Background The reproductive performance of chickens mainly depends on the development of follicles.Abnor-mal follicle development can lead to decreased reproductive performance and even ovarian disease among chick-ens.Chicken is the only non-human animal with a high incidence of spontaneous ovarian cancer.In recent years,the involvement of circRNAs in follicle development and atresia regulation has been confirmed.Results In the present study,we used healthy and atretic chicken follicles for circRNA RNC-seq.The results showed differential expression of circRALGPS2.It was then confirmed that circRALGPS2 can translate into a protein,named cir-cRALGPS2-212aa,which has IRES activity.Next,we found that circRALGPS2-212aa promotes apoptosis and autophagy in chicken granulosa cells by forming a complex with PARP1 and HMGB1.Conclusions Our results revealed that circRALGPS2 can regulate chicken granulosa cell apoptosis and autophagy through the circRALGPS2-212aa/PARP1/HMGB1 axis. 展开更多

The physical and chemical properties of the air-dried residual coal after soaking in the goaf will change,resulting in an increase in its spontaneous combustion tendency.This study aimed to look into the features and mechanism of soaked-dried coal's spontaneous combustion.Five samples of coal were dried to various degrees,and the weight loss features during thermal processing were examined.Based on this,the pore structure and chemical structure characteristics of the coal samples with the highest tendency to spontaneous combustion were quantitatively examined,and the mechanism by which soaking-drying afected the spontaneous combustion heating process of the remaining coal in goaf was investigated in turn.The results show that T1 decreases with the increase of drying time,T2–T6 shows a fuctuating change,and the ignition activation energy of 36-S-Coal is smaller than that of other coal samples.The pore type of 36-S-Coal changes from a oneend closed impermeable pore to an open pore,and the pore group area is large.During the 36 h drying process,the internal channels of the coal were dredged,and a large number of gravels and minerals were precipitated from the pores with the air fow.A large number of gravels were around the pores to form a surface structure that was easy to adsorb various gases.Furthermore,infrared spectroscopy was used to analyze the two coal samples.It was found that soaking and drying did not change the functional group types of coal samples,but the fatty chain degree of 36-S-Coal was reduced to 1.56.It shows that the aliphatic chain structure of coal is changed after 36 h of drying after 30 days of soaking,which leads to the continuous shedding of aliphatic chain branches of residual coal,and the skeleton of coal is looser,which makes the low-temperature oxidation reaction of 36-S-Coal easier.Based on the above results,the coal-oxygen composite mechanism of water-immerseddried coal is obtained,and it is considered that the key to the spontaneous combustion oxidation process of coal is to provide oxygen atoms and accelerate the formation of peroxides. 展开更多

Optical thermometry as an important local temperature-sensing technique,has received increasing attention in scientific and industrial areas.However,it is still a big challenge to develop luminescent materials with self-activated dual-wavelength emissions toward high-sensitivity optical thermometers.Herein,a novel ratiometric thermometric strategy of Bi^(3+)-activated dual-wavelength emission band was realized in the same lattice position with two local electronic states of La_(3)Sb_(1-x)Ta_xO_(7):Bi^(3+)(0≤x≤1.0)materials based on the different temperature-dependent emission behaviors,benefiting from the highlysensitive and regulable emission to the coordination environment of Bi^(3+).The structural and spectral results demonstrate that the emission tremendously shifted from green to blue with 68 nm and the intensity was enhanced 2.6 times.Especially,the visual dual-wavelength emitting from two emission centers was presented by increasing the Ta^(5+)substitution concentration to 20%or 25%,mainly originating from the two local electronic states around the Bi^(3+)emission center.Significantly,the dual-wavelength with different thermal-quenching performance provided high-temperature sensitivity and good discrimination signals for optical thermometry in the range between 303 and 493 K.The maximum relative sensitivity reached 2.64%/K(La_(3)Sb_(0.8)Ta_(0.2)O_(7):0.04Bi^(3+)@383 K)and 1.91%/K(La_(3)Sb_(0.75)Ta_(0.25)O_(7):0.04Bi^(3+)@388 K).This work reveals a rational design strategy of different local electronic states around the singledoping multiple emission centers towards practical applications,such as luminescence thermometry and white LED lighting. 展开更多

