[Methods]Eucalyptus oil samples with different dilution ratio were d...[Objectives] To explore whether the thin layer chromatography( TLC) can be used to guide the preparation of drug containing serum of eucalyptus oil. [Methods]Eucalyptus oil samples with different dilution ratio were detected by TLC. Eucalyptus oil was intragastrically administered to mice,serum samples of different eucalyptus oil doses,different intragastric administration methods and different blood sampling times were collected. The above samples were detected by TLC,and the above results were verified by gas chromatography. [Results] TLC can detect concentration difference of eucalyptus oil samples with different dilution ratio. TLC can provide qualitative and semi-quantitative detection of eucalyptus oil in serum. High,medium and low dose of eucalyptus oil in serum had different effects on the growth of MCF-7 cells in the SD rats( P < 0. 05). [Conclusions] TLC can be used to guide the preparation of drug containing serum of eucalyptus oil.展开更多

We investigated the significance of procalcitonin(PCT) and C-reactive protein(CRP) in evaluating the s...The severity of systemic lupus erythematosus(SLE) patients with pulmonary bacterial infection varies widely. We investigated the significance of procalcitonin(PCT) and C-reactive protein(CRP) in evaluating the severity of pulmonary infection in SLE patients. This retrospective study contained a total of 117 patients(107 women and 10 men) with SLE from January 2010 to June 2011. Serum levels of PCT and CRP were measured by enzyme-linked immunosorbent assay. The severity of pulmonary bacterial infection(PBI) was evaluated using the pneumonia severity index(PSI). SLE patients with PBI, particularly those with bacterial isolates, had significantly higher levels of serum PCT and CRP than those without PBI. Serum PCT and CRP were not associated with SLE disease activity, but positively with the values of PSI in active SLE patients with PBI. Serum levels of PCT and CRP may be additional biomarkers in evaluating the severity of PBI in lupus patients.展开更多
The aim of the paper is to explore whether AMPK/U1K1 signaling pathway is involved in regulating autophagy and alleviating cerebral ischemia/reperfusion(I/R)injury.On this basis,study the regulatory effect of Buyang H...The aim of the paper is to explore whether AMPK/U1K1 signaling pathway is involved in regulating autophagy and alleviating cerebral ischemia/reperfusion(I/R)injury.On this basis,study the regulatory effect of Buyang Huanwu Decoction(BHD)on AMPK/U1K1 signaling pathway,aiming to clarify the molecular mechanism of BHD in alleviating cerebral I/R injury and the protective effect of BHD containing serum(BHDS)on OGD/R neurons.The research group adopted the following methods:Neurons cultured for 7 days in vitro were randomly divided into normal control group,model group(OGD/R),AMPK activator(AICAR)group(3 mmol/L),Buyang Huanwu Decoction medicated serum(BHDS)group(3%),Buyang Huanwu Decoction medicated serum+AMPK activator(BHDS+AICAR)group.The protein expressions of p-AMPK,AMPK,p-UlKl,U1K1,Beclin-1 and LC3B in neurons were detected by Western blot,the fluorescence intensity and apoptosis rate of neuron autophagosome CYTO-ID staining were detected by flow cytometry,the fluorescence intensity and morphological changes of neuron LC3B were detected by immunofluorescence double labeling method,the morphological changes of neurons were observed by inverted microscope,and the survival rate of neurons was detected by CCK-8 method.Neuron OGD/R model and the optimal concentration of BHDS.The neurons in the model group were subjected to hypoxia and glucose deprivation for 1 h,2 h,3 h,4 h and reoxygenation and glucose deprivation for 24 h.The survival rate of neurons and the release rate of 1DH to determine whether the model was successfully established.And the results show that BHDS group,the survival rate of OGD/R neurons was significantly increased,and the apoptosis rate was significantly decreased.Among them,3%BHDS intervention at the same time of hypoxia glucose deprivation for 3 h and reoxygenation glucose for 24 h was the best condition.In BHDS group,the levels of p-AMPK/AMPK,p-UlKl/UlKl,beclin-1,CYTO-ID and LC3B fluorescence intensity,and the transformation from LC3B-I to LC3B-II of OGD/R neurons were significantly decreased.So we can draw a conclusion that BHD can inhibit autophagy and alleviate cerebral I/R injury through AMPK/U1K1 signaling pathway.展开更多

Methods: Human lung adenocarcinoma A549/DD...Objective: To explore the pharmacologic effects of Chinese medicine Bushen Huayu Jiedu Compound Recipe (补肾化瘀解毒复方,BSHYJDR) in drug-resistance cells of lung cancer. Methods: Human lung adenocarcinoma A549/DDP cell strain was selected, serum pharmacology and flow cytometer (FCM) method were adopted, $180 tumor-bearing mice and normal mice were given, through gastrogavage, different doses of a decocted concentration of BSHYJDR. Serum from the abdominal aorta was taken to observe the effect of drug-serum on cisplatin (DDP) concentration, free Ca^2+ concentration and the expression of lung drug-resistance protein LRP-56 in A549/DDP cells. Results: Compared with the drug-resistance group, the intracellular DDP concentration in the group taking a high dose and the normal group of Chinese medicine showed significant difference (P〈0.05), while no significant difference was found in the low-dose group (P〉0.05). Compared with the drug-resistance group, the Ca^2+ concentration in cells and the expression of LRP in lung cancer drug-resistance cells A549/DDP of the high-dose group, the low-dose group and the normal group of Chinese medicine were significantly different (all P〈0.01), the LRP expression of the normal group was obviously higher than that of the drug-resistance group (P〈0.05). Merrill(P.scandens) in rat plasma.The bioactive constituents in plasma samples from rats administrated orally with P.scandens extract were analyzed by Ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS).Four prototype compounds were identified in rat serum as potential bioactive components of P.scandens by comparing their retention times and mass spectrometry data or by mass spectrometry analysis and retrieving the reference literatures.Glucuronidation after deglycosylation was the major metabolic pathway for the iridoid glycosides in P.scandens.These results showed that the methods had high sensitivity and resolution and were suitable for identifying the bioactive constituents in plasma after oral administration of P.scandens.providing helpful chemical information for further pharmacological and mechanistic researched on the P.scandens.展开更多
