The role of Toll-like receptor 4 (TLR4) and nuclear factor κB p65 (NF-κB p65) proteins in the pathogenesis of otitis media is explored. In recent years, the incidence of otitis media has been rising globally, becoming a significant threat to human health. More and more studies have found that Toll-like receptor 4 (TLR4), as a member of the Toll-like receptor family, can promote the generation of inflammatory factors and is closely related to the body’s immune response and inflammatory response. Nuclear factor-κB p65 (NF-κB p65) is a nuclear transcription factor that can interact with various cytokines, growth factors, and apoptotic factors, participating in processes such as oxidative stress, apoptosis, and inflammation in the body [1]. This article elaborates on the structure, function, and signaling pathways of TLR4 and NF-κB p65 proteins in the pathogenesis of otitis media, aiming to provide more precise targets and better therapeutic efficacy for the diagnosis and treatment of otitis media. 目的探讨垂体泌乳素瘤组织中核受体亚家族2c成员2(NR2C2)和细胞外基质金属蛋白酶诱导因子(EMMPRIN)的水平变化及其与肿瘤侵袭性、临床预后的关系。方法选择2019年6月—2021年6月本院收治的78例垂体泌乳素瘤患者为研究对象,患者均行手术... 目的探讨垂体泌乳素瘤组织中核受体亚家族2c成员2(NR2C2)和细胞外基质金属蛋白酶诱导因子(EMMPRIN)的水平变化及其与肿瘤侵袭性、临床预后的关系。方法选择2019年6月—2021年6月本院收治的78例垂体泌乳素瘤患者为研究对象,患者均行手术切除,术后随访时间1年。采用免疫组化法检测NR2C2和EMMPRIN表达情况,对比癌组织、癌旁组织中NR2C2和EMMPRIN表达,分析垂体泌乳素瘤患者癌组织中NR2C2和EMMPRIN表达情况与临床病理参数的关系,分析影响垂体泌乳素瘤患者预后的因素。结果癌组织中NR2C2阳性表达率高于癌旁组织(P<0.05),EMMPRIN阳性表达率低于癌旁组织(P<0.05),NR2C2、EMMPRIN表达均是预后不良的独立危险因素(P<0.05);受试者工作特征曲线(ROC)分析显示,垂体泌乳素瘤组织NR2C2、EMMPRIN两者联合预测垂体泌乳素瘤患者预后不良的曲线下面积(AUC)为0.886,高于NR2C2、EMMPRIN单独预测的AUC(Z=5.245,P=0.014;Z=3.652,P=0.028)。结论垂体泌乳素瘤患者癌组织中NR2C2和EMMPRIN表达与肿瘤侵袭性、临床预后有关,NR2C2阳性患者预后不良风险高。 展开更多

目的探究C-C趋化因子受体10(C-C chemokine receptor 10,CCR10)在结直肠癌(colorectal cancer,CRC)中对细胞增殖和铁死亡的影响,并探究其影响铁死亡的机制。方法在GEPIA数据库(Gene Expression Profiling Interactive Analysis)共表达分... 目的探究C-C趋化因子受体10(C-C chemokine receptor 10,CCR10)在结直肠癌(colorectal cancer,CRC)中对细胞增殖和铁死亡的影响,并探究其影响铁死亡的机制。方法在GEPIA数据库(Gene Expression Profiling Interactive Analysis)共表达分析CCR10与铁死亡关键基因的相关性。构建慢病毒介导的CCR10过表达(CCR10-OE)、敲除(CCR10-KO)及其阴性对照(NC)稳转HCT116细胞,采用细胞活力(CCK8)、克隆形成和裸鼠成瘤实验检测CCR10过表达或敲除后的细胞增殖。利用CCK8法、流式细胞术检测细胞铁死亡和脂质过氧化,评估CCR10过表达及敲除后细胞对铁死亡诱导剂erastin和rsl3的耐受。利用WB检测CCR10过表达和敲除后铁死亡关键蛋白质的表达。在CCR10敲除细胞株瞬时转染核因子E2相关因子2(NF-E2-related factor 2,NRF2)质粒,鉴定铁死亡表型以及其下游分子溶质载体家族7成员11(solute Carrier Family 7 Member 11,SLC7A11)、谷胱甘肽过氧化物酶4(glutathione Peroxidase 4,GPX4)、铁重链蛋白1(ferritin Heavy Chain 1,FTH1)表达水平。结果CCR10表达与铁死亡抑制基因BCL-2(r=0.54,P<0.001)、FTH1(r=0.18,P<0.001)和NRF2(r=0.31,P<0.001)呈正相关,与铁死亡驱动基因TP53(r=-0.46,P<0.001)、TFRC(r=-0.45,P<0.001)和KEAP1(r=-0.29,P<0.001)呈负相关。CCR10过表达促进CRC细胞增殖(P<0.01),而CCR10缺失抑制CRC细胞增殖(P<0.01)和小鼠皮下肿瘤生长(P<0.001)。CCR10过表达或缺失分别抑制或促进其对铁死亡的敏感性(P<0.01),并影响转录因子NRF2表达。结论CCR10基因可以通过上调转录因子NRF2表达而抑制erastin和rsl3诱导的CRC细胞铁死亡。 展开更多

