目的观察葛根素对糖尿病大鼠视网膜细胞间黏分子-1(ICAM-1)表达的影响。方法利用链脲佐菌素腹腔注射法建立糖尿病大鼠模型,将实验用Wistar大鼠随机分为正常对照组、葛根素治疗组和糖尿病组,治疗12周。观察治疗期间及治疗后各组大鼠的一... 目的观察葛根素对糖尿病大鼠视网膜细胞间黏分子-1(ICAM-1)表达的影响。方法利用链脲佐菌素腹腔注射法建立糖尿病大鼠模型,将实验用Wistar大鼠随机分为正常对照组、葛根素治疗组和糖尿病组,治疗12周。观察治疗期间及治疗后各组大鼠的一般情况、尿糖、血糖、血NO及血SOD值,HE染色及免疫组化染色法观察大鼠视网膜形态及ICAM-1表达的不同。结果和正常对照组相比,造模组大鼠血糖均升高,血SOD和血NO值降低(P<0.05)。葛根素能改善大鼠基本情况,降低血糖(P<0.05),升高血SOD及血NO值(P<0.05),并减少视网膜组织细胞间ICAM-1表达,其差异有统计学意义。结论葛根素对糖尿病大鼠视网膜组织间ICAM-1的表达具有抑制作用,从而对高血糖引起的视网膜损伤有一定的保护作用。 展开更多

目的咀嚼槟榔与口腔黏膜下纤维性变(OSF)有关,但其相关机制尚不清楚。观察槟榔提取物(ANE)对体外培养的人类口腔黏膜成纤维细胞(FB)表达细胞间黏附分子-1(ICAM-1)的影响,探讨FB与ICAM-1在OSF发生中的作用。以槟榔提取物对体外人口腔黏... 目的咀嚼槟榔与口腔黏膜下纤维性变(OSF)有关,但其相关机制尚不清楚。观察槟榔提取物(ANE)对体外培养的人类口腔黏膜成纤维细胞(FB)表达细胞间黏附分子-1(ICAM-1)的影响,探讨FB与ICAM-1在OSF发生中的作用。以槟榔提取物对体外人口腔黏膜成纤维细胞表达细胞间黏附分子的影响,证明咀嚼槟榔与OSF发病机制相关的若干假设成立。方法所有口腔黏膜标本都采自1997-07/1998-04在中南大学湘雅二医院口腔科就诊的患者,经临床及病理确诊为OSF者(男8例,女3例)作为实验组。另外,从同时间来院的手术患者中,选择无槟榔、烟酒及辣椒嗜好,年龄与实验组匹配者(男6例,女5例),作为对照组。分别从正常(NM)及OSF患者的口腔黏膜中培养出FB,向培养基中加入不同浓度的ANE培养48h后,用细胞酶联免疫方法检测FB表达的ICAM-1水平。结果表示ICAM-1水平高低的OD值在OSF-FB为0.386±0.099,高于NM-FB的OD值0.324±0.030(F=18.177,P=0.001);ANE在50~150μg/ml内以浓度-效应依赖关系刺激FB产生ICAM-1。结论ANE能上调FB表达ICAM-1的水平,从而间接促进纤维化过程,引起OSF的发生。 展开更多

