Parkinson’s disease is a neurodegenerative disease characterized by motor and gastrointestinal dysfunction.Gastrointestinal dysfunction can precede the onset of motor symptoms by several years.Gut microbiota dysbiosis is involved in the pathogenesis of Parkinson’s disease,whether it plays a causal role in motor dysfunction,and the mechanism underlying this potential effect,remain unknown.CCAAT/enhancer binding proteinβ/asparagine endopeptidase(C/EBPβ/AEP)signaling,activated by bacterial endotoxin,can promoteα-synuclein transcription,thereby contributing to Parkinson’s disease pathology.In this study,we aimed to investigate the role of the gut microbiota in C/EBPβ/AEP signaling,α-synuclein-related pathology,and motor symptoms using a rotenone-induced mouse model of Parkinson’s disease combined with antibiotic-induced microbiome depletion and fecal microbiota transplantation.We found that rotenone administration resulted in gut microbiota dysbiosis and perturbation of the intestinal barrier,as well as activation of the C/EBP/AEP pathway,α-synuclein aggregation,and tyrosine hydroxylase-positive neuron loss in the substantia nigra in mice with motor deficits.However,treatment with rotenone did not have any of these adverse effects in mice whose gut microbiota was depleted by pretreatment with antibiotics.Importantly,we found that transplanting gut microbiota derived from mice treated with rotenone induced motor deficits,intestinal inflammation,and endotoxemia.Transplantation of fecal microbiota from healthy control mice alleviated rotenone-induced motor deficits,intestinal inflammation,endotoxemia,and intestinal barrier impairment.These results highlight the vital role that gut microbiota dysbiosis plays in inducing motor deficits,C/EBPβ/AEP signaling activation,andα-synuclein-related pathology in a rotenone-induced mouse model of Parkinson’s disease.Additionally,our findings suggest that supplementing with healthy microbiota may be a safe and effective treatment that could help ameliorate the progression of motor deficits in patients with Parkinson’s disease. 展开更多

目的:探究单纯疱疹病毒(HSV)感染后脑脊液(CSF)中S100B、Cys-C、MMP-9水平对自身免疫性脑炎(AE)的预测价值。方法:选取2016年1月至2021年3月河北中石油中心医院收治的200例HSV感染患者为研究对象,根据是否继发AE分为研究组(继发AE,35例... 目的:探究单纯疱疹病毒(HSV)感染后脑脊液(CSF)中S100B、Cys-C、MMP-9水平对自身免疫性脑炎(AE)的预测价值。方法:选取2016年1月至2021年3月河北中石油中心医院收治的200例HSV感染患者为研究对象,根据是否继发AE分为研究组(继发AE,35例)和对照组(未继发AE,165例)。多因素Logistic回归分析HSV感染患者继发AE的独立影响因素。Spearman法分析脑脊液中Cys-C、MMP-9与S100B水平的相关性。受试者工作特征(ROC)曲线分析S100B、Cys-C、MMP-9对AE的预测价值。构建风险预测模型并进行评价。结果:多因素Logistic回归分析显示,MRI异常、脑脊液S100B、MMP-9升高、EEG异常是HSV感染患者继发AE的独立危险因素,脑脊液Cys-C是其保护因素(P<0.05)。Spearman分析显示,HSV感染患者Cys-C浓度与S100B水平呈负相关(r=-0.83,P<0.05),MMP-9浓度与S100B水平呈正相关(r=0.88,P<0.05)。构建的联合预测因子pre1诊断HSV患者继发AE的AUC明显大于S100B、Cys-C、MMP-9单独预测的AUC(0.876 vs 0.827、0.787、0.750)。构建的风险预测模型具有良好的区分度和一致性。结论:脑脊液中S100B、Cys-C、MMP-9水平均可对HSV感染患者诱发AE的可能性进行有效预测,且三项指标联合预测价值最大,其次是S100B蛋白、Cys-C、MMP-9。 展开更多