Background:In many cancer types,aryl hydrocarbon receptor nuclear translocator 2(ARNT2)has been found to be associated with tumor cell proliferation and prognosis.However,the role of ARNT2 in clear cell renal cell carcinoma(ccRCC)has not been completely elucidated.In this study,the potential role of ARNT2 in ccRCC development was characterized.Methods:A pan-cancer dataset(TCGA-TARGET-GTEx)was accessed from UCSC Xena Data Browser.ARNT2 expression in normal and tumor samples was compared.Univariate Cox regression was performed to evaluate the prognostic value of ARNT2.Single sample gene set enrichment analysis(ssGSEA)was used to estimate the enrichment of functional pathways and gene signatures.CIBERSORT and ESTIMATE methods evaluated the immune infiltration.The ARNT2 expression was determined in ccRCC tissue and cell lines using RT-qPCR and Western blot.Results:ARNT2 expression was significantly dysregulated in 23 out of 30 cancer types.Pan-cancer data revealed a strong correlation between ARNT2 expression and immune modulators,immune cell infiltration,and genomic alternations.In ccRCC patients,the low-ARNT2 expression group had higher immune infiltration,CD8 T cells,and programmed cell death ligand 1 expression,as well as higher enrichment score of immunotherapeutic predictors than those in the high-ARNT2 expression group.Low-ARNT2 expression group was more responsive to immunotherapy.Moreover,low ARNT2 expression was observed in ccRCC tissue and cell lines.Conclusions:Dysregulated ARNT2 expression is involved in cancer development and the modulation of the immune microenvironment.ARNT2 can be potentially used as a prognostic indicator and an immunotherapeutic indicator for ccRCC. 展开更多

Soluble receptor for advanced glycation end products(sRAGE)acts as a decoy sequestering of RAGE ligands,thus preventing the activation of the ligand-RAGE axis linking human diseases.However,the molecular mechanisms underlying sRAGE remain unclear.In this study,THP-1 monocytes were cultured in normal glucose(NG,5.5 mmol/L)and high glucose(HG,15 mmol/L)to investigate the effects of diabetesrelevant glucose concentrations on sRAGE and interleukin-1β(IL-1β)secretion.The modulatory effects of epigallocatechin gallate(EGCG)in response to HG challenge were also evaluated.HG enhanced intracellular reactive oxygen species(ROS)generation and RAGE expression.The secretion of sRAGE,including esRAGE and cRAGE,was reduced under HG conditions,together with the downregulation of a disintegrin and metallopeptidase 10(ADAM10)and nuclear factor erythroid 2-related factor 2(Nrf2)nuclear translocation.Mechanistically,the HG effects were counteracted by siRAGE and exacerbated by siNrf2.Chromatin immunoprecipitation results showed that Nrf2 binding to the ADAM10 promoter and HG interfered with this binding.Our data reinforce the notion that RAGE and Nrf2 might be sRAGE-regulating factors.Under HG conditions,the treatment of EGCG reduced ROS generation and RAGE activation.EGCG-stimulated cRAGE release was likely caused by the upregulation of the Nrf2-ADAM10 pathway.EGCG inhibited HG-mediated NLRP3 inflammasome activation at least partly by stimulating sRAGE,thereby reducing IL-1βrelease. 展开更多