Preeclampsia is a serious obstetric complication.Currently,there is a lack of effective preventive approaches for this disease.Recent studies have identified transcutaneous auricular vagus nerve stimulation(taVNS)as a potential novel non-pharmaceutical therapeutic modality for preeclampsia.In this study,we investigated whether taVNS inhibits apoptosis of placental trophoblastic cells through ROS-induced UPRmt.Our results showed that taVNS promoted the release of acetylcholine(ACh).ACh decreased the expression of UPRmt by inhibiting the formation of mitochondrial ROS(mtROS),presumably through M3AChR.This reduced the release of pro-apoptotic proteins(cleaved caspase-3,NF-kB-p65,and cytochrome C)and helped preserve the morphological and functional integrity of mitochondria,thus reducing the apoptosis of placental trophoblasts,improving placental function,and relieving preeclampsia.Our study unravels the potential pathophysiological mechanism of preeclampsia.In-depth characterization of the UPRmt is essential for developing more effective therapeutic strategies for preeclampsia targeting mitochondrial function. 展开更多

Background:Cancer stem-like cells(CSCs)are a small subset of cells in tumors that exhibit self-renewal and differentiation properties.CSCs play a vital role in tumor formation,progression,relapse,and therapeutic resistance.B7-H3,an immunoregulatory protein,has many protumor functions.However,little is known about the mechanism underlying the role of B7-H3 in regulating gastric cancer(GC)stemness.Our study aimed to explore the impacts of B7-H3 on GC stemness and its underlying mechanism.Methods:GC stemness influenced by B7-H3 was detected both in vitro and in vivo.The expression of stemness-related markers was examined by reverse transcription quantitative polymerase chain reaction,Western blotting,and flow cytometry.Sphere formation assay was used to detect the sphere-forming ability.The underlying regulatory mechanism of B7-H3 on the stemness of GC was investigated by mass spectrometry and subsequent validation experiments.The signaling pathway(Protein kinase B[Akt]/Nuclear factor erythroid 2-related factor 2[Nrf2]pathway)of B7-H3 on the regulation of glutathione(GSH)metabolism was examined by Western blotting assay.Multi-color immunohistochemistry(mIHC)was used to detect the expression of B7-H3,cluster of differentiation 44(CD44),and Nrf2 on human GC tissues.Student’s t-test was used to compare the difference between two groups.Pearson correlation analysis was used to analyze the relationship between two molecules.The Kaplan-Meier method was used for survival analysis.Results:B7-H3 knockdown suppressed the stemness of GC cells both in vitro and in vivo.Mass spectrometric analysis showed the downregulation of GSH metabolism in short hairpin B7-H3 GC cells,which was further confirmed by the experimental results.Meanwhile,stemness characteristics in B7-H3 overexpressing cells were suppressed after the inhibition of GSH metabolism.Furthermore,Western blotting suggested that B7-H3-induced activation of GSH metabolism occurred through the AKT/Nrf2 pathway,and inhibition of AKT signaling pathway could suppress not only GSH metabolism but also GC stemness.mIHC showed that B7-H3 was highly expressed in GC tissues and was positively correlated with the expression of CD44 and Nrf2.Importantly,GC patients with high expression of B7-H3,CD44,and Nrf2 had worse prognosis(P=0.02).Conclusions:B7-H3 has a regulatory effect on GC stemness and the regulatory effect is achieved through the AKT/Nrf2/GSH pathway.Inhibiting B7-H3 expression may be a new therapeutic strategy against GC. 展开更多

The components of the oil bleed and the effect of the silicone chemical parameters on the amount ofoil bleed have been determined. The main components of oil bleeds are the uncrosslinked silicones in the cured resins, which include the unreacted silicone materials and the macromolecular substances produced by the hydrosilylation reaction. Cured resins with a high crosslinking density and a high molecular weight of vinyl silicone residues had a lower amount of oil bleed. In addition, a low Si-H content also reduced the amount ofoil bleed. 展开更多

The detection and analysis of circulating tumor cells (CTCs) from patients' blood is important to assess tumor status; however, it remains a challenge. In the present study, we developed a programmable DNA-responsive microchip for the highly efficient capture and nondestructive release of CTCs via nucleic acid hybridization. Transparent and patternable substrates with hierarchical architectures were integrated into the microchip with herringbone grooves, resulting in greatly enhanced cell-surface interaction via herringbone micromixers, more binding sites, and better matched topographical interactions. In combination with a high-affinity aptamer, target cancer cells were specifically and efficiently captured on the chip. Captured cancer cells were gently released from the chip under physiological conditions using toehold-mediated strand displacement, without any destructive factors for cells or substrates. More importantly, aptamercontaining DNA sequences on the surface of the retrieved cancer cells could be further amplified by polymerase chain reaction (PCR), facilitating the detection of cell surface biomarkers and characterization of the CTCs. Furthermore, this system was extensively applied to the capture and release of CTCs from patients' blood samples, demonstrating a promising high-performance platform for CTC enrichment, release, and characterization. 展开更多

