OBJECTIVE:To explore the protective mechanisms of the Traditional Chinese Medicine Bushenhuoxue(BSHX)in a rat model of vascular dementia(VD).METHODS:A rat model of VD was developed using bilateral common carotid artery occlusion(BCCAO).Rats were administered BSHX(10.14 or 5.07 g/kg),nimodipine(11.06 mg/kg;positive control),or saline(control)by gavage daily for 30 d post-surgery.Learning and memory abilities were assessed using the Morris water maze.Morphological changes in the hippocampus were observed using light microscopy(hematoxylin and eosin staining)and transmission electron microscopy(TEM).The m RNA and protein expression levels of brain-derived neurotrophic factor(BDNF),tyrosine receptor kinase B(Trk B),phosphatidyl inositol 3-kinase(PI3 K),serine/threonine kinase(AKT),and c AMP response element binding protein(CREB)were measured by real-time polymerase chain reaction(RT-PCR)and Western blot,respectively.RESULTS:Compared with the sham group,rats with BCCAO exhibited impaired learning and memory abilities(Morris water maze)and showed abnormalities in neuronal morphology(light microscopy)and ultrastructure(TEM)in the hippocampus.They also had decreased m RNA and protein expressions of BDNF,Trk B,PI3 K,AKT,and CREB in hippocampal tissue(all P<0.05).In rats with BCCAO,administration of BSHX attenuated deficits in learning and memory,improved the morphology and ultrastructure of hippocampal neurons,and enhanced m RNA and protein expression levels of BDNF,Trk B,PI3 K,AKT,and CREB(all P<0.05).CONCLUSION:BSHX may protect hippocampal neurons and improve learning and memory abilities,at least in part via the activation of BDNF/Trk B/PI3 K/AKT/CREB signaling. 展开更多

OBJECTIVE:To determine the effects of Bushenhuoxue formula(BHF) on interleukin-1 beta(IL-1β),transforming growth factor beta 1(TGF-β1),discoidin domain receptor 2(DDR2) and matrix metalloproteinase-1(MMP-1) levels in a rat model of osteoarthritis(OA).METHODS:Sprague-Dawley rats were used to establish an OA model and subjected to various treatments over 6 weeks.Rats were treated with BHF,glucosamine sulfate(G5),or starch as a control.Serum levels of IL-1β and MMP-1 and joint fluid levels of IL-1 β were determined by means of ELISAs.We used immunohistochemistry to determine DDR2 levels in knee cartilage.Gene expression levels of MMP-1 in joint synovial tissue were assessed using reverse transcription polymerase chain reaction assays.RESULTS:Serum IL-1β levels were unchanged throughout the study.Levels of IL-1β in joint fluid and MMP-1 in sera from the BHF- and GS-treated groups were significantly reduced.DDR2 levels in knee cartilage were also significantly reduced in the BHF group.Expression of the MMP-1 gene was significantly reduced by BHF treatment.CONCLUSION:BHF might be beneficial in the inhibition and alleviation of local inflammatory responses and cartilage degeneration in OA. 展开更多

OBJECTIVE: To observe the effects and mechanisms of Bushenhuoxue on desmin and nephrin expression in mice podocytes, and to investigate its effects on wt1 expression in Wilms' tumor.METHODS: Adriamycin(ADR) was used to induce focal segmental glomerulous sclerosis(FSGS) in mice. Bushenhuoxue was used to treat FSGS for 6 weeks. We measured body mass and right renal mass, and determined serum albumin(ALB) levels,protein content in urine, and urinary protein and albumin creatinine ratio(UACR). Changes in renal tissue morphology were evaluated by microscopy.wt1 and nephrin expression in podocytes were detected using immunofluorescence. Expression levels of desmin, wt1 and nephrin m RNAs in renal tissue were determined using reverse transcription polymerase chain reaction assays.RESULTS: Protein levels in urine and UACR were significantly increased in FSGS model mice compared with Bushenhuoxue-treated and control mice.Body mass and ALB levels were decreased in FSGS mice compared with control and Bushenhuoxue-treated mice. Expression of the wt1 protein was observed in control mice. Compared with controls,wt1 expression levels were reduced in Bushenhuoxue-treated mice, and to a greater extent in FSGS mice. Nephrin protein expression was widespread in FSGS mice, and significantly reduced in control and Bushenhuoxue mice. Expression levels of wt1 and nephrin m RNAs in FSGS mice were lower compared with those in control and Bushenhuoxue-treated mice. Desmin m RNA levels in FSGS mice were reduced compared with those in control and Bushenhuoxue-treated mice.CONCLUSION: Bushenhuoxue ameliorated albuminuria in FSGS mice; this was possibly related to the up-regulation of wt1 and nephrin, and down-regulation of desmin. 展开更多

Sixty-eight patients suffering from diabetes mellitus (DM) type with Kidney Deficiency andblood stasis Syndrome were enrolled and divided randomly into treatment group (34 cases) and placebogroup (34 cases) . The contents of serum lipid peroxide ( LPO) , blood lipid and blood sugar were deter-mined. The results showed that the serum LPO in treatment group was higher than that in piacebo group ( P<0.05) , it increased significantly in DM patients with vascular disease or with uncontrolled blood sugar andwas positively correlated with level of triglyceride ( TG, r = 0. 25, P< 0. 05) . After using Bushen Huoxuetablet ( BSHXT) serum LPO lowered ( P< 0 . 05 ) , blood sugar decreased ( P< 0 . 05) and high density lipopro-tein-cholesterol ( HDL-C) increased ( P< 0 . 05) in treatment group, but in placebo group these three parame-ters were not changed significantly. These results indicated that LPO reaction in type DM patients in-creased. When the blood sugar and TG were higher or complicated with vascular disease, the LPO reactionwould be enhanced more markedly. BSHXT will have a good prospect in the prevention and treatment of DMpatients complicated with vascular disease . 展开更多
