Inducible nitric oxide synthase (iNOS), bone morphogenetic protein-7 (BMP-7) and B-cell lymphoma 2 (Bcl-2) are surrogate markers of renal tubular epithelial regeneration and subsequent recovery of renal function following ATN. Methods: Serum creatinine (Scr) and blood urea nitrogen (BUN), as well as expression of iNOS, BMP-7 and Bcl-2 in gentamycin-induced ATN rat kidneys was investigated after human umbilical cord-derived mesenchymal stem cell (HUC-MSC) transplantation. Immunohistochemical staining was performed in 3 groups of rats: gentamycin-induced ATN treated with HUC-MSC, gentamycin-induced ATN without HUC-MSC, and untreated rats not receiving any treatments. Results: HUC-MSC transplantation led to a reduction in Scr and BUN in the kidneys of rats with gentamycin-induced ATN. Expression of iNOS in the HUC-MSC treated group occurred later and the expression levels were much lower during gentamycin-induced ATN compared to rats with ATN that were not treated with HUC-MSC. The expression of BMP-7 and Bcl-2 in the MSC-transplanted group was significantly increased compared to both control groups of rats with injured and healthy renal tubules. Conclusions: HUC-MSCs induce renal protection in a rat model of gentamycin-induced ATN, which is associated with reduced iNOS expression and up-regulation of Bcl-2 and BMP-7.展开更多

Aqueous zinc-ion batteries(ZIBs) are attracting considerable attention because of their low cost,high safety and abundant anode material resources.However,the major challenge faced by aqueous ZIBs is the lack of stabl...Aqueous zinc-ion batteries(ZIBs) are attracting considerable attention because of their low cost,high safety and abundant anode material resources.However,the major challenge faced by aqueous ZIBs is the lack of stable and high capacity cathode materials due to their complicated reaction mechanism and slow Zn-ion transport kinetics.This study reports a unique 3 D ’flower-like’ zinc cobaltite(ZnCo_(2)O_(4-x)) with enriched oxygen vacancies as a new cathode material for aqueous ZIBs.Computational calculations reveal that the presence of oxygen vacancies significantly enhances the electronic conductivity and accelerates Zn^(2+) diffusion by providing enlarged channels.The as-fabricated batteries present an impressive specific capacity of 148.3 mAh g^(-1) at the current density of 0.05 A g^(-1),high energy(2.8 Wh kg^(-1)) and power densities(27.2 W kg^(-1)) based on the whole device,which outperform most of the reported aqueous ZIBs.Moreover,a flexible solid-state pouch cell was demonstrated,which delivers an extremely stable capacity under bending states.This work demonstrates that the performance of Zn-ion storage can be effectively enhanced by tailoring the atomic structure of cathode materials,guiding the development of low-cost and eco-friendly energy storage materials.展开更多
Tree peony is a unique traditional f lower in China,with large,fragrant,and colorful f lowers.However,a relatively short and concentrated f lowering period limits the applications and production of tree peony.A genome...Tree peony is a unique traditional f lower in China,with large,fragrant,and colorful f lowers.However,a relatively short and concentrated f lowering period limits the applications and production of tree peony.A genome-wide association study(GWAS)was conducted to accelerate molecular breeding for the improvement of f lowering phenology traits and ornamental phenotypes in tree peony.A diverse panel of 451 tree peony accessions was phenotyped for 23 f lowering phenology traits and 4 f loral agronomic traits over 3 years.Genotyping by sequencing(GBS)was used to obtain a large number of genome-wide single-nucleotide polymorphisms(SNPs)(107050)for the panel genotypes,and 1047 candidate genes were identified by association mapping.Eighty-two related genes were observed during at least 2 years for f lowering,and seven SNPs repeatedly identified for multiple f lowering phenology traits over multiple years were highly significantly associated with five genes known to regulate f lowering time.We validated the temporal expression profiles of these candidate genes and highlighted their possible roles in the regulation of f lower bud differentiation and f lowering time in tree peony.This study shows that GWAS based on GBS can be used to identify the genetic determinants of complex traits in tree peony.The results expand our understanding of f lowering time control in perennial woody plants.Identification of markers closely related to these f lowering phenology traits can be used in tree peony breeding programs for important agronomic traits.展开更多