金盆梁是南秦岭柞水-山阳矿集区勘查的一处微细浸染型金矿床,矿床成因与成矿机制尚不清楚。矿体产于上泥盆统桐峪寺组粉砂质板岩和钙质板岩中,以浸染状、脉状金锑矿化为主,成矿过程可划分为3个阶段:毒砂-黄铁矿-硅化阶段(Ⅰ)、石英-辉锑... 金盆梁是南秦岭柞水-山阳矿集区勘查的一处微细浸染型金矿床,矿床成因与成矿机制尚不清楚。矿体产于上泥盆统桐峪寺组粉砂质板岩和钙质板岩中,以浸染状、脉状金锑矿化为主,成矿过程可划分为3个阶段:毒砂-黄铁矿-硅化阶段(Ⅰ)、石英-辉锑矿-白铁矿±锑氧化物阶段(Ⅱ)和方解石-石英阶段(Ⅲ)。流体包裹体及C-H-O-S-Pb同位素研究结果显示,Ⅱ阶段主要为金锑矿化,以H_(2)O-NaCl两相包裹体占绝对优势,成矿流体属于中温(200~290℃)、低盐度(w(NaCl_(eq))为0~6.0%)、低密度(0.64~0.99 g/cm^(3))的H_(2)O-NaCl±CO_(2)体系,以循环大气降水为主。无矿化的Ⅲ阶段主要发育H_(2)O-NaCl两相包裹体,含少量CO_(2)-H_(2)O-NaCl±CH_(4)、纯CO_(2)±CH_(4)及含子晶多相包裹体,流体以中低温(140~280℃)、低盐度(w(NaCl_(eq))为2.0%~8.0%)、低密度(0.68~1.02 g/cm^(3))的富CO_(2)-H_(2)O-NaCl±CH_(4)体系为主,或存在少量高温、高盐度、高密度H_(2)O-NaCl体系的岩浆热液混入。硫化物δ^(34)S值为较大负值(−12.50‰~−10.20‰),Pb同位素组成具上地壳源铅特征,成矿物质主要来源于围岩地层。综合研究表明,金盆梁金矿的成因类型属于卡林型金矿,水-岩反应(围岩硫化作用)是金富集沉淀的主要机制。 展开更多

老年脊柱骨折伴脊髓损伤患者早期血清pNF-H、NSE、S100B、C反应蛋白动态变化及意义

目的探讨老年脊柱骨折伴脊髓损伤患者早期血清神经丝蛋白H磷酸化亚型(pNF-H)、神经元特异性烯醇化酶(NSE)、S100B钙结合蛋白(S100B)、C反应蛋白动态变化特点及意义。方法老年脊柱骨折患者120例根据脊髓损伤神经功能评估结果分为脊髓未... 目的探讨老年脊柱骨折伴脊髓损伤患者早期血清神经丝蛋白H磷酸化亚型(pNF-H)、神经元特异性烯醇化酶(NSE)、S100B钙结合蛋白(S100B)、C反应蛋白动态变化特点及意义。方法老年脊柱骨折患者120例根据脊髓损伤神经功能评估结果分为脊髓未损伤组83例,脊髓不完全损伤组24例,脊髓完全损伤组13例。均行前路减压内固定术治疗,分别于脊柱骨折后12、24 h、7、14 d抽取空腹外周静脉血5 ml,酶联免疫吸附试验检测血清pNF-H、NSE、S100B、C反应蛋白水平。结果脊髓完全损伤组和脊髓不完全损伤组脊髓损伤后12、24 h、7 d血清pNF-H、NSE、S100B、C反应蛋白浓度逐渐升高,脊髓损伤后14 d下降;脊髓未损伤组脊髓损伤后12、24 h、7、14 d血清pNF-H、NSE、S100B、C反应蛋白浓度无明显变化。脊髓完全损伤组脊髓损伤后12、24 h、7、14 d血清pNF-H、NSE、S100B、C反应蛋白浓度明显高于脊髓不完全损伤组和脊髓未损伤组,脊髓不完全损伤组明显高于脊髓未损伤组(P<0.05)。受试者工作特征(ROC)曲线分析显示,血清pNF-H、NSE、S100B、C反应蛋白对老年脊柱骨折伴脊髓损伤均具有较高的诊断价值,其中血清pNF-H、NSE、S100B、C反应蛋白联合检测的诊断价值最高[曲线下面积(AUC)=0.857],灵敏度87.348%、特异度83.581%,高于各指标单独检测。结论pNF-H、NSE、S100B、C反应蛋白4项血清学指标联合检测在脊柱骨折伴脊髓损伤早期诊断和病情严重程度评估中具有较高的应用价值。 展开更多

