Effect of Mg^(2+)level on the functions of CD8^(+)T lymphocytes and NK cells in patients with COVID-19

Objective:To investigate the changes of Mg^(2+) levels in serum and peripheral blood mononuclear cells(PBMCs)of patients with COVID-19 and its effects on the functions of CD8^(+)T lymphocytes and NK cells.Methods:A total of 165 COVID-19 patients hospitalized in Ezhou Central Hospital from January 20 to February 20,2020 were divided into mild/common group(98 cases)and severe/critical group(67 cases).At the same time,34 healthy persons were selected as the control group.Peripheral blood was collected and PBMCs were isolated,the level of Mg^(2+) in serum and PBMCs was detected.The subsets of CD8^(+)T lymphocytes and NK cell and the expression levels of their surface inhibitory molecular PD-1 and activator molecular NKG2D were detected by flow cytometry.The correlation between Mg^(2+) concentration and the expression levels of PD-1 and NKG2D was also analyzed.Results:Compared with the control group,the concentration of Mg^(2+) in serum and PBMCs,the counts of CD8^(+)T lymphocytes and NK cell in patients with mild/common and severe/critical groups were significantly reduced(P<0.05),while the expression level of surface inhibitory molecular PD-1 were significantly increased(P<0.05),while the expression level of the activation molecule NKG2D were significantly decreased(P<0.05).However,the changes of the above indicators in patients with severe/critical group were greater than those in the mild/common group(P<0.05).In addition,the Mg^(2+) concentration in COVID-19 patients was negatively correlated with the expression level of PD-1 on CD8^(+)T lymphocytes and NK cells(P<0.05),and positively correlated with the expression levels of NKG2D(P<0.05).Conclusion:The concentration of Mg^(2+) in the serum and PBMCs of COVID-19 patients is significantly reduced,which may cause the function of CD8^(+)T lymphocytes and NK cells to be inhibited. 展开更多

Autoimmune pancreatitis(AIP)is a rare form of pan-creatitis characterized by prominent lymphocyte inf iltration and pancreatic f ibrosis resulting in organ dysfunc-tion.The pathogenesis and pathology of AIP remain unknown.A 64-year-old Chinese man presented with symptoms and signs of bile duct obstruction diffuse enlargement of the head of pancreas,elevated IgG levels,and negative autoimmune antibody responses.A pylorus-preserving pancreatoduodenectomy was per-formed and a pancreatic tumor was suspected.Howev-er,periductal lymphoplasmacytic inf iltration and f ibrosis were found in the head of pancreas and nearby organs instead of tumor cells.Four months after surgery,the patient was readmitted because of reoccurrence ofsevere jaundice and sustained abdominal distension.Prednisone 30 mg/d was administered orally as an AIP was suspected.One and a half months later,the symp-toms of the patient disappeared,and globulin,amino-transferase and bilirubin levels decreased signif icantly.Over a 9-mo follow-up period,the dose of prednisone was gradually decreased to 10 mg/d and the patient remained in good condition.We further demonstrated dominant CD3+/CD8+ populations,CD20+ cells and a few CD4+ cells in the pancreatic parenchyma,duo-denum and gallbladder wall by immunohistochemical assay.This AIP case presented with signif icant CD8+ T lymphocyte inf iltration in the pancreas and extra-pan-creatic lesions,indicating that this cell population may be more important in mediating AIP pathogenesis than previously known and that AIP might be a poorly defined autoimmune disease with heterogeneous pathogenesis. 展开更多

Objective: To determine the role of CD4+ and CD8+ T lymphocytes in the onset of stroke in people living with HIV. Methodology: This was a descriptive, cross-sectional study from January to July 2019, in the neurology department of loandjili general hospital, including any patient hospitalized for a first episode of stroke confirmed by brain scan. The study variables were: age, sex, CRP value, serum T cell CD4+, CD8+. The statistical analysis was carried out using the EPI info 7 software. Results: Twenty stroke patients were included. The relative frequency of HIV was 20%. The risk factors were potentiated by immunosuppression of CD4+ T cells. Sixty percent (60%) of the patients had a CD4+ count < 200/mm<sup>3</sup> and the mean CD4+ count was ±191/mm<sup>3</sup>. Stroke was the predominant mechanism of injury with a frequency of 70%, the only injury mechanism of stroke in patients with CD8+ T cell count > 800/mm<sup>3</sup> (p = 0.04). Conclusion: Risk factors are potentiated by TCD4+ lymphocyte immunosupression, also CD8+ lymphocytes of immune system activation marker are a cardiovascular risk factor for living people with HIV. 展开更多