The incidence of lung cancer in cases of colorectal cancer is rare and synchronous rectal cancer and lung cancer is even rare. A 61-year-old man was referred to our hospital with a 2-month history of blood in his stool, tenesmus, and mucous discharge in July 2010. Colonoscopy showed an irregular ulcerated rectal mass and histological examination of biopsy material showed a poorly differentiated adenocarcinoma. Computed tomography (CT) scan of the chest and abdomen showed a mass in the posterior segment of the right upper lobe of the lung and a mass in the right rectal wall of upper rectum. The rectal tumor was diagnosed as primary cancer based on the findings of immunohistochemical stain. An anterior resection (AR) and video assisted thoracoscopic (VAT) wedge resection were performed and histological findings of resected rectal and lung tumor specimen showed synchronous primary rectal cancer and lung cancer. A combination chemotherapy regimen with docetaxel and Iobaplatin was used and the patient was successfully discharged from hospital in August 2010. Although the incidence of synchronous multiple primary cancers is very low, we need to remain suspicious, when faced with two or even multiple organ lesions, and employ the necessary examination methods to confirm the diagnosis. For synchronous multiple primary cancers, if conditions allow, surgical resection for all the cancers can be performed in a single operation.展开更多
[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performanc...[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performance of Sichuan white rabbits and reveal the regulatory role of miRNAs in their muscle growth and meat quality formation.[Methods]Three constructed skeletal muscle libraries of Sichuan white rabbits aged six months were sequenced by the solexa technology to identify known miRNAs,predict new miRNAs and construct an expression profile of muscle miRNAs.[Results]A total of 511 known miRNAs and 42 miRNAs were detected in 34089472 pure sequences,and the proportion of miRNAs with a length of 22 nt was the highest.The number of known miRNA sequences accounted for 71.38%of pure sequences,which was much higher than the proportion of other types of RNAs.The proportion of sequences from exons was 0.38%,indicating a low degree of mRNA degradation in the samples.Base U had the highest proportion at the first position,and the bases with the highest proportions at positions 8 and 10 were U and A,respectively.Muscle-specific miRNAs(miR-1,miR-133,and miR-206)ranked in the top 10 in terms of expression level.The number and expression levels of new miRNAs were lower than those of known miRNAs.The length distribution,base bias at different positions and expression profile characteristics of miRNAs might be related to the biological function of miRNAs in regulating muscle proliferation and differentiation and the action mechanisms with target genes.[Conclusions]The identification and expression of miRNAs in muscle tissues of Sichuan white rabbits will help to understand the complex molecular mechanisms of meat production performance and provide a theoretical basis for the functional research of miRNAs in meat rabbits.展开更多

Previous studies have indicated an association of fat mass and obesity-associated(FTO)with nonalcoholic fatty liver disease(NAFLD),the most common chronic liver disease worldwide.This study aimed to decipher the compl...Previous studies have indicated an association of fat mass and obesity-associated(FTO)with nonalcoholic fatty liver disease(NAFLD),the most common chronic liver disease worldwide.This study aimed to decipher the complex role of FTO in hepatic lipid metabolism.We found that a decrease in N^(6)-methyladenosine(m^(6)A)RNA methylation in the liver of mice fed with a high-fat diet(HFD)was accompanied by an increase in FTO expression.Overexpression of FTO in the liver promoted triglyceride accumulation by upregulating the expression of lipogenic genes.Mechanistical studies revealed that FTO could stabilize the mRNAs of sterol regulatory element binding transcription factor 1(SREBF1)and carbohydrate responsive element binding protein(ChREBP),two master lipogenic transcription factors,by demethylating m^(6)A sites.Knockdown ofeither SREBF1 or ChREBP attenuated the lipogenic effect of FTO,suggesting that they are bona fide effectors for FTO in regulating lipogenesis.Insulin could stimulate FTO transcription through a mechanism involving the action of intranuclear insulin receptor beta,while knockdown of FTO abrogated the lipogenic effect of insulin.Inhibition ofFTo by entacapone decreased the expression of SREBF1,ChREBP,and downstream lipogenic genes,ameliorating liver steatosis in HFD-fed mice.Thus,our study established a critical role of FTO in both the insulin-regulated hepatic lipogenesis and the pathogenesis of NAFLD and provided a potential strategyfor treating NAFLD.展开更多