中重度颅脑创伤患者血清STC1 S100B和NETRIN-1的表达及对预后的预测价值 被引量:2

目的探讨斯钙素抗体-1(STC1)、S100钙结合蛋白B(S100B)、轴突生长诱向因子-1(NETRIN-1)在中重度创伤性颅脑损伤(TBI)术后患者血清中的表达水平及预测预后的价值。方法收集74例TBI患者及40名健康者的血清样本,使用q RT-PCR检测健康者与TB... 目的探讨斯钙素抗体-1(STC1)、S100钙结合蛋白B(S100B)、轴突生长诱向因子-1(NETRIN-1)在中重度创伤性颅脑损伤(TBI)术后患者血清中的表达水平及预测预后的价值。方法收集74例TBI患者及40名健康者的血清样本,使用q RT-PCR检测健康者与TBI患者术前及术后第1天、第3天、第5天、第7天的血清STC1、S100B、NETRIN-1表达水平。收集TBI患者的一般临床资料,使用扩展版格拉斯哥结局量表(EGOS)评估患者预后,根据EGOS评分将患者分为预后良好组(EGOS≥5分,n=40)及预后不良组(EGOS<5分,n=34)。分析TBI患者术后血清STC1、S100B、NETRIN-1表达水平与预后的关系。采用受试者工作特征(ROC)曲线分析STC1、S100B、NETRIN-1表达水平对患者预后的预测价值,多因素Logistic分析评估引起TBI患者预后不良的因素。结果q RT-PCR分析显示,与健康者相比,TBI患者术前血清STC1、S100B水平升高,NETRIN-1水平降低,术后血清STC1、S100B水平降低,NETRIN-1水平升高(P<0.05)。TBI患者术后第1、3、5、7天血清STC1、S100B水平逐渐降低,NETRIN-1水平逐渐升高,术后第5天最显著(P<0.05)。与术后第5天预后良好组患者相比,预后不良组患者血清STC1、S100B水平显著升高,NETRIN-1水平显著降低(P<0.05)。皮尔逊相关性分析显示术后第5天TBI患者血清STC1、S100B水平与APACHEⅡ评分呈正比,NETRIN-1水平与APACHEⅡ评分呈反比(P<0.05)。ROC曲线分析发现术后第5天血清STC1、S100B、NETRIN-1水平对TBI患者预后具有预测价值,多因素Logistic回归分析显示仅STC1水平与患者预后相关(P<0.05)。结论STC1、S100B、NETRIN-1在TBI患者血清中表达水平异常,对TBI患者预后分析具有较高价值。 展开更多