AIM: To assess if a specific cytotoxic T cell response can be induced in patients with malignant liver tumors treated with radio-frequency ablation (RFA). METHODS: Six Patients with liver metastases of colorectal cancer and 6 with hepatocellular carcinoma (HCC) underwent RFA. Blood was sampled before, 4 and 8 wk after RFA. Test antigens were autologous liver and tumor lysate obtained from each patient by biopsy. Peripheral T cell activation was assessed by an interferon gamma (IFNγ) secretion assay and flow cytometry. T cells were double-stained for CD4/CD8 and IFNγ to detect cytotoxic T cells. The ratio of IFNγ positive and IFNγ negative T cells was determined as the stimulation index (SI). To assess cytolytic activity, T cells were co-incubated with human CaCo colorectal cancer and HepG2 HCC cells and release of cytosolic adenylate kinase was measured by a luciferase assay. RESULTS: Before RFA SI was 0.021 (±0.006) for CD4^+ and 0.022 (± 0.004) for CD8^+T cells against nonmalignant liver tissue and 0.018 (± 0.005) for CD4^+ and 0.021 (± 0.004) for CD8^+ cells against autologous tumor tissue. Four weeks after RFA SI against tumor tissue increased to 0.109 (± 0.005) for CD4+ and 0.11 (± 0.012) for CD8+ T cells against HCC, and to 0.115 (± 0.031) for CD4^+ and 0.15 (± 0.02) for CD8^+ cells for colorectal metastases (P 〈 0.0001). No increased SI was observed with nonmalignant tumor tissue at all time points. Before RFA cytolytic activity against the respect(ve cancer cells was low with 2.62 (± 0.37) relative luminescence units (RLU), but rose more than 100 fold 4 and 8 wk after RFA. Spontaneous release was 〈 2% of maximum release in all experiments. CONCLUSION: Patients with primary and secondary tumors of the liver show a significant tumor-specific cytotoxic T-cell stimulation with a dramatically increased tumor specific cytolytic activity of CD8^+ T cells after RFA. 展开更多

Hepatocellular carcinoma (HCC), chronic hepatitis B (CHB) and chronic hepatitis C (CHC) are characterized by exhaustion of the specific CD8<sup>+</sup> T cell response. This process involves enhancement of negative co-stimulatory molecules, such as programmed cell death protein-1 (PD-1), cytotoxic T-lymphocyte antigen-4 (CTLA-4), 2B4, Tim-3, CD160 and LAG-3, which is linked to intrahepatic overexpression of some of the cognate ligands, such as PD-L1, on antigen presenting cells and thereby favouring a tolerogenic environment. Therapies that disrupt these negative signalling mechanisms represent promising therapeutic tools with the potential to restore reactivity of the specific CD8<sup>+</sup> T cell response. In this review we discuss the impressive in vitro and in vivo results that have been recently achieved in HCC, CHB and CHC by blocking these negative receptors with monoclonal antibodies against these immune checkpoint modulators. The article mainly focuses on the role of CTLA-4 and PD-1 blocking monoclonal antibodies, the first ones to have reached clinical practice. The humanized monoclonal antibodies against CTLA-4 (tremelimumab and ipilimumab) and PD-1 (nivolumab and pembrolizumab) have yielded good results in testing of HCC and chronic viral hepatitis patients. Trelimumab, in particular, has shown a significant increase in the time to progression in HCC, while nivolumab has shown a remarkable effect on hepatitis C viral load reduction. The research on the role of ipilimumab, nivolumab and pembrolizumab on HCC is currently underway. 展开更多