Dielectric barrier discharge(DBD)plasma applied as surface treatment technology was employed for the modification of Ag_(2)O and graphitic carbon nitride(g-C_(3)N_(4))powders.Subsequently,the pretreated powders were sequentially loaded onto TiO_(2)nanorods(TiO_(2)-NRs)via electro-deposition,followed by calcination at N_(2)atmosphere.The results indicated that at the optimal plasma discharge time of 5 min for modification of g-C_(3)N_(4)and Ag_(2)O,photocurrent density of ternary composite was 6 times to bare TiO_(2)-NRs under UV-visible light irradiation.Phenol was degraded by using DBD plasma-modified g-C_(3)N_(4)/Ag_(2)O/TiO_(2)-NRs electrode to analyze the photoelectrocatalytic performance.The removal rate of phenol for g-C_(3)N_(4)-5/Ag_(2)O-5/TiO_(2)-NRs electrode was about 3.07 times to that for TiO_(2)-NRs electrode.During active species scavengers'analysis,superoxide radicals and hydroxyl radicals were the main oxidation active species for pollutants degradation.A possible electron-hole separation and transfer mechanism of ternary composite with high photoelectrocatalytic performance was proposed. 展开更多

Effective and low toxicity delivery of siRNA is of great importance for clinical gene therapy. Herein, self-assembled DNA nanoparticles (NPs) based on rolling circle amplification (RCA) with a small interfering RNA (siRNA) payload were successfully developed as a facile and efficient siRNA delivery strategy. This intracellular gene silencing strategy exhibits various advantages including low toxicity, high efficiency, and good stability. The synthesized DNA NPs serve as siRNA carriers, protecting the siRNA against nuclease degradation. We demonstrate that the obtained self-assembled siRNA/NP/PEI system can successfully deliver enhanced green fluorescent protein (EGFP)-siRNA into HeLa cells, realizing the same EGFP knockdown efficiency with less toxicity as that of commercial Lipofectamine 2000. 展开更多

The availability and reliability of strategies for molecular biosensing over a finely adjustable dynamic range is essential to enhance the understanding and control of vital biological process. To expand the versatility and utility of nucleic acid- related enzymes, we demonstrated a rational approach to acquiring tunable, pH-dependent deoxyribozymes (DNAzymes) with catalytic activities and response sensitivities that can be tuned through a simple change in solution pH. To do this, we capitalized upon the pH dependence of Hoogsteen interactions and designed i-motif- and triplex-based DNAzymes that can be finely regulated with high precision over a physiologically relevant pH interval. The modified DNAzymes are dependent upon pH for efficient cleavage of substrates, and their catalytic performance can be tuned by regulating the sequence of inserted i-motif/triplex structures. The principle of tunable, pH-dependent DNAzymes provides the opportunity to engineer pH-controlled DNA-machinery devices with unprecedented sensitivity to pH changes. For example, we constructed a DNA-walker device, the stepping rate of which could be adjusted by simply modulating solution pH within an interval of 5.6 to 7.4, as well as a DNA tetrahedron that can be opened at pH 6.4 and kept closed at pH 7.4. The potential of this approach is not limited to serve as pH-dependent devices, but rather may be combined with other elements to expand their practical usefulness. 展开更多

The development of multifunctional theranostic nano-agents is an important resolution for personalized treatment of cancer.In this work,we synthesized a new kind of gadolinium boride nanoparticles(GBN)by a microwave-assisted chemical etching method,and discovered their optical characteristics including fluorescence imaging and near-infrared(NIR)photothermal conversion capability.Bright greenishyellow fluorescence enabled for intracellular localization,while effective NIR-photothermal conversion supported photothermal therapy(PTT).In vitro and in vivo results indicated that GBN exhibited a superior antitumor performance and high biocompatibility.This study demonstrated a promising multifunctional theranostic nanoplatform for cancer treatment. 展开更多

The design and development of multifunctional nano-drug delivery systems(NDDSs)is a solution that is expected to solve some intractable problems in traditional cancer treatment.In particular,metal-organic frameworks(MOFs)are novel hybrid porous nanomaterials which are constructed by the coordination of metal cations or clusters and organic bridging ligands.Benefiting from their intrinsic superior properties,MOFs have captivated intensive attentions in drug release and cancer theranostic.Based on what has been achieved about MOF-based DDSs in recent years,this review introduces different stimuli-responsive mechanisms of them and their applications in cancer diagnosis and treatment systematically.Moreover,the existing challenges and future opportunities in this field are summarized.By realizing industrial production and paying attention to biosafety,their clinical applications will be enriched. 展开更多