内蒙古根河三道桥大型铅锌银矿床位于大兴安岭得尔布干成矿带中北段,矿体主要呈脉状赋存于火山岩地层中。氢氧同位素研究表明,成矿期石英和绢云母的δD值变化范围为-149.1‰~-156.7‰,δ^(18)OH_(2)O值变化范围为-13.6‰~3.4‰;成矿后... 内蒙古根河三道桥大型铅锌银矿床位于大兴安岭得尔布干成矿带中北段,矿体主要呈脉状赋存于火山岩地层中。氢氧同位素研究表明,成矿期石英和绢云母的δD值变化范围为-149.1‰~-156.7‰,δ^(18)OH_(2)O值变化范围为-13.6‰~3.4‰;成矿后期石英δD值变化范围为-131.9‰~-147.7‰,δ^(18)OH_(2)O值变化范围为-16.5‰~-18.2‰。碳同位素分析结果表明,与矿化有关的方解石δ^(13)C值变化范围为-1.8‰~-3.1‰,δ^(18)O值变化范围为5.3‰~8.6‰。原位S同位素分析结果表明,硫化物的δ^(34)S值变化范围为2.3‰~5.6‰,与其西南侧下护林矽卡岩型铅锌银矿床中的硫化物的δ^(34)S值(1.2‰~5.9‰)基本一致。上述同位素组成特征指示成矿物质主要来源于岩浆热液,在上升到地壳浅部时有一定量的大气降水混入。锆石U-Pb年代学研究表明,矿化的闪长玢岩脉年龄为(136.0±0.7)Ma(MSWD=0.44);未矿化、穿切硫化物微细脉的闪长玢岩脉的锆石U-Pb年龄为(120.8±0.6)Ma(MSWD=0.49)。结合前人相关研究进展,认为三道桥铅锌银矿床形成于136.0~120.8 Ma期间(早白垩世),为伸展构造背景下与浅成侵入岩有关的中温热液型铅锌银矿床。 展开更多

Epiphytic zooplankton community profiles in a typical urban wetland as revealed by DNA metabarcoding

Zooplankton,a crucial component of urban wetland,are one of the effective bioindicators for monitoring the feeding stocks of organisms at higher trophic levels and assessing the ecological quality of ecosystems.However,information about the characteristics of epiphytic zooplankton community structure resulted from traditional methods is limited and hindered by the large amount of detritus and sludge attached to the macrophytes.We investigated the epiphytic zooplankton communities associated with macrophytes(Vallisneria,Nymphaea,and Thalia dealbata)in a subtropical wetland using as DNA markers of the 18 S rRNA gene and the mitochondrial cytochrome c oxidase subunit I(COI)gene.A total of 241 OTUs of zooplankton were obtained from COI amplicons,including 194 OTUs of Rotifera,22 of Cladocera,and 25 of Copepoda,while only 62 OTUs of zooplankton were obtained from 18 S rDNA amplicons including 34 OTUs of Rotifera and 28 of Copepoda.The zooplankton communities associated with the three macrophytes were similar,but they differed significantly from those in the open waters.However,there were no significant temporal differences among the zooplankton communities.Epiphytic zooplankton communities were dominated by littoral zooplankton such as Testudinella,Lecane,and Philodina.Microzooplankton,especially littoral species,utilize macrophytes as food sources and as refuges against predation.This further led to an increase inαandβdiversity of zooplankton communities in urban wetlands.Our result suggests that the joint use of multiple molecular markers could improve the taxonomic resolution and generate a comprehensive biodiversity profile of zooplankton. 展开更多

In this editorial,we discussed the apparent discrepancy between the findings described by Colapietro et al,in their case report and data found in the literature.Colapietro et al reported a case of hepatitis B virus(HBV)-related hepatic decompensation in a patient with chronic myeloid leukemia and a previously resolved HBV infection who was receiving Bruton’s tyrosine kinase(BTK)inhibitor therapy.First of all,we recapitulated the main aspects of the immune system involved in the response to HBV infection in order to underline the role of the innate and adaptive response,focusing our attention on the protective role of anti-HBs.We then carefully analyzed literature data on the risk of HBV reactivation(HBVr)in patients with previous HBV infection who were treated with either tyrosine kinase inhibitors or BTK inhibitors for their hematologic malignancies.Based on literature data,we suggested that several factors may contribute to the different risks of HBVr:The type of hematologic malignancy;the type of therapy(BTK inhibitors,especially second-generation,seem to be at a higher risk of HBVr than those with tyrosine kinase inhibitors);previous exposure to an anti-CD20 as first-line therapy;and ethnicity and HBV genotype.Therefore,the warning regarding HBVr in the specific setting of patients with hematologic malignancies requires further investigation. 展开更多