Recently, the human cochlea has been shown to contain numerous resident macrophages under steady-state. The macrophages accumulate in the stria vascularis, among the auditory nerves, and are also spotted in the human organ of Corti. These macrophages may process antigens reaching the cochlea by invasion of pathogens and insertion of CI electrode. Thus, macrophages execute an innate, and possibly an adaptive immunity. Here, we describe the molecular markers CD4 and CD8 of T cells, macrophage markers MHCⅡ and CD11b, as well as the microglial markers TEME119 and P2Y12, in the human cochlea. Immunohistochemistry and the advantageous super-resolution structured illumination microscopy(SR-SIM) were used in the study. CD4^+ and CD8^+ cells were found in the human cochleae. They were seen in the modiolus in a substantial number adjacent to the vessels, in the peripheral region of the Rosenthal's canal, and occasionally in the spiral ligament. While there are a surprisingly large number of macrophages in the stria vascularis as well as between the auditory neurons,CD4^+ and CD8^+ cells are hardly seen in these areas, and neither are seen in the organ of Corti. In the modiolus,macrophages, CD4^+ and CD8^+ cells appeared often in clusters. Interaction between these different cells was easily observed with SR-SIM, showing closely placed cell bodies, and the processes from macrophages reaching out and touching the lymphocytes. Otherwise the CD4^+ and CD8^+ cells in human cochlear tissue are discretely scattered. The possible roles of these immune cells are speculated. 展开更多

Objective To investigate the expression and regulation of programmed cell death protein 1(PD1),B lymphocyte and T lymphocyte attenuator(BTLA)in peripheral blood of patients with non-small cell lung cancer(NSCLC);to examine the correlation of the mRNA levels between PD and BTLA in NSCLC.Methods Flow cytometry was used to detect the expression of PD1 and BTLA on the surfaces of CD8^+T cells andγδ+T cells in the peripheral blood samples collected from 32 in-patients with stage IV NSCLC and 30 healthy individuals.We compared the expression of PD1 and BTLA on the surfaces ofγδ+T cells in the NSCLC patients with bone metastasis before and after the treatment of zoledronic acid.The correlations of PD1 and BTLA,as well as their ligands were analyzed using Pearson correlation analysis with the cBioPortal data platform.Results The frequency of PD1 on the surfaces of CD8^+T cells was significantly higher than that of theγδT cells in both healthy controls(t=2.324,P=0.024)and NSCLC patients(t=2.498,P=0.015).The frequency of PD1 on CD8^+T cells,rather than onγδ+T cells,was significantly upregulated in advanced NSCLC patients compared with that in healthy controls(t=4.829,P<0.001).The PD1+BTLA+γδT cells of the healthy controls were significantly lower than that of the NSCLC patients(t=2.422,P=0.0185).No differences in percentage of PD1+γδ+and BTLA+γδ+T cells were observed in 7 NSCLC patients with bone metastasis before and after zoledronic acid treatment.PD1 was positively correlated with BTLA in both lung adenocarcinoma(r=0.54;P<0.05)and lung squamous cell carcinoma(r=0.78;P<0.05).Conclusions The upregulation of co-inhibitory molecules occurs on the surfaces of both CD8^+T cells andγδT cells in advanced NSCLC,suggesting that these molecules were involved in regulating the inactivation of CD8^+T cells andγδ+T cells,immune escape and tumor invasion. 展开更多

BACKGROUND: The effect of pituitary adenylate cyclase activating polypeptide (PACAP) during traumatic brain injury (TBI) and whether it can modulate secondary injury has not been reported previously. The present study evaluated the potential protective effects of ventricular infusion of PACAP in a rat model of TBI.METHODS: Male Sprague Dawley rats were randomly divided into 3 treatment groups (n=6, each): sham-operated, vehicle (normal saline)+TBI, and PACAP+TBI. Normal saline or PACAP (1 μg/5 μL) was administered intracerebroventricularly 20 minutes before TBI. Right parietal cortical contusion was produced via a weight-dropping method. Brains were extracted 24 hours after trauma. Histological changes in brains were examined by HE staining. The numbers of CD4+ and CD8+ T cells in blood and the spleen were detected via flow cytometry.RESULTS: In injured brain regions, edema, hemorrhage, inflammatory cell infiltration, and swollen and degenerated neurons were observed under a light microscope, and the neurons were disorderly arrayed in the hippocampi. Compared to the sham group, average CD4+ CD8+ lymphocyte counts in blood and the spleen were significantly decreased in rats that received TBl+vehicle, and CD4- CD8+ were increased. In rats administered PACAP prior to TBI, damage was attenuated as evidenced by significantly increased CD4+, and decreased CD8+, T lymphocytes in blood and the spleen.CONCLUSION: Pretreatment with PACAP may protect against TBI by influencing periphery T cellular immune function. 展开更多