In this paper, we study the potential use of the realized volatility as a proxy in a stochastic volatility model estimation. We estimate the leveraged stochastic volatility model using the realized volatility computed from five popular methods across six sampling-frequency transaction data (from 1-min to 60- min) based on the trust region method. Availability of the realized volatility allows us to estimate the model parameters via the MLE and thus avoids computational challenge in the high dimensional integration. Six stock indices are considered in the empirical investigation. We discover some consistent findings and interesting patterns from the empirical results. In general, the significant leverage effect is consistently detected at each sampling frequency and the volatility persistence becomes weaker at the lower sampling frequency.展开更多
In a tritrophic context of plant-insect-entomopathogen,plants play important roles in modulating the interaction of insects and their pathogenic viruses.Currently,the influence of plants on the transmission of insect ...In a tritrophic context of plant-insect-entomopathogen,plants play important roles in modulating the interaction of insects and their pathogenic viruses.Currently,the influence of plants on the transmission of insect viruses has been mainly studied on baculoviruses and some RNA viruses,whereas the impact of plants on other insect viruses is largely unknown.Here,we identified a new densovirus infecting the green peach aphid Myzus persicae and tested whether and how host plants influence the transmission of the aphid densovirus.The complete single-stranded DNA genome of the virus,M.persicae densovirus 2,is 5727 nt and contains inverted terminal repeats.Transcription and phylogenetic analysis indicated that the virus was distinct from other a few identified aphid densoviruses.The virus abundance was detected highly in the intestinal tract of aphids,compared with the lower level of it in other tissues including head,embryo,and epidermis.Cabbage and pepper plants had no obvious effect on the vertical transmission and saliva-mediated horizontal transmission of the virus.However,the honeydew-mediated horizontal transmission among aphids highly depended on host plants(65%on cabbages versus 17%on peppers).Although the virus concentration in the honeydew produced by aphids between 2 plants was similar,the honeydew production of the infected aphids reared on peppers was dramatically reduced.Taken together,our results provide evidence that plants influence the horizontal transmission of a new densovirus in an aphid population by modulating honeydew secretion of aphids,suggesting plants may manipulate the spread of an aphid-pathogenic densovirus in nature.展开更多

文摘Aqueous zinc-ion batteries(ZIBs) are attracting considerable attention because of their low cost,high safety and abundant anode material resources.However,the major challenge faced by aqueous ZIBs is the lack of stable and high capacity cathode materials due to their complicated reaction mechanism and slow Zn-ion transport kinetics.This study reports a unique 3 D ’flower-like’ zinc cobaltite(ZnCo_(2)O_(4-x)) with enriched oxygen vacancies as a new cathode material for aqueous ZIBs.Computational calculations reveal that the presence of oxygen vacancies significantly enhances the electronic conductivity and accelerates Zn^(2+) diffusion by providing enlarged channels.The as-fabricated batteries present an impressive specific capacity of 148.3 mAh g^(-1) at the current density of 0.05 A g^(-1),high energy(2.8 Wh kg^(-1)) and power densities(27.2 W kg^(-1)) based on the whole device,which outperform most of the reported aqueous ZIBs.Moreover,a flexible solid-state pouch cell was demonstrated,which delivers an extremely stable capacity under bending states.This work demonstrates that the performance of Zn-ion storage can be effectively enhanced by tailoring the atomic structure of cathode materials,guiding the development of low-cost and eco-friendly energy storage materials.

文摘Tree peony is a unique traditional f lower in China,with large,fragrant,and colorful f lowers.However,a relatively short and concentrated f lowering period limits the applications and production of tree peony.A genome-wide association study(GWAS)was conducted to accelerate molecular breeding for the improvement of f lowering phenology traits and ornamental phenotypes in tree peony.A diverse panel of 451 tree peony accessions was phenotyped for 23 f lowering phenology traits and 4 f loral agronomic traits over 3 years.Genotyping by sequencing(GBS)was used to obtain a large number of genome-wide single-nucleotide polymorphisms(SNPs)(107050)for the panel genotypes,and 1047 candidate genes were identified by association mapping.Eighty-two related genes were observed during at least 2 years for f lowering,and seven SNPs repeatedly identified for multiple f lowering phenology traits over multiple years were highly significantly associated with five genes known to regulate f lowering time.We validated the temporal expression profiles of these candidate genes and highlighted their possible roles in the regulation of f lower bud differentiation and f lowering time in tree peony.This study shows that GWAS based on GBS can be used to identify the genetic determinants of complex traits in tree peony.The results expand our understanding of f lowering time control in perennial woody plants.Identification of markers closely related to these f lowering phenology traits can be used in tree peony breeding programs for important agronomic traits.