结直肠癌组织中NR2F1的表达及其与预后的相关性

目的检测结直肠癌中核受体亚家族2组F成员1(NR2F1)的蛋白表达水平,并探讨其与患者预后的关系。方法选择2012年8月至2013年9月在苏州市立医院进行根治性切除术的56例结直肠癌患者作为研究对象,采集患者的结直肠癌组织标本及对应的癌旁组... 目的检测结直肠癌中核受体亚家族2组F成员1(NR2F1)的蛋白表达水平,并探讨其与患者预后的关系。方法选择2012年8月至2013年9月在苏州市立医院进行根治性切除术的56例结直肠癌患者作为研究对象,采集患者的结直肠癌组织标本及对应的癌旁组织标本。采用EnVision法测定结直肠癌组织和癌旁组织的NR2F1的蛋白表达水平,比较不同NR2F1蛋白表达水平结直肠癌患者的临床病理特征。采用Kaplan-Meier法绘制结直肠癌患者的生存曲线,分析NR2F1蛋白表达水平与结直肠癌的预后的相关性。结果结直肠癌组织的NR2F1高表达率为48.21%,明显高于癌旁组织的7.14%,差异有统计学意义(P<0.05);NR2F1高表达者的无脉管侵犯率为85.19%,明显高于NR2F1低表达者的58.62%,差异有统计学意义(P<0.05);NR2F1高表达患者突破浆膜层的占比为48.15%,明显低于NR2F1低表达者的65.52%,且随着浸润深度从固有肌层到突破浆膜层,NR2F1低表达增加,差异均具有统计学意义(P<0.05);NR2F1高表达和低表达患者在性别、年龄、肿块大小、分化程度、淋巴结转移方面比较,差异均无统计学意义(P>0.05);经Kaplan-Meier生存曲线分析结果显示,结直肠癌的NR2F1高表达患者的10年总生存率为26.79%,明显高于NR2F1低表达患者的17.86%,差异有统计学意义(P<0.05)。结论结直肠癌的NR2F1表达升高,NR2F1可能参与了结直肠癌的发生、侵袭,对患者的预后造成一定的影响。 展开更多

Activated G-protein-coupled receptor 39(GPR39)has been shown to attenuate inflammation by interacting with sirtuin 1(SIRT1)and peroxisome proliferator-activated receptor-γcoactivator 1α(PGC-1α).However,whether GPR39 attenuates neuropathic pain remains unclear.In this study,we established a Sprague-Dawley rat model of spared nerve injury-induced neuropathic pain and found that GPR39 expression was significantly decreased in neurons and microglia in the spinal dorsal horn compared with sham-operated rats.Intrathecal injection of TC-G 1008,a specific agonist of GPR39,significantly alleviated mechanical allodynia in the rats with spared nerve injury,improved spinal cord mitochondrial biogenesis,and alleviated neuroinflammation.These changes were abolished by GPR39 small interfering RNA(siRNA),Ex-527(SIRT1 inhibitor),and PGC-1αsiRNA.Taken together,these findings show that GPR39 activation ameliorates mechanical allodynia by activating the SIRT1/PGC-1αpathway in rats with spared nerve injury. 展开更多

