was significantly higher than that in the healthy control,chronic,and inapparent groups(P<0.05).Finally,the highest expression of TLR4 in lymphocytes,monocytes and granulocytes was observed in the recovery group,and this was followed by the acute,chronic,healthy control,and inapparent groups.There was a significant difference between the recovery group and the other groups,except for the acute group(P<0.05).Moreover,a correlation in TLR4 expression was observed in lymphocytes,monocytes and granulocytes among the five groups(r>0.5),except for the inapparent group between lymphocytes and granulocytes(r=0.34).Conclusions: Two key factors(CD8^(+)T cells and TLR4)in human immune profiles may closely correlate with the progression of brucellosis.The detailed function of TLR4 in the context of a greater number of cell types or tissues in human or animal brucellosis and in larger samples should be further explored in the future.展开更多

Background COVID-19 pandemic continues,clarifying signatures in clinical characters and antibody responses between severe and non-severe COVID-19 cases would benefit the prognosis and treatment.Methods In this study,1...Background COVID-19 pandemic continues,clarifying signatures in clinical characters and antibody responses between severe and non-severe COVID-19 cases would benefit the prognosis and treatment.Methods In this study,119 serum samples from 37 severe or non-severe COVID-19 patients from the First People's Hospital of Yueyang were collected between January 25 and February 182020.The clinical features,antibody responses targeting SARS-CoV-2 spike protein(S)and its different domains,SARS-CoV-2-specific Ig isotypes,IgG subclasses,ACE2 competitive antibodies,binding titers with FcγIIa and FcγIIb receptors,and 14 cytokines were comprehensively investigated.The differences between severe and non-severe groups were analyzed using Mann–Whitney U test or Fisher’s exact test.Results Severe group including 9 patients represented lower lymphocyte count,higher neutrophil count,higher level of LDH,total bile acid(TBA)(P<1×10–4),r-glutaminase(P=0.011),adenosine deaminase(P<1×10–4),procalcitonin(P=0.004),C-reactive protein(P<1×10–4)and D-dimer(P=0.049)compared to non-severe group(28 patients).Significantly,higher-level Igs targeting S,different S domains(RBD,RBM,NTD,and CTD),FcγRIIa and FcγRIIb binding capability were observed in a severe group than that of a non-severe group,of which IgG1 and IgG3 were the main IgG subclasses.RBD-IgG were strongly correlated with S-IgG both in severe and non-severe group.Additionally,CTD-IgG was strongly correlated with S-IgG in a non-severe group.Positive RBD-ACE2 binding inhibition was strongly associated with high titers of antibody(S-IgG1,S-IgG3,NTD-IgG,RBD-IgA,NTD-IgA,and CTD-IgA)especially RBD-IgG and CTD-IgG in the severe group,while in the non-severe group,S-IgG3,RBD-IgG,NTD-IgG,and NTD-IgM were correlated with ACE2 blocking rate.S-IgG1,NTD-IgM and S-IgM were negatively associated with illness day in a severe group,while S-IgG3,RBD-IgA,CTD-IgA in the severe group(r=0.363,P=0.011)and S-IgG1,NTD-IgA,CTD-IgA in the non-severe group were positively associated with illness day.Moreover,GRO-α,IL-6,IL-8,IP-10,MCP-1,MCP-3,MIG,and BAFF were also significantly elevated in the severe group.Conclusion Antibody detection provides important clinical information in the COVID-19 process.The different signatures in Ig isotypes,IgG subclasses,antibody specificity between the COVID-19 severe and non-severe group will contribute to future therapeutic and preventive measures development.展开更多
Numerical simulation of heat transfer in a high aspect ratio rectangular microchannel with heat sinks has been conducted,similar to an experimental study.Three channel heights measuring 0.3 mm,0.6 mm and 1 mm are cons...Numerical simulation of heat transfer in a high aspect ratio rectangular microchannel with heat sinks has been conducted,similar to an experimental study.Three channel heights measuring 0.3 mm,0.6 mm and 1 mm are considered and the Reynolds number varies from 300 to 2360,based on the hydraulic diameter.Simulation starts with the validation study on the Nusselt number and the Poiseuille number variations along the channel streamwise direction.It is found that the predicted Nusselt number has shown very good agreement with the theoretical estimation,but some discrepancies are noted in the Poiseuille number comparison.This observation however is in consistent with conclusions made by other researchers for the same flow problem.Simulation continues on the evaluation of heat transfer characteristics,namely the friction factor and the thermal resistance.It is found that noticeable scaling effect happens at small channel height of 0.3 mm and the predicted friction factor agrees fairly well with an experimental based correlation.Present simulation further reveals that the thermal resistance is low at small channel height,indicating that the heat transfer performance can be enhanced with the decrease of the channel height.展开更多