文摘The effect of N_(2)discharge products on cyclohexane degradation over a MnO_(2)/γ-Al_(2)O_(3)catalyst has been evaluated by feeding N_(2)discharge products to the catalyst using a specially designed dielectric barrier discharge reactor.At a reaction temperature of 100℃,the cyclohexane conversion increased from 2.46%(without N_(2)discharge products)to 26.3%(with N_(2)discharge products).N-and O-containing by-product(3,4-dehydroproline)was found on the catalyst surface using gas chromatograph-mass spectrometry identification,in which C=N–C and C=N–H bonds were also confirmed from x-ray photoelectron spectroscopy analysis results.Operando analysis results using diffuse reflectance infrared Fourier transform spectroscopy revealed that N atoms can react with surface H_(2)O possibly to NH and OH reactive species that have reactivities to promote CO oxidation to CO_(2).The mechanism of N-atom-driven cyclohexane degradation to CO and CO_(2)is proposed.

文摘To identify industrial control equipment is often a key step in network mapping,categorizing network resources,and attack defense.For example,if vulnerable equipment or devices can be discovered in advance and the attack path canbe cut off,security threats canbe effectively avoided and the stable operationof the Internet canbe ensured.The existing rule-matching method for equipment identification has limitations such as relying on experience and low scalability.This paper proposes an industrial control device identification method based on PCA-Adaboost,which integrates rule matching and machine learning.We first build a rule base from network data collection and then use single andmulti-protocol rule-matchingmethods to identify the type of industrial control devices.Finally,we utilize PCA-Adaboost to identify unlabeled data.The experimental results show that the recognition rate of this method is better than that of the traditional Nmap device recognitionmethod and the device recognition accuracy rate reaches 99%.The evaluation effect of the test data set is significantly enhanced.
文摘Multiple primary cancers refer to the condition where more than two cancers occur independently in an individual. The incidence of lung cancer in cases of colorectal cancer is rare and synchronous rectal cancer and lung cancer is even rare. A 61-year-old man was referred to our hospital with a 2-month history of blood in his stool, tenesmus, and mucous discharge in July 2010. Colonoscopy showed an irregular ulcerated rectal mass and histological examination of biopsy material showed a poorly differentiated adenocarcinoma. Computed tomography (CT) scan of the chest and abdomen showed a mass in the posterior segment of the right upper lobe of the lung and a mass in the right rectal wall of upper rectum. The rectal tumor was diagnosed as primary cancer based on the findings of immunohistochemical stain. An anterior resection (AR) and video assisted thoracoscopic (VAT) wedge resection were performed and histological findings of resected rectal and lung tumor specimen showed synchronous primary rectal cancer and lung cancer. A combination chemotherapy regimen with docetaxel and Iobaplatin was used and the patient was successfully discharged from hospital in August 2010. Although the incidence of synchronous multiple primary cancers is very low, we need to remain suspicious, when faced with two or even multiple organ lesions, and employ the necessary examination methods to confirm the diagnosis. For synchronous multiple primary cancers, if conditions allow, surgical resection for all the cancers can be performed in a single operation.

文摘[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performance of Sichuan white rabbits and reveal the regulatory role of miRNAs in their muscle growth and meat quality formation.[Methods]Three constructed skeletal muscle libraries of Sichuan white rabbits aged six months were sequenced by the solexa technology to identify known miRNAs,predict new miRNAs and construct an expression profile of muscle miRNAs.[Results]A total of 511 known miRNAs and 42 miRNAs were detected in 34089472 pure sequences,and the proportion of miRNAs with a length of 22 nt was the highest.The number of known miRNA sequences accounted for 71.38%of pure sequences,which was much higher than the proportion of other types of RNAs.The proportion of sequences from exons was 0.38%,indicating a low degree of mRNA degradation in the samples.Base U had the highest proportion at the first position,and the bases with the highest proportions at positions 8 and 10 were U and A,respectively.Muscle-specific miRNAs(miR-1,miR-133,and miR-206)ranked in the top 10 in terms of expression level.The number and expression levels of new miRNAs were lower than those of known miRNAs.The length distribution,base bias at different positions and expression profile characteristics of miRNAs might be related to the biological function of miRNAs in regulating muscle proliferation and differentiation and the action mechanisms with target genes.[Conclusions]The identification and expression of miRNAs in muscle tissues of Sichuan white rabbits will help to understand the complex molecular mechanisms of meat production performance and provide a theoretical basis for the functional research of miRNAs in meat rabbits.