Antigen containing, allogeneic cells secreting the genetically modified protein and peptide-chaperone gp96-Ig cross, prime and expand antigen specific CD8 T cells with therapeutic antitumor activity in mice. In a first in man phase I study, we now report the results of therapeutic vaccination of non-small cell lung cancer (NSCLC) patients with an established, allogeneic non-small cell lung adenocarcinoma cell line secreting gp96-Ig. Advanced NSCLC-patients stage IIIB or IV of any histological subtype were enrolled and treated with up to 36 vaccinations over the course of 18 weeks. Primary endpoint was safety, secondary endpoints tumor response and overall survival. Measurement of tumor antigen specific CD8 CTL responses is precluded by the lack of known NSCLC associated antigens. Therefore, we measured patient CD8 T cell-IFN-γ responses to allo-antigens of the vaccine cells as surrogate for tumor antigen specific CD8 CTL. In 7 of 18 treated patients tumor growth was stabilized, however none of the 18 patients had an objective tumor response by RECIST criteria. Of 15 patients evaluable for immune response, 11 responded to vaccination with more than twofold increase in CD8-IFN-γ frequency above baseline. These patients had a median survival time of 16.5 months. Four patients who had no CD8 response above base line had survival times from 2.1 to 6.7 months. Our data are consistent with the concept that generation of CD8 CTL by therapeutic vaccination may delay tumor growth and progression and mediate prolonged survival even in the absence of objective tumor responses. Further studies will be required to test this concept and promising result. 展开更多

Background:CD8 positive T lymphocytes and natural killer(NK)cells in the peripheral blood of cervical cancer patients exhibit varying sensitivities to radiotherapy and chemotherapy.Methods:A total of 50 healthy peoples and 60 cervical cancer patients were recruited.The patients with cervical cancer were separated into two groups:radiation and chemotherapy,and blood sample were collected before and after treatment.Data on the proportion of CD8 positive T lymphocytes and NK cells were gathered for analytical evaluation.Results:Compared to healthy individuals,patients with cervical cancer exhibit a reduced proportion of CD8 positive T cells within their peripheral blood.And for patients with cervical cancer,radiation therapy has been found to be more effective than chemotherapy in increasing the proportion of CD8 positive T lymphocytes and NK cells.Conclusions:These results suggest that radiation therapy increases the levels of CD8 positive T lymphocytes and NK cells within the peripheral blood of patients with cervical cancer.The study hypothesis that the changes in the percentage of CD8 positive T lymphocytes may serve as a potential indicator for predicting treatment efficacy. 展开更多

