In this experiment, a 316 bp and 382 bp fragments of TLR4 gene named T4CRBR1 and T4CRBR2, of Chinese Holstein, Sanhe cattle, and Chinese Simmental was amplified by polymerase chain reaction (PCR), respectively. The genetic polymorphisms in the three populations were detected by Single-Strand Conformational Polymorphism (SSCP) in the first locus and by digesting the fragments with restriction endonuclease Alu I in the second one. Results showed that both alleles (A and B) of two loci were found in all the three populations and the value of polymorphism information content (PIC) indicated that these were a moderate polymorphism. Statistical results of X^2 test indicated that two polymorphism sites in the three populations fitted with Hardy-Weinberg equilibrium (P 〉 0.05). After sequencing, A-G single nucleotide polymorphism (SNP) was identified at nucleotide 4,525 in intron 1 of TLR4 gene and C-T SNP was identified at nucleotide 1,397 in exon 3 of TLR4 gene. Meanwhile, the effect of polymorphism of TLR4 gene on somatic cell score (SCS) was analyzed, the results indicated that the cattle with allele A in T4CRBR1 showed lower somatic cell score than that of allele B (P 〈 0.05). This tumor can proliferate rapidly and cause cystic changes because of internal tissue necrosis. We evaluated a 54-year-old woman with right breast lump. Mammography showed a category four mass with a diameter of 2.5 cm. Ultrasonography(US) revealed a complex cystic lesion, and fine-needle aspiration(FNA) cytology demonstrated bloody fluid and malignant cells. Partial breast resection and sentinel lymph node biopsy were performed. Immunohistology revealed spindle cells with positive results for cytokeratin(AE1/AE3) and vimentin, partially positive results for s-100, and negative results for desmin and α-actin. The pathological stage was IIA, and biochemical characterization showed that the tumor was triple negative. Six courses of FEC-100 chemotherapy(5-fluorouracil 500 mg/m2, epirubicin 100 mg/m2, and cyclophosphamide 500 mg/m2) were administered. Radiotherapy was performed. This case is discussed with reference to the literature.展开更多

MicroRNA (miRNA) is a large class of small noncoding RNA of about 22 nt involved in the regulation of various biological processes. This study aims to identify the miRNA expression profile in PMBC. Methods: MiRNA expression profiles in 11 PMBCs were analyzed by miRNA-microarray and real-time polymerase chain reaction (PCR). Thirty-one PMBCs and 27 invasive ductal carcinoma of no special types (IDC-NSTs) were assessed by immunohistochemistry using antibodies against ER, PR-progesterone receptor, HER2, Ki-67, Bcl-2, p53, PCNA, and CK5 and 6. Results: We analyzed the miRNA expression in 11 PMBCs and corresponding normal tissues using miRNA-microarray and real-time PCR, and found that miR-143 and miR-224-5p were significantly downregulated in mucinous carcinoma tissue. Compared with IDC-NSTs, PMBC showed a significantly higher ER positive rate, lower HER-2 positive rate, and lower cell proliferation rates. Conclusions: To our knowledge, this is the first study to demonstrate the miRNA expression profile of PMBC, and our findings may lead to further understanding of this type of breast cancer.展开更多

The present study explores the ICs0 value of chitosan-poly (vinyl alcohol) hydrogel (Cs/PVA) and Ag-doped chitosan-poly (vinyl alcohol) (Cs/PVA/Ag) nanocomposite in view of their anticancer application. The aim was to develop (Cs/PVA) based hydrogel synthesized by gamma radiation which could behave both as a nanoreactor for Ag nanoparticle with promising anticancer applications. The (Cs/PVA/Ag) nanocomposite was confirmed by FTIR (Fourier transform infrared) spectroscopy, XRD (X-ray diffraction) and EDX (energy dispersive X-ray) analysis. The anti-cancer activity of the prepared nanocomposites was demonstrated in human liver cancer cell line (HEPG2) and breast cancer cell lines (MCF7). It has significant effects against human liver cancer cell line HEPG2 compared to breast cancer cell line MCF7. Further quantitative analysis on the molecular and protein levels is still required to confirm the impact of chitosan on genotoxic effect before reaching a final conclusion and starting its biomedical application.展开更多

Methods: A 47-year-old female patient was admitted to our center (Department of General Surgery, the First Affiliated Hospital, Luzhou Medical College, China) due to a large left breast mass for about 2.5 years. Core tissue biopsy showed mixed epithelial-stromal proliferation suggestive of a phyllodes tumor. Left modified radical mastectomy with dissection of level Ⅰ and level Ⅱ lymph nodes was performed. Results: The pathologic findings of this proce- dure were consistent with borderline phyllodes tumor. The tumor measured 47.5 cm× 37,0 cm× 28.0 cm and weighed 9.79 kg ex vivo. Conclusion: To our knowledge, this patient presented one of the biggest phyllodes tumors of breast in all cases reported in English-language publications so far.展开更多

The rs2294021 Foxp3 polymorphism contributes to Foxp3 down-regulation, thereby weakens its tumor suppressing activity. The aim of our study was to evaluate the potential influence of Foxp3 polymorphism on breast cancer, we conducted a case-control study in Han Chinese women. Methods: Foxp3 genotyping was conducted in 677 breast carcinoma patients and 828 age-frequency matched cancer-free controls. Genotypes were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Experiment data was analyzed using Chi-square test and SPSS software. Results: The T/C genotype was found to be significantly associated with increased risk of breast carcinoma occurrence (OR = 1.462; 95% CI 1.165-1.833, P = 0.001) compared with the T/T or C/C (OR 1.143; 95% CI 0.838-1.559, P = 0.397) genotype. The increased risk for breast carcinoma related to heterozygous genotype was more pronounced in subjects over 50 years (OR 1.631; 95% CI 1.116-2.383, P = 0.011). No significant association was found between the polymorphism and the ER/PR status, metastasis or tumor stage of breast cancer. Conclusion: Our findings suggest that rs2294021 Foxp3 polymorphism may be a potential contributor for development of breast carcinoma in Han Chinese women.展开更多