文摘Chiroptical switches based on circularly polarized luminescence(CPL)have shown the promising applications in advanced information technologies.Herein,a pair of lanthanide coordination polymer enantiomers[Eu_(2)(L^(R))_(3)(BTFPO)_(2)]_(n) and[Eu_(2)(L^(S))_(3)(BTFPO)_(2)]_(n) with light-regulated CPL property are designed,which are assembled by a chiral binuclear triple-stranded Eu^(3+) helicates[Eu_(2)(L^(R/S))_(3)]coordinated with two photochromic triphenylphosphine oxides(BTFPO).Upon the alternative UV and 526 nm light irradiation,the complexes show the reversible photochromism,PL and CPL responses.Notably,the luminescence dissymmetry factor,g lum of ^(5)D_(0)→^(7)F_(1)(591 nm)transition shows an obvious increase from 0.19 to 0.29 before and after 275 nm light irradiation.Additionally,the emission from Eu^(3+) center is not completely quenched in closed-ring state due to the low photocyclization(Ф_(o-c))quantum yield of the polymer.The partial maintenance of emissive intensity is of essential importance for the monitor of CPL signal.More importantly,the CPL photo-switching property of the complexes in solid hybrid film is maintained,and still displays the enhanced CPL emission in photostationary state.Further,the potential applications of the doping film in logic gate and anti-counterfeiting were investigated.

文摘Excessive use of antibiotics poses a threat to public health and the environment.In ecosystems,such as the marine environment,antibiotic contamination has led to an increase in bacterial resistance.Therefore,the study of bacterial response to antibiotics and the regulation of resistance formation have become an important research field.Traditionally,the processes related to antibiotic responses and resistance regulation have mainly included the activation of efflux pumps,mutation of antibiotic targets,production of biofilms,and production of inactivated or passivation enzymes.In recent years,studies have shown that bacterial signaling networks can affect antibiotic responses and resistance regulation.Signaling systems mostly alter resistance by regulating biofilms,efflux pumps,and mobile genetic elements.Here we provide an overview of how bacterial intraspecific and interspecific signaling networks affect the response to environmental antibiotics.In doing so,this review provides theoretical support for inhibiting bacterial antibiotic resistance and alleviating health and ecological problems caused by antibiotic contamination.

文摘Previous studies have indicated an association of fat mass and obesity-associated(FTO)with nonalcoholic fatty liver disease(NAFLD),the most common chronic liver disease worldwide.This study aimed to decipher the complex role of FTO in hepatic lipid metabolism.We found that a decrease in N^(6)-methyladenosine(m^(6)A)RNA methylation in the liver of mice fed with a high-fat diet(HFD)was accompanied by an increase in FTO expression.Overexpression of FTO in the liver promoted triglyceride accumulation by upregulating the expression of lipogenic genes.Mechanistical studies revealed that FTO could stabilize the mRNAs of sterol regulatory element binding transcription factor 1(SREBF1)and carbohydrate responsive element binding protein(ChREBP),two master lipogenic transcription factors,by demethylating m^(6)A sites.Knockdown ofeither SREBF1 or ChREBP attenuated the lipogenic effect of FTO,suggesting that they are bona fide effectors for FTO in regulating lipogenesis.Insulin could stimulate FTO transcription through a mechanism involving the action of intranuclear insulin receptor beta,while knockdown of FTO abrogated the lipogenic effect of insulin.Inhibition ofFTo by entacapone decreased the expression of SREBF1,ChREBP,and downstream lipogenic genes,ameliorating liver steatosis in HFD-fed mice.Thus,our study established a critical role of FTO in both the insulin-regulated hepatic lipogenesis and the pathogenesis of NAFLD and provided a potential strategyfor treating NAFLD.

文摘Muscle regeneration after damage or during myopathies requires a fine cooperation between myoblast proliferation and myogenic differentiation.A growing body of evidence suggests that microRNAs play critical roles in myocyte proliferation and differentiation transcriptionally.However,the molecular mechanisms underlying the orchestration are not fully understood.Here,we showed that miR-130b is able to repress myoblast proliferation and promote myogenic differentiation via targeting Sp1 transcription factor.Importantly,overexpression of miR-130b is capable of improving the recovery of damaged muscle in a freeze injury model.Moreover,miR-130b expression is declined in the muscle of muscular dystrophy patients.Thus,these results indicated that miR-130b may play a role in skeletal muscle regeneration and myopathy progression.Together,our findings suggest that the miR-130b/Sp1 axis may serve as a potential therapeutic target for the treatment of patients with muscle damage or severe myopathies.
文摘Increasing attention has been focused on the analysis of the realized volatil- ity, which can be treated as a proxy for the true volatility. In this paper, we study the potential use of the realized volatility as a proxy in a stochastic volatility model estimation. We estimate the leveraged stochastic volatility model using the realized volatility computed from five popular methods across six sampling-frequency transaction data (from 1-min to 60- min) based on the trust region method. Availability of the realized volatility allows us to estimate the model parameters via the MLE and thus avoids computational challenge in the high dimensional integration. Six stock indices are considered in the empirical investigation. We discover some consistent findings and interesting patterns from the empirical results. In general, the significant leverage effect is consistently detected at each sampling frequency and the volatility persistence becomes weaker at the lower sampling frequency.