NR1D1和NR1D2转录因子属于核受体转录因子家族,二者通过调控靶基因表达参与众多生物学过程,尤其在昼夜节律调节中发挥重要作用。本研究通过分析NR1D1和NR1D2的蛋白质分子特征以及在冷胁迫下珍珠龙胆石斑鱼(Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂)的基因调控关系以及它们所参与的通路,揭示它们在鱼类低温环境中的功能作用。生物信息学比较分析显示NR1D1和NR1D2基因序列均有23个开放阅读框,编码的蛋白质均属于不稳定的亲水性蛋白质。NR1D1和NR1D2蛋白质均含有ZF-C4类锌脂蛋白结构域和Hormone_Recep核激素受体的配体结合域,主要配体为含铁原卟啉IX。这两种蛋白主要在细胞核中发挥调控基因表达的作用,且与昼夜节律通路的多个关键基因有直接作用关系。结果表明,在冷胁迫下NR1D1和NR1D2作为转录因子可通过调控靶基因的表达,参与昼夜节律调节并促进糖脂代谢、线粒体氧化等过程,为珍珠龙胆石斑鱼提供必要的能量,有助于增强其耐寒性。 展开更多

目的:探究核受体亚家族4A组成员1(nuclear receptor subfamily 4 group A member 1,Nr4a1)Nr4a1激动剂胞孢子酮B(cytosporone B,Csn-B)对小鼠噪声暴露后听力损失的治疗作用。方法:采用双氧水刺激HEI-OC1毛细胞系的方法构建氧化应激细胞... 目的:探究核受体亚家族4A组成员1(nuclear receptor subfamily 4 group A member 1,Nr4a1)Nr4a1激动剂胞孢子酮B(cytosporone B,Csn-B)对小鼠噪声暴露后听力损失的治疗作用。方法:采用双氧水刺激HEI-OC1毛细胞系的方法构建氧化应激细胞模型;通过实时荧光定量PCR(quantitative real-time PCR,q PCR)检测细胞中Nr4a1的mRNA表达水平;分别通过细胞计数试剂盒(cell counting kit-8,CCK8)及流式细胞术的方法检测细胞活力和细胞凋亡水平以评估Csn-B预处理后经双氧水刺激的细胞状态。构建小鼠噪声性听力损失模型,运用qPCR和免疫荧光技术检测噪声暴露后Nr4a1在小鼠耳蜗中的表达;通过检测听性脑干反应(auditory brainstem response,ABR)评估噪声暴露后以及Csn-B连续治疗13 d后小鼠听力情况。结果:双氧水刺激后HEI-OC1毛细胞中Nr4a1表达上升,细胞活力显著下降,凋亡水平显著升高;Csn-B预处理HEI-OC1毛细胞经双氧水刺激,细胞活力显著高于对照组而凋亡水平则显著低于对照组。在体研究结果显示,噪声暴露后小鼠听力显著降低,Nr4a1在小鼠耳蜗中的表达水平显著升高。噪声暴露后经Csn-B治疗小鼠听力得到改善,主要表现为Click-ABR以及Tone Burst-ABR(4000、8000Hz处)阈值下降。结论:Nr4a1激动剂Csn-B增强内耳毛细胞对氧化应激损伤的抵御能力,部分改善噪声暴露后的小鼠听力。 展开更多

Traumatic brain injury involves complex pathophysiological mechanisms,among which oxidative stress significantly contributes to the occurrence of secondary injury.In this study,we evaluated hypidone hydrochloride(YL-0919),a self-developed antidepressant with selective sigma-1 receptor agonist properties,and its associated mechanisms and targets in traumatic brain injury.Behavioral experiments to assess functional deficits were followed by assessment of neuronal damage through histological analyses and examination of blood-brain barrier permeability and brain edema.Next,we investigated the antioxidative effects of YL-0919 by assessing the levels of traditional markers of oxidative stress in vivo in mice and in vitro in HT22 cells.Finally,the targeted action of YL-0919 was verified by employing a sigma-1 receptor antagonist(BD-1047).Our findings demonstrated that YL-0919 markedly improved deficits in motor function and spatial cognition on day 3 post traumatic brain injury,while also decreasing neuronal mortality and reversing blood-brain barrier disruption and brain edema.Furthermore,YL-0919 effectively combated oxidative stress both in vivo and in vitro.The protective effects of YL-0919 were partially inhibited by BD-1047.These results indicated that YL-0919 relieved impairments in motor and spatial cognition by restraining oxidative stress,a neuroprotective effect that was partially reversed by the sigma-1 receptor antagonist BD-1047.YL-0919 may have potential as a new treatment for traumatic brain injury. 展开更多