In reptiles,such as the red-eared slider turtle(Trachemys scripta elegans),gonadal sex determination is highly dependent on the environmental temperature during embryonic stages.This complex process,which leads to differentiation into either testes or ovaries,is governed by the finely tuned expression of upstream genes,notably the testis-promoting gene Dmrt1 and the ovary-promoting gene Foxl2.Recent studies have identified epigenetic regulation as a crucial factor in testis development,with the H3K27me3 demethylase KDM6B being essential for Dmrt1 expression in T.s.elegans.However,whether KDM6B alone can induce testicular differentiation remains unclear.In this study,we found that overexpression of Kdm6b in T.s.elegans embryos induced the male development pathway,accompanied by a rapid increase in the gonadal expression of Dmrt1 at 31°C,a temperature typically resulting in female development.Notably,this sex reversal could be entirely rescued by Dmrt1 knockdown.These findings demonstrate that Kdm6b is sufficient for commitment to the male pathway,underscoring its role as a critical epigenetic regulator in the sex determination of the red-eared slider turtle. 展开更多

Pancreatic surgery units undertake several complex operations,albeit with consi-derable morbidity and mortality,as is the case for the management of complicated acute pancreatitis or chronic pancreatitis.The centralisation of pancreatic surgery services,with the development of designated large-volume centres,has contribu-ted to significantly improved outcomes.In this editorial,we discuss the complex associations between diabetes mellitus(DM)and pancreatic/periampullary disease in the context of pancreatic surgery and overall management of complex pancreatitis,highlighting the consequential needs and the indispensable role of specialist diabetes teams in support of tertiary pancreatic services.Type 3c pan-creatogenic DM,refers to DM developing in the setting of exocrine pancreatic disease,and its identification and management can be challenging,while the glycaemic control of such patients may affect their course of treatment and outcome.Adequate preoperative diabetes assessment is warranted to aid identification of patients who are likely to need commencement or escalation of glucose lowering therapy in the postoperative period.The incidence of new onset diabetes after pancreatic resection is widely variable in the literature,and depends on the type and extent of pancreatic resection,as is the case with pancreatic parenchymal loss in the context of severe pancreatitis.Early involvement of a specialist diabetes team is essential to ensure a holistic management.In the current era,large volume pancreatic surgery services commonly abide by the principles of enhanced recovery after surgery,with inclusion of provisions for optimisation of the perioperative glycaemic control,to improve outcomes.While various guidelines are available to aid perioperative management of DM,auditing and quality improvement platforms have highlighted deficiencies in the perioperative management of diabetic patients and areas of required improvement.The need for perioperative support of diabetic patients by specialist diabetes teams is uniformly underlined,a fact that becomes clearly more prominent at all different stages in the setting of pancreatic surgery and the management of complex pancreatitis.Therefore,pancreatic surgery and tertiary pancreatitis services must be designed with a provision for support from specialist diabetes teams.With the ongoing accumulation of evidence,it would be reasonable to consider the design of specific guidelines for the glycaemic management of these patients. 展开更多