目的观察血管紧张素(1-7)[Ang(1-7)]对氧化低密度脂蛋白(ox-LDL)诱导的大鼠主动脉内皮细胞(SVAREC)血凝素样氧化型低密度脂蛋白受体1(LOX-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)表达水平的影响,探讨其可能作用机制... 目的观察血管紧张素(1-7)[Ang(1-7)]对氧化低密度脂蛋白(ox-LDL)诱导的大鼠主动脉内皮细胞(SVAREC)血凝素样氧化型低密度脂蛋白受体1(LOX-1)、细胞间黏附分子-1(ICAM-1)、血管细胞黏附分子-1(VCAM-1)表达水平的影响,探讨其可能作用机制。方法将大鼠随机分为ox-LDL组和ox-LDL+Ang(1-7)组。实验通过Cell Counting Kit-8(简称CCK-8)检测细胞增殖情况。通过荧光实时定量PCR法检测LOX-1、VCAM-1、ICAM-1mRNA的表达水平。结果 ox-LDL可以抑制SVAREC细胞的增殖,并呈剂量依赖性,与对照组比较有统计学意义(P<0.05)。ox-LDL+Ang(1-7)组可以使SVAREC细胞生长抑制率降低,并呈剂量依赖性,与对照组比较有统计学意义(P<0.05)。随着ox-LDL浓度的升高,LOX-1、ICAM-1及VCAM-1 mRNA表达量上升,并呈剂量依赖性,与对照组比较有统计学意义(P<0.05),加入Ang(1-7),随着Ang(1-7)的浓度升高,LOX-1、ICAM-1及VCAM-1mRNA表达量下降。结论 ox-LDL可以损伤SVAREC细胞,并使LOX-1、ICAM-1及VCAM-1mRNA表达量上升,Ang(1-7)抑制ox-LDL的上述反应,并且可能通过LOX-1、ICAM-1及VCAM-1mRNA发挥作用。 展开更多

目的探讨肺炎衣原体(CPn)感染高脂饮食小鼠后血清细胞间黏附分子-1(sICAM-1)、血管细胞黏附分子-1(sVCAM-1)、E选择素(sE-slectin)的变化及黄芩苷调节作用。方法将50只小鼠随机分为黄芩苷高剂量组、黄芩苷低剂量组、阿奇霉素组、模型组... 目的探讨肺炎衣原体(CPn)感染高脂饮食小鼠后血清细胞间黏附分子-1(sICAM-1)、血管细胞黏附分子-1(sVCAM-1)、E选择素(sE-slectin)的变化及黄芩苷调节作用。方法将50只小鼠随机分为黄芩苷高剂量组、黄芩苷低剂量组、阿奇霉素组、模型组、正常对照组。除正常对照组外均于喂饲高脂饲料1周后,经一侧鼻腔吸入含CPn的培养液,每周1次,共3次。最后1次接种5d后开始给药至实验结束,第1次接种后第18周处死全部动物。结果模型组血清sICAM-1、sVCAM-1、sE-slectin均较正常对照组升高(P<0.01);与模型组比较,黄芩苷高剂量组、低剂量组和阿奇霉素组3个指标均有不同程度下降,其中黄芩苷低剂量组和阿奇霉素组sICAM-1和sE-slectin显著降低(P<0.01)。结论CPn感染高脂饮食小鼠可使血清sICAM-1、sVCAM-1、sE-slectin水平升高,黄芩苷高剂量、低剂量和阿奇霉素均具有不同程度的调节作用,从而在一定程度上抑制动脉粥样硬化的形成。 展开更多

Objective To investigate effects of electroacupuncture (EA) on expression of intercellular adhesion molecule-1 (ICAM-1) in the rat of local cerebral ischemia-reperfusion. Methods Eighty SD rats were randomly divided into a normal control group, a sham operation group, a model group and an EA treatment group, 20 rats in each group. The thread-obstruction method was used for preparation of ischemia-reperfusion model. Zea-Longa rating criteria were used for evaluation of nervous function disorder; Immunohistochemical SABC method was used for detection of ICAM-1 expression in the microvascular endothelial cell of the ischemic brain region, and ELISA method for the soluble ICAM-1 (slCAM-1) content in peripheral blood. Re. suits After cerebral ischemia-reperfusion, both ICAM-1 expression level in the microvascular endethelium cell of the ischemic brain region and slCAM-1 content in the peripheral blood significantly increased in the model group as compared with the normal group and the sham operation group (P〈0.01); After EA treatment, the ICAM-1 expression level in the microvascular endothelial cell of the ischemic brain region and slCAM-1 content in the peripheral blood were significantly down-regulated in the EA treatment group as com- pared with the model group (P〈 0.05). Conclusion After cerebral ischemia-reperfusion, the microvascular endothelial cell of the ischemic brain region releases ICAM-1, which induces inflammatory injury of cerebral tissues; EA treatment can decease the expression of ICAM-1, so as to prevent the brain from the injury. 展开更多