目的观察桃红四物汤对糖尿病大鼠心脏的保护作用,同时基于核因子E2相关因子2(nuclear factor-E2-related factor 2,Nrf2)/血红素加氧酶-1(heme oxygenase-1,HO-1)和NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein d... 目的观察桃红四物汤对糖尿病大鼠心脏的保护作用,同时基于核因子E2相关因子2(nuclear factor-E2-related factor 2,Nrf2)/血红素加氧酶-1(heme oxygenase-1,HO-1)和NOD样受体热蛋白结构域相关蛋白3(NOD-like receptor thermal protein domain associated protein 3,NLRP3)通路探讨其作用机制。方法腹腔注射链脲佐菌素复制糖尿病心肌病大鼠模型,将模型复制成功的大鼠分为模型组,桃红四物汤低、中、高剂量组,每组10只,另设10只正常大鼠作为对照组。比较各组大鼠血糖、超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)和B型利钠肽(B-type natriuretic peptide,BNP)、肌钙蛋白I(cardiac troponin I,cTnI)水平。采用苏木精—伊红染色和Masson染色观察大鼠心肌组织病理变化。制备心肌组织匀浆,使用流式细胞术进行线粒体膜电位检测。Western blot法检测Nrf2、HO-1、Kelch样环氧氯丙烷相关蛋白1(Kelch-like ECH-associated protein 1,Keap1)、NLRP3、半胱氨酸天冬氨酸酶1剪切体(cleaved-caspase-1)、消皮素D(gasdermin D,GSDMD)和消皮素D蛋白N端结构域(N-terminal gasdermin D-N,GSDMD-N)蛋白表达水平。结果与对照组比较,模型组大鼠血糖升高,心肌纤维断裂、排列紊乱,炎症细胞浸润明显,胶原沉积明显;与模型组比较,桃红四物汤低、中、高剂量组大鼠心肌纤维化明显改善。与对照组比较,模型组SOD活性显著降低(P<0.05),MDA、BNP及cTnI水平显著升高(P<0.05);与模型组比较,桃红四物汤低、中、高剂量组SOD活性显著升高(P<0.05),MDA、BNP、cTnI水平显著降低(P<0.05)。与对照组比较,模型组心肌组织线粒体膜电位显著降低(P<0.05);与模型组比较,桃红四物汤低、中、高剂量组心肌组织线粒体膜电位显著升高(P<0.05)。与对照组比较,模型组心肌组织中Nrf2、HO-1、NLRP3、cleaved-caspase-1、GSDMD和GSDMD-N蛋白表达水平均显著升高(P<0.05);与模型组比较,桃红四物汤低、中、高剂量组心肌组织中Nrf2和HO-1蛋白表达水平均显著升高(P<0.05),Keap1、NLRP3、cleaved-caspase-1、GSDMD和GSDMD-N蛋白表达水平均显著降低(P<0.05)。结论桃红四物汤改善糖尿病心肌病的机制可能与调节Nrf2-NLRP3途径,抗氧化应激及抑制细胞焦亡相关。 展开更多