Background: The World Health Organization (WHO) has set a goal to eradicate or at least significantly reduce the prevalence the human immunodeficiency virus (HIV), hepatitis B Virus (HBV) and Hepatitis C Virus (HCV) by 2030. The main objective was to provide an evolving overview of the prevalence of HIV, HBV and HCV infection between 2003 and 2022 in Burkina Faso. Methods: It was a retrospective cross-sectional study based on data from 2003 to 2022. The data were collected using information available in the databases of the HOSCO and CERBA laboratories and included all individuals who underwent HIV and/or HBV and/or HCV testing. Data analysis was performed using SPSS version 20.0, EpiInfo 7, and R version 4.1.0. Results were considered statistically significant if p Results: The study recorded 7432 samples and the mean age of the subjects was 27.98 ± 8.50 years. During this period, the respective prevalence of HIV, HBV, and HCV were 4.66% (346/7432), 8.77% (582/6636) and 5.54% (322/5816). However, from 2003 to 2022, there was a significant decrease (P y=−1.75x+12.59;y=−0.24x+10.01and y=−0.11x+6.02, with “y” corresponding to prevalence and “x” to the years. Background:Ziwan and Taoren(ZT)is a classic medicine pair in the formula of Mai Dong Di Shao Decoction,has been used to treat primary Sjogren’s syndrome(pSS)for more than 20 years.But its action mechanism is still unknown.This study is aimed to reveal the potential mechanism of ZT treated pSS and discover its active compounds of ZT and therapeutic target for pSS.Methods:Firstly,the potential pathways of ZT for pSS treatment were predicted through network pharmacology and GO and KEGG enrichment analysis.Secondly,the inter-structural relationships between active compounds of ZT and target proteins were visualized using molecular docking techniques.Finally,efficacy and mechanism were conducted through in vivo experiments,such as water intake,spleen index,hematoxylin-eosin staining pathological changes,ELISA,Western Blot analysis,and immunofluorescence staining.Results:Nine active compounds were extracted from network pharmacology,including quercitrin,luteolin,kaempferol,β-sitosterol,isorhamnetin,galangin,hederagenin,diosmetin and gibberellin 7.Seven disease targets were identified:RELA,TP53,AKT1,interleukin(IL)6,MAPK1,ESR1,IL10;with RELA being the most core target.KEGG and GO enrichment analysis indicated that ZT may act through the TLR/NF-κB/RELA inflammatory mechanism process.preliminary results of molecular docking showed that ZT’s active compounds bind well to the RELA(p65)receptor.In vivo results demonstrated that a high dose of ZT significantly improved water intake and reduced lymphocytes infiltration in submandibular gland pathology in NOD mice.The expression content of AQP5 and vasoactive intestinal peptide in the submaxillary gland was significantly increased,while levels of inflammatory factors such as tumor necrosis factor-α,IL-6,and IL-1βalong with protein expressions including toll-like receptor4,p-p65 and p-IKKα/βin NF-κB pathway were reduced.Conclusions:The ZT treatment exhibits a promising efficacy in mitigating dryness symptoms of pSS,potentially attributed to its capacity for suppressing the TLR/NF-κB inflammatory signaling pathway. 展开更多

Objective Alzheimer’s disease(AD)has become a significant global concern,but effective drugs able to slow down AD progression is still lacked.Electroacupuncture(EA)has been demonstrated to ameliorate cognitive impairment in individuals with AD.However,the underlying mechanisms remains poorly understood.This study aimed at examining the neuroprotective properties of EA and its potential mechanism of action against AD.Methods APP/PS1 transgenic mice were employed to evaluate the protective effects of EA on Shenshu(BL 23)and Baihui(GV 20).Chemogenetic manipulation was used to activate or inhibit serotonergic neurons within the dorsal raphe nucleus(DRN).Learning and memory abilities were assessed by the novel object recognition and Morris water maze tests.Golgi staining,western blot,and immunostaining were utilized to determine EA-induced neuroprotection.Results EA at Shenshu(BL 23)and Baihui(GV 20)effectively ameliorated learning and memory impairments in APP/PS1 mice.EA attenuated dendritic spine loss,increased the expression levels of PSD95,synaptophysin,and brain-derived neurotrophic factor in hippocampus.Activation of serotonergic neurons within the DRN can ameliorate cognitive deficits in AD by activating glutamatergic neurons mediated by 5-HT1B.Chemogenetic inhibition of serotonergic neurons in the DRN reversed the effects of EA on synaptic plasticity and memory.Conclusion EA can alleviate cognitive dysfunction in APP/PS1 mice by activating serotonergic neurons in the DRN.Further study is necessary to better understand how the serotonergic neurons-related neural circuits involves in EA-induced memory improvement in AD. 展开更多