文摘In a tritrophic context of plant-insect-entomopathogen,plants play important roles in modulating the interaction of insects and their pathogenic viruses.Currently,the influence of plants on the transmission of insect viruses has been mainly studied on baculoviruses and some RNA viruses,whereas the impact of plants on other insect viruses is largely unknown.Here,we identified a new densovirus infecting the green peach aphid Myzus persicae and tested whether and how host plants influence the transmission of the aphid densovirus.The complete single-stranded DNA genome of the virus,M.persicae densovirus 2,is 5727 nt and contains inverted terminal repeats.Transcription and phylogenetic analysis indicated that the virus was distinct from other a few identified aphid densoviruses.The virus abundance was detected highly in the intestinal tract of aphids,compared with the lower level of it in other tissues including head,embryo,and epidermis.Cabbage and pepper plants had no obvious effect on the vertical transmission and saliva-mediated horizontal transmission of the virus.However,the honeydew-mediated horizontal transmission among aphids highly depended on host plants(65%on cabbages versus 17%on peppers).Although the virus concentration in the honeydew produced by aphids between 2 plants was similar,the honeydew production of the infected aphids reared on peppers was dramatically reduced.Taken together,our results provide evidence that plants influence the horizontal transmission of a new densovirus in an aphid population by modulating honeydew secretion of aphids,suggesting plants may manipulate the spread of an aphid-pathogenic densovirus in nature.

文摘Fat is one of the three macronutrients and a significant energy source for piglets.It plays a positive role in maintaining intestinal health and improving production performance.During the weaning period,physiological,stress and diet-related factors influence the absorption of fat in piglets,leading to damage to the intestinal barrier,diarrhea and even death.Signaling pathways,such as fatty acid translocase(CD36),pregnane X receptor(PXR),and AMP-dependent protein kinase(AMPK),are responsible for regulating intestinal fat uptake and maintaining intestinal barrier function.Therefore,this review mainly elaborates on the reasons for diarrhea induced by insufficient fat absorption and related signaling pathways in weaned-piglets,with an emphasis on the intestinal fat absorption disorder.Moreover,we focus on introducing nutritional strategies that can promote intestinal fat absorption in piglets with insufficient fat absorption-related diarrhea,such as lipase,amino acids,and probiotics.

文摘Potato(Solanum tuberosum)is the most consumed non-cereal food crop.Most commercial potato cultivars are autotetraploids with highly heterozygous genomes,severely hampering genetic analyses and improvement.By leveraging the state-of-the-art sequencing technologies and polyploid graph binning,we achieved a chromosome-scale,haplotype-resolved genome assembly of a cultivated potato,Cooperation-88(C88).lntra-haplotype comparative analyses revealed extensive sequence and expression differences in this tetraploid genome.We identified haplotype-specific pericentromeres on chromosomes,suggesting a distinct evolutionary trajectory of potato homologous centromeres.Furthermore,we detected double reduction events that are unevenly distributed on haplotypes in 1021 of 1034 selfing progeny,a feature of autopolyploid inheritance.By distinguishing maternal and paternal haplotype sets in C88,we simulated the origin of heterosis in cultivated tetraploid with a survey of 3110 tetra-allelic loci with deleterious mutations,which were masked in the heterozygous condition bytwo parents.This study provides insights into the genomic architecture of autopolyploids and will guide their breeding.

文摘D-aspartate, a natural and endogenous amino acid, widely exists in animal tissues and can be synthesized through aspartate racemase and transformed by D-aspartate oxidase(DDO). D-aspartate mainly serves as a neurotransmitter and has been demonstrated to exhibit various physiological functions,including nutritional potential, regulation on reproduction and hormone biology, and neuron protection.This article mainly reviews the synthesis, racemization, and physiological functions of D-aspartate with emphasis on the potential in diseases.
