Xibei tree peony is a distinctive cultivar group that features red–purple blotches in petals.Interestingly,the pigmentations of blotches and non-blotches are largely independent of one another.The underlying molecula...Xibei tree peony is a distinctive cultivar group that features red–purple blotches in petals.Interestingly,the pigmentations of blotches and non-blotches are largely independent of one another.The underlying molecular mechanism had attracted lots of attention from investigators,but was still uncertain.Our present work demonstrates the factors that are closely related to blotch formation in Paeonia rockii‘Shu Sheng Peng Mo’.Non-blotch pigmentation is prevented by the silencing of anthocyanin structural genes,among which PrF3H,PrDFR,and PrANS are the three major genes.We characterized two R2R3-MYBs as the key transcription factors that control the early and late anthocyanin biosynthetic pathways.PrMYBa1,which belongs to MYB subgroup 7(SG7)was found to activate the early biosynthetic gene(EBG)PrF3H by interacting with SG5 member PrMYBa2 to form an‘MM’complex.The SG6 member PrMYBa3 interacts with two SG5(IIIf)bHLHs to synergistically activate the late biosynthetic genes(LBGs)PrDFR and PrANS,which is essential for anthocyanin accumulation in petal blotches.The comparison of methylation levels of the PrANS and PrF3H promoters between blotch and non-blotch indicated a correlation between hypermethylation and gene silencing.The methylation dynamics of PrANS promoter during flower development revealed a potential early demethylating reaction,which may have contributed to the particular expression of PrANS solely in the blotch area.We suggest that the formation of petal blotch may be highly associated with the cooperation of transcriptional activation and DNA methylation of structural gene promoters.展开更多
Chlorophylls and carotenoids are essential and bene fi cial substances for both plant and human health.Identifying the regulatory network of these pigments is necessary for improving fruit quality.In a previous study,...Chlorophylls and carotenoids are essential and bene fi cial substances for both plant and human health.Identifying the regulatory network of these pigments is necessary for improving fruit quality.In a previous study,we identi fi ed an R2R3-MYB transcription factor,SlMYB72,that plays an important role in chlorophyll and carotenoid metabolism in tomato fruit.Here,we demonstrated that the SlMYB72-interacting protein SlZHD17,which belongs to the zinc-fi nger homeodomain transcription factor family,also functions in chlorophyll and carotenoid metabolism.Silencing SIZHD 17 in tomato improved multiple bene fi cial agronomic traits,including dwar fi sm,accelerated fl owering,and earlier fruit harvest.More importantly,downregulating SIZHD17 in fruits resulted in larger chloroplasts and a higher chlorophyll content.Dual-luciferase,yeast one-hybrid and electrophoretic mobility shift assays clari fi ed that SlZHD17 regulates the chlorophyll biosynthesis gene SIPOR-B and chloroplast developmental regulator SITKN2 in a direct manner.Chlorophyll degradation and plastid transformation were also retarded after suppression of SIZHD17 in fruits,which was caused by the inhibition of SISGR1,a crucial factor in chlorophyll degradation.On the other hand,the expression of the carotenoid biosynthesis genes SIPSY1 and SIZISO was also suppressed and directly regulated by SlZHD17,which induced uneven pigmentation and decreased the lycopene content in fruits with SIZHD17 suppression at the ripe stage.Furthermore,the protein-protein interactions between SlZHD17 and other pigment regulators,including SlARF4,SlBEL11,and SlTAGL1,were also presented.This study provides new insight into the complex pigment regulatory network and provides new options for breeding strategies aiming to improve fruit quality.展开更多

The pen shell(Atrina pectinata) is a large wedge-shaped bivalve, which belongs to family Pinnidae. Due to its large and nutritious adductor muscle, it is the popular seafood with high commercial value in Asia-Pacific ...The pen shell(Atrina pectinata) is a large wedge-shaped bivalve, which belongs to family Pinnidae. Due to its large and nutritious adductor muscle, it is the popular seafood with high commercial value in Asia-Pacific countries. However, limiting genomic and transcriptomic data have hampered its genetic investigations. In this study, the transcriptome of A. pectinata was deeply sequenced using Illumina pair-end sequencing technology. After assembling, a total of 127263 unigenes were obtained. Functional annotation indicated that the highest percentage of unigenes(18.60%) was annotated on GO database, followed by 18.44% on PFAM database and 17.04% on NR database. There were 270 biological pathways matched with those in KEGG database. Furthermore, a total of 23452 potential simple sequence repeats(SSRs) were identified, of them the most abundant type was mono-nucleotide repeats(12902, 55.01%), which was followed by di-nucleotide(8132, 34.68%), tri-nucleotide(2010, 8.57%), tetra-nucleotide(401, 1.71%), and penta-nucleotide(7, 0.03%) repeats. Sixty SSRs were selected for validating and developing genic SSR markers, of them 23 showed polymorphism in a cultured population with the average observed and expected heterozygosities of 0.412 and 0.579, respectively. In this study, we established the first comprehensive transcript dataset of A. pectinata genes. Our results demonstrated that RNA-Seq is a fast and cost-effective method for genic SSR development in non-model species.展开更多