Up-regulation of intestinal nuclear factor kappa B and intercellular adhesion molecule-1 following traumatic brain injury in rats 被引量:16

AIM: Nuclear factor kappa B (NF-κB) regulates a large number of genes involved in the inflammatory response to critical illnesses, but it is not known if and how NF-KB is activated and intercellular adhesion molecule-1 (ICAM-1) expressed in the gut following traumatic brain injury (TBI). The aim of current study was to investigate the temporal pattern of intestinal NF-κB activation and ICAM-1 expression following TBI. METHODS: Male Wistar rats were randomly divided into six groups (6 rats in each group) including controls with sham operation and TBI groups at hours 3, 12, 24, and 72, and on d 7. Parietal brain contusion was adopted using weight-dropping method. All rats were decapitated at corresponding time point and mid-jejunum samples were taken. NF-KB binding activity in jejunal tissue was measured using EMSA. Immunohistochemistry was used for detection of ICAM-1 expression in jejunal samples. RESULTS: There was a very low NF-κB binding activity and little ICAM-1 expression in the gut of control rats after sham surgery. NF-KB binding activity in jejunum significantly increased by 160% at 3 h following TBI (P<0.05 vs control), peaked at 72 h (500% increase) and remained elevated on d 7 post-injury by 390% increase. Compared to controls, ICAM-1 was significantly up-regulated on the endothelia of microvessels in villous interstitium and lamina propria by 24 h following TBI and maximally expressed at 72 h post-injury (P<0.001). The endothelial ICAM-1 immunoreactivity in jejunal mucosa still remained strong on d 7 post-injury. The peak of NF-κB activation and endothelial ICAM-1 expression coincided in time with the period during which secondary mucosal injury of the gut was also at their culmination following TBI. CONCLUSION: TBI could induce an immediate and persistent up-regulation of NF-κB activity and subsequent up-regulation of ICAM-1 expression in the intestine. Inflammatory response mediated by increased NF-κB activation and ICAM-1 expression may play an important role in the pathogenesis of acute gut mucosal injury following TBI. 展开更多

Cardioprotective effects of Guanxinshutong (GXST) against myocardial ischemia/ reperfusion injury in rats 被引量:14

Background The protective effects against reperfusion injury of cardioprotective drugs have recently been evaluated and found to be inadequate. Guanxinshutong (GXST), a combination of the traditional herb and Mongolian medicine, is effective and safe in treating angina pectoris in clinical trials. We assess the cardioprotective effects of GXST against myocardial ischemia and reperfusion (MI/R) injury in rats and explore its possible mechanism. Methods Forty-five male Sprague Dawley rats were randomized into three groups: non-MlfR group (Sham, n = 15), MUR group treated with vehicle (Control, n = 15) and MI/R group treated with GXST (Drug, n = 15). MI/R was induced by ligation of the left anterior descending coronary artery (LAD) for 30 minutes, followed by 2/24 hour reperfusion in the Control and Drug groups. In the Sham group, the LAD was exposed without occlusion. GXST powder (in the Drug group) or saline (in the Control and Sham groups) were administered via direct gastric gavage from 7 day prior to surgery. Blood samples were collected from the carotid artery (10 rats each group) after 2 hours of reperfusion, to determine the levels of tumor necrosis factor-or (TNF-ct), interleukin-1 ~ (IL-113), interleukin-6 (IL-6) and intercellular adhesion molecule-1 (ICAM-1) using enzyme-linked immunosorbent assays. The animals were then sacrificed and the hearts were harvested for histopathology and western blot analysis. Infarct size was measured in the remaining five rats in each group after 24 hours reperfusion. Results GXST significantly decreased levels of TNF-ct, IL-1β, IL-6, ICAM-1, apoptosis index (AI) and infarct size. GXST also obviously inhibited nuclear factor kappa B (NF.r,B) activity when compared with the Control group (all P 〈 0.05). Conclusions GXST is effective in protecting the myocardium against MI/R injury in rats. Its possible cardioprotective mechanism involves inhibition of the inflammatory response and apoptosis following MI/R injury. 展开更多

