【目的】观察金丝桃苷对过敏性哮喘小鼠辅助性T(Th)9、Th17、调节性T(Treg)细胞分化及气道炎症的影响。【方法】将60只雄性BALB/c小鼠随机分为5组,即正常组,模型组,金丝桃苷低、高剂量组,地塞米松组,每组12只。除正常组,其他各组小鼠采... 【目的】观察金丝桃苷对过敏性哮喘小鼠辅助性T(Th)9、Th17、调节性T(Treg)细胞分化及气道炎症的影响。【方法】将60只雄性BALB/c小鼠随机分为5组,即正常组,模型组,金丝桃苷低、高剂量组,地塞米松组,每组12只。除正常组,其他各组小鼠采用卵清蛋白(OVA)诱导法构建过敏性哮喘模型。给药结束后,观测吸气阻力(Ri)、呼气阻力(Re)和肺动态顺应性(Cldyn),评估肺功能,采用苏木素-伊红(HE)以及高碘酸希夫(PAS)染色法评估肺部组织病理学变化,流式细胞术分析脾脏Th9细胞、Th17细胞和Treg细胞比例,采用酶联免疫吸附分析(ELISA)检测血清和支气管肺泡灌洗液(BALF)中白细胞介素(IL)-9、IL-10、IL-17A和转化生长因子(TGF)-β1水平,采用实时定量聚合酶链反应(PCR)法检测肺组织中IL-9、IL-10、IL-17A和TGF-β1 mRNA表达水平,通过免疫组织化学法和Western Blot法检测肺组织中PU.1、维甲酸相关孤核受体γt(RORγt)和叉头状转录因子3(Foxp3)蛋白表达水平。【结果】与模型组比较,金丝桃苷高剂量组小鼠哮喘发作症状减轻,Ri和Re显著降低、Cldyn显著升高,BALF中白细胞总数、EOS数量显著降低,Th9和Th17细胞比例降低、Treg细胞比例升高,IL-9和IL-17A在肺组织中的mRNA水平和在血清、BALF中的含量显著降低,IL-10和TGF-β1在肺组织中的mRNA水平和在血清、BALF中的含量显著升高,肺组织中PU.1和RORγt蛋白表达水平显著降低,Foxp3蛋白表达水平显著升高,差异均有统计学意义(P<0.05)。【结论】金丝桃苷可抑制过敏性哮喘小鼠Th9、Th17细胞分化、促进Treg细胞分化,减轻气道炎症。 展开更多

目的:探讨Th9细胞和Bregs细胞在溃疡性结肠炎(UC)免疫学致病中的作用及其与患者疾病活动度的相关性。方法:前瞻性纳入2020年6月至2022年3月海安市人民医院收治的UC患者105例作为观察对象(UC组),其中48例处于疾病缓解期(缓解期UC组),57... 目的:探讨Th9细胞和Bregs细胞在溃疡性结肠炎(UC)免疫学致病中的作用及其与患者疾病活动度的相关性。方法:前瞻性纳入2020年6月至2022年3月海安市人民医院收治的UC患者105例作为观察对象(UC组),其中48例处于疾病缓解期(缓解期UC组),57例处于疾病活动期(活动期UC组),并根据活动期UC组患者病情的严重程度划分亚组:轻型组(n=20)、中型组(n=24)和重型组(n=13),另选取同期健康志愿者50例作为对照组。应用流式细胞术对两组患者外周血中Th9和Bregs细胞比例进行分析,ELISA检测两组血清IL-9、IL-10、IL-35及C反应蛋白(CRP)水平,并采用Pearson相关性分析法分析Th9、Bregs细胞水平与疾病活动度和炎症程度的相关性。结果:UC组患者Bregs细胞比例、Th9细胞比例均明显高于对照组,活动期UC组CAI评分、Bregs细胞比例及Th9高于缓解期UC组(P<0.05)。重型活动期UC患者Th9细胞和Bregs细胞比例明显高于中型和轻型活动期UC患者,其中中型活动期组高于轻型活动期组(P<0.05)。活动期和缓解期UC组患者CRP、IL-9水平均明显高于对照组,且活动期高于缓解期(P<0.05);活动期和缓解期IL-10、IL-35水平均显著低于对照组,其中活动期低于缓解期(P<0.05)。重型组的CRP、IL-9水平高于轻型、中型组,其中中型组高于轻型组(P<0.05);重型组IL-10及IL-35水平明显低于中型和轻型组,其中中型组低于轻型组(P<0.05)。Pearson分析结果显示,Th9、Bregs细胞均与疾病活动度、CRP、IL-9水平呈正相关,与IL-10、IL-35水平呈负相关(P<0.05)。结论:活动期UC患者外周血中Th9、Bregs细胞比例均明显升高,均与其疾病的发生发展密切相关,有望成为辅助临床早期诊断、评估病情的良好指标。 展开更多

目的 探讨润燥灵对干燥综合征(SS)NOD小鼠颌下腺及血清Th9、Th17、Treg表达的影响。方法 选取75只8周龄的雌性NOD小鼠,随机分为5组:模型组、羟氯喹组、润燥灵高、中、低剂量组,每组15只。另抽取15只同龄BALB/c小鼠作为空白组。羟氯喹组... 目的 探讨润燥灵对干燥综合征(SS)NOD小鼠颌下腺及血清Th9、Th17、Treg表达的影响。方法 选取75只8周龄的雌性NOD小鼠,随机分为5组:模型组、羟氯喹组、润燥灵高、中、低剂量组,每组15只。另抽取15只同龄BALB/c小鼠作为空白组。羟氯喹组每天4 mg/mL羟氯喹片灌胃,润燥灵高、中、低组分别按1.8、0.9、0.45 g/mL润燥灵方灌胃。连续灌胃8周后处死NOD小鼠,计算颌下腺指数和HE染色检测;ELISA测定血清中IL-10、IFN-γ、IL-17A和IL-23的水平;RT-PCR检测颌下腺组织IL-17A、IL-17F、IL-23、IL-9、IL-10和PU.1mRNA水平;流式细胞仪检测外周血中Th9、Th17和Treg的表达。结果 给药干预后,与模型组比较,润燥灵方低、中、高剂量组和羟氯喹组在日饮水量均有显著减少。与模型组比较,润燥灵方低、中、高剂量组和羟氯喹组颌下腺病理显示淋巴细胞浸润减少,颌下腺组织IL-17A、IL-17F、IL-23、IL-9和PU.1的mRNA表达降低(P<0.05),血清及颌下腺中IL-10表达升高(P<0.05);润燥灵方高剂量组和羟氯喹组NOD小鼠的颌下腺指数升高(P<0.05);润燥灵方中、高剂量组和羟氯喹组外周血中Treg细胞表达增加(P<0.05),Th9、Th17细胞表达降低(P<0.05),血清IFN-γ、IL-17A和IL-23含量降低(P<0.05)。结论 润燥灵方对NOD小鼠唾液腺损伤有改善作用,可能通过上调Treg的分化,抑制Th17、Th9细胞表达,改善颌下腺淋巴细胞浸润,从而改善SS疾病进展。 展开更多