目的探讨丁酸钠(sodium butyrate,SB)对脂多糖(lipopolysaccharide,LPS)诱导肺泡巨噬细胞极化的影响及其作用机制。方法小鼠肺泡巨噬细胞(MH-S细胞)随机分为对照(Control)组、LPS组、SB组、LPS+SB(LB)组、LPS+SB+腺苷酸活化蛋白激酶(AM... 目的探讨丁酸钠(sodium butyrate,SB)对脂多糖(lipopolysaccharide,LPS)诱导肺泡巨噬细胞极化的影响及其作用机制。方法小鼠肺泡巨噬细胞(MH-S细胞)随机分为对照(Control)组、LPS组、SB组、LPS+SB(LB)组、LPS+SB+腺苷酸活化蛋白激酶(AMPK)抑制剂(Compound C)(LC)组、LPS+SB+核因子E2相关因子2(Nrf2)抑制剂(ML385)(LM)组。通过CCK8检测MH-S细胞活力,筛选出最佳的1000 ng/mL LPS、1 mmol/L SB、10μmol/L Compound C、5μmol/L ML385药物浓度进行后续实验;实时荧光定量(qRT-PCR)检测MH-S细胞白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)、白细胞分化抗原86(CD86)、巨噬细胞甘露糖受体(CD206)、AMPK、Nrf2和血红素加氧酶1(HO-1)的mRNA表达水平;酶联免疫吸附试验(ELISA)检测培养基上清IL-6、TNF-α、IL-1β和IL-10蛋白含量;流式细胞术测定M1和M2型巨噬细胞相关标记物CD86和CD206的表达。各组数据通过单因素方差分析和Tukey法进行检验。结果通过CCK8选取了1000 ng/mL LPS、1 mmol/L SB、10μmol/L Compound C和5μmol/L ML385进行造模和干预。qRT-PCR和ELISA结果一致显示,与LPS组比较,LB组M1型巨噬细胞相关促炎细胞因子IL-6、TNF-α、IL-1β显著降低(均P<0.01),但M2型巨噬细胞相关抑炎细胞因子IL-10显著升高(均P<0.01)。qRT-PCR和流式细胞术结果一致显示,与Control组比较,LPS组CD86水平显著升高(均P<0.01),SB组差异无统计学意义;与LPS组比较,LB组CD86表达水平显著降低(均P<0.01),但M2型巨噬细胞标记物CD206的变化趋势与CD86相反。qRT-PCR结果显示,与LPS组比较,LB组促进AMPK/Nrf2/HO-1的表达(均P<0.05);与LB组比较,LC组降低了AMPK/Nrf2/HO-1的表达(均P<0.05),LM组降低了Nrf2/HO-1的表达(均P<0.05)。流式细胞术结果显示,与LB组比较,LC组和LM组逆转了SB对CD86水平的抑制作用(均P<0.01);M2型巨噬细胞标记物CD206的表达趋势与M1型巨噬细胞标记物CD86相反。结论SB通过激活AMPK/Nrf2/HO-1信号通路,抑制LPS诱导的M1型、促进M2型肺泡巨噬细胞极化,改善了炎症反应。 展开更多

乳腺癌组织NRIP1、DUSP14表达及其与患者预后的关系

目的:探讨血清核因子E2相关因子2(Nrf2)、转化生长因子结合蛋白2(LTBP2)、血小板内皮细胞黏附分子1(PECAM1)与非小细胞肺癌(NSCLC)患者表皮生长因子受体(EGFR)靶向治疗反应性关系及意义。方法:选取我院2021年1月—2023年1月收治的95例NS... 目的:探讨血清核因子E2相关因子2(Nrf2)、转化生长因子结合蛋白2(LTBP2)、血小板内皮细胞黏附分子1(PECAM1)与非小细胞肺癌(NSCLC)患者表皮生长因子受体(EGFR)靶向治疗反应性关系及意义。方法:选取我院2021年1月—2023年1月收治的95例NSCLC患者为研究对象,根据EGFR靶向治疗3个月后的治疗反应性分为有效组(54例)、无效组(41例)。比较2组治疗前及治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平,分析治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平与NSCLC患者EGFR靶向治疗反应性相关性,偏回归分析治疗无效的影响因素,受试者工作特征曲线(ROC)分析治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平联合检测对NSCLC患者EGFR靶向治疗效果的预测价值。结果:治疗1个月、2个月后无效组血清Nrf2、LTBP2、PECAM1水平均高于有效组(P<0.05);相关性分析发现,治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平与治疗反应性均呈负相关(P<0.05);Logistic回归分析发现,治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平为NSCLC患者EGFR靶向治疗效果的影响因素(P<0.05);ROC曲线分析发现,治疗1个月、2个月后血清Nrf2、LTBP2、PECAM1水平联合预测治疗无效的曲线下面积(AUC)分别为0.761、0.816,最佳预测敏感度、特异度分别为(85.37%、66.67%)、(92.68%、70.37%),高于单一指标预测(P<0.05)。结论:血清Nrf2、LTBP2、PECAM1水平与NSCLC患者EGFR靶向治疗反应性密切相关,并在预测治疗效果方面具有较高效能。 展开更多