AIM: To generate dendritic cells (DCs) from human peripheral blood and to detect the expression of dendritic cell-specific intercellular adhesion molecule 3 grabbing nonintegrin (DC-SIGN; CD209) for the further study of DC-SIGN in hepatitis C virus (HCV) transmission. METHODS: Peripheral blood monocytes were isolated from blood of healthy individuals by Ficoll--Hypaque sedimentation and cultured in complete medium containing rhGM-CSF and rhIL-4. Cells were cultured for seven days, with cytokine addition every two days to obtain immature DCs. Characteristics of the cultured cells were observed under light and scanning microscope, and the expression of DC-SIGN was detected by immunofluorescence staining. RESULTS: After seven-day culture, a large number of cells with typical characteristics of DCs appeared. Their characteristics were observed under light and scanning electron microscope. These cells had a variety of cell shapes such as those of bipolar elongate cells, elaborate stellate cells and DCs. DC-SIGN was detected by immunofluorescence staining and its expression level on cultivated dendritic cells was high. CONCLUSION: DCs with a high expression of DC-SIGN can be generated from human peripheral blood monocytes in complete medium containing rhGM-CSF and rhIL-4. 展开更多

OBJECTIVE: To explore the effect of kidney-rein- forcing, blood-activating and stasis-removing recipes on adhesion molecule expression of bone mar- row mesenchymal stem cells (MSCs) from patients with chronic aplastic anemia (CAA). METHODS: We used three Traditional Chinese Medicine recipes, namely a kidney-reinforcing recipe (KRR), blood-activating and stasis-removing recipe (BASRR), and kidney-reinforcing, blood-activating and stasis-removing recipe (KRBASFIR), and a nor- mal saline control to prepare herbal medicine se- rum in Sprague Dawley rats. Thirty CAA patients were enrolled in the experimental group, including 17 kidney-Yang deficient patients and 13 kidney-Yin deficient patients. Ten healthy individuals were included in the control group. MSCs were isolated from bone marrow samples, and the cell density was observed to measure their proliferation ability by microscopy on days 2, 7, and 14 after isolation. In addition, the expression of adhesion molecules of bone marrow MSCs (CD106, CD49d, CD31 and CD44) were detected by flow cytometry after 48 h of treatment with the four different herbal medi- cine serums. RESULTS: The proliferation of MSCs from kid- ney-Yang deficient and kidney-Yin deficient pa- tients was weaker than that of MSCs from the con- trol group. The expression of all adhesion mole- cules of bone marrow MSCs from CAA patients was obviously lower than that in the control group (P〈 0.01). The expression of CD49d and CD31 in MSCs from patients with a kidney-Yin deficiency was low- er than in those with a kidney-yang deficiency (P〈 0.05 and P〈O.01, respectively). For kidney-Yang defi- cient patients, CD31 expression in the KRBASRR group was significantly higher than that in the BASRR group (P〈O.01), while CD44 in the KRBASRR group was significantly higher than that in both KRR and BASRR groups (P〈O.01). For kidney-Yin defi- cient patients, CD106 and CD49d expression in the KRBASRR group was obviously higher than that in the KRR group (P〈0.05), while CD31 and CD44 ex- pression in the KRBASRR group was significantly higher than that in both KRR and BASRR groups (P〈 0.05 and P〈0.01, respectively). CONCLUSION: The bone marrow microenviron- ment in CAA patients is abnormal. The effect of KRBASRR may be better than that of KRR and BASRR for kidney-Yang deficient and kidney-Yin de- ficient patients by improving the expression levels of MSC adhesion molecules. 展开更多

Objective: To study the influence of different methods on serum soluble intercellular adhesion molecule1 (sICAM1) in patients with acute cerebral infarction. Methods: Double antibody enzyme-linked immunoadsorbent assay was used. Results and Conclusion: Scalp point-to-point acupuncture has a significant reducing effect on abnormally elevated serum sICAM1 in patients with acute cerebral infarction. Its curative effect is superior to that of simple Western medicine. There is a significant difference between them (P〈 0,05). 展开更多