目的探讨S100b联合CRP在诊断早产儿脑损伤(Brain injury in premature infants,BIPI)中的临床效能。方法选择2022年1月—2024年1月于本院儿科确诊的125例BIPI患儿作为BIPI组,另选择同期收治的85例非BIPI早产儿作为对照组。根据BIPI患儿... 目的探讨S100b联合CRP在诊断早产儿脑损伤(Brain injury in premature infants,BIPI)中的临床效能。方法选择2022年1月—2024年1月于本院儿科确诊的125例BIPI患儿作为BIPI组,另选择同期收治的85例非BIPI早产儿作为对照组。根据BIPI患儿病情严重程度分为轻度BIPI组和重度BIPI组两组亚组。收集所有研究对象的临床资料并检测血清中S100b联合CRP表达水平。Logistic回归分析发生BIPI的危险因素,S100b联合CRP在诊断BIPI的临床效能采用ROC曲线分析。结果对照组、轻度BIPI组和重度BIPI组三组间S100b、CRP表达水平存在统计学差异(P<0.05)。与对照组相比,轻度和重度BIPI组S100b、CRP表达水平均明显升高(P<0.05);与轻度BIPI组相比,重度BIPI组S100b、CRP表达水平均明显升高(P<0.05)。与对照组相比,宫内窘迫、出生后窒息、有创机械通气、新生儿呼吸窘迫综合征发生率及血清中S100b、CRP表达水平在BIPI组中明显升高(P<0.05)。Logistic回归分析结果显示,有创机械通气(OR=1.432)、新生儿呼吸窘迫综合征(OR=2.543)、S100b(OR=3.782)和CRP表达(OR=3.449)是发生BIPI的危险因素。S100b及CRP在诊断BIPI的曲线下面积分别为0.895和0.812,S100b联合CRP在诊断BIPI的曲线下面积为0.931,诊断敏感度和特异度分别为97.43%和92.25%。结论BIPI患儿外周血中S100b和CRP表达水平显著升高,S100b联合CRP在诊断BIPI中具有显著的临床价值。 展开更多

冷凝集素病(Cold agglutinin disease,CAD)是一种临床少见的由补体经典途径介导的一种明确的、克隆性的低级别的骨髓淋巴增生性疾病,主要以自身免疫性溶血性贫血(Autoimmune hemolytic anemia,AIHA)为特征[1],通常由免疫球蛋白M抗体(冷... 冷凝集素病(Cold agglutinin disease,CAD)是一种临床少见的由补体经典途径介导的一种明确的、克隆性的低级别的骨髓淋巴增生性疾病,主要以自身免疫性溶血性贫血(Autoimmune hemolytic anemia,AIHA)为特征[1],通常由免疫球蛋白M抗体(冷凝集素)与红细胞结合并固定补体引起。目前,CAD无明确的统一治疗方案,主要包括药物治疗和非药物治疗。临床常用的药物中,传统类固醇药仅在高不可接受的剂量下有效;B细胞靶向药物主要包括利妥昔单抗,其单药或是联合苯达莫司汀或氟达拉滨或硼替佐米治疗,具有一定的疗效。虽然利妥昔单抗与细胞抑制剂的组合增加了反应率和反应持续时间,但利妥昔单抗不但会消耗正常B细胞,导致机体免疫功能低下,且有在一年内疾病复发的高风险,并常伴随着明显药物毒性的不良反应[2],用药过程中需要及时监测其药物不良反应,以及时调整治疗剂量,尤其当治疗过程中出现对病人弊大于利的情况时,需及时停用。因此,其临床应用也具有一定的局限性。此外,目前已上市的C5抑制剂依库珠单抗,也是一种可选择的CAD药物[3-4],但经其治疗后血红蛋白上升的水平有限。非药物治疗措施主要包括输血、血浆置换、脾切除及注意保暖等,临床疗效也不尽人意,尤其是反复输血有异体免疫或铁超载的风险。因此,开发新的治疗CAD的药物是临床所迫切需要的。 展开更多