补肾方对慢性乙型肝炎T细胞亚群及其功能的影响 被引量:18

目的观察补肾方对慢性乙型肝炎患者免疫功能状态的调节作用。方法收集HBeAg阳性ALT异常者30例,给予补肾方治疗,用药6个月,于用药前、用药3个月、6个月时分别检测下列指标。采用罗氏荧光定量PCR法检测HBVDNA ,外周血单个核细胞(periphera... 目的观察补肾方对慢性乙型肝炎患者免疫功能状态的调节作用。方法收集HBeAg阳性ALT异常者30例,给予补肾方治疗,用药6个月,于用药前、用药3个月、6个月时分别检测下列指标。采用罗氏荧光定量PCR法检测HBVDNA ,外周血单个核细胞(peripheralbloodmononuclearcell,PBMC)加入HBeAg、HBcAg与植物血凝素(Phytohemagglutinin ,PHA)培养48h ,检测培养上清液中IL 10、IFN -γ,血液中CD4+、CD8+、CD8+CD2 8+、CD8+CD2 8-和CD2 8+T细胞的比例及加入HBeAg、HBcAg与PHA培养48h后培养细胞中CD8+CD2 8+的比例。结果补肾方治疗有效组在治疗3个月后,PBMC培养中IFN- γ较治疗前明显上升,IL- 10明显下降(P <0 . 0 5,P <0. 0 1) ;CD8+CD2 8+T、CD2 8+T、培养后CD8+CD2 8+T细胞较治疗前明显上升,CD8+CD2 8-T细胞明显下降(P <0 .0 5或P <0 .0 1)。结论补肾方能明显增强Th1型细胞因子的表达,降低Th2型细胞因子的表达,促进CD8+(CytotoxicTlymphocyte ,CTL)的表达,这可能是使HBVDNA复制得到抑制的机制之一。 展开更多

目的探讨微波辐射对雷达作业人员外周血T淋巴细胞免疫活性的影响。方法采取30例雷达作业人员和20例健康人的外周血,对外周血T淋巴细胞进行银染,检测淋巴细胞核仁银染强度;另外采用微量全血直接荧光法标记CD3+、CD4+、CD8+,用流式细胞仪... 目的探讨微波辐射对雷达作业人员外周血T淋巴细胞免疫活性的影响。方法采取30例雷达作业人员和20例健康人的外周血,对外周血T淋巴细胞进行银染,检测淋巴细胞核仁银染强度;另外采用微量全血直接荧光法标记CD3+、CD4+、CD8+,用流式细胞仪检测T淋巴细胞亚群变化。结果巨嗜银核仁银染面积与细胞核银染面积的比值(I.S%),对照组为6.06±0.32,雷达作业组为5.12±0.57;对照组外周血CD3+、CD4+、CD8+百分比分别为(65.81±1.60)%、(32.58±0.71)%、(25.07±1.46)%,雷达作业组为(59.20±3.07)%、(28.32±2.66)%、(27.24±1.72)%。与对照组相比,雷达作业组外周血T淋巴细胞的I.S%明显降低(P<0.05),T淋巴细胞亚群CD3+、CD4+、CD4+/CD8+比值明显降低(P<0.05)。结论微波辐射对雷达作业人员的免疫活性产生明显影响,提示应加强雷达作业人员的防护措施。 展开更多

目的:了解不同类型乙型肝炎患者T淋巴细胞亚群的演变规律及意义。方法:我院2002年7月~2004年6月收治的16例急性肝炎、40例慢性肝炎及46例重型肝炎病例的抗凝血,通过流式细胞仪检测其T淋巴细胞亚群,对不同类型肝炎进行统计学分析。结果... 目的:了解不同类型乙型肝炎患者T淋巴细胞亚群的演变规律及意义。方法:我院2002年7月~2004年6月收治的16例急性肝炎、40例慢性肝炎及46例重型肝炎病例的抗凝血,通过流式细胞仪检测其T淋巴细胞亚群,对不同类型肝炎进行统计学分析。结果:慢性肝炎组与急性肝炎组比较CD3+,CD8+细胞计数降低且差异有显著性(P<0.05)。重型肝炎组与非重型肝炎组(急性肝炎组与慢性肝炎组)比较CD3+,CD4+,CD8+细胞计数明显降低(P<0.05),幼稚的CD4+,CD8+细胞计数明显升高(P<0.05)。结论:不同类型乙型肝炎的T淋巴细胞亚群有明显差异,乙型肝炎随着病情严重及病程延长,CD3+,CD4+,CD8+淋巴细胞逐渐减少,重型肝炎出现大量幼稚的CD4+CD8+淋巴细胞,T淋巴细胞亚群的变化可以反映病情严重程度及免疫状态,对免疫调节治疗有指导意义。 展开更多

