金黄色葡萄球菌蛋白A串联Z结构域的表达、纯化及其亲和填料制备

目的构建金黄色葡萄球菌蛋白A(Staphylococcal Protein A,简称SPA或蛋白A),串联Z结构域的重组表达克隆,并实现在大肠杆菌中诱导表达,随后纯化获得重组蛋白,制备亲和填料.方法对目的基因进行元和表达的密码子优化,利用PCR扩增方法获得SPA... 目的构建金黄色葡萄球菌蛋白A(Staphylococcal Protein A,简称SPA或蛋白A),串联Z结构域的重组表达克隆,并实现在大肠杆菌中诱导表达,随后纯化获得重组蛋白,制备亲和填料.方法对目的基因进行元和表达的密码子优化,利用PCR扩增方法获得SPA的3个串联Z结构域,并在蛋白的N端加入组氨酸标签.将优化后的基因序列克隆到p ET43.1a(+)表达载体上,获得p ET43.1a(+)-SPA-3Z重组质粒;测序正确的重组质粒转化大肠杆菌感受态细胞BL21(DE3),IPTG诱导表达,通过亲和层析方法纯化表达的重组蛋白,并使用SDS-PAGE鉴定表达蛋白.将纯化获得的SPA纯品偶联到经环氧氯丙烷活化的Sepharose 6B Fast Flow凝胶上.使用兔多抗和腹水单抗对填料进行验证.结果成功构建p ET43.1a(+)-SPA-3Z克隆,纯化的SPA蛋白纯度高.成功将重组SPA偶联到经环氧氯丙烷活化的Sepharose 6B Fast Flow凝胶上.制备的填料在0.1mol/L的Na OH溶液里浸泡30小时,填料亲和性能未见明显变化,这为填料的长期使用提供了可能.将该填料用于抗体纯化,湿胶的多抗结合量可达22.7mg/ml,纯化的腹水单抗纯度在98%以上.结论本研究为SPA的大规模制备提供了基础. 展开更多

The nanoparticles of the hydrophobic drug of danazol with narrow size distribution are facilely prepared by controlled high-gravity anti-solvent precipitation (HGAP) process. Intensified micromixing and uniform nucleation environment are created by the high-gravity equipment (rotating packed bed) in carrying out the anti-solvent precipitation process to produce nanoparticles. The average particle size decreases from 55 μm of the raw danazol to 190 nm of the nanoparticles. The Brunauer-Emmett-Teller (BET) surface area sharply increases from 0.66 m^2·g^-1 to 15.08 m^2·g^-l. Accordingly, the dissolution rate is greatly improved. The molecular state, chemical composition, and crystal form of the danazol nanoparticles remains unchanged after processing according to Fourier transform infrared (FTIR) and X-ray diffraction (XRD), The high recovery ratio and continuous production capacity are highly appreciated in industry. Therefore, the HGAP method might offer a general and facile platform for mass production of hydrophobic pharmaceutical danazol particles in nanometer range. 展开更多

Co0.6Cu0.16Ni0.24Fe2O4/multi-walled carbon nanotube nanocomposites (CCNF/MWCNTs) were synthesized by solution filling method.The phase structure,thermal stability,morphology and electrical-magnetic properties of the samples were characterized by means of modern testing technology.The effect of iron concentration,filling time,sintering temperature on their electrical and magnetic performance was discussed.The results indicated that conductivity was related to the content of MWCNTs,while the magnetism correlated with the volume fraction of the filled CCNF in the composites.When the optimal condition satisfied the filling time of 18 h,ferric concentration of 0.25 mol L-1 and sintering temperature of 350°C,the prepared composite had the best magnetic loss performance,and its minimum reflection loss reached-22.47 dB on 9.76 GHz,the available bandwidth was beyond 2.0 GHz.Hence,the obtained composite can be used as advancing absorption and shielding material due to its favorable microwave absorbing property. 展开更多

Facile synthesis of an interfacial layer in organic solar cells (OSCs) is important for broadening material designs and upscaling photovoltaic conversion efficiency (PCE).Herein,a mild solution process of spin-coating In(acac)3and Ga(acac)3isopropanol precursors followed by low-temperature thermal treatment was developed to fabricate In_(2)O_(3)and Ga2O3cathode buffer layers (CBLs).The introduction of In_(2)O_(3)or Ga2O3CBLs endows PM6:Y6-based OSCs with outstanding performance and high PCEs of 16.17%and 16.01%,respectively.Comparison studies present that the In_(2)O_(3)layer possesses a work function (WF) of 4.58 eV,which is more favorable for the formation of ohmic contact compared with the Ga2O3layer with a WF of 5.06 eV and leads to a higher open circuit voltage for the former devices.Electrochemical impedance spectroscopy was performed to reveal how In_(2)O_(3)and Ga2O3affect the internal charge transfer and the origin of their performance difference.Although In_(2)O_(3)possesses lower series resistance loss,Ga2O3has a higher recombination resistance,which enhances the device fill factor and compensates for its series resistance loss to some extent.Comparative analysis of the photo-physics of In_(2)O_(3)and Ga2O3suggests that both are excellent CBLs for highly efficient OSCs. 展开更多