In patients with Alzheimer’s disease,gamma-glutamyl transferase 5(GGT5)expression has been observed to be downregulated in cerebrovascular endothelial cells.However,the functional role of GGT5 in the development of Alzheimer’s disease remains unclear.This study aimed to explore the effect of GGT5 on cognitive function and brain pathology in an APP/PS1 mouse model of Alzheimer’s disease,as well as the underlying mechanism.We observed a significant reduction in GGT5 expression in two in vitro models of Alzheimer’s disease(Aβ_(1-42)-treated hCMEC/D3 and bEnd.3 cells),as well as in the APP/PS1 mouse model.Additionally,injection of APP/PS1 mice with an adeno-associated virus encoding GGT5 enhanced hippocampal synaptic plasticity and mitigated cognitive deficits.Interestingly,increasing GGT5 expression in cerebrovascular endothelial cells reduced levels of both soluble and insoluble amyloid-βin the brains of APP/PS1 mice.This effect may be attributable to inhibition of the expression ofβ-site APP cleaving enzyme 1,which is mediated by nuclear factor-kappa B.Our findings demonstrate that GGT5 expression in cerebrovascular endothelial cells is inversely associated with Alzheimer’s disease pathogenesis,and that GGT5 upregulation mitigates cognitive deficits in APP/PS1 mice.These findings suggest that GGT5 expression in cerebrovascular endothelial cells is a potential therapeutic target and biomarker for Alzheimer’s disease. 展开更多

In this editorial,we comment on the article by Wang et al.This manuscript explores the potential synergistic effects of combining zanubrutinib,a novel oral inhibitor of Bruton’s tyrosine kinase,with high-dose methotrexate(HD-MTX)as a therapeutic intervention for primary central nervous system lymphoma(PCNSL).The study involves a retrospective analysis of 19 PCNSL patients,highlighting clinicopathological characteristics,treatment outcomes,and genomic biomarkers.The results indicate the combination’s good tolerance and strong antitumor activity,with an 84.2%overall response rate.The authors emphasize the potential of zanubrutinib to modulate key genomic features of PCNSL,particularly mutations in myeloid differentiation primary response 88 and cluster of differentiation 79B.Furthermore,the study investigates the role of circulating tumor DNA in cerebrospinal fluid for disease surveillance and treatment response monitoring.In essence,the study provides valuable insights into the potential of combining zanubrutinib with HD-MTX as a frontline therapeutic regimen for PCNSL.The findings underscore the importance of exploring alternative treatment modalities and monitoring genomic and liquid biopsy markers to optimize patient outcomes.While the findings suggest promise,the study’s limitations should be considered,and further research is needed to establish the clinical relevance of this therapeutic approach for PCNSL. 展开更多

E-learning platforms support education systems worldwide, transferring theoretical knowledge as well as soft skills. In the present study high-school pupils’, and adult students’ opinions were evaluated through a modern structured MOODLE interactive course, designed for the needs of the laboratory course “Automotive Systems”. The study concerns Greek secondary vocational education pupils aged 18 and vocational training adult students aged 20 to 50 years. The multistage, equal size simple random cluster sample was used as a sampling method. Pupils and adult students of each cluster completed structured 10-question questionnaires both before and after attending the course. A total of 120 questionnaires were collected. In general, our findings disclosed that the majority of pupils and adult students had significantly improved their knowledge and skills from using MOODLE. They reported strengthening conventional teaching, using the new MOODLE technology. The satisfaction indices improved quite, with the differences in their mean values being statistically significant. 展开更多