Objective To explore Effects of marine collagen peptides (MCPs) on markers of metablic nuclear receptors, i.e peroxisome proliferator-activated receptor (PPARs), liver X receptor (LXRs) and farnesoid X receptor (FXRs) in type 2 diabetic patients with/without hypertension. Method Study population consisted of 200 type 2 diabetic patients with/without hypertension and 50 healthy subjects, all of whom were randomly assigned to MCPs-treated diabetics (n=50), placebo-treated diabetics (n=50), MCPs-treated diabetics with hypertension (n=50), placebo-treated diabetics with hypertension (n=50), and healthy controls (n=50). MCPs or placebo (water-soluble starch) were given daily before breakfast and bedtime over three months. Levels of free fatty acid, cytochrome P450, leptin, resistin, adiponectin, bradykinin, NO, and Prostacyclin were determined before intervention, and 1.5 months, and 3 months after intervention. Hypoglycemia and the endpoint events during the study were recorded and compared among the study groups. Result At the end of the study period, MCPs-treated patients showed marked improvement compared with patients receiving placebo. The protection exerted by MCPs seemed more profound in diabetics than in diabetics with hypertension. In particular, after MCPs intervention, levels of free fatty acid, hs-CRP, resistin, Prostacyclin decreased significantly in diabetics and tended to decrease in diabetic and hypertensive patients whereas levels of cytochrome P450, leptin, NO tended to decrease in diabetics with/without hypertension. Meanwhile, levels of adiponectin and bradykinin rose markedly in diabetics following MCPs administration. Conclusion MCPs could offer protection against diabetes and hypertension by affecting levels of molecules involved in diabetic and hypertensive pathogenesis. Regulation on metabolic nuclear receptors by MCPs may be the possible underlying mechanism for its observed effects in the study. Further study into its action may shed light on development of new drugs based on bioactive peptides from marine sources. 展开更多

Patients with fatty liver display features of metabolic syndrome(Met S), like insulin resistance(IR), glucose intolerance, hypertension and dyslipidemia. Recently, epidemiological studies have linked obesity, Met S, and NAFLD to decreased bone mineral density and osteoporosis, highlighting an intricate interplay among bone, adipose tissue, and liver. Osteoprotegerin(OPG), an important symbol of the receptor activator of nuclear factor-B ligand/receptor activator of nuclear factor kappa B/OPG system activation, typically considered for its role in bone metabolism, may also play critical roles in the initiation and perpetuation of obesityrelated comorbidities. Clinical data have indicated that OPG concentrations are associated with hypertension, left ventricular hypertrophy, vascular calcification, endothelial dysfunction, and severity of liver damage in chronic hepatitis C. Nonetheless, the relationship between circulating OPG and IR as a key feature of Met S as well as between OPG and NAFLD remains uncertain. A well defined progression model of accumulation of genetic alterations ranging from single point mutations to gross chromosomal abnormalities has been introduced to describe the origin of this disease. However, due to the its subtle nature and concurring events PDAC cure remains elusive. Nuclear receptors (NR) are members of a large superfamily of evolutionarily conserved ligand-regulated DNA-binding transcription factors functionally involved in important cellular functions ranging from regulation of metabolism, to growth and development. Given the nature of their ligands, NR are very tempting drug targets and their pharmacological modulation has been widely exploited for the treatment of metabolic and inflammatory diseases. There are now clear evidences that both classical ligand-activated and orphan NR are involved in the pathogenesis of PDAC from its very early stages; nonetheless many aspects of their role are not fully understood. The purpose of this review is to highlight the striking connections that link peroxisome proliferator activated receptors, retinoic acid receptors, retinoid X receptor, androgen receptor, estrogen receptors and the orphan NR Nur, chicken ovalbumin upstream promoter transcription factor II and the liver receptor homologue-1 receptor to PDAC development, connections that could lead to the identification of novel therapies for this disease. 展开更多
