MicroRNAs (miRNAs) belong to a class of noncoding, regulatory RNAs that are involved in oncogenesis and show remarkable tissue specificity.miRNAs are approximately 22 nt non-coding RNAs, which regulate gene expression in a sequence-specific manner via translational inhibition or messenger RNA (mRNA) degradation, thus affecting various cellular processes.Since the discovery of their fundamental mechanisms of action, the field of miRNAs has opened a new era in the understanding of small noncoding RNAs.Recent evidence has shown that miRNA controls cell growth, apoptosis, and differentiation.Cancer is a complex genetic disease caused by abnormalities in gene structure and expression, moreover, miRNA expression correlates with cancers and could have a crucial function in tumor progression.Bioinformatic data indicate that each miRNA can control hundreds of target genes, but identification of the accurate miRNA targets will be crucial to exploit the emerging knowledge of miRNA contribution to cancer process. 展开更多

Role of Notoginsenoside Rg1 in Improving Spatial Cognitive Ability and Lowering Phosphorylation Level of Tau Protein in AD Model Rats 被引量:1

[Objectives] To study the effects and mechanism of notoginsenoside Rg1 on the spatial learning and memory and phosphorylated tau protein in the AD( Alzheimer's Disease) model rat. [Methods]The AD model rat was replicated by injection of Aβ_(25-35) in the left lateral ventricles of SD rats. The low dose( 25 mg/kg),middle dose( 50 mg/kg) and high dose( 100 mg/kg) notoginsenoside Rg1 was used for intragastric administration,respectively,two times every day. After 4 weeks,the Morris water maze test was done to detect the learning and memory capacity,and the immunoblotting,immunohistochemical methods were used to detect the changes in the phosphorylation level and distribution of tau protein in hippocampus of the rats. [Results] After the intracerebroventricular injection of Aβ_(25-35),the learning and memory capacity of the model rats was significantly lower than the learning and memory capacity of the normal control rats. The immunoblotting test results showed that the phosphorylation level of tau protein threonine 231 site( Thr231) in hippocampus was significantly increased,and the nonphosphorylation level was significantly decreased. The morphological testing results showed that the phosphorylation level of tau protein Thr231 of AD model rats was increased markedly in region of DG,CA1 and CA3 of the hippocampus. The intervention of the middle dose notoginsenoside Rg1 could significantly improve the learning and memory capacity of the model rats in Morris water maze. The notoginsenoside Rg1 in three different doses could all reduce the phosphorylation level of tau protein Thr231 in the hippocampal DG,CA1,CA3 regions,and there were no significant differences among the three doses. [Conclusions]The notoginsenoside Rg1 could improve Aβ_(25-35)-induced spatial learning and memory impairment of the AD model rats,and decreased the phosphorylation level of tau protein in hippocampus. 展开更多

The material feeding changing of combine harvester is easy to cause accumulation and blockage of the vibrating screen,which seriously affects the harvest operation.In order to alleviate such accumulation and blockages on the vibrating screen surface,the guide chute rotation angle of the improved variable amplitude screening mechanism was selected as the target variable,and EDEM-RecurDyn was employed to simulate the anti-blocking process of the variable amplitude under a changing feeding quantity(0.5 kg/s abnormal,0.2 kg/s normal)of materials(rice grain and stem mixture).A BP(an error back propagation algorithm)neural network was designed and the prediction model of the material distribution was subsequently constructed on the variable screening surface under different chute angles during abnormal feeding.The results revealed a continuous decrease in the quality and time of the material blockage at the front end of the screen surface with the increasing guide chute angle.At the guide chute angle of 20°-45°and adjustment time of 3-6 s,the blocked and accumulated materials at the front-end screen surface was be moved back to Grid 6 for screening.However,overtime,the screen surface materials continued to move back under the chute angle of 40°-45°,which had a great impact on the screening performance.At the guide chute angle of 30°-35°and adjustment time of 4 s,the materials on the screen surface were evenly distributed in Grid 1-6.This was able to alleviate the accumulation and blockage of the screen surface materials.The R of the material distribution prediction model(BP neural network)on the screen surface was determined as 0.97,indicating the high reliability and accuracy of the material distribution model on the screen surface based on the BP neural network.This work provides an important reference for the variable amplitude intelligent control of screen surface material anti-blocking. 展开更多

The Sulu orogenic belt is an uplift zone that was formed in the Late Triassic.Several Jurassic to Cretaceous sedimentary successions have been recognized within the Sulu orogenic belt in recent studies,including outcrops that are considered to be related to the newly discovered Riqingwei Basin.This basin has been the focus of extensive study due to its continuous Cretaceous rock sequence,geological location and petroleum resource potential.However,the lack of a consolidated chronology for the strata has precluded a better understanding of stratigraphy,tectonic evolution and resource potential of the Riqingwei Basin.Here,we present the results of a new magnetostratigraphic study of the continental scientific drilling borehole LK-1,which is located on Lingshan Island,offshore Shandong province,eastern China.The goals of this study are to(1)refine the Late Jurassic to Early Cretaceous chronostratigraphic framework of the Riqingwei Basin,and(2)investigate the location of the J/K boundary in the Borehole Core LK-1.The observed patterns of the paleomagnetic polarity zone in the LK-1 borehole correlate well with the geomagnetic polarity time scale(GPTS),and the continuous magnetostratigraphy profile defined in this core indicates an age ranging from 146.5 to 125.8 Ma for the samples interval.The sediment accumulation rates(SAR)of LK-1 show one period of high SAR(~10.5 cm kyr^(-1))at 135.3–130.6 Ma and two periods of low SAR(~4.8 and~2.2 cm kyr^(-1))at145.7–135.3 and 130.6–125.8 Ma,respectively.In addition,the magnetostratigraphic results suggest that the Jurassic-Cretaceous(J/K)boundary of the LK-1 is located within the magnetozone N21.2 n(~1254 m).This comprehensive geochronologic framework provides a good correlation of the marine Upper Jurassic to Lower Cretaceous strata in the Riqingwei Basin to other marine strata and continental sequences,in addition to providing a foundation for the study of the structural evolution of eastern China. 展开更多