CD8^(+) T lymphocytes are pivotal cells in the host response to antitumor immunity.Tumordriven microenvironments provide the conditions necessary for regulating infiltrating CD8^(+) T cells in favor of tumor survival,including weakening CD8^(+) T cell activation,driving tumor cells to impair immune attack,and recruiting other cells to reprogram the immune milieu.Also in tumor microenvironment,stromal cells exert immunosuppressive skills to avoid CD8^(+) T cell cytotoxicity.In this review,we explore the universal function and fate decision of infiltrated CD8^(+) T cells and highlight their antitumor response within various stromal architectures in the process of confronting neoantigen-specific tumor cells.Thus,this review provides a foundation for the development of antitumor therapy based on CD8^(+)T lymphocyte manipulation. 展开更多

目的观察人表皮生长因子受体通路底物8(Eps8)抗原改造表位是否有人白细胞抗原A(HLA-A)*0201限制性抗肿瘤能力。方法替换Eps8抗原锚定位点氨基酸获得改造肽,用BIMAS、SYFPEITHI在线数据库及Aotodock 4.2软件预测改造表位与HLA-A*0201分... 目的观察人表皮生长因子受体通路底物8(Eps8)抗原改造表位是否有人白细胞抗原A(HLA-A)*0201限制性抗肿瘤能力。方法替换Eps8抗原锚定位点氨基酸获得改造肽,用BIMAS、SYFPEITHI在线数据库及Aotodock 4.2软件预测改造表位与HLA-A*0201分子的结合力,筛选出稳定结合的改造表位E1-9V(ILDDIEFFV)、E1-1Y9V(YLDDIEFFV)。用经典肽结合力实验检测各改造表位与HLA-A*0201分子的亲和力。制备天然表位(E1)、E1-9V、E1-1Y9V特异性杀伤性T淋巴细胞(CTLs)。用乳酸脱氢酶(LDH)释放试验检测并比较E1-9V、E1-1Y9V特异性CTLs对MCF-7细胞、沉默Eps8的MCF-7细胞(MCF-7/shRNA)和抗HLA-A2抗体预孵育的MCF-7细胞(MCF-7/A2Ab)的杀伤率,E1-9V、E1-1Y9V特异性CTLs对SW480、U251、K562、IM-9细胞的杀伤率,E1、E1-9V、E1-1Y9V特异性CTLs对T2细胞的杀伤率。结果 E1-9V、E1-1Y9V均可与HLA-A*0201分子B、F对接口袋的氨基酸形成稳定氢键。肽结合力实验结果显示E1-9V、E1-1Y9V与HLA-A*0201均具有高亲和力,且高于天然表位E1,P均<0.05。E1-9V、E1-1Y9V特异性CTLs对MCF-7/shRNA、MCF-7/A2Ab细胞杀伤率明显低于MCF-7细胞,P均<0.05。E1-9V、E1-1Y9V特异性CTLs对SW480、U251细胞杀伤率明显高于K562、IM-9细胞(P均<0.05)。E1-1Y9V特异性CTLs对T2细胞的杀伤率显著高于E1、E1-9V特异性CTLs,P均<0.05。结论 Eps8抗原改造表位E1-9V、E1-1Y9V与天然表位E1相比具有更高的HLA-A*0201分子亲和力,保留了原有的免疫原性,并且E1-1Y9V抗肿瘤免疫效应强于天然表位E1。 展开更多

Among them human leukocyte antigen(HLA) locus plays a key role at different levels. In fact, genes of the HLA locus have shown the peculiar capability to modulate both innate and adaptive immune responses. In particular, HLA class Ⅰmolecules are recognized by CD8+ T-cells and natural killers(NK) cells towards the interaction with T cell receptor(TCR) and Killer Immunoglobulin Receptor(KIR) 3DL1 respectively. Polymorphisms within the different HLA alleles generate structural changes in HLA classⅠpeptide-binding pockets. Amino acid changes in the peptide-binding pocket lead to the presentation of a different set of peptides to T and NK cells. This review summarizes the role of HLA in HIV progression toward acquired immunodeficiency disease syndrome and its receptors. Recently, many studies have been focused on determining the HLA binding-peptides. The novel use of immune-informatics tools, from the prediction of the HLA-bound peptides to the modification of the HLAreceptor complexes, is considered. A better knowledge of HLA peptide presentation and recognition are allowing new strategies for immune response manipulation to be applied against HIV virus. 展开更多
